Study on the Organoleptic Quality Characteristics of Cassia tora teas by Roasting Conditions (볶음조건에 따른 결명자차의 관능적 품질특성에 관한 연구)

  • The roasting condition and organoleptic characteristics in Cassia tora tea were investigated. Intact Cassia tora seeds were composed of water 11.6%, crude protein 13.1%, crude fat 4.4%, crude fiber 13.8%, N-free extract 47.2% and ash 4.9%. Organoleptic qualities in Cassia tora tea were sweetness, astringency, tartness, bitterness, roasted coffee like, roasted barley like and burnt smell. Organoleptic qualities were investigated by descriptive analysis method, too. Overall acceptability was increased by roasting but it was low because of formation of bitterness and burnt smell at excessive roasting conditions. Sweetness was the most important factor in organoleptic quality of Cassia tora seeds and the optimum condition for the best quality was $210^{\circ}C$, 20 minutes.

Roles of Carbohydrate-Binding Module (CBM) of an Endo-β-1,4-Glucanase (Cel5L) from Bacillus sp. KD1014 in Thermostability and Small-Substrate Hydrolyzing Activity

  • An endo-${\beta}$-1,4-glucanase gene, cel5L, was cloned using the shot-gun method from Bacillus sp.. The gene, which contained a predicted signal peptide, encoded a protein of 496 amino acid residues, and the molecular mass of the mature Cel5L was estimated to be 51.8 kDa. Cel5L contained a catalytic domain of glycoside hydrolase (GH) family 5 and a carbohydrate-binding module family 3 (CBM_3). Chromatography using HiTrap Q and CHT-II resulted in the isolation of two truncated forms corresponding to 50 (Cel5L-p50) and 35 kDa (Cel5L-p35, CBM_3-deleted form). Both enzymes were optimally active at pH 4.5 and $55^{\circ}C$, but had different half-lives of 4.0 and 22.8 min, respectively, at $70^{\circ}C$. The relative activities of Cel5L-p50 and Cel5L-p35 for barley ${\beta}$-glucan were 377.0 and 246.7%, respectively, compared to those for carboxymethyl-cellulose. The affinity and hydrolysis rate of pNPC by Cel5L-p35 were 1.7 and 3.3 times higher, respectively, than those by Cel5L-p50. Additions of each to a commercial enzyme set increased saccharification of pretreated rice straw powder by 17.5 and 21.0%, respectively. These results suggest CBM_3 is significantly contributing to thermostability, and to affinity and substrate specificity for small substrates, and that these two enzymes could be used as additives to enhance enzymatic saccharification.

Evaluation of Forage Yield and Feed Value of Winter Crops Following Rice Harvest at Paddy Field in the Southern Region of Korea

  • This study was carried out to evaluate the forage yield and feed value of winter crops such as Italian ryegrass (IRG), oat, rye, barley, and hairy vetch. Forage crops were sown on 15th October 2018 in a paddy field after harvesting rice and forage crops were harvested at two times of 25th April 2019 and 18th May 201 9. The highest dry matter (DM) yield was harvested from rye among the five species of forage crop on 18th May. DM yield of all forages was increased 14.4% on 18th May compared to 25th April. The highest yield of crude protein (CP) was obtained from hairy vetch on 25th April, whereas it was decreased (38.1%) on 18th May. Moreover, the highest total digestible nutrient (TDN) yield and relative feed value (RFV) was harvested respectively from rye and hairy vetch on 25th April. Delay of harvest time considerably increased DM yield but significantly decreased CP and RFV. In conclusion, rye was the best source of DM and TDN and hairy vetch was the high-quality feed. Rye harvested on 25th April is the proper forage for resolving Korea's problem of insufficient forage yield.

