Spontaneous ophthalmic diseases of Beagles in Korea (국내 Beagle견의 안과질환)

  • Beagle is one of the most popular laboratory animal in a dog. We investigated the incidence of spontaneous ophthalmic disease in Beagles in order to make basic data for quality control of Beagles in Korea. We surveyed 389 beagles from 3 different farms. The average prevalence rate of ophthalmic diseases was 19.3%. The prevalence rate of cataracts was higher than any other ophthalmic diseases, 7.5% and then in order of corneal scar(2.1%), retinal hemorrhage(1.5%), post-inflammatory retinal change(1.5%), cherry eye(1.0%), distichiasis(0.8%), entropion(0.8%), hypoplastic optic disc(0.8%), conjunctivitis(0.5%), retinal dysplasia(0.5%), asteroid hyalosis(0.3%), congenital eye anomaly(0.3%), corneal lipidosis(0.3%), conjunctival hemorrhage(0.3%), lens capsule pigmentation(0.3%), persistent hyperplastic primary vitreous (0.3%), posterior synechia(0.3%), eyelid tumor(0.3%) and uveal cyst(0.3%) were diagnosed. There was a slight difference between male(24.8%) and female(17.4%) in prevalence rate of ophthalmic disease. The incidence of spontaneous ophthalmic disease was increased with age. These data would be useful for using Beagles as laboratory animals and standard reference for quality control of dogs.

  • The general pharmacological profiles of SKI 2053R were investigeted on the central nervous system, autonomic nervous system, respiratory-cardiovascular system, digestive system and other systems. SKI 2053R had no significant pharmacological effects. Pharmacokinetic studies on time-course of blood levels, tissue distribution and excretion of SKI 20S3R were performed in rats and beagle dogs after intravenous administration of $^{14}$ C-labeled SKI 2053R. The blood level of radioactivity decreased in bi-or tri-exponential manners: rapidly decreased at $\alpha$-phase but slowly decreased at $\beta$-or ${\gamma}$-phase. $^{14}$ C SKI 2053R was well distributed to all tissues except central nervous system. Tissue concentration profiles of radioactivity were almost consistent wi th those of blood, but higher than those of plasma from 1 to 168 hrs after administration. Also, these results were consistent wi th those of whole body ARG study. The urinary and fecal excretions of radioactivity within 168 hr after administration were 84-87 and 9-11 % of total radioactivity of $^{14}$ C-SKI 2053R administered. In lactating rats, the levels of radioactivity in the milk were significantly lower than that in the blood, but slightly higher than that in the plasma. The disapperance of the radioactivity from the milk was similar as that in the plasma.

Histomorphometric Study of Implants Initially Stabilized through Bone Graft Packing into the Osteotomy before Implant Placement in Case of Wide Defects

  • Purpose: This study sought to evaluate the effects of bone graft wedging on the initial stability of implants in bone sites of unfavorable quality. Materials and Methods: Three male beagle dogs were used in this study. Osteotomies were performed with parallel drills (${\O}4.1{\times}10mm$), and fixtures (${\O}3.3{\times}8mm$) were placed. The control group was given implants without bone graft. Experiment group A was given implants with minimal initial stability using autobone grafts, whereas experiment group B was given xenografts. Groups were also divided by healing times at 4, 8, and 12 weeks. Results: All implants in the control group failed to osseointegrate. On the other hand, all implants in the experiment groups were clinically well-maintained during the entire experiment period. After 4, 8, and 12 weeks, bone-to-implant contact (BIC) ratio and implant stability quotient (ISQ) increased in the experiment groups. The differences between experiment groups A and B were not statistically significant, however. Conclusion: In unfavorable bone regions for dental implants, bone graft packing into the osteotomy prior to implant placement secured minimal initial stability and showed reasonable BIC ratios and ISQ values throughout the study period.

