  • Entomopathogenic bacteria (Xenorhabdus nematophila, X. sp. and Photorhabdus temperata subsp. temperata) isolated from entomopathogenic nematodes express potent insecticidal activity in insect hemocoel. They are also known to suppress insect immune mediation by inhibiting phospholipase $A_2$, leading to host immunosuppression. This study analyzed effects of their cultured broths on inhibiting insect immunosuppression. For this, we removed all bacterial cells using $0.2\;{\mu}m$ pore sized membrane from the bacteria-cultured broth. All three sterilized cultured media, in dose-dependent manners, significantly inhibited hemocyte-spreading behavior of 5th instar larvae of Spodoptera exigua. However, they showed differential inhibitory activities among different bacterial species, in which X. nematophila showed the most potent inhibitory activity. This immunosuppressive effect was applied to increase the pathogenicity of Bacillus thuringiensis (Bt). All three bacterial cultured broths including bacterial cells significantly potentiated Bt pathogenicity against young S. exigua larvae when each of them was orally administered in a mixture of low dose of Bt. Finally, we tested the effect of oral administration of the cultured media containing the immunosuppressive compound(s) secreted by the bacteria. The membrane-sterilized cultured broths were mixed with the low dose of Bt and then orally administered to the young S. exigua. Only the cultured medium of X. nematophila showed increase of Bt pathogenicity. These results indicated that the; cultured media of the three bacteria possessed immunosuppressive factor(s), which may act to potentiate Bt toxicity to young S. exigua larvae.

  • During summer in Korea, the excessively high temperature causes growth retardation and quality reduction in cut roses grown in greenhouse. Mist treatments were conducted to reduce the temperature and avoid quality reduction of cut roses. The temperature change in the greenhouse, growth and quality of cut roses, and injuries caused by insects or fungi were investigated during mist treatment. Daily maximum temperature reduced as the number of mist treatment increased, resulting in $6^{\circ}C$ reduction by mist treatment for 10 seconds at 5 min interval. This temperature reduction occurred only when maximum temperature was over $40^{\circ}C$ in greenhouse, and not when it was less than $40^{\circ}C$ or rainy and/or cloudy day. Plant height and fresh weight of the cut roses were increased at the range of 10-20% by mist treatment. As frequency of mist treatment increased, however, malformed flowers increased and vase life of cut rose was largely shortened. The injuries by insects like as beet armyworm larvae and scale insect increased as well. In conclusion, it is recommended that mist treatment must be used when the daily maximum temperature in a green house is over $40^{\circ}C$ and forecasting for disease or insects should be conducted as well.

  • The objective was to know if chinese cabbage growers in Gangwon alpine farmland control agricultural pests including weeds effectively and use pesticide properly. Examiners visited 185 farmers at Taebaek, Pyongchang, and Jeongseon and surveyed 33 questions on pest control methods pesticide use. Chinese cabbage farmers have noxious plant diseases such as clubroot, bacterial soft rot, downy mildew, anthracnose, and mosaic disease, and also noxious insects such as diamondback moth, aphid, beet armyworm, common cabbage worm, and Japanese native slug. In addition, farmers have noxious weeds such as common chickweed, marsh pepper, hairy crabgrass, common purslane, and horseweed. To control diseases and insects, 51.3% of farmers used many chemical agents, while 20.7% of farmers used chemical agents with too much emphasis on paraquat and glyphosate to control weeds: 87.2% of the answered farmers have a preference of the both non-selective herbicides. Farmers in the survey area selected pesticides on the basis of their own experience and sales managers' recommendation (84.2%) which results in the use of inappropriate pesticides such diniconazole. Many farmers have experienced phytotoxicities (46.7%) and pesticide poisoning (51.2%). We conclude that a systematic educational program for the proper selection and use of pesticides should be conducted for chinese cabbage growers in Gangwon alpine farmland.

  • Insecticidal activities of imidacloprid to sucking type insect pests, brown planthopper (BPH, Nilaparvata lugens) and peen peach aphid (GPA Myzus persicae), to chewing type insect pests, tobacco cut worm (TCW; Spodoptera litura) and beet armyworm (BAW, Spodoptera exigua) and to spiders, Pirata subpiraticus, Pachygnata clercki and Ummeliata insecticeps, as natural enemies were investigated by several bioassay methods. $LD_{50}$ of the chemical by topical treatment to BPH was 0.015 ${\mu}g/g$(48 hrs), while $LC_{50}s$ by leaf dipping and root zone treatment were 18.1 and 21.5 ppm, respectively. There was no difference in insecticidal activities between leaf dipping and root zone treatment. Imidacloprid also showed ovicidal effect of root zone treatment and its $LC_{50}$ was 6.8ppm $LD_{50}$ (48 hrs after treatment) of imidacloprid to GPA was $0.4{\mu}g/g$ in case of topical application and $LC_{50}s$ by leaf dipping and root zone treatment were 1.9 ppm and 13.7 ppm respectively. Leaf dipping was more effective than root zone treatment in GPA At topical application $LD_{50}$ (48 hrs after treatment) of imidacloprid to chewing type insect pests, TCW and BAW, were greater than 1,492 and $312{\mu}g/g$ and $LC_{50}s$ by leaf dipping method were 4,803 and Heater than 5,000ppm respectively. This means that imidacloprid has much less effect on chewing type insect pests, TCW and BAW. $LD_{50}$(48 hrs after treatment) of imidacloprid to wandering spiders, Pirata subpiraticus, Pachygnata clercki at topical application were greater dan $2941{\mu}g/g$ and greater than $2,976{\mu}g/g$ respectively and that to webbing spider, Ummeliata insecticeps, was 357 ${\mu}g/g$. Imidacloprid showed very low toxicity to the spiders and its selective toxicity ratios between spiders and BPH were greater than 19,600, greater than 19,800 and 23,800, respectively.