Characterization of a Thermostable Lichenase from Bacillus subtilis B110 and Its Effects on β-Glucan Hydrolysis

  • Lichenase is an enzyme mainly implicated in the degradation of polysaccharides in the cell walls of grains. Emerging evidence shows that a highly efficient expression of a thermostable recombinant lichenase holds considerable promise for application in the beer-brewing and animal feed industries. Herein, we cloned a lichenase gene (CelA203) from Bacillus subtilis B110 and expressed it in E. coli. This gene contains an ORF of 729 bp, encoding a protein with 242 amino acids and a calculated molecular mass of 27.3 kDa. According to the zymogram results, purified CelA203 existed in two forms, a monomer, and a tetramer, but only the tetramer had potent enzymatic activity. CelA203 remained stable over a broad pH and temperature range and retained 40% activity at 70℃ for 1 h. The Km and Vmax of CelA203 towards barley β-glucan and lichenan were 3.98 mg/ml, 1017.17 U/mg, and 2.78 mg/ml, 198.24 U/mg, respectively. Furthermore, trisaccharide and tetrasaccharide were the main products obtained from CelA203-mediated hydrolysis of deactivated oat bran. These findings demonstrate a promising role for CelA203 in the production of oligosaccharides in animal feed and brewing industries.

Increasing sustainability in pork production by using high inclusion levels of co-products distillers dried grains with solubles, wheat middling and canola meal doesn't affect pig growth performance and meat quality but reduces boar taint

  • Objective: The present study is to examine the effect of high inclusion of co-products in pig diets (referred to as an alternative diet) during the finishing stage on pig growth performance, meat quality and boar taint compounds. Methods: Growing pigs were fed an alternative diet made with distillers dried grains with solubles (25%), canola meal (20%), and wheat middling (15%) or a control diet based on barley and soybean meal to investigate the impact of co-products on pig performance and meat quality. Sixteen female and sixteen entire male Duroc×(Large White×Landrace) pigs (22.6±2.07 kg, body weight±standard error) were equally allocated to the diets. Results: Pigs fed the alternative diet had a lower feed intake; however, growth rate and feed conversion efficiency were unaffected by diet. A diet by sex interaction was found for gain:feed whereby males fed the alternative diet had the best feed conversion (p<0.01). Pork from pigs fed the alternative diet had lower a* and Chroma and protein % (p<0.05), while other meat quality characteristics were unaffected. The alternative diet reduced backfat skatole levels (p<0.001). Conclusion: A diet containing high inclusion levels of co-products can be fed to pigs during the finishing stage without detrimental effects on pig performance or meat quality and with the potential to enhance pork flavour. This finding suggests a solution to increase the sustainable development of pig production.

Development of Dietary Fiber Enriched Menu for the Aged and Analysis of Sensory and Physicochemical Characteristics (고령 소비자를 위한 식이섬유 강화식단 개발 및 관능적 이화학적 특성 분석)

  • Four dietary fiber enriched menus for the aged were developed and physicochemical were analyzed. Major foods enriching the dietary fiber were as following; kimchi & bean sprouts soup and sweet potato for menu 1, chamcheunamul and grape for menu 2, mushroom and kimchi for menu 3, dried radish leaves and green pepper for menu 4. One portion of calorie, carbohydrate, protein and fats for the aged were calculated as 567 kcal, 92.8 g, 21.3 g and 12.6 g, respectively based on recommended daily allowances for adults. Dietary fiber content increased in the menus 1, 2, 3 and 4 were 17.27%, 20.84%, 16.91%, and 23.64%, respectively. The menu 1 had significantly the highest acceptance scores among the rice, soups, side dishes and overall desirability with the acceptance sores of 4.77, 4.58, 4.62 and 4.69, respectively(p<0.05). Female showed significantly the higher acceptance scores of all of the enriched fiber menu than those of male. Moisture contents of the boiled rice with barely in menu 3 had significantly the highest value of all(p<0.05) and the side dishes of all menus did not show any significant differences at p<0.05. The pH value of boiled rice with barely in menu 3 was significantly the highest and menu 1 of kimchi & bean sprouts soup had significantly the lowest pH values(p<0.05). Hunter L value was significantly the highest in boiled rice with barley. The dietary fiber enriched menus were successfully developed for application and standardization in the meal service planning for the aged as one of trials of basic data collection.