  • The purpose of this study was to analyze comparatively the effect of various root pretreatments prior to delayed replantation. Total 6 beagle dogs were used for the experiment and 8 teeth per each animal were treated by several prepared regimens before replantation. After 3 & 6 weeks of experimental periods, animals were sacrificed by perfusion method. The results obtained from the present study can be summarized as follows: 1. All root pretreatment regimen used in this experiment showed effectiveness in the periodontal repair of delayed replanted teeth. 2. The teeth treated by the regimen of stannous fluoride combined with tetracycline revealed more favorable tissue response and less frequent root resorption or ankylosis than other groups. 3. The long term effect of Group IV-regimen seems to be worth further study since the result at 6-weeks showed the significant progress in periodontal healing when compared to 3-week result which was not indicated in any other group

Oblique Single-Cut Rotation Osteotomy for Correction of Femoral Varus-Torsional Deformities in 3D-Reconstructed Canine Bone Models

  • The purpose of this study was to report the reliability and validity of oblique single-cut rotation osteotomy (OSCRO) in 3D-reconstructed canine bone models with femoral varus and torsional deformities. A healthy adult male beagle was recruited to create a 3D bone model, and this bone model was modified by using a 3D program. Fifteen bone models were constructed for this study. OSCRO simulation was performed in accordance with the plan after printing using a 3D printing machine. The anatomical lateral distal femoral angle (aLDFA), anteversion angle (AA), anatomical caudo-distal femoral angle (aCdDFA), mechanical caudo-distal femoral angle (mCdDFA) and pre- and postoperative bone length were calculated. There were no significant differences between the target values and postoperative values. In addition, the difference between pre- and postoperative bone length was small (p = 0.001). Our findings suggest that OSCRO could be an effective surgical option for MPL with bone deformities in small-breed dogs that often undergo conventional distal femoral osteotomy.

Assessment on Gastrointestinal Transit Movement of Capsule Endoscopy in Beagle Dogs (비글견에서 캡슐 내시경의 위장관 이동분석 평가)

  • The object of this study is to measure the transit time and passage rate of capsule endoscopy (CE) in the gastrointestinal tract in medium sized beagle dogs ($7{\sim}13\;kg$). Animals were divided into four groups: only capsule (group 1, n=10), capsule+water (group 2, n=10), mettoclopramide+capsule (group 3, n=10), metoclopramide +capsule+water (group 4, n=10). The capsule transit times through the stomach and small bowel were evaluated by radiography findings. Gastric transit time (GTT), small intestinal transit time (SITT) and complete passage rate were measured in four groups. GTT's for each group were as follows; $45{\pm}20\;min$ (group 1), $117{\pm}35\;min$ (group 2), $150{\pm}40\;min$ (group 3), and $154{\pm}65\;min$ (group 4), while SITT's were $75{\pm}20\;min$ (group 1), $195{\pm}55\;min$ (group 2), $70{\pm}15\;min$ (group 3), and $76{\pm}15\;min$ (group 4). The complete passage rates were 20% (group 1), 40% (group 2), 20% (group 3), 50% (group 4). In all groups, if CE could pass through the pylorus, it passed all small intestinal tracts within 8 hours (battery life). Administration of water helped CE to pass pylori, except in case of metoclopramide administration. These results indicate that CE could be an useful tool for examining gastrointestinal diseases in the veterinary medicine.

Periodontal regenerative effect of a bovine hydroxyapatite/collagen block in one-wall intrabony defects in dogs: a histometric analysis

  • Purpose: The aim of this study was to elucidate the effect of a bovine hydroxyapatite/collagen (BHC) block in one-wall intrabony periodontal defects in dogs. Methods: A one-wall intrabony periodontal defect (4 mm wide and 5 mm deep) was prepared bilaterally at the mesial side of the mandibular fourth premolar in five beagle dogs. After thorough root planing, block-type BHC ($4{\times}5{\times}5$ mm) was placed on one side. The contralateral defect area did not receive any material as a sham-surgery control. Histological analysis of the sites was performed after an 8-week healing period. Results: Two of five samples in the experimental group healed well without dissipation of the graft materials, and histological analysis revealed excellent regeneration of the periodontal tissues. However, most of the grafted materials had been displaced in the other three samples, leaving only a small portion of the graft. The measured parameters exhibited large standard deviations, and the mean values did not differ significantly between the experimental and sham-surgery control sides. Conclusions: The application of BHC alone-without a barrier membrane-to wide, one-wall intrabony periodontal defects yielded inconsistent results regarding both periodontal regeneration and substantivity of the graft materials. Thus, the use of a barrier membrane for noncontained-type defects is recommended to improve the stability of the grafted material, and to condense it.

