Quality Properties and Antioxidant Activities of Chocolate with Variable Leaf Powder (잎채소 분말을 첨가한 초콜릿의 품질특성 및 항산화 활성)

  • Purpose: The aim of this study was to investigate the effects of various leaf powders on chocolate by analyzing quality characteristics, antioxidant activities, and sensory characteristics. Methods: Pine needles, mulberry leaves, ramie leaves, barley leaves, and squash leaves were freeze-dried, powdered, and added to the chocolate. Results: Water contents of all leaf powder-added groups were significantly higher than that of the control (15.78%). The pH values of chocolate with leaf powder were lower than that of the control (6.33) while that of mulberry leaf chocolate (6.56) was higher than that of the control. In color measurement, L-value, a-value, and b-value were highest in pine needle chocolate (30.31, 3.52, and 4.78, respectively). The ${\Delta}E$ value (total color difference compared to white board) was the highest in squash leaf chocolate (69.51) and significantly different from that of the control (67.69). The hardness of leaf powder-added chocolate was lower than that of the control (6.84 N), except for pine needles. In the antioxidant activity analysis, chocolate with pine needles and squash leaves had higher polyphenols, flavonoids, and ABTS radical scavenging antioxidant activity than those of the control. Overall consumer acceptability was the most preferred in the control compared to the other groups. However, chocolate with mulberry and barley leaves also received high scores (5.60 and 5.40, respectively) for overall acceptability. Conclusion: It can be concluded that chocolate with barley leaves is desirable for making functional chocolate due to its high consumer preference and positive effects on improving antioxidant activities.

Structural Properties of Naked and Covered Barley Starches (쌀보리와 겉보리 전분의 분자구조적 성질)

  • The structural properties of covered and naked barley starches and those components were investigated. ${\beta}-amylosis\;limit(%)$ of covered and naked barley starches were 58.6 and 56.3%, respectively and those of their amyloses and amylopectins were 87.0, 77.7, 57.6 and 52.0%, respectively. The ratios of outer chain length ${\overline{OCL}}$ and inner chain length${\overline{ICL}}$ for amylopectins of covered and naked barley were about 2.2:1 and 2.0:1, respectively. The elution curves by Sephadex G-75 after debranching starches with pullulanase were similar patterns for two starches and yielded two peaks consisting of amylose component${\overline{dp}}>55$ and bimodal amylopectin components of ${\overline{dp}_(35-45)$ and ${\overline{dp}}(10-20)$ hydrolysates. Also, hydrolysis products by sequential hydrolysis with pullulanase and ${\beta}-amylase$ contained 0.03-0.5% non-hydrolyzed peak components of ${\overline{dp}}>55$. The elution profiles of ${\beta}-amylase$ hydrolysates yielded two peaks consisting of the inner components${\overline{dp}}>55$ and the outer chain components of amylopectin${\overline{dp}}>10$ regardless of samples. However, ${\overline{dp}}$ distributions of ${\overline{dp}>55}$ hydrolysates exhibited the significant differences due to the varieties.

Effect of Cropping System and Application of Cattle Slurry on Forage Production and Environmental Pollution in Paddy Land (논에서 경작형태와 우분액비 시용이 사초생산성 및 환경오염에 미치는 영향)

  • This study was performed to investigate the effects of forage cropping system and cattle slurry on productivity of whole crop rice, whole crop barley and Sorghum-Sudangrass hybrid and environmental pollution in paddy land. Forage cropping system used in this study was consisted of double-cropping whole crop barley followed by whole crop rice applied with cattle slurry (DWBRC) and double-cropping whole crop barley followed by Sorghum-Sudangrass hybrid applied with cattle slurry (DSSBC). The field experiments were conducted on the clay loam at Backsanmyun, Kimje, Chunlabukdo province in Korea for three years (May 2006 to Apr. 2009). This study was arranged in completely randomized design with three replicates. The field had been sown with whole crop rice 'Nampyung', Sorghum-Sudangrass hybrid 'Sordan79' and whole crop barley 'Younyang'. The yields of whole crop barley in DWBRC and DSSBC were 7,515 kg/ha and 8,515 kg/ha, respectively. The yields of whole crop barley in DSSBC significantly increased as compared with that of DWBRC (p<0.05). The contents of crude protein, neutral detergent fiber (NDF), acid detergent fiber (ADF), total digestible nutrient (TDN) of whole crop barley in DWBRC were not difference as compared with those of DSSBC. The pH, and contents of total nitrogen and organic matter in soil samples collected at the end of the experiment increased as compared with those at the beginning of the experiment (p<0.05). However, The content of phosphate in DWBRC was no difference as compared with DSSBC. In addition, after the end of experiment, the concentrations of exchangeable cations (Ca, Na, Mg and K) in soil samples collected at the end of the experiment were remarkably higher than those at the beginning of the experiment (p<0.05). The concentrations of $NH_4$-N, $NO_3$-N, $PO_4$-P, Cl, Ca, K, Mg and Na in leaching water were hardly influenced by the cropping system and application of cattle slurry.

