Effect of Fermented Small Soybean Powder Mixed with Mulberry Leaf on Metabolic Improvement and Hexokinase Activity in Streptozotocin-induced Diabetic Rats

  • Beans are well known to be high-protein diets. Bean seeds contain arginine, lysine, or glycine-rich proteins which are effective to maintain lower glucose levels. In this study, the synergistic effect of fermented small soybean (Chounggukjang) and mulberry leaf on metabolism and hexokinase activity was investigated in streptozotocin (STZ)-induced diabetic rats. We divided 8 groups as follows: non-diabetic rat group fed with only water diet (NC: control), and STZ-induced diabetic rat groups fed with water (DC), fermented Rhynchosia Nulubilis (Bbc), fermented Glycine max Merr (Ybc), Bbc and YBc (BYbc), mulberry leaf and Bbc (MBbc), mulberry leaf and Ybc (MYbc), or the mulberry leaf, Bbc, and Ybc (MBYbc). Diabetes mellitus was induced in rats by subcutaneous STZ administration (70 mg/kg of body weight). All diet groups were fed with Chounggukjang in a powder form. Three ml of Chounggukjang solution (0.75 mg per gram of body weight) dissolved in distilled water was orally administered to all rat groups after STZ administration except for NC rat group. In groups fed with fermented soybeans, the body weight (increased), food efficiency ratio (FER) (increased), glucose level (decreased) and hexokinase (HK) activity (increased) significantly differed to NC. Among them, particularly in the groups fed with both fermented soybeans and mulberry leaf, kidney weight significantly decreased, whereas HK activity significantly increased compared to DC. These results suggest that Chounggukjang of both fermented soybeans and mulberry leaf is potentially used as an effective functional food to prevent diabetes complications.

  • This study was designed to comparse the metabolic effects of Korean diet pattern which contained all-polished rice and 70% polished rice, and to compare dietary restriction. The results were as follows. 1. Food intake survey to catch the Korean diet pattern The calory intake showed women took much more than men and farm village was higher than city, However, protein intake showed men were higher than women and decrease gradually from city, fishing village, and farm village. Calcium intake showed Seoul was the first rank of all, but was merely 70% of recommended quantities. Fe intake was greatly short especially in women and vitamin C also showed same situation. In the view of food group intake, grains and potatoes were 70.06%, vegetables and fruits 13.05%, meats fishes and beans 11.99%, respectivelly. Oil and fat intake showed lowest percentage as 0.77%. 2. Nutritional experiments albino rats have been fed for 7 weeks with three different diet: all-polished rice diet, 70% polished rice diet, and the standard diet for contral group. Dietary restriction were at 4 levels: 5% , 10%, 15%, 20% in 70% polished rice det. Body weight of 70% polished rice group gained value compare to all polished rice group. In the final organ weight all-polished rice group and 70% polished rice group and 70% polished rice group revealed similar results, but organ weight almost decreased dued to dietary restriction and statistical data showed significant differences between dietary restriction group and none-restriction group. In the femur length, 70% polished rice group was longer than all polished rice group but there was no significant differences. The nitrogen content of liver and muscle of 70% polished rice group was higher than all-polished rice group. Furthermone there was significant differences in the female (P<0.1). In the 10% restriction group(female) and 15% restriction group(male), nitrogen retention was higher than 0% restriction group. The other biochemical analysis such as liver lipid, serum cholesterol, glucose of urine, feces and serum were not revealed any significant differences. As a result of studying, it seems clear that 70% polished rice intake influenced much nourshment to white rat growth than all-porished rice intake, and there is no significant influence to animal growth and metabolic effect even if it was restricted $5{\sim}10%$ of diet.

