Effects of Activated Carbon on the Reduction of Benzo(a)pyrene in Artemisia argyi Extract

  • Background and objective: Artemisia argyi has a long history as an effective treatment for various diseases. The detection of environmental pollutant benzo(a)pyrene, a known human carcinogen, in the leaves of Artemisia argyi is cause for concern. For medicinal plant extracts, both a reduction of benzo(a)pyrene as well as the maintained effectiveness of the compound are important. Therefore, in this study, we propose an optimized process for the addition and filtration of activated carbon to reduce benzo(a)pyrene and change the contents of the indicating substance(jaceosidine and eupatilin). Methods: Artemisia argyi EtOH extract containing 36 ppb of benzo(a)pyrene was added to 0.1, 0.5, 1.0, and 1.5% (w/w) of activated carbon for 120 min and filtered using an activated carbon filter 1, 2, 3, and 5 times respectively. The content of benzo(a)pyrene and indicating substances in Artemisia argyi extract were then measured with high performance liquid chromatography (fluorescence and UV detectors). Results: As the amounts of activated carbon powder and filtering cycles increased, the content of benzo(a)pyrene in the Artemisia argyi extract decreased. However, when activated carbon powder 1.5% was added to the extract, and when the activated carbon filter was filtered five times, the results were reduced by 15% and 30~40% respectively. The optimal extraction condition for reducing benzo(a)pyrene was adding 1.5% of activated carbon powder. This resulted in reducing benzo(a)pyrene by 83% and indicating substances by about 4%. Conclusions: Here we present a process for reducing benzo(a)pyrene in Artemisia argyi extract using activated carbon to reduce toxicity and minimize the loss of active ingredients. This approach has potential application within a manufacturing process of various medicinal plant extracts.

Determination of polycyclic aromatic hydrocarbons in processed foods (가공식품 중 다환방향족탄화수소 분석)

  • In this study, the following concentrations of some PAHs (Polycyclic Aromatic Hydrocarbons) were investigated; [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in processed foods (n=165) and cooked meats (n=45) and established the analytical method by characteristics of processed foods. The methodology involved extraction (alkali digestion, liquid-liquid extraction, microwave extraction), clean-up on Sep-Pak Florisil Cartridges and determination by HPLC/FLD (High Performance Liquid Chromatography/Fluorescence Detector). The recovery of overall method for 8 PAHs spiked into these products ranged from 92 to 103%. The mean level of detected foods was found to be benzo(b)fluoranthene $0.9\;{\mu}g/kg$ in smoked salmon, benzo(b)fluoranthene $1.0\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in dried banana, benzo(b)fluoranthene $0.2\;{\mu}g/kg$, benzo(k)fluoranthene $0.1\;{\mu}g/kg$, benzo(a)pyrene $0.2\;{\mu}g/kg$ in smoked chicken, benzo(b)fluoranthene $1.3\;{\mu}g/kg$, benzo(k)fluoranthene $0.3\;{\mu}g/kg$, benzo(a)pyrene $0.9\;{\mu}g/kg$ in charcoal grilled pork, respectively.

  • To evaluate the preventive effects of methanolic extract of Angelica koreana(MEAK), this extract was given to rats orally at various doses of 10, 50 and 100 mg/kg before hepatotoxicant, benzo$(\alpha)pyrene$ treatment The increased serum enzyme levels of aspartate aminotransferase (AST), alanine aminotransferase(ALT) and alkaline phosphatase(ALP) by benzo$(\alpha)pyrene$ induction were significantly lowered in a dose dependent manner after pretreatment with MEAK. Furthermore, MEAK also decreased the elevated lipid levels after benzo$(\alpha)pyrene$ administration. These results revealed that MEAK could afford a significant protective action in the alleviation of benzoBenzo$(\alpha)pyrene$ induced hepatocellular injury.

Suppressive effect of Petroleum Ether Extract of Panax ginseng against Benzo(a)pyrene induced Micronuclei in Mice (인삼 석유에텔 추출물이 흰쥐에서 Benzo(a)pyrene에 의해 유도된 소핵생성의 억제효과)

  • Petroleum ether extracts of panax ginseng C.A. Meyer (GPEE) were tested for the evaluation of anticlastogenic effects against benzo(a)pyrene-induced micronucleated polychromatic erythrocytes using mouse bone marrow micronucleus test. When the GPEE was singly administered before benzo(a)pyrene injection, GPEE showed significant anticlastogenic effect at $50{\sim}200\;mg/kg$. When the GPEE was multiply administered for 5 consecutive days before benzo(a)pyrene injection, GPEE showed potent anticlastogenic effect, even at the low doses, $5{\sim}50\;mg/kg/day$. As a control experiment, GPEE was administered without benzo(a)pyrene injection to demonstrate a clastogenic effect of this extract. When the range of $1{\sim}200\;mg/kg/day$ for 5 consecutive days was administered to mice, it was found that there was no increase of MNPCEs frequency.

  • Concentrations of PAHs [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in vegetable oils and fats available in Korean market were estimated. Involved methodology were liquid-liquid partition, purification on Sep-Pak Florisil Cartridges, and high performance liquid chromatography using fluorescence detector. Overall recoveries for eight PAHs spiked into vegetable oils and fats ranged from 68.2 to 101.5%, averaging 85.4%. Mean levels of benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, and indeno(1,2,3-c,d)pyrene in vegetable oils and fats were 0.53, 0.82, 0.50, 0.18, 0.35, 0.16, 0.31, and $0.44{\mu}g/kg$, respectively.

