Effects of the special media on the mycelium growth in Agaricus campestris were studied. The results might be summarized as follows: 1. The mycelium growth fo Agaricus campestris were scarecely stimulated on the Peptone basal medium which was added 0.5gr. of Peptone and Dextrose basal medium which was added 1.5gr. of dextrose during the culture for 144 hours. 2. The mycelium of Agaricus campestris on the media which was added the several kinds of vegetable extracts showed a considerable growth for 144 hours. The order is as follows; Carrot-basal medium(4ml./100ml.)>Tomato-basal medium(2ml./100ml.)>Spinach-basal medium(3ml./100ml.). However, the spinach-basal medium among these three media were no significant difference.

However, the Lettuce (Lactuca scariota L.) basal medium and the Cabbage (Brassica chinensis L.) basal medium among these five media are no significant differences.
The study was conducted to determine the optimal basal media and the concentration of plant growth regulator for germination of seeds and growth of plantlet from Dendrobium monile. The results obtained were summarized as follows. Germination was similar in light and dark condition, but the growth of plantlet after germination was better under dark than under light condition in several media. Germination was best in Hyponex and Kyoto solution medium among the 9 media tested. The number of roots/shoot was most in the Hyponex medium containing 0,1ppm NAA and 1.Oppm BA.

The eleven strains, which could be used lactic and acetic acids as carbon sources, were isolated from kimchi and identified; the strains were facultative microorganisms which could be grown at low temperature (10$\circ$C) and around pH 3.2. As results of morphological, biochemical and physiological tests, 5 species of 3 genera were identified as Debaryomyces coudertii, Pichia edia, Pichia chambardii, Pichia haplophilia and Saccharomyces fermentati. Each strain was grown in basal media. In acidic resistance and acid utilization test, Saccharomyces sp. YK- 17 and Saccharomyces fermentati YK-19 were grown well in basal and YM media containing 0.3% lactic acid. And two strains were grown in basal and YM media containing O.3% lactic acid and 0.6% acetic acid. Since strain YK-19 was grown better at 10$\circ$C than that in 25$\circ$C, strain YK-19 was known to be a psychrophilic strain.

Objective: We examined the effect of different culture media on oocyte maturation. Methods: Four groups of media, (1) 0.3% BSA mBASAL-XI-HTF, (2) 0.3% BSA mBASAL-XI-HTF with FSH, (3) 10% FBS mBASAL-XI-HTF and (4) 10% FBS mBASAL-XI-HTF with FSH were prepared. Mouse cumulus enclosed oocytes (CEOs) were incubated in each group of medium. Hypoxanthine (Hx) was mixed to each group of medium in increasing concentrations of 1 mM, 2 mM and 4 mM. CEOs were incubated and assessed for GVBD and MII development at 3, 6, 18 hours. Results: CEOs maturation to GVBD was seen in all four groups during 3 hours of culture, however MII stage of oocytes was seen after 6 hours. Complete arrest of GV stage in 4 mM Hx media without FSH and partial arrest in 2 mM Hx media without FSH were seen during 18 hours of culture but development was not suppressed in 1 mM Hx media without FSH. More prominent GVBD suppression was noted at early 3, 6 hours culture in 1 mM, 2 mM Hx media with FSH compared to media without FSH. But the suppression was recovered at 18 hours. This result suggests that low concentrated Hx and FSH supplemented media can suppress CEOs maturation during early culture period but recovery is resumed or even stimulated at late period. 1 mM, 2 mM Hx 10% FBS medium with FSH had significantly higher rates of MII development (71.7%, 66.7%) at 18 hours compared to other media. Conclusion: Our results show that low concentrated Hx and FSH supplemented 10% FBS media may stimulate MII development after an initial inhibitory effect.

The effects of basal media, carbon, nitrogen, phosphate and some major macro elements on growth and shikonin production in Lithospermum erythrorhizon hairy root culture were studied. Among examined media, growth of hairy root cultured in B5 liquid medium was rapid, whereas shikonin production was high in MS liquid medium. Under B5 basal medium, sucrose concentration for optimal growth and shikonin production was 9% and 4% respectively. The growth and shikonin production on pH changes in B5 medium resulted little effect in pH 5.8 to pH 8.8 ranges, whereas growth was decreased dramatically in both above 8.8 and under 5.8. Nitrogen source and concentration effected on the growth and shikonin production. The highest growth rate was in B5 medium (50 mM $KNO_3$ and 1 mM $NaH_2PO_4)$, whereas the highest shikonin production was in the condition supplemented with 5 mM $KNO_3$ and 10 mM $NaH_2PO_4$.

