Understanding the Protox Inhibition Activity of Novel 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene Derivatives Using Holographic Quantitative Structure-Activity Relationship (HQSAR) Methodology (홀로그램(H) QSAR 방법에 따른 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene 유도체들의 Protox 저해 활성에 관한 이해)

  • Holographic quantitative structure activity relationships (HQSAR) as 2D QSAR between the herbicidal activities against root and shoot of rice plant (Orysa sativa L.) and barnyardgrass (Echinochloa crus-galli), and structures of A=3,4,5,6-tetra-hydrophthalimino, B = 3-chloro-4,5,6,7-tetrahydro-2H-indazolyl and C = 3,4-dimethylmaleimino substituents in 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene derivatives were studied and discussed. The statistical results of four HQSAR models for the herbicidal activities against root and shoot of the two plants showed the best predictability of the herbicidal activities based on the cross-validated $r^2\;_{cv}\;(q^2=\;0.760{\sim}0.924)$, non cross-validated conventional coefficient $(r^2\;_{ncv}\;=\;0.868{\sim}0.970)$ and PRESS values $(0.123{\sim}0.261)$. The results indicated that the qualities of HQSAR models for barnyardgrass were slightly higher than that of rice plant. And also, the predictability of HQSAR models were higher $(q^2\;=\;HQSAR\;>\;CoMFA)$ than CoMFA but the conventional coefficients of HQSAR models lower $(r^2\;=\;HQSAR\;<\;CoMFA)$ than CoMFA. Moreover, from the contribution maps, it was founded that the selectivity between the two plants depends upon the 2-fluoro-4-chloro-5-alkoxyanilino and $R_3$ substituent on the C-phenyl ring. These features suggest where to modify a molecular structure in order to improve its selective of herbicidal activities against barnyardgrass.

  • The seventeen barnyardgrass [Echinochloa crus-galli (L.) Beauv.] accessions, which were collected national-widely in 1986 and selected two times through 1987, were experimented at 1988. To identify the ecological properties of the collected accessions of native barnyardgrass species as a weed, the experiment was conducted with Wagner pots in size of I/500a and under PE film house. 1. Accessions were classified into 5 specific groups in plant type properties by use of data from plant height, number of maximum tillers, erectness, culm length and panicle type, among others. 2. As for species identification, they were clustered into 3 similar groups and 2 individual species by use of data from color, first-glumer type, and erectness. 3. Four groups were identified for elongational properties by plant height of 22 days old seedling, length of culm, panical, leaf length and width, and inter-node and spikelet, among others. 4. Properties on quanititative growth were classified into 4 groups and 1 individual accession corresponding to differential plant height of 22 days old seedling, length of culm, panical, inter-node, leaf-sheath, spikelet, first-glumes length, grain, number of tillers, spike, and grain weight. 5. Due to different daily increasing rate in seedling height, dry weight, number of tillers and ratio in dry weight to plant height, the growth rate properties were clustered into 4 groups and one individual accession. 6. Properties on seedling growth were classified into 4 groups by use of differential date in length and width of first-leaf, plant height, number of tillers, and dry weight of young and medium aged seedling. 7. Responding to heading date, the accessions were classified into 3 groups : temperative sensitive, medium, and short-day length sensitive types, respectively. 8. By integrating of all quanititative and attributable characters, the seventeen accessions were clustered into 4 groups and 2 individual accessions.

Production of a Phytotoxic Substance by Exserohilum monoceras, the Causal Fungus of Barnyardgrass Leaf Blight, and its Response on Host Plants (논피잎마름병균(Exserohilum monoceras)의 독소생산과 그 기주반응)

  • Phytotoxicity of the culture filtrate and culture conditions for the production of the phytotoxin by Exserohilum monoceras 92-044 were examined. The necrotic lesions were developed on the leaves of Echinochloa crus-galli within 48 hrs after inoculation of the culture filtrate, and the leaves were completely blighted within 5∼7 days. Maximum toxicity was found in the culture broth containing 20% V-8 juice. Phytotoxin accumulation and fungal growth reached their highest peak at around 11 days. Typical symptom appeared on the leaf of E. crus-galli within 48 hrs. Only a weak chlorosis appeared on rice, Arundinella hirta (THUNB) and henry crabgrass (Digitaria sanguinalis SCOP.), but no further symptom developed.

Diallel Analysis for Rice Allelopathic Potential against Barnyardgrass Assessed in Field Condition

  • A complete diallel cross was performed to determine the inheritance of allelopathic potential and combining ability of six rice germplasm that showed different level of allelopathic potential against barnyardgrass. Parents and F1 of 30 crosses were assessed for allelophatic potential in field condition. Barnyardgrass (Echinochloa crus-galli (L.) Beauv. var. frumentaceae) was used for rice allelopathic potential indicator which was observed in height and shoot dry weight, then suppression percentage were calculated by comparing to barnyardgrass grown in control plot (no rice). It was found that general combining ability showed no significant effect, whereas specific combining ability and reciprocal effect were significant. Variance analysis confirmed that genetic control of allelopathic potential in rice against barnyardgrass was mostly directed by dominance gene effect. The dominant genes were associated with high allelopathic potential and recessive genes were associated with low allelopathic potential. Heritability in broad sense for barnyardgrass dry weight suppression was around 41%; whereas for barnyardgrass height suppression was 51%. However, heritability in narrow sense was very low. The finding suggests that accumulation of genes associated with allelopathic potential would be a strategy to develop promising varieties in reference with the specific combining ability of cross combinations.