Estimating Human Exposure to Benzo(a)pyrene through Multimedia/Multiroute Exposure Scenario (다매체/다경로 노출을 고려한 benzo(a)pyrene의 총 인체 노출량 예측)

  • The objective of this study was to estimate human exposure to benzo (a)pyrene through multimedia/multi-pathway exposure scenario. The human exposure scenario for benzo(a)pyrene was consisted of 12 multiple exposure pathways, and the multipathway human exposure model based on this scenario constituted. In this study, the multipathway human exposure model was used to estimate the concentrations in the exposure contact media, human intake factors and lifetime average daily dose (LAD $D_{model}$) of benzo(a)pyrene in the environment. Sensitivity analysis was performed to identify the important parameters and Monte-Carlo simulation was undertaken to examine the uncertainty of the model. The total LAD $D_{model}$ was estimated to be 5.52${\times}$10$^{-7}$ mg/kg-day (2.06${\times}$10$^{-7}$ -8.65${\times}$10$^{-7}$ mg/kg-day) using the multipathway human exposure model. The inhalation dose accounted for 78% of the total LADD, whereas ingestion and dermal contact intake accounted for 20.2% and 1.8% of the total exposure, respectively. Based on the sensitivity analysis, the most significant contributing input parameter was benzo (a)pyrene concentration of ambient air. Consequently, exposure via inhalation in outdoor/indoor air was the highest compared with the exposure via other medium/pathways.

Use of the Comet Assay to Assess DNA Damage in Hemocytes and Gill of Oyster(Crassostrea gigas) Exposed to Pyrene and Benzo(a)pyrene (Pyrene과 Benzo(a)pyrene에 노출된 굴의 혈구세포과 아가미 세포에서의 DNA손상 측정을 위한 Comet assay의 이용)

  • Sessile organisms such as the oyster Crassostrea gigas have been given much attention as a potential biomonitoring indicator to assess the impact of toxicants on aquatic organism. In this study, we exposed cells isolated from gill of oyster (Crassostrea gigas) to hydrogen peroxide in vitro. In addition oysters were in vivo exposed to pyrene and benzo(a)pyrene at various concentrations for 2 weeks. Comet assay was used to detect DNA single strand breaks and to investigate the application of this technique as a tool for aquatic biomonitoring. Hydrogen peroxide increased DNA single strand break with increasing concentration after 30 minutes exposure in vitro. Pyrene and benzo(a)pyrene caused DNA damage only at very high concentration (100 $\mu\textrm{g}$/L or 1000 $\mu\textrm{g}$/L) at two week exposure in vivo. DNA damage was relatively higher at hemocyte than at gill. It suggested that metabolized PAHs are transferred to hemolymph from digestive gland which have a relatively high enzyme activity, and attacked the DNA of hemocyte, while gill accumulated PAHs without degrading them to their metabolites due to low enzyme activity at gill. Both in vitro and in vivo exposure experiments showed that the comet assay is an effective tool on screening whether the organism are exposed to genotoxic contaminants.

Analysis of Benzo[a]pyrene and Bisphenol A in lakes of Kyonggi-Do Province (환경홀몬 Benzo[a]pyrene및 Bisphenol A의 경기도 일부호소에서의 요염실태 및 위해성 평가 방안)

  • Benzo(a)pyrene and bisphenol A have been classified as endocrine disrupting chemicals (EDCs), which have been of concern in toxicology and environmental sciences. Benzo(a)pyrene and bisphenol A were monitored by HPLC or GC-MS in Baekwoon and Ilwirl lakes of Kyonggi-Do province to investigate contamination levels of EDCs. During the period between June, 2000 and August, 2000, water samples were collected from four different sites of each lake once per month. Contamination levels of benzo(a)pyrene were 3.27~4.25 ppb in Ilwirl lake and 2.00~2.33 ppb in Baekwoon lake, respectively. Bisphenol A levels were detected with the range of 0.33~7.94 and 0.43~4.71 for Baekwoon lake and Ilwirl lake, respectively. pH levels were higher in Ilwirl lake than in Baekwoon lake, where the contamination was relatively lower. These data suggest that lakes in Kyonggi-Do province could be contaminated with EDCs and be subjected to the routine monitoring and water quality control.

The mean level of benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k) fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene and indeno(1,2,3-c,d)pyrene in cooked fish was ND, ND, 0.0009, ND, 0.01, ND, ND, ND and in cooked shellfish was 1.84, 3.51, 0.81, 0.38, 0.39, 0.04, 0.20, ND, respectively.

Photodegradation of Pyrene, Chrysene and Benzo[a]pyrene in Water (II) (수중의 Pyrene, Chrysene 및 Benzo[a]pyrene의 광분해(II))

  • The photodegradations of pyrene, chrysene and benzo[a]pyrene that were similar in structure among polycyclic aromatic hydrocarbons (PAHs) were investigated with a low-pressure mercury lamp(the wavelength of 253.7 nm and UV output of 1.35${\times}$10$\^$-3/J/s). The optimum concentrations of TiO$_2$ and H$_2$O$_2$ on the photodegradation of pyrene, chrysene and benzo[a]pyrene were 1 g/L and 1.5${\times}$10$\^$-3/ M, respectively. By these optimum concentrations, their rates increased with increasing the concentration of TiO$_2$ and H$_2$O$_2$ because the amounts of OH radical formed increased, but for the higher concentrations than the optimum, their rates decreased with increasing those concentrations because the white turbidity phenomena occurs in case of TiO$_2$ and H$_2$O$_2$ acts as an OH radical inhibitor. The photodegradation rates among the photodegradation processes such as UV, UV/TiO$_2$, UV/H$_2$O$_2$, and UV/H$_2$O$_2$/TiO$_2$ decreased in the following sequences.: UV/H$_2$O$_2$/TiO$_2$> UV/H$_2$O$_2$> UV/TiO$_2$> UV.