A study on dietary habits and food intakes in adults aged 50 or older according to depression status (50세 이상 성인과 노인의 우울여부에 따른 식습관 및 식품섭취상태에 관한 연구)

  • Purpose: This study was conducted in order to analyze the dietary habits and food intake status according to the prevalence of depression in adults. Methods: A total of 496 subjects, 224 men and 272 women aged 50 to 100 years were recruited from more than 13 cities in Korea. The questionnaires consisted of general characteristics, dietary life status, food intakes, the mean mini dietary assessment and satisfaction with food-related life questionnaire of the subjects to ask according to the degree of depression. Depression status was evaluated according to the Korean form of the Geriatric Depression Scale (K-GDS). Results: All subjects were divided into depression or normal groups; the prevalence of depression was 13.8% (male) and 23.9% (female). People aged 65 or older were more likely to have higher prevalence of depression and to be bereaved. People in the depression group had poor appetite, almost never dined-out, and ate alone. The food intake of miscellaneous cereals, beans, tofu, chicken/duck meat, cabbage, carrot/amber, fungi, apple, pear, strawberry, grapes, and banana was significantly higher in the normal group compared with the depression group. In particular, significantly higher intake of vegetables and fruits was observed in the normal group. In addition, subjects in the depression group ate more fatty meat and were more dissatisfied with food-related life than those in the normal group. Conclusion: The results of this study imply that the prevalence of depression may affect the dietary habits, food intake, and dietary behaviors in the older adult population.

Development of Analytical Method and Monitoring for Bifenazate in Commercial Agricultural Products (농산물 중 Bifenazate의 분석법 개선 및 모니터링)

  • The analytical method for bifenazate was developed using a HPLC (UVD). Also, analytical condition of LC/MS was set up for bifenazate. We validated the method for the precision and the reproducibility. The correlation coefficient of bifenazate ranged from 0.05 to 2.5 mg/kg was 1.0. Limit of quantitation (LOQ) was 0.01 mg/kg. To measure recoveries from agricultural products such as foxtail millet (cereal grains), kidney bean (beans), orange (fruits), perilla leaves (vegetables) and oak mushroom (mushrooms), bifenazate was spiked. Mean recoveries of bifenazate for each sample were 82.7~104.1% at the level of 0.1 mg/kg and 73.1~104.3% at the level of 0.5 mg/kg. The relative standard deviations (n=3) were 0.2~9.7%. Pesticide residues for bifenazate were investigated in 16 commodities (rice, foxtail millet, buckwheat, kidney bean, peanut, sesame, orange, grapefruit, kiwifruit, spinach, perilla leaves, leek, garlic stem, garlic, ginger and oak mushroom) collected from 22 provinces in 2009. Bifenazate was analyzed using analytical method by HPLC from 304 samples, and residue was not detected.

Development of Han-sik Database Utilizing an Expert Focus Group and Assessment of Han-sik Effects on Diet Quality (전문가 포커스 그룹을 활용한 한식 데이터베이스 작성과 한식 섭취 수준에 따른 식품군 섭취 균형도 평가)

  • This study was conducted to develop a Han-sik database as well as to assess the effects of Han-sik on dietary quality among Koreans. The Han-sik database was developed by a focus group composed of food and nutrition specialists considering the results of Han-sik perception surveys conducted in previous studies for frequently consumed dishes. Among the 1,322 dish items identified in the $4^{th}$ (2007-2009) Korea National Health and Nutrition Examination Survey (KNHANES), 973 items (73.6%) were classified as Han-sik. Han-sik usage was defined as the percentage of Han-sik items of all individually consumed dish items in the dietary data of 22,113 subjects who participated in a 24-hour diet recall in the $4^{th}$ (2007-2009) KNHANES. Dietary quality was evaluated based on adherence to the Korean Food Guidance System (KFGS), which was calculated as the percentage of the number of servings consumed in each food group out of recommended servings according to age and sex. Mean usage of Han-sik was 80.1% and was higher in older age groups, rural areas, as well as in households at the lowest income level. Han-sik usage was also higher on weekdays as well as for breakfast. Adherence to the KFGS was significantly higher for grains, meat fish egg beans, vegetables, and fruits (p<0.001) but lower for milk dairy products and oils fats sugars (p<0.001) across the quartiles of Han-sik usage. The results of this study indicate that Han-sik usage is high among Koreans. Further studies are needed to update the Han-sik database as well as investigate the association between health-related factors and Han-sik use.