  • Barley embryoless half seeds were incubated in medium containing $10{\mu}M$ GA. Time course activity changes of ${\alpha}-amylase$ were studied in extract and medium seperately by the addition of $0.1{\mu}M,\;5{\mu}M,\;and\;10{\mu}M$ ABA in midcourse incubation of 10 hours after GA treatment. MAK profiles of nucleic acids in embryoless half seeds were compared either with $10{\mu}M$ GA treatment or concomitant treatment with $10{\mu}M$ GA and $10{\mu}M$ ABA after 10 hours incubation, Time course changes of weight increase, chlorophyll, protein and RNA consent in addition to RNase activity were studied in the presence of $10{\mu}M$ GA or $10{\mu}M$ ABA in barley coleoptile sections. After 20 hours incubation in the presence of plant hormones, MAK profiles of nucleic acids and reactive distribution of polysome and monosome were investigated. The above results were summarized as follows. 1) The production of ${\alpha}-amylase$ by treatment with GA alone increased at a linear rate in the incubation period and the active secretion of ${\alpha}-amylase$ began from 18 hours incubation in embryoless half seeds. 2) On the contrary to the partial inhibition by addition of $0.1{\mu}M$ ABA, the production of ${\alpha}-amylase$ was completely inhibited by both $5{\mu}M$ and $10{\mu}M$ ABA within 4 hours. Regardless of concentration of GA, the addition of $5{\mu}M$ ABA in midcourse completely inhibited the production of ${\alpha}-amylase$ 3) ABA treatment gave no effect on the secretion of ${\alpha}-amylase$. 4) There were no differences in RNA fractions between GA treatment and concomitant treatment with GA and ABA in the barlye embryoless half seeds. 5) While GA treatment increased the r-RNA fraction, ABA treatment decreased it and increased the s-RNA fraction in the coleoptile sections. 6) GA treatment increased RNA-DNA fraction best ABA treatment decreased it in the coleoptile sections. 7) While GA treatment suppressed RNase activity, ABA treatment increased it in the coleoptile sections. 8) GA treatment gave no great effect on the total RNA but ABA treatment remarkably diminished it in the coleoptile sections. 9) While GA treatment increased the growth and chlorophyll content, ABA treatment decreased them in the coleoptile sections. 10) GA treatment increased the protein synthesis and polysome formation but ABA treatment decreased them in the coleoptile sections. 11) The inhibition effect of ABA on polysome formation seemed to be resulted from the inhibition of r-RNA synthesis by ABA.

Effect of Mixed Sowing Ratios Between Whole Crop Barley with Hooded Type and Forage Pea on the Forage Yield and Quality (삼차망 청보리와 사료용 완두의 혼파재배가 수량 및 사료가치에 미치는 영향)

  • The study was conducted to clarify the mixed seeding rate of whole crop barley with hood type and forage pea for using of forage crops and to compare the forage yield and quality. At a mixed seeding rate between the whole crop barley (WCB) and forage pea, The heading date and plant height of WCB were not a difference according to mixed seeding rate of forage pea. The tillers of the WCB were a decrease and plant of the forage pea were a increase according to increased seeding rate of forage pea. The lodging index of the WCB was a appearance with distribution of $0{\sim}3$, The lodging index of WCB with a 20kg/10a seeding rate of a only WCB without seeding of the forage pea was 3. The overwintering rate of forage pea was a appearance more than 90% at all treatment. The plant height of forage pea was a increase according to increased seeding rate of forage pea at 14 kg/10a and 20 kg/10a plots of WCB. At a mixed seeding between the WCB and forage pea, The fresh weight was a increase according to increased seeding rate of forage pea and was a appearance more than 3,000 kg at all treatment plot. But the dry matter weight was decrease according to increased seeding rates of forage pea. The dry matter weight of 20 kg/10a seeding rate of a only WCB without seeding of the forage pea showed the most amount with 1,266 kg. The crude protein (CP) content was a tendency to increase according to increased seeding rates of forage pea. But, the relative feed value (RFV) was a tendency to decrease according to increased seeding rate of forage pea. The highest RFV was 183.8 at 14 kg/10a seeding rate of a only WCB without seeding of the forage pea. The acid detergent fiber (ADF) and neutral detergent fiber (NDF) were a increase according to increased seeding rate of forage pea at 14 kg/10a and 20 kg/10a plots of WCB. The highest content of ADF and NDF were 23.9% and 46.3% at mixed seeding rate of 20 kg/10a of WCB with 10 kg/10a of forage pea, respectively. The highest sum of standardized score by fresh weight, dry matter weight, CP, ADF, NDF and RFV was 2.309 at mixed seeding rate of 20 kg/10a of WCB with 7.5 kg/10a of forage pea. The optimum mixed seeding rate was a considered judgment in the order of mixed seeding rate of 20 kg/10a of WCB with 7.5 kg/10a of forage pea, mixed seeding rate of 20 kg/10a of WCB with 5.0 kg/10a of forage pea.

