Non-Invasive Sex Determination of Asiatic Black Bear (Ursus thibetanus) via Sex-Specific Amplification of the Amelogenin Gene

  • The Asiatic black bear, Ursus thibetanus, is among the most threatened or endangered species in Asia. For its conservation and management, sex identification of U. thibetanus using non-invasive samples (e.g., hair and/or feces) is potentially valuable. In this study, a non-invasive molecular method for sex identification of U. thibetanus samples collected from various countries was first utilized, and it was based on polymerase chain reaction (PCR) amplification of the amelogenin gene via PCRs. Thirty-three bear DNA samples, extracted not only from blood (n=9) but also from hair (n=18) and feces (n=6), were used. We performed sex-specific PCR amplifications of the amelogenin gene using a primer set, SE47 and SE48. The primer set could successfully amplify a single X-specific band for females and both X- and Y-specific bands for males from all blood (100%) and hair (100%) samples. In addition, the primer set could distinguish the sex of bears in four out of a total of six fecal samples (approximately 67%). This study's findings suggest that this molecular method can be applied to sex identification of Asiatic black bears from various Asian regions using non-invasive samples, such as hair and feces.

Observations of Solar Filaments with Fast Imaging Solar Spectrograph of the 1.6 meter New Solar Telescope at Big Bear Solar Observatory

  • Fast Imaging Solar Spectrograph (FISS) is an instrument developed by Seoul National University and Korea Astronomy and Space Science Institute and installed at the 1.6 meter New Solar Telescope of Big Bear Solar Observatory. Using this instrument, we observed solar filaments and analyzed the data focusing on determining the temperature and non-thermal velocity. We inferred the Doppler absorption widths of $H{\alpha}$ and Ca II 8542$\bar{A}$ lines from the line profiles using the cloud model. From these values, we separately determined temperature and non-thermal velocity. Our first result came from a solar filament observed on 2010 July 29th. Temperature inside a small selected region of this ranges from 4500K to 12000K and non-thermal velocity, from 3.5km/s to 7km/s. We also found temperature varied a lot with time. For example temperature at a fixed point varied from 8000K to 18000K for 40 minutes, displaying an oscillating pattern with a period of about 8 minutes and amplitude of about 2000K. We will also present new results from filaments observed in 2011 summer.

  • Nine trace elements in Bear Galls(Fel Ursi) studied in this paper are Na, Mg, K, Ca, Cu, Zn, Fe, Pb and Ge. The contents of Na, Mg, K, Ca, Cu, Zn, Fe, Pb are determined by atomic absorption spectrometry and Ge is determined by graphite atomic absorption spectrometry. In both methods, the sample is digested with nitric-perchloric acid and then ashed. The results of analysis are shown in table and the result obtained from this study are as follows. a. The highest content of trace element of Bear Gall is Na and other elements are getting lower in order of K, Mg, Ca, Ge, Zn, Fe, Cu, and Pb. b. Particularly the content of Ge in Bear Gall was determined.

Intestinal rupture due to small bowel volvulus in a captive American black bear (Ursus americanus) (아메리카 흑곰에서 발생한 소장 염전에 의한 장 파열 증례)

  • Intestinal rupture caused by small bowel volvulus was diagnosed in a captive 13-year-old male American black bear. The animal presented with decreased appetite, depression, lethargy, and mild abdominal distention for 3 days. The animal was treated with antibiotics, hypermetabolites, and digestive medicine daily; however, it died on the third day of treatment. The clinical symptoms included hemorrhagic ascites, gaseous extension of the small intestine, and intestinal rupture caused by small bowel volvulus. Hemorrhagic signs were observed in the lungs and heart. This is the first case to describe small bowel volvulus in mammals of the family Ursidae.