A Study to Determine the Consumptive Use of Water for Upland Crops (전작물의 필요수량 결정을 위한 연구)

  • This study was carried out to investigate the consumptive use of water for red peppers and soy beans. The correlation between the soil moisture contents and the selected meteorological factors during the growing season was analyzed. Characteristics of the drought at Jinju, Yeosu, Gwangju, and Mokpo area were figured out in view of frequency analysis. The results obtained from this study could be used as a reasonable criteria for the estimation of the duty of water in the design of upland irrigation systems. Obtained results are summarized as follows: 1. Red peppers were grown at the three levels of soil moisture contents; 75 percent, 50 percent, and 25 percent, respectively. The red pepper grown at the 75 percent of soil moisture content showed the highest yield. The total evapotranspiration during the growing season from red peppers was 471. lmm, which was 86.6mm less than the pan evaporation. 2. The soy bean grown at 75 percent soil moisture content showed the highest yield, although there was no signicant difference in yields among treatments. The total evapotranspiration during the growing season from the soy bean was 342.8 mm, which was 119.2mm less than the pan evaporation. 3. Coefficients of consumptive use(k) and meteorological data are shown on Table-9. 4. The significant correlations between the evapotranspiration and the humidity and daily temperature range were observed. Results are shown on Table-11.. The results of the correlation analysis between the evapotranspiration from the crops and the soil moisture content are shown on Table-12. Development of PCR Primers to Detect Pseudomonas savastanoi pv. phaseolicola from the Bean Seeds (강낭콩 종자에서 Pseudomonas savastanoi pv. phaseolicola의 검출을 위한 PCR 프라이머의 개발)

  • PCR primers were developed to detect Pseudomonas savastanoi pv. phaseolicola, a causal agent of halo blight that occurs in all species of common bean (Phaseolus vulgaris L.), from the bean seeds. A primer set, Psp-JHF and Psp-JH-R, specifically amplified 513 bp fragment from Pseudomonas savastanoi pv. phaseolicola only. A nested primer set, psp-JH-F-ne and psp-JH-R-ne, designed from the $1^{st}$ PCR amplicon, amplified 169 bp fragment. The primer sets did not amplify any non-specific DNA from the seed extracts of Fabaceae including 4 beans, 2 soybeans, and 2 peas. The detection sensitivity of the nested PCR method developed in this study was much higher than that of ELISA and selective medium. PCR assays developed in this study should be useful to detect Pseudomonas savastanoi pv. phasolicola from the bean seeds.

Life Cycle Assessment (LCA) for Calculation of the Carbon Emission Amount of Organic Farming Material -With Emphasis on Hardwood Charcoal, Grass Liquid and Microbial Agents- (유기농자재의 탄소배출량 산정을 위한 전과정평가(LCA) -참숯, 목초액, 미생물제재를 중심으로-)

  • Since 1997, Korean Ministry of Knowledge Economy and Ministry of Environment have established data on some 400 basic raw and subsidiary materials and process like energy, petro-chemical, steel, cement, glass, paper, construction materials, transportation, recycling and disposal etc by initiating establishment of LCI database. Regarding agriculture, Rural Development Administration has conducted establishment of LCI database for major farm products like rice, barley, beans, cabbage and radish etc from 2009, and released that they would establish LCI database for 50 items until 2020 later on. The domestic LCI database for seeds, seedling, agrochemical, inorganic fertilizer and organic fertilizer etc is only at initial stage of establishment, so overseas LCI databases are brought and being used. However, since the domestic and overseas natural environments differ, they fall behind in reliability. Therefore, this study has the purpose to select organic farming materials, survey the production process for various types of organic farming materials and establish LCI database for the effects of greenhouse gas emitted during the process in order to select carbon basic units for agricultural production system compliant in domestic situation instead of relying on overseas data and apply life cycle assessment of greenhouse gas emitted by each crop during the process. As for selecting methods, in this study organic farming materials were selected in the method of direct observation of material and bottom-up method a survey method with focus on the organic farming materials admitted into rice production. For the basic unit of carbon emission amount by the production of 1kg of organic farming material, the software PASS 4.1.1 developed by Korea Accreditation Board under Ministry of Knowledge Economy was used. The study had the goal to ultimately provide basic unit to calculate carbon emission amount in executing many institutions like goal management system and carbon performance display system etc in agricultural sector to be conducted later on. As a result, emission basic units per 1kg of production were calculated to be 0.0088kg-$CO_2$ for charcoal, 0.1319kg-$CO_2$ for grass liquid, and 0.2804kg-$CO_2$ for microbial agent.

