Purification and Characterization of NAD-Dependent n-Butanol Dehydrogenase from Solvent-Tolerant n-Butanol-Degrading Enterobacter sp. VKGH12

  • The solvent-tolerant bacterium Enterobacter sp. VKGH12 is capable of utilizing n-butanol and contains an $NAD^+$-dependent n-butanol dehydrogenase (BDH). The BDH from n-butanol-grown Enterobacter sp. was purified from a cell-free extract (soluble fraction) to near homogeneity using a 3-step procedure. The BDH was purified 15.37-fold with a recovery of only 10.51, and the molecular mass estimated to be 38 kDa. The apparent Michaelis-Menten constant ($K_m$) for the BDH was found to be 4 mM with respect to n-butanol. The BDH also had a broad range of substrate specificity, including primary alcohols, secondary alcohols, and aromatic alcohols, and exhibited an optimal activity at pH 9.0 and $40^{\circ}C$. Among the metal ions studied, $Mg^{2+}$ and $Mn^{2+}$ had no effect, whereas $Cu^{2+},\;Zn^{2+}$, and $Fe^{2+}$ at 1 mM completely inhibited the BDH activity. The BDH activity was not inhibited by PMSF, suggesting that serine is not involved in the catalytic site. The known metal ion chelator EDTA had no effect on the BDH activity. Thus, in addition to its physiological significance, some features of the enzyme, such as its activity at an alkaline pH and broad range of substrate specificity, including primary and secondary alcohols, are attractive for application to the enzymatic conversion of alcohols.

  • The bi-directional high pile load test(BDH PLT) does not have weaknesses found in the existing bi-directional low pile load test(BDL PLT); it has strong economics, is unbound by load capacity limit and secures quality stability of working piles. In this study, Verification the field found a very high capacity level of stability and reliability of the BDH PLT, as well as outstanding field applicability. Field verifications reaffirmed the advantage of the BDH PLT device, which was capable of loading 90 MN capacities as maximum. It was also found to be durable enough to load high capacity with ease.

Benzyldihydroxyoctenone, a Novel Nonsteroidal Antiandrogen, Shows Differential Apoptotic Induction in Prostate Cancer Cells in Response to Their Androgen Responsiveness

  • The molecular mechanisms of apoptotic induction by benzyldihydroxyoctenone (BDH), a nonsteroidal antiandrogen, isolated from the culture broth of Streptomyces sp., have been previously published in prostate cancer LNCaP cells. Apoptotic induction of BDH-treated LNCaP cells was associated with downregulation of Bcl-xL that caused, in turn, cytochrome c release from mitochondria, and activation of procaspases and specific proteolytic cleavage of poly(ADP-ribose) polymerase (PARP). The purpose of the present study was to investigate the patterns of apoptotic induction by BDH in non-prostate, ovarian cancer PA-1 (androgen-independent and -insensitive) cells and prostate cancer cells with different androgen responsiveness, such as C4-2 (androgen-independent and -sensitive), 22Rv1 (androgen-dependent and -low sensitive), and LNCaP (androgen-dependent and -high sensitive) cells. We found that BDH-treated LNCaP cell proliferation was significantly inhibited in a time-dependent manner and induced apoptosis via downregulation of the androgen receptor (AR) and prostate-specific antigen (PSA), as well as antiapoptotic Bcl-xL protein. However, the levels of BDH-mediated apoptotic induction and growth inhibition in 22Rv1 cells were apparently lower than those of LNCaP cells. In contrast, the induction of apoptosis and antiproliferative effect in BDH-treated non-prostate cancer PA-1 and hormone refractory C4-2 cells were not detectable and marginal, respectively. Therefore, BDH-mediated differential apoptotic induction and growth inhibition in a cell type seem to be obviously dependent on its androgen responsiveness; primarily on androgen-dependency, and then on androgensensitivity.

  • The bi-directional high pressure pile load test(BDH PLT) which is a kind of pile load tests was conducted to find out a reasonable design procedure of large-diameter drilled shafts of large-size building structures. The behaviors of bearing capacity and settlement of the large-diameter drilled shafts were analyzed and the results obtained from BDH PLT were also compared with those obtained from the equations suggested in the specification. In case of the reasonable design procedure adopted, the construction cost could be saved at least 15 ~ 28%. It could be concluded that BDH PLT should be needed for the foundation construction cost reduction of the high-rise building structures.

  • The purpose of this study is to search the characteristics of PVA degrading enzymes which were obtained from Microbacterium barkeri KCCM 10507 and Paenibacillus amylolyticus KCCM 10508, respectively. As a result of the PVA degrading test using crude enzymes, the activity of SAO (secondary alcohol oxidase) was maximized after 2 or 3 days from start of the test, while the activity of BDH (${\beta}$-diketone hydrolase) was gradually increased during the test. Activities of them were maintained in the presence of PVA, but as PVA was gradually degraded, their activity was decreased. PVA was inoculated again into the media, their activity was revealed. This result indicated that above two different enzymes were closely connected with PVA degradation and PVA was degraded by activity of SAO and BDH. Maximum activity of them was 1.5-1.8 unit for SAO and 1.5-2.0 unit for BDH under $35^{\circ}C$ and pH 7.8-8.8, respectively.