Changes of Sucrose Content and Invertase Activity in Leaves of Barley Seedlings under Low Temperature (저온 처리한 보리 유식물 잎의 설탕함량과 Invertase의 활성변화)

  • Changes of contents of reducing sugar and sucrose and activities of sucrose-phosphate synthase, sucrose synthease and invertase from the leaves of barley (Hordeum vulgare L. cv. Chalssal) seedlings grown at $4^{\circ}C$ were investigated, and the property of acid invertase were also examined. In the seedlings grown at $4^{\circ}C$ for 3 days, the contents of reducing sugar and sucrose were increased to 1.3 and 2.4 times, respectively. Activity of acid invertase was decreased markedly by cold treatment while the activities of sucrosephosphate synthase, sucrose synthase, and alkaline invertase were not changed. In acid phosphatase purified partially by ammonium sulfate fractionation and DEAE-Sephacel column chromatography, the $K_m$ value for sucrose was 9.5 mM and the optimum pH and temperature was 5.5 and $35^{\circ}C$ respectively. This enzyme was supposed to be ${\beta}-fructosidase$ by studies on the substrate specificity and the molecular weight was estimated to be 63 Kd by Sephadex G-200 gel chromatography.graphy.

Studies on Rheological Characterization of Barley ${\beta}-Glucan$ [mixed-linked $(1-3),(1-4)-{\beta}-D-Glucan$] (보리 ${\beta}-Glucan$ [mixed-linked $(1-3),(1-4)-{\beta}-D-Glucan$의 리올로지 특성)

  • Crude ${\beta}-glucan$ extracted from Barley was purified by stepwise enzyme treatment (Thermostable ${\alpha}-amylase$, amyloglucosidase, protease). The Intrinsic Viscosity $[{\eta}]$ of the purified ${\beta}-glucan$ was determined by Cannon Fenske Capillary Viscometer (size 50, Cannon Instruments, State, College pa.) at different pH (2, 4, 7, 9, 11) and various salt concentration (0.01 M, 0.03 M, 0.05 M, 0.07 M, 0.1 M and 0.2 M). The $[{\eta}]$ of purified ${\beta}-glucan$ was ranged from $0.997{\sim}2.290\;dl/g$. The $[{\eta}]$ of purified ${\beta}-glucan$ at both alkali, acid condition were lower than those at pH 7. However, the alkali condition of puified ${\beta}-glucan$ solution showed less $[{\eta}]$ than the acid condition of this solution. From 0 M to 0.2 M salt concentration, the $[{\eta}]$ of purified ${\beta}-glucan$ solution was decreased to 0.03 M then increased to 0.05 M NaCl and remained constant to 0.2 M NaCl. The chain stiffness parameter of purified ${\beta}-glucan$ was not affected by temperature from $15^{\circ}C$ to $65^{\circ}C$. The shear rates of various ${\beta}-glucan$ conditions were determined by Bohlin Rheometer (Lund, Sweden). The ${\beta}-glucan$ concentration of 1.0 g/dl and 2.0 g/dl behaved as Newtonian fluid. However, above the concentration of 3.0 g/dl ${\beta}-glucan$ solution, it showed thixotropic and psedoplastic characteristics. Barley ${\beta}-glucan$ appears a damping at 0.5 frequency for the 4.0 g/dl solution. Below 0.5 frequency, it appears a viscous behavior property and above 0.5 frequency, it appears a elastic behavior property.

Hypoglycemic Effects of Basidiomycetes Mycelia and Cereals Fermented with Basidiomycetes (버섯 균사체 및 배양곡물의 혈당강하 효과)

  • To develop basidiomycetes-fermented cereals with hypoglycemic property, inhibitory effects of basidiomycetes mycelia and basidiomycetes-fermented cereals on postprandial glucose were investigated. In vitro effect of basidiomycetes mycelia on retarding the membrane transport of glucose was compared with pectin. For basidiomycetes mycelia, $13.1{\pm}3.6{\sim}41.8{\pm}8.0%$ of total glucose remained in inner solution of dialysis membrane after dialysis for 120 min, indicating that most of basidiomycetes mycelia might effectively retard membrane transport of glucose. Glucose tolerance of basidiomycetes mycelia and basidiomycetes-fermented cereals was tested on streptozotocin-induced diabetic rats administrated with maltose. Postprandial glucose levels of basidiomycetes mycelia, $389.4{\pm}43.8{\sim}426.3{\pm}49.4mg/dL$, were considerably lower than that of control, $535.3{\pm}78.6mg/dL$, at 30 minutes after maltose administration. Namely, basidiomycetes mycelia showed better postprandial glucose lowering effect than pectin. Brown rice and barley fermented with Paecilomyces japonica showed much lower postprandial glucose level than raw brown rice and barley, especially hypoglycemic effect of barley fermented with Paecilomyces japonica was significant.