Effect of Recombinant Lactobacillus Expressing Canine GM-CSF on Immune Function in Dogs

  • Many Lactobacillus strains have been promoted as good probiotics for the prevention and treatment of diseases. We engineered recombinant Lactobacillus casei, producing biologically active canine granulocyte macrophage colony stimulating factor (cGM-CSF), and investigated its possibility as a good probiotic agent for dogs. Expression of the cGM-CSF protein in the recombinant Lactobacillus was confirmed by SDS-PAGE and Western blotting methods. For the in vivo study, 18 Beagle puppies of 7 weeks of age were divided into three groups; the control group was fed only on a regular diet and the two treatment groups were fed on a diet supplemented with either $1\times10^9$ colony forming units (CFU)/day of L. casei or L. casei expressing cGM-CSF protein for 7 weeks. Body weight was measured, and fecal and blood samples were collected from the dogs during the experiment for the measurement of hematology, fecal immunoglobulin (Ig)A and IgG, circulating IgA and IgG, and canine corona virus (CCV)-specific IgG. There were no differences in body weights among the groups, but monocyte counts in hematology and serum IgA were higher in the group receiving L. casei expressing cGM-CSF than in the other two groups. After the administration of CCV vaccine, CCV-specific IgG in serum increased more in the group supplemented with L. casei expressing cGM-CSF than the other two groups. This study shows that a dietary L. casei expressing cGM-CSF enhances specific immune functions at both the mucosal and systemic levels in puppies.

Quantitative Comparison of Computed Radiography and Film Radiography in Detection of Peritoneal Effusion in Dogs (개에서 실험적으로 복수를 유발한 후, 컴퓨터 촬영술과 필름 촬영술을 이용한 복수량의 정량적 비교)

  • The aim of this report is to compare quantitatively computed radiography (CR) and screen-film radiography (SFR) in the detection of peritoneal effusion in dogs. Normal four beagle dogs and one Maltese dog were used. Each five CR and SFR abdominal images of right lateral and ventro-dorsal position were obtained after lodge of 6 ml, 8 ml, 12 ml, 15 ml, and 18 ml of normal saline by intraperitoneal injection within the abdomen. The reviewers were asked to evaluate each SFR and CR images for the presence of peritoneal effusion using the score by the presence of a peritoneal effusion on a five-point ordinal scale. A receiver operating curve (ROC) analysis compared the two imaging modalities. The present study showed that there was no statistical difference between SFR and CR in the detecting peritoneal effusion, but CR was relatively more sensitive based on the increased area under its ROC analysis. Moreover, Readers were more likely to detect peritoneal effusion on CR images than SFR.

Autonomic Nervous Properties of Atropine and Glycopyrrolate on Heart Rate Variability during Anesthesia with Ketamine-Xylazine in Dogs (개에서 케타민-자일라진 마취동안 심박변이도에 대한 아트로핀과 글리코피롤레이트의 자율신경적 특성)

  • Anticholinergics, which are commonly given as a pre-anesthetic medication to prevent adverse effects in canine anesthesia, can cause cardiac adverse effects. To determine the effects of atropine and glycopyrrolate on the balance of sympathetic nervous tone and parasympathetic nervous tone of the heart during ketamine anesthesia in beagle dogs, heart rate variability(HRV), duration of anesthesia and behavioral changes were evaluated. There were no significant temporal domain differences between atropine and glycopyrrolate. Concerning the frequency domain component, atropine and glycopyrrolate effects were significantly lower(P<0.05) than the control saline-treated group. However, the root mean square of the interval differences between consecutive R peaks(RMSSD) and the standard deviation of Poincare plot perpendicular to the line-of-identity(SD1) in atropine were significantly decreased(P<0.05) from the baseline value, and the low frequency/high frequency ratio(LF:HF ratio) in glycopyrrolate was significantly increased from baseline value(P<0.05). The change of SD1 agreed with that of the high frequency(HF) in the frequency domain component and also with those of respiratory rate and $SpO_2-R$. Our results prove that glycopyrrolate is more suitable as a pre-anesthetic anticholinergic in ketamine anesthesia of dogs with respect to safety and duration of action.