  • This experiment was conducted to investigate effects of different water depths on emergence, emerged soil depth and growth of E. crus-galli in Seoul, Korea. Plant number of emergence was decreased with deepened water depth, and dry weight of plant was especially negatively affected. However, a few E. crus-galli were emerged under 24 cm water depth. Soil depth of germination was increased as water depth decreased, and a few E. crus-galli were emerged below 10 cm soil depth under - 12 ㎝ water depth. But most E. crus-galli were emerged in ca. 2 cm soil depth under water depth over 0 cm.

  • The seventeen barnyardgrass [Echinochloa crus-galli (L.) Beauv.] accessions, which were collected national-widely in 1986 and selected two times through 1987, were experimented at 1988. To study the degree of ecological variations of the collected accessions of native barngardgrass species as a weed, the experiment was conducted with Wagner pots in size of 1/5000a and under P.E. film house. Among 57 characters, 100 grains weight, rate of tiller increase from transplanting to 30 days, the number of the maximum tillers and rate of dry weight increase from 15 DAS to 22 DAS were become intra-specific variarions as higher coefficient of varation than 30%, the size of the first leaf, fresh weight and dry 22 seedlings, rate of productive tiller, panicle number and tillering divergence made intra-specific variations as more and less C. V. than 25%, while the required days upto heading since seeding at June 6. rate of fresh/dry weight and plant height of 22 days old seedling made intra-specific variations as the coefficient of variation of about 50%. Among the attributes the color in leaf, leaf sheath, mid-ribs, node, spike, grain, and plant type, length of leaf marginal tooth, grain shape, purple bend on leaf were seemed useful to indentify the accessible specific.

Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

Assessment of Allelopathic Potentiality and Identification of Allelopathic Compounds on Korean Local Rice Varieties

  • This study was conducted to assess the allelopathic potentiality of Korea traditional seventy-nine rice (Oryza sativa L.) cultivars on barnyardgrass (Echinochloa crus-galli P. Beauv. var. oryzicola Ohwi) and to identify possible allelopathic compounds from selected rice cultivars, such as Seogandodobyeo, Huadobyeo and Heugbalbyeo. In the straw mixture, Seogandodobyeo showed the greatest inhibition (67.07%) on total emergence percentage. The greatest inhibition on total seedling length and dry weight of barnyardgrass occurred in Huadobyeo (58.32 %) and Heugbalbyeo (81.20%), respectively. An HPLC analysis with nine standard compounds showed that the concentrations and compositions of allelopathic compound depend upon the cultivars. Four compounds including Ο-coumaric acid in Seogandodobyeo extracts, four compounds including p-coumaric acid in Huadobyeo extracts, and seven compounds including Ο-coumaric acid in Heugbalbyeo were detected, respectively. Ο-coumaric acid (0.97 mg/g) in Seogandodobyeo, p-coumaric acid (0.92mg/g) in Huadobyeo and Ο-coumaric acid (1.02 mg/g) in Heugbalbyeo was detected as the highest amounts, respectively. The preliminary identification by HPLC analysis resulted in peaks with retention times close to those of standards which were confirmed with EI/MS. The mass spectra of p-coumaric, and Ο-coumaric acids showed molecular ions (m/z) at 164 and 164, and their base peaks (m/z) at 164 and 118, respectively.

  • The comparative molecular field analyses (CoMFA) models for the herbicidal activities against barnyardgrass (Echinochloa crus-galli) and rice plant (Orysa sativa L.) by the substituent (R) on the hexahydroisoindol-1-one ring in a series of new 2-(4-chloro-5-(2-chloroallyloxy)-2-fluorophenyl)-3-thioalkoxy-2,3,4,5,6,7-hexahydroisoindol-1-one derivatives were conducted and discussed for selectivity between both plants. The statistical results of the two best models (B2 & R7) showed the best predictability for the herbicidal activities based on the cross-validated value $q_2(r^2cv.=0.529{\sim}0.755)$ and none cross-validated value $({r^2}_{ncv.}=0.937{\sim}0.945)$, respectively. Based on the findings, the predictability and fitness of the model (B2) for barnyard grass was better than that of the model (R7) for rice plant. From the two models and contour maps, it is revealed that the novel selective character for herbicidal activity between the two plants depend on the electrostatic field and steric field for the substituent of ortho-positions on the S-phenyl group as R-substituent in hexahydroisoindol-1-one ring.