Studies on the Response of Isogenic lines of Compact and lax Type to Fertilization and Plant Density in Malting Barley (맥주맥에 있어서 직두형 및 수두형 Isogenic line의 시비량 및 재식밀도에 대한 반응)

  • Taiwan land race, Taichung 2 rowed barley #1 was crossed with England cultivar, Deba abed in 1972, and two isogenic lines-lax and compact heads-were made in 1982. So as to three fertilizer and three seeding density levels were treated at Dan-Kook Agricultural College farm located in Cheonan in 1984-1985. Lax head type had longer spike length and rachis than compact type. The traits uninfluenced by different head types were heading time, number of grains per spike, yield and soluble nitrogen content. However, the traits related to malting quality such as 1,000 grain weight, assortment ratio, protein content of grain and malt, malt extract, malt total nitrogen content, Kolbach index and diastatic power were significantly affected by head types. The number of spikes per m2, yield, assortment ratio, crude protein content of grain and malt yield ratio, extract yield ratio, malt total nitrogen, souble nitrogen content, Kolbach index and diastatic power were affected by fertilizer and seeding density levels in both isogenic lines. The malting quality was best at 12-12-9 kg/l0a fertilizer level and the yield was greatest at 15-12-9 kg/l0a level. Narrow-spaced or drilling seeding had greater yield and better malting quality than conventional row seeding. The plant with lax head type had some advantages in malting quality, suggesting that this trait would be considered as an selection criterion fer good malting quality lines.

Growth Performance, Carcass and Meat Characteristics of Black Goat Kids Fed Sesame Hulls and Prosopis juliflora Pods

  • Finding alternative feeds, such as sesame hulls and Prosopis juliflora species can attenuate difficulties of feed shortage and reduce the cost of animal feed in arid and semi-arid countries. Thirty-two Black male kids with similar initial weights (BW = $16.7{\pm}0.80\;kg$) and $120{\pm}5\;d$ of age, were used to evaluate the effect of replacing barley grains and soybean meal with Prosopis juliflora pods (PJP) and sesame hulls (SH) on growth performance, digestibility and carcass and meat characteristics. Kids were equally divided into four dietary treatment groups for an 84-d fattening period. Treatment diets had similar crude protein (CP) and metabolizable energy (ME). The treatment groups were: (T1) no PJP nor SH, (T2) 10% PJP and 20% SH, (T3) 15% PJP and 15% SH, and (T4) 20% PJP and 10% SH. A tendency was detected (p<0.08) for dry matter (DM), crude protein (CP), organic matter (OM) and neutral detergent fiber (NDF) intakes that were greater for T2 than T3 and T4 while T1 was not different from all other treatment groups. Ether extract (EE) intake was the greatest (p<0.05) for T2 and the lowest for T1. Acid detergent fiber (ADF) intake was greater (p<0.05) for T2 than T1 while T3 and T4 were intermediate. Final live weight, average daily gain (ADG) and feed conversion ratio (FCR) were comparable among different treatment groups. Digestibility of DM, OM, CP, NDF and ADF were similar among all treatment groups, however, EE digestibility was the lowest (p<0.05) for T1 when compared to other treatments. In addition, nitrogen intake, nitrogen in urine and retained and retention percentages were similar among all treatment groups. However, N loss in feces was higher (p<0.05) for T2 than T3 and T4 while T1 was intermediate. No differences were observed among treatment groups with respect to fasting live weight, hot and cold carcass weights, dressing-out percentages, mesenteric fat, visceral organs, carcass cuts percentages and carcass linear dimensions. No differences were also observed for dissected loin, leg, rack and shoulder tissues except in the total bone % for loin cuts and in the meat to bone ratio for rack cuts. T3 has the greatest total bone % and the lowest meat to bone ratio when compared to all other treatment groups. No differences were observed between treatment groups in all quality characteristics of the longissimus muscle. The present study demonstrates the potential of using PJP and SH for growing kids without adverse effects on growth performance, carcass characteristics and meat quality.
