근적외 분광법을 응용한 휴대용 벌꿀 품질 평가 장치 개발 (Development of a Portable Quality Evaluation System for Bee-honeys by Using Near Infrared Spectroscopy)

  • This study was conducted to develop a portable quality evaluation system of bee-honey by near infrared spectroscopic technique. Two kinds of bee-honeys from acacia and polyflower sources were tested in this study. The system consists of power supply, tungsten-halogen lamp, detector, and optical fiber probe. Performance of the system was analyzed by comparing the prediction accuracy of the laboratory spectrophotometer. Total of 346 spectra was divided into a calibration set and a prediction set. The PLS (Partial Least Squares) models were developed to predict the quality parameters of bee-honeys. Reflectance spectra, moisture contents, ash, invert sugar, sucrose, F/G ratio, HMF(hydroxy methyl furfural), and $C^{12}/C^{13}$ ratio of honeys were measured. The PLS models of the laboratory spectrophotometer showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~2.200 nm. The PLS analysis of the portable quality evaluation system showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~1.300 nm and 1.400~1.700 nm. The results showed the feasibility of the portable quality evaluation system to determine the quality parameters of bee-honey in the field during harvesting.

  • Objective : To study anti-inflammatory, analgesic effect and molecular biological mechanism of honey bee venom for aqua-acupuncture, human mast cell line(HMC-1) and human glioma cell line(HS683) were treated with bee venom. Methods : Cell viability of bee venom was tested by 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) asssay. To explore whether anti-inflammatory, analgesic effects of bee venom are associated with the control of gene expression, quantitative RT-PCR analysis of inflammation and pain related genes was performed. Results : The MTT assay demonstrated that cell viability was not decreased by treatment with 10-9 ug/ml bee venom in comparison with 10-2, 10-3, 10-4, 10-5, 10-6, 10-7, 10-8, 10-9, 10-10 and 10-11 ug/ml. sPLA2 and COX-l were down-regulated by treatment with 10-9 ug/ml bee venom in HS683 Cell line in comparison with control. COX-2 was up-regulated by treatment with 10-9 ug/ml bee venom in HS683 Cell line and HSP-2 was up-regulated by treatment with 10-9 ug/ml bee venom in HMC-1 Cell line in comparison with control. sPLA2, COX-1 and COX-2 showed no significant regulation in HMC-1 Cell line and cPLA2 also showed no significant regulation in both HMC-l and HS683 Cell line between control and bee venom treated group.

  • Honey is one of the most significant ingredients in conventional food production in different regions of the world. Honey is commonly used as an ingredient in ethnic food. Beekeeping is performed in various locations as part of the local food culture and an occupation related to pollinator production. It is important to conduct beekeeping so that it generates food culture and helps regulate the regional environment in an integrated manner in preserving and improving local food culture. This study analyzes different types of environmental factors of a smart bee farm. The major goal of this study is to determine the best prediction model between the linear regression model (LM) and the support vector regression model (SVR) based on the environmental factors of a smart bee farm. The performance of prediction models is measured by R2 value, root mean squared error (RMSE), and mean absolute error (MAE). From all analysis reports, the best prediction model is the support vector regression model (SVR) with a low coefficient of variation, and the R2 values for Farm inside temperature, bee box inside temperature, and Farm inside humidity are 0.97, 0.96, and 0.44.

  • The wintering honey bee chambers were constructed and inside environment factors were analyzed to provide high technology for efficient wintering of honeybee colonies. As a result of this study, inside air temperature of yongin chamber under the wide range of outside temperature condition was found to be appropriated for wintering of bee, and inside temperature of beehive was about 2$^{\circ}C$ higher than the air temperature in the chamber. The fan operation schedule did not control the high temperature down properly in the chamber, It was necessary to find a new fan operation schedule to be able to provide the optimum temperature for wintering honey bee colonies. inside humidity variation of Yongin chamber was very much depended on outside humidity.

Effects of Flower Thinning Formulation on Activities of Digestive Enzymes and Acetylcholine Esterase in Honey bee Apis mellifera

  • The effects of a newly developed flower thinning formulation (FTF) on the vitality of the honey bee Apis mellifera were examined by measuring the activities of various digestive enzymes in adult worker bees. First, direct spraying of the FTF solution did not cause any behavioral changes or lethal effects for the honey bees based on 24 h observation. Second, oral ingestion of a sugar solution containing the FTF did not produce any significant change in the activities of amylase, proteinases, lipase, or acetylcholine esterase (AChE) in the worker bees 6 h or 24 h after treatment. Meanwhile, a commercial formulation containing sulfur compounds showed slightly reduced activities for several digestive enzymes and AChE, although no behavioral disturbance. Thus, the results of the present study suggest that the FTF is not toxic for honey bees, in terms of contact and ingestion. Therefore, this newly developed FTF can be used for flower thinning without any detrimental effects on pollinating insects.