  • Climate change has been regarded as one of main factors to change Korean insect pest fauna. Especially, a global warming model predicts to expand habitat for insect pests originated from tropical or subtropical regions. Two insect pests, the beet armyworm (Spodoptera exigua) and the diamondback moth (Plutella xylostella), are known to overwinter in some greenhouse conditions without diapause induction in Korea. There was a clear difference between these two insects in seasonal occurrence. P. xylostella occurred only at early spring and fall seasons, but did not occur during summer. In contrast, S. exigua maintained their occurrence from late spring to fall seasons. This study set up a hypothesis that the difference in the seasonal occurrence may be resulted from variation in susceptibility to high temperature. To test the hypothesis, heat tolerance was compared between these two insects. Exposure to $42^{\circ}C$ for 40 min killed 100% individuals of P. xylostella larvae. However, most larvae of S. exigua survived in response to $42^{\circ}C$ even for 80 min. Heat tolerance varied among developmental stages in both insects. Highest tolerant stages were $4^{th}$ instar larvae and adults for P. xylostella, but $1^{st}$ instar larvae for S. exigua. Pre-exposure to $37^{\circ}C$ for 30 min significantly increased heat tolerance in both insects. Induction of heat tolerance accompanied with significant increase of glycerol contents in the hemolymph in both insects and up-regulation of three heat shock protein expressions in S. exigua. These results suggest that the differential susceptibility to high temperature explains the disappearance of P. xylostella during summer, at which S. exigua maintains its occurrence.

  • Bacillus thuringiensis subsp. kurstaki KB100 isolated from the domestic soil have the most effective activity against the beet armyworm, Spodoptera exigua larva. The tannic acid as protease inhibitor might be increased the efficacy of sublethal concentrations of B. thuringiensis. The tannic acid was identified as a protease inhibitor that could increased the efficacy of sublethal concentrations of B. thuringiensis. Mixture of B. thuringiensis and tannic acid was investigated the mortality of S. exigua larva in the laboratory and field. When B. thuringiensis treated to 2nd larva of S. exigua, mortality was shown 54.4%. However, mixtures of B. thuringiensis with 4 and 40 mM tannic acid were increased mortalities to 2nd larva of S. exigua as 64.0 and 95.5%, respectively. Also, synergy effect of mixture of B. thuringiensis and 40 mM tannic acid was increased the mortality of S. exigua 3rd larva to 93.3%, even though 60.0% mortality with only B. thuringiensis treatment. On the other hand, the mortality of mixture with B. thuringiensis and 80 mM tannic acid was 53.3% lower than B. thuringiensis single treatment. In the welsh onion field, the accumulated mortalities of 3 times replicated with mixture of B. thuringiensis and 40 mM tannic acid were 83.9, 89.4 and 66.8% compare with 61.8, 80.4 and 47.3% as only B. thuringiensis treatment, respectively.

  • Samples of water, soil, and sediment were taken from 10 streams near Andong, Korea in May 2004. To assess the degree of environmental pollution of each stream, chemical pollutants such as total notrogen (T-N), total phosphorus (T-P), chemical oxygen demand (COD), heavy metals, organophosphorus pesticides, organochlorine pesticides, and dioxin-like PCB congeners were analyzed by standard process tests or U.S. EPA methods. In addition, biomarkers originated from insect immune systems of beet armyworm, Spodoptera exigua, were used to analysis of the environmental samples. Except Waya-chun stream showing T-N content of 9.12 mg/L, most streams were contaminated with relatively low levels of overall pollutants in terms of T-N, T-P, and COD, compared to their acceptable environmental levels designated by the Ministry of Environment. Contents of Pb and Cd in samples of each stream were much lower than environmentally permissible levels. However, several times higherconcentrations of Pb and Cd were found in locations at Mi-chun, Kilan-chun, and Hyunha-chun streams, in comparison with other streams. Diazinon, parathion, and phenthoate compounds among organophosphorus pesticides were detected as concentrations of 0.19, 0.40, and $1.13\;{\mu}g/g$, respectively, from soil sample collected in the vicinity of Mi-chun stream. On the other hand, 16 organochlorine pesticides and 12 dioxin-like PCB congeners, known as endocrine disrupting chemicals, selected in this study were not found above the limit of detection. Biomarker analyses using insect immune responses indicated that Waya-chun stream was suspected as exposure to environmental pollutants. Limitation and compensation of both environmental analysis techniques are discussed.