Chromospheric Canopy Fields over a Flux Emergence Region as a Key Condition for Formation of the Sunspot Penumbra

  • A presence of a penumbra is one of the main properties of a mature sunspot, and its formation mechanism has been elusive due to a lack of observations that fully cover the formation process. Utilizing the New Solar Telescope at the Big Bear Solar Observatory, we observed the formation of a partial penumbra for about 7 hours simultaneously at the photospheric (TiO; $7057{\AA}$) and the chromospheric ($H{\alpha}$, $-1{\AA}$) spectral lines with high spatial and temporal resolution. From this uninterrupted, long observational sequence, we found that flux emergence under the stable chromospheric canopy fields resulted in penumbra formation, while emerging flux under the expanding chromospheric fields appeared as transient elongated granules. Based on these findings, we suggest a possible scenario for penumbra formation in which a penumbra forms when the emerging flux is constrained from continuing to emerge, but rather is trapped at the photospheric level by the overlying chromospheric canopy fields.

  • Edwardsiella (E) tarda belongs to the Enterobacteriaceae family and is a motile, gram-negative, rod-shaped, facultative anaerobe regarded as an opportunistic and food-borne pathogen in animals and humans. A 21-year-old male Eurasian brown bear (Ursus arctos arctos) died suddenly without any preliminary signs. Necropsy performed according to standard protocol revealed swollen abdomen with hemorrhagic congestions of the gastroenteric organs, ascites, and hemorrhagic exudates around the mouth. The liver showed discoloration, along with a severely swollen and multiple hemorrhages of the spleen, an elongated gallbladder, and a congested cortex and medullar lesion of kidneys. The stomach contained semi-liquid exudates and undigested chicken exuding a decayed odor. The stomach membranes were dark-gray in color with several cysts in the fundus lesions. Rod-shaped bacteria were found in the major organs by Giemsa staining, identified as E. tarda using a biochemical rapid diagnostic identification kit.

  • Teddy bear is a typical rag doll which has been loved by people all over the world for more than 100 years based on its cute and friendly image. In addition, as it has been together for a long time with us, it is considered as a friend of people with memories of all ages and sexes, not just animal doll. Teddy bear has been developing its appearance and character continually playing a role as the symbol of society and issues of an era beyond toys, however it still remains in the image of stuffed toys. Therefore, more advanced teddy bear characters should be created in line with the current environment and market conditions that are undergoing major changes based on the Internet and smart phones. Thus, the concept of the character and the recent development of the market were reflected and the meaning and current image of teddy bears were analyzed to develop new teddy bear stories, worldviews, and characters through design process. And it was created 3D characters, videos, and SNS channels through the developed 3D character design and motion design. Furthermore, we want to take a look at the direction in which Korea's character business can develop in accordance with global changes and suggest the possibility of entering as a character powerhouse.

MAGNETIC HELICITY PUMPING BY TWISTED FLUX TUBE EXPANSION

  • Recent observations have shown that coronal magnetic fields in the northern (southern) hemisphere tend to have negative (positive) magnetic helicity. There has been controversy as to whether this hemispheric pattern is of surface or sub-surface origin. A number of studies have focused on clarifying the effect of the surface differential rotation on the change of magnetic helicity in the corona. Meanwhile, recent observational studies reported the existence of transient shear flows in active regions that can feed magnetic helicity to the corona at a much higher rate than the differential rotation does. Here we propose that such transient shear flows may be driven by the torque produced by either the axial or radial expansion of the coronal segment of a twisted flux tube that is rooted deeply below the surface. We have derived a simple relation between the coronal expansion parameter and the amount of helicity transferred via shear flows. To demonstrate our proposition, we have inspected Yohkoh soft X-ray images of NOAA 8668 in which strong shear flows were observed. As a result, we found that the expansion of magnetic fields really took place in the corona while transient shear flows were observed in the photosphere, and the amount of magnetic helicity change due to the transient shear flows is quantitatively consistent with the observed expansion of coronal magnetic fields. The transient shear flows hence may be understood as an observable manifestation of the pumping of magnetic helicity out of the interior portions of the field lines driven by the expansion of coronal parts as was originally proposed by Parker (1974).
