The mushroom takes in charge of decomposer in ecosystem and its production is important indicator for sounded ecosystem function. To determine standing crop of basidiocarps(fruit body of mushroom), a weight must be measured by harvesting mushroom individual in the field. But this method has profound affection on the basidiocarps population or its surrounding condition due to habitat destruction. Thus, in this study, without harvesting any mushroom in the field, we developed allometric equation using some morphological parameters to estimate standing crop biomass of basidiocarps. Lentinula edodes, Pleurotus ostreatus, Flammulina velutipes and Conocybe tenera were used for allometry. Morphological variables of the mushroom were pileus diameter, pileus area, stipe length and stipe thickness. Consequently, all the experimental mushrooms species showed significantly correlation in biomass estimation of basidiocarps from allometric equation (p<0.05). As a result of this research, the standing biomass of the basidiocarps could be indirectly estimated with proportional expression, allometric equation drived from morphological characters.
Five basidiocarps of Pholiota species have been collected from the areas of BubJu Temple for last two years, and identified to those of P. adiposa or Pholiota species. The taxonomy of these basidiocarps with the morphological aspects was compared with the analysis obtained from the polymorphisms of PCR-RAPD bands made after reacted with the random primers. The polymorphic variations were observed within the species of the basidiocarps, but not between genomic DNA's of the mycelia obtained and the basidiocarps. Several different bands made from the primers (28 and 36) in PCR-RAPD reactions were identified within the genus of Pholiota and speculated to be specific for the individual basidiocarp of P. adiposa collected. The primers employed here were considered to be very useful for distinguishing the individual isolates or basidiocarps collected from the fields. Also, the basidiospores were obtained from the sporeprints of the above basidiocarps as a simple agar and confirmed with observations of clamp connection under microscopes. The matings of them indicated the 'tetrapolar' type, being different from the 'bipolar' type reported by Japanese basidiocarps of P. adiposa or P. nameko. Based on our work, the edible fungi collected were speculated to be a new breeding resource for those of Pholiota commercialized in Japan.
Agricultural and forest wastes were tested as substrate for the production of Flammulina velutipes. Among the meranti, pine tree, mixed sawdust, coffee waste and peanut hull tested, coffee waste was the best basal substrate for the production of basidiocarps. When various supplements, such as ricebran, brewer's grain, defatted corn flour, defatted soybean flour and defatted rapeseed flour were tested, defatted corn flour was chosen for the production of basidiocarps. Maximum yield of basidiocarps(129.38 g/bottle) was obtained from the mixed medium at 4 : 1 ratio of coffee waste and defatted corn flour.
Tricholoma matsutake (TM=T. caligatum var. nauseoum) is, for an agricultural income earned by Forestry by-products, a very important mushroom in Korea. The mycelia isolated from the basidiocarps were compared with basidiocarps of TM by the random amplified polymorphisms of RAPD-DNA bands. The mycelia were confirmed to be originated from the basidiocarps of TM by cluster analyses of the DNA-bands made from RAPDs and Southern blotting with the band (0.75 kb) identified. The mycelia defined were observed to grow very slowly at the rate of 10 cm per month at $25^{\circ}C$ and also to be semi-transparent and submerged in on PDA. The method developed in this work was considered to be very useful for confirming the mycelia originated from the ectomycorrhizal mushrooms and also to be applied for the fungal mycelia isolated from the commercial useful mushrooms.
To find antitumor components in the hot water extract from the basidiocarps of Hypsizigus mamomus. protein-bound polysaccharides were purified and fractionated by DEAE-cellulose ion exchange column chromatography and Sepharose CL-4B gel filtration chromatography. When a dose of 20 mg/kg/day was injected intraperitoneally into ICR mice. fraction IV of the component showed the highest inhibition ratio of 73.8% against the count of hemolytic plaque forming cells in mice to 3.2 times. when IV was about 30 KD and the fraction was composed of 76.1% polysaccharide and 4.9% protein. The hexosamine was detected in all the fractions, showing that the polysaccharide and protein moieties were bound each other.
The 18S rDNA sequences of Tricholoma matsutake (TM=T. caligatum var. nauseoum) collected in Korea were analyzed for the ectomycorrhizal fungi in the roots of Pinus densiflora. The 514 base pairs of rDNA region were synthesized by UF-5 and UR-6 primers, and double checked in the base pair. The sequence of four strains synthesized were all identical in this work, but different from those done by the previous workers. The basidiocarps collected in this work. were identified to T. matstake after searching the 18s rDNA by the BLAST in NCBI. Only several base pairs of 18S rDNA analyzed from other related basidiocarps were different from our analyses of 18S rDNA. The dendrogram were made based on the sequences of the 514 bp 18S rDNA by CLUSTAL-X alignment program. The groupings of the species at the level of genus in the dendrogram were well constructed.
The ectomycorrhizal roots were collected from the soils around the bases of basidiocarps of the four edible mushrooms in the stands of Pinus densiflora or Querus acutissima communities (Mt. Wol-Ak in Eastern Chung-Puk): The basidiocarps of Tricholoma matsutake (TM), Sarcodon asparatus (SA), S. imbricatum (SI), and Polyozellus multiplex (PM) are usually collected. The ectomycorrhizal roots of TM, PM and SI were related to the roots of P. densiflora, but the other to the roots of Q. acutissima in Korea. Particularly, the basidiocarps of PM were collected in the mixed stand of both P. densiflora and Q. acutissima. The morphologies of the ectomycorrhizal roots were observed to be the yellowish brown coral (dichromatous) or pyramid types in the roots of the pine, but dark brown un-branched sticks (roots) in the ends of ectomycorrhizal roots of Querus plants. The un-branched roots were covered with the dark mycelia (rhizomorph) around them. Therefore, the ectomyorrhizal roots of PM were observed to have two kinds of types; The single blackish un-branched roots were observed to be attached to the yellowish coral type roots. The bundles of TM mycelia were filled with cortical cells (in the roots of P. densiflora), but the mycelia of the other fungi (Aphylloporales) were massed between the cortical cells of P. densiflora or Q. acutissima. Their anatomical and gross features were considered to be simailar but very important in the ectomycorrhizal roots for these edible mushrooms.
We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.
Artificial cultivation of Phellinus linteus was studied. The best yield (B.E. $9.4{\pm}1.6$) of basidiocarps including basidiospore was obtained using Quercus acutissima among the logs of Morus alba, Quercus acutissima, Alnus japonica, Castanea crenata. Optimum temperature and required periods for the basidiocarps formation were $25^{\circ}C$ and about 10 months, respectively.
Two new species of Fulvifomes are described from specimens collected in rainforests of Nonggang Nature Reserve of southern China, based on morphological characteristics and molecular phylogenetic analysis of the internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA (nLSU) sequences. Fulvifomes nonggangensis sp. nov. is characterized by perennial, sessile and solitary basidiocarps, applanate pileus, small cystidioles of 9.9-15.4 × 2.9-3.5 ㎛, large pores of 5-6 per mm, a dimitic hyphal system, and broadly ellipsoid basidiospores of 4.3-5.3 × 3.3-4.2 ㎛. F. tubogeneratus sp. nov. is characterized by perennial, sessile, and imbricate basidiocarps, a duplex context, small pores of 7-8 per mm, a dimitic hyphal system, and ovoid to subglobose basidiospores of 5.72 × 5.00 ㎛.