  • To overcome the limits of the static pile load test for large diameter drilled shafts, bi-directional low pressure pile load test (BDL PLT) has been used but this test method containes some problems that has not been solved basically. That is, BDL PLT has some problems: difficulty in jack (or cell) arrangement for large test capacity, void remain inside jack (or cell) due to the unrecovery of piston after test etc. In this study, bi-directional double-acting high pressure pile load test (BDH PLT W/DOJ) was developed and confirmed for a in-situ large diameter drilled shaft. At present, test specification of bi-directional pile load test (BDPLT) is being made, and severed main issues (such as, test kinds, test capacity, necessity of use of double-acting and attention of application to service pile) will be contained at the specification.

A Numerical Study on the Estimation Method of the Results of Static Pile Load Test Using the Results of Bi-directional Pile Load Test of Barrette Piles (바렛말뚝의 양방향재하시험을 이용한 정적압축재하시험 결과 추정방법에 관한 수치해석적 연구)

  • Bi-directional pile load test (briefly called 'BDH PLT') cannot be performed at loading levels where ultimate bearing capacity could be assessed in field, it is not possible to precisely determine both ultimate load and yield load and under loading. Since the load is transmitted separately to the skin and the end unlike the static pile load test (briefly called 'SPLT') and the direction of loading on the skin is opposite, such methods could have a result different from actual movements of shafts. In this study, three-dimensional finite element method (briefly called '3D FEM') analysis was conducted from results of the BDH PLT, made with barret piles, which were large-diameter cast-in-place concrete piles, and the calculated design constants were applied to the 3D FEM analysis of the SPLT to interpret them numerically and then, actual behaviors of cast-in-place concrete piles were estimated. First, using the results of the BDH PLT with cast-in-place concrete piles, behaviors of the piles made by loading upwards and downwards were analyzed to calculate load-displacement. Second, the design constants, calculated by the 3D FEM analysis and the back analysis, were applied on the 3D FEM analysis for the SPLT, and from these results, behaviors of the SPLT through the BDH PLT was analyzed. Last, the results of the 3D FEM analysis of the SPLT through the BDH PLT was expressed in relationships as {A ratio of bearing capacity of the SPLT and of the BDH PLT (y)} ~ {A ratio of reference displacement and pile circumference (x)}, and they were all classified by reference displacement at 10.0 mm, 15.0 mm, and 25.4 mm.

  • In this paper, a case of a high-rise building supported by a large-diameter drilled shaft socketed in a weathered ground has been studied. The design case of pile foundation presented in this paper could be a representative example of execution of pile design through three(3) design stages of "preliminary design-proof test-supplementary design". As proof test, two(2) BDH PLT (Bi-directional High Pressure Pile Load Test) were performed at the early construction stage. By following the design process of this way, the economic pile design could be carried out and a large amount of construction cost saving could be realized.

  • Purpose : Catecholamines are the neuro-transmitters in the sympathetic nervous system (SNS) and are activated by stress stimulus. Tyrosine hydroxylase (TH) and Dopamine-${\beta}$-Hydroxylase (DBH) are very important enzymes in the catecholamine synthesis. Corticotropin releasing hormone (CRH) is released in the process of reacting to stresses. The aim of this study is to find out what effects immobilization stresses have on the expression of TH, BDH and CRH mRNA in a rat's brains. Methods : We compare expression levels in rat's brains of TH, DBH and CRH mRNA induced by immobilization stresses between the test group and controled group. The expression levels of TH, DBH and CRH mRNA are measured by RT-PCR and the Western Blotting Analysis (WBA). Results : In brains and adrenal glands of the immobilization stress group, the expression levels of TH and DBH mRNAs are significantly two to three times higher (P<0.01), and CRH mRNAs are approximately one and a half times higher (P<0.05) than those of controlled group. Conclusion : This study suggest that the expression levels of TH, DBH and CRH mRNAs are activated by stress stimulus in a rat's brains and adrenal glands.

Poly(3-hydroxybutyrate) Degradation by Bacillus infantis sp. Isolated from Soil and Identification of phaZ and bdhA Expressing PHB Depolymerase

  • Poly(3-hydroxybutyrate) (PHB) is a biodegradable and biocompatible bioplastic. Effective PHB degradation in nutrient-poor environments is required for industrial and practical applications of PHB. To screen for PHB-degrading strains, PHB double-layer plates were prepared and three new Bacillus infantis species with PHB-degrading ability were isolated from the soil. In addition, phaZ and bdhA of all isolated B. infantis were confirmed using a Bacillus sp. universal primer set and established polymerase chain reaction conditions. To evaluate the effective PHB degradation ability under nutrient-deficient conditions, PHB film degradation was performed in mineral medium, resulting in a PHB degradation rate of 98.71% for B. infantis PD3, which was confirmed in 5 d. Physical changes in the degraded PHB films were analyzed. The decrease in molecular weight due to biodegradation was confirmed using gel permeation chromatography and surface erosion of the PHB film was observed using scanning electron microscopy. To the best of our knowledge, this is the first study on B. infantis showing its excellent PHB degradation ability and is expected to contribute to PHB commercialization and industrial composting.