Detection of Korean Native Honey and European Honey by Using Duplex Polymerase Chain Reaction and Immunochromatographic Assay

  • Korean native honey (KNH) is much more expensive than European honey (EH) in Korea, because KNH is a favored honey which is produced less than EH. Food fraud of KNH has drawn attention of the government office concerned, which is in need of a method to differentiate between KNH and EH which are produced by the Asiatic honeybee, Apis cerana and the European honeybee, Apis mellifera, respectively. A method to discriminate KNH and EH was established by using duplex polymerase chain reaction (PCR) in this study. Immunochromatographic assay (IC) was examined to analyze the duplex PCR product. The DNA sequences of primers for the duplex PCR were determined by comparing cytochrome C oxidase genes of the two honey bee species. Chelex resin method was more efficient in extracting genomic DNA from honey than the other two procedures of commercial kits. The duplex PCR amplifying DNA of 133 bp were more sensitive than that amplifying DNA of 206 bp in detecting EH in the honey mixture of KNH and EH. Agarose gel electrophoresis and IC detected the DNA of 133 bp at the ratios of down to 1% and 5% EH in the honey mixture, respectively and also revealed that several KNH products distributed by internet shopping sites were actually EH. In conclusion, the duplex PCR with subsequent IC could also discriminate between KNH and EH and save time and labor.

Prevalence of Nosema and Virus in Honey Bee (Apis mellifera L.) Colonies on Flowering Period of Acacia in Korea

  • Honey production from approximately 1.6 million colonies owned by about 199,000 Korean beekeepers was almost 23,000 metric tons in 2009. Nosema causes significant losses in honey production and the virus decreases population size. We initiated a survey of honey bee colonies on the blooming period of Acacia to determine the prevalence of Nosema and virus in 2011. Most Korean beekeepers have moved from the south to north of Korea to get Acacia nectar for 2 mon. This provided a valuable opportunity to sample bees originating from diverse areas in one location. Twenty hives owned by 18 beekeepers were sampled in this year. Nosema spore counts ranged from zero to 1,710,000 spores per bee. The average number of nosema spores per bee was 580,000. Approximately 95% of the colonies were infected with Nosema, based on the presence of spores in the flowering period of Acacia. This indicates that Nosema is the predominant species affecting honeybee colonies. Also, the seven most important honeybee viruses were investigated by reverse transcription-PCR. Among them, four different viruses were detected in samples. Black queen cell virus was present in all samples. Chronic bee paralysis virus was detected in 10% of samples. Deformed wing virus was present in only 5% of the samples. Prevalence of Sacbrood virus was 15%. However, Cloudy wing virus, Israel acute paralysis virus and kashmir bee virus were not detected in any of samples.

근 골격계질환에 대한 Bee Venom 요법 (Bee Venom Therapy for Musculoskeletal Disorders)

  • Bee Venom therapy, as bee honey has long contributed to human life as a valuable fancy food since human history began. It can be found in the literature back up to a thousand years as a form of therapy for management of apin of various origin. Even to the present, there are some physicians and medical personnel who believe in the effects of bee sting therapy throughout the world. However Bee Venom therapy remains somewhat controversial and there are very few controlled studies available to guide clinical practice. We administered apitherapy(Bee venom therapy) against pain of musculoskeletal origin in thirty one patients from october 1988 to october 1990 and got successful results in seventy one percent of patients. Our results suggest that Bee Venom therapy can be one of the methods for conquering pain caused by true inflamation.

벌꿀중의 잔유항생물질 및 Propionic Acid 분석011 관한 조사연구 (A Study on Analysis of Residual Antibiotics and Prop Acid in Honey)

  • A sensitive and simple analytical system for the simultaneous determination of residual oxytetracycline, tetracycline, and doxycycline in honey was described, and that the analytical method for determination of residual propionic acid in honey was established. Experimental subjects were purchased four kinds of honey, native kind honey, acaccia honey, mixed floral honey, chestnut honey in Kyung Sang Nam Do. Several microbiological methods are available to determine tetracycline antibiotecs(TCs) in foods but their precision apears to be variable and the specificity is questionable. These methods are considered to be not suitable for analysis of tetracycline antibiotics in honey because honey itself has bacteriostatic action. For determination of tetracycline antibiotics in honey, therefore the High Performance Liquid Chromatography(HPLC) method was applied, and the propionic acid were determined by Gas Chromatography(5.C). Ethylacetate, as an extract solvent, was found to be suitable for seperation of TCs in honey, but methanol and acetone were not. The recoverly rate of Oxytetracycline(OTC), Tetracycline(TC), Doxycycline(DC) from honey spiked at a level of 10 $\mu $g/g were 97%, 89%, and 91%, respectively. The cailbration curve in TCs was linear expression from 2$\mu $g/ml to 10$\mu $g/ml. As the results of analysis, the residual tetracycline antibiotics were not detected in the 100 samples of honey. The recovery rate of propionic acid from honey spiked at level of 10$\mu $g/g was 98.3% , and the calibra lion curves were linear expression from 21$\mu $g/ml to 101$\mu $g/ml. As the results of analysis, the residual propionic acid was not detected in the 100 samples of honey. Retention time(min) of OTC, DC, and TC were 3.35, 4.61, and 5.30 minutes at the conditions of table 2, respectively, and retention time(min) of propionic acid was 3.50 minutes at the conditions of table 3. The residual TCs and propionic acid were not detected in the 100 samples of honey, but there is a possibility that antibiotics or propionic acid will be to remain in honey if they are used during product period in order to prevent putrefaction of honey-bee.