Sensory and Chemical Characteristics of Worts Fermented by Leuconostoc citreum and Saccharomyces cerevisiae and Consumer Acceptability with Added Flavorings

  • This study was conducted to examine the chemical and sensory characteristics of fermented worts and consumer acceptability according to added flavorings. The worts were fermented by yeast (Saccharomyces cerevisiae) following fermentation by lactic acid bacteria (Leuconostoc citreum) at different aeration conditions. Chemical and sensory descriptive analyses were conducted to examine the effects of the fermentation conditions. The consumer acceptability of the worts with added flavorings was also examined. Organic acids, functional sugars, and ethanol were produced by L. citreum and S. cerevisiae, respectively. Ethanol concentrations ranged from 10 to 25 g/L depending on the fermentation conditions. The sensory characteristics of the fermented worts were clearly differentiated by the fermentation conditions. Yeast fermentation resulted in high intensities for certain sensory attributes such as 'alcohol', 'fermented barley', 'fermented white grape', and 'grassy'. Consumer acceptability changed with different levels of sugar and lemon flavoring, and the optimum levels were determined as 14.08% sugar and 0.98% lemon flavoring. Under these conditions, it was shown that a relatively acceptable fermented wort beverage containing functional materials can be produced.

Cultural Characteristics of Rhizoctonia cerealis Isolated from Diseased Wheat Fields and Evaluation of the Resistance of Korean Winter Cereal Crops (밀 잎집눈무늬병원균(Rhizoctonia cerealis)의 배양적 특성과 국내육성 맥류 품종의 저항성 평가)

  • It was identified as a sharp eyespot (Rhizoctonia cerealis) that the isolates from abnormal symptoms in wheat that showed yellowing leaves, necrotic spot on stem base and dead tillers. These isolates have slower growth property and fewer mycelia than Rhizoctonia solani AG-1(1A) (KACC 40106). They showed binuclear cell, same media cultural and DNA characteristics to R. cerealis. They caused same symptoms on leaves and stem base appeared in artificial inoculation test, comparing to diseased wheat fields and also affect to maturing of kernels. They have optimal growth temperature and acidity on the artificial media as $20{\sim}25^{\circ}C$ and pH 5~7, respectively. In the investigation of varietal resistance of Korean winter cereal crops to sharp eyespot, there was no resistant in wheat cultivars that all materials infected over 20% diseased ratio. 12 cultivars including 'Anbaekmil', however, considered to moderate resistance with 20 to 30% infection ratio. The others crops using in feeding, whole crop barley, oat, rye and triticale were resistant below 15% diseased degree except the rye that showed over 50% infection rate. It was the first evaluation to sharp eyespot resistance for the Korean feeding crop cultivars. Most tested Korean barley cultivars for malting and food were moderate and susceptible to the sharp eyespot. Only 3 hulled barley, 'Tapgolbori', 'Albori' and 'Seodunchalbori', showed resistance with less than 10% diseased ratio. All tested naked barley cultivars showed susceptible response to the disease.

  • This study was conducted to investigate the effects of cattle slurry on productivity of whole crop barley and Italian ryegrass and environmental pollution in paddy land. This study was arranged in completely randomized design with three replicates. The yields of WCB and IRG were 7,520 kg/ha and 10,320 kg/ha, respectively. The yields of IRG significantly increased as compared with that of WCB (p<0.05). The yield of Italian ryegrass in 1st cutting time was about 2-fold higher than that of 2nd cutting time. The contents of crude protein of IRG were higher than that of WCB. However, the contents of NDF, ADF and TDN were no difference between WCB and IRG treatments. The pH, and contents of T-N, $P_2O_5$ and organic matter (OM) in soil samples collected at the end of the experiment increased as compared with those at the beginning of the experiment (p<0.05). After the end of experiment, the concentrations of CEC (Ca, Na, Mg and K) in soil samples collected at the end of the experiment were remarkably higher than those at the beginning of the experiment (p<0.05). The concentrations of $NH_4$-N, $NO_3$-N, $PO_4$-P, Cl, Ca, K, Mg and Na in leaching water in paddy land cultivated with WCB and IRG were no difference between WCB and IRG treatments.

Emulsifying Properties of Octenyl Succinic Anhydride Modified β-Glucan from Barley (옥테닐 호박산 베타글루칸의 유화 특성)

  • We investigated the emulsifying properties of barley octenyl succinic anhydride (OSA)-${\beta}$-glucan, such as changes in the fat globule size and zeta-potential as influenced by pH or the addition of NaCl. Additional experiments to fabricate a suitable co-surfactant system were also performed. We found that the fat globule size in OSA-${\beta}$-glucan emulsions increased upon lowering the pH (i.e., under acidic conditions) or increasing the NaCl concentration. These results were confirmed through microscopic observation. Co-surfactant hydrophilic Tween 20 was found to be suitable for the OSA-${\beta}$-glucan emulsion, which facilitated the formation of smaller fat globules and enhanced the creaming stability when it was added in >0.2 wt% concentration. From the results of the surface load of OSA-${\beta}$-glucan in emulsions, Tween 20 addition enhanced the stability probably by the co-adsorption of the two surfactants at the droplet surface.