The Comparison in Daily Intake of Nutrients and Dietary Habits of College Students in Busan (부산지역 일부 대학생의 식습관 및 영양소 섭취상태에 관한 연구)

  • The purpose of this study was to investigate the dietary habits and daily intake of nutrients in college students. This survey was conducted using a self-administered questionaire. The average heights and weights were 173.5 cm and 72.3 kg of male students and 161.8 cm and 57.2 kg of female students. The average of BMI was $24.2kg/m^2$ of male and $21.9kg/m^2$ of female, and the value of male students was higher than the value of female students. The response to the daily meals was 54.6% for '$2{\sim}3$ times/week'. The regularity of mealtime was 41.7% for irregular and the frequency eating after nine was 45.7% for '5-6 times/week', respectively. The repast was 72.2% for 'overeating and little eating' and was a significant difference of male and female students (p<0.05). The eating rate was higher '$10{\sim}20min$'. As for breakfast food eaten, skipping breakfast was 23.6% for 'no/week' and female students were higher than male students (p<0.05). The frequency of snacks was 36.0% for 'nothing' of males students and 34.8% for '3-4 times/week' of female students (p<0.05). The type of snack was a significant difference of males and females students (p<0.01), and was the highest 75.0% for carbonated drinks of males and 37.5% for snacks of females. The eating due to stress solution was a significant difference of male and female students (p<0.01), and was the highest 23.0% for 'frequency' of males and 44.7% for 'sometime' of females. As for food intake of male and female students, the meat intake was 66.7% for 'everything of male and female students. The fish intake was 68.1 % for '1-2times/week'. The milk, milk products, eggs and beans were each 40.3%, 58.3%, 56.9%, 47.2% for '1-2 times/week' (p<0.05). The fat intake was 55.6% for '$1{\sim}2$ times/week'. The average consumption of energy was 58% of male and 67% of female of estimated energy requirement (EER). Their mean ratio of carbohydrate: protein: fat was 57 : 15 : 28 of all subjects. The mean intakes of vitamin C and folic acid were 70% and 51% of males and 62% and 52% of females of recommended intake (RI). The mean intakes of Ca, P, Fe and Na were 71%, 140%, 146% of males and 72%, 122%, 76% of female of RI and 273% of males and 233% of females of adequate intake (AI). Therefore, nutritional education is necessary for college students to establish physicall and mentall optimal health conditions though nutritional intervention.

Associations of Cognitive Function and Dietary Factors in Elderly Patients with Alzheimer's Disease (알쯔하이머병 노인들의 인지기능과 관련된 식이 요인)

  • The purpose of this study was to investigate nutrients or food factors related to cognitive function of elderly having Alzheimer's disease. In this study 38 subjects who were over 65 years old have participated in dementia clinic at A medical center. After they were diagnosed to Alzheimer's Disease (AD) through blood analysis, neuropsychological test, brain image and interview by medical specialist, we examined for their general information, anthropometry, blood pressure and dietary intakes. Dietary intakes were investigated using the 24-hour recall record. Energy intake was adequate and the energy composition of carbohydrate, protein and fat was 60.8 : 16.2 : 23.0, but dietary intakes of calcium, vitamin A and folate were less than 75% of the recommended intake levels for Koreans. The multiple regression analysis adjusted with age, sex and educational level showed that cognitive function was positively related to intakes of zinc, fishes and shellfishes, beans & nuts, sugars and fats, and negatively related to intakes of plant calcium and eggs. These results indicate that intakes of specific nutrients or food groups are associated with the specific domains of cognitive function in elderly with AD.