A protocol for in vitro propagation from pseudobulb sections of Lycaste armomatica (Graham ex Hook) Lindl., an ornamental and fragrant orchid, was developed. The effect of four cytokinins: kinetin (K), metatopolin (mT), $N^6$-benzyladenine (BA), and thidiazuron (TDZ), in equimolar concentrations, was investigated. Shoot formation from apical and basal pseudobulb sections was obtained in all treatments. A few medial sections cultured in media supplemented with BA formed protocorm-like bodies. Shoot formation was greater from the basal section than the apical, and mainly occurred in explants cultured in media containing TDZ. The highest average numbers of shoots per explant were achieved from basal sections cultured in media supplemented with TDZ at 4.4, 8.87 and 2.2 ${\mu}M$, forming on average 9.9, 8.6 and 7.3 shoots per explant, respectively. Since the medial pseudobulb section was the worst explant for propagation of L. aromatica, we recommend that pseudobulbs be divided into two sections; the basal half should be cultured in MS medium supplemented with TDZ at 4.4 ${\mu}M$ and the apical half with TDZ at 2.2 ${\mu}M$. Subculturing individual shoots in MS medium without plant growth regulators allows further development and rooting. A survival rate of more than 90% under greenhouse conditions was achieved. This research represents a direct contribution to the conservation and sustainable use of this valuable natural resource.

This study was conducted to find out the commercial in vitro propagation methods through protocom-like bodies (PLB) induced from shoot tip culture of Miltonia spp. Among several culture media for induction PLB from shoot tip in Miltonia spp., MS basal medium was better than Hyponex, Vacin & Went basal medium and other media supplemented with natural additives. PLB's proliferation and differentiation in Hyponex medium including $20g{\cdot}L^{-1}$ banana + $20g{\cdot}L^{-1}$ sucrose(pH 5.2) was better than in MS medium. It was tendency that solid media showed higher PLB fresh weight than liquid medium or other cotton bridge culture. The dark culture for 1~2 weeks and adding $10{\sim}20g{\cdot}L^{-1}$ sucrose on hyponex basal medium was the most effective to increase the PLBs growth and shoot number.
Although the efforts to establish fish embryonic stem cells (ESCs) have been made for a long time, derivation of authentic ESCs that possess pluripotency is still difficult suggesting a need for the stepwise optimization of the methods to establish fish ESCs. Primary culture of the blastomeres from the embryos at blastula stage is a critical step for establishing continuous ESC lines. Here, we evaluated the effects of temperatures and basal media on primary culture of blastula embryo-derived blastomeres in marine medaka (Oryzias dancena). The blastomeres were isolated from the blastula embryos and cultured in various conditions designed by the combination of 4 temperatures including $28^{\circ}C$, $31^{\circ}C$, $34^{\circ}C$, and $37^{\circ}C$ and 2 basal media including Dulbecco's modified eagle's medium (DMEM) and Leibovitz's L-15 medium (L15). With the exception of a case cultured in L15 at $31^{\circ}C$, the rate of primary cell adherence reached 100% when the blastomeres were cultured over $31^{\circ}C$. The period for primary adherence was significantly shorter in the groups cultured in $34^{\circ}C$ and $37^{\circ}C$ than in the ones in $28^{\circ}C$ and $31^{\circ}C$. The proportion of subculture was significantly high in the group cultured in DMEM at $31^{\circ}C$ compared to the other groups. Collectively, we demonstrated that the culture in DMEM at $31^{\circ}C$ was effective to primary culture of the blastomeres derived from blastula embryos.

We examined the effects of various basal media and exogenous auxins on the growth and saikosaponin production in hairy root cultures of Bupleurum falcatum. Of the various media tested, 2RCM medium was superior for growth and saikosaponin content, in which saikosaponin a, c, and d was produced, however MS medium was resulted in extremely low level in saikosaponin production. By the treatment of 0.5 mg/L IBA the growth rate was increased by about 60% in 2RCM medium, whereas both IAA and IBA showed a potent inhibition effect in saikosaponin biosynthesis by the treatment of 0.01 mg/L~5mg/L.
