Activities and Isozyme Profiles of Antioxidant Enzymes in Intercellular Compartment of Overwintering Barley Leaves

  • Overwintering plants should survive the various biotic and abiotic stresses that occur during winter. Previous studies indicated that active oxygen species are involved in freezing, dehydration, anoxia and pathogen infections. As the importance of the events that occur in the intercellular compartment became apparent in disease resistance, we examined the nature of intercellular antioxidant enzymes in order to access their possible involvement in the winter hardiness of barley. The levels of intercellular peroxidase, catalase, and SOD activities on the unit protein basis were 394, 18, and 9% of those of cellular activities, respectively. Major intercellular peroxidase isoforms consisted of four neutrals and four basic forms; whereas major cellular isoforms were two basic forms. Out of the two major catalase isoforms a higher molecular weight form was predominantly abundant in both cellular and intercellular compartments. Among the five major cellular SOD isoforms, three were also present in the intercellular compartment. The presence of substantial amounts of intercellular antioxidant enzymes in overwintering barley leaves may suggest the involvement of these enzymes in the tolerance mechanism to the various stresses that occur during winter.

  • This experiment was carried out to show how to make the silage of raddish and Chinese cabbage leaves remained after pickling vegetable in autumn. The moisture of raw materials was controlled by the rice straw and barley bran. The results obtaind were as follows; 1. the silage composed of vegetable leaves 70%, rice straw 20% and barley bran 10% showed the best quality and desirable acid content. 2. Adding barley bran increased the crude protein, crude fat, and crude fiber in the silage. 3. 10% of barley bran made the silage more palatable and also have a good colour.

Genetic Analysis of Apoplastic Proteins in Barley Crosses

  • Antifreeze proteins (AFPs) accumulate in the leaves of barley during cold acclimation, where they may inhibit ice recrystallization and produce freezing resistance of the plant. Four parental diallel crosses of the barley varieties were used to determine the heritability of AFPs and the relationship between the accumulation level of AFPs and freezing resistance. The concentration of apoplastic proteins in the cold-acclimated leaves was increased in the mean by four-fold over as compared with that of nonacclimated. The diallel cross analyses revealed that the gene of Sacheon 6 was dominant and those of Reno and Dongbori 1 were recessive. The AFPs had high narrow-sense heritabilities. The general combining ability effects of Reno and Dongbori 1 were much higher than the other parents. The bands of 32-kD for GLP, 35-& 28-kD for CLP and 25-, 22- & 16-kD for TLP were observed in the apoplastic extracts from cold-acclimated plants, but there were no clear differences between the parents and Fl hybrids. The concentrations of AFPs were significantly correlated with the degree of freezing resistance, indicating that the concentration of AFPs in the plant is the very important factor for freezing resistance.

Greenhouse Method for Assessing Spot Blotch Resistance in Barley

  • New sources of barley (Hordeum vulgare L.) resistant to spot blotch, caused by Cochliobolus sativus, are needed to provide effective resistance because of the rapid change pathotype patterns of C. sativus in fields. The purposes of our study were to develop a method to screen barley for resistance to spot blotch disease and then use this methodology to screen barley genotypes for resistance to the major virulent pathotype Pt4 in barley populations in Syria. A transparent tape method, in which a conidial suspension of C. sativus was dropped onto transparent tape and placed, treated-side down, on the second leaf surface of barley plants. Disease symptoms of fungus were easily detected on the leaves covered by the transparent tape after 48h of inoculation. The transparent tape method was repeatable and the disease scores obtained were correlated (r = 0.91, P = 0.001) with those obtained by the seedling assay. This method may be beneficial in various plant pathology breeding programs.

Functional Characteristics from the Barley Leaves and its antioxidant mixture - Study on the Nitrite Scavenging Effect - (맥엽 및 항산화 조성물의 생리 기능적 특성에 관한 연구 - 아질산염 소거능을 중심으로 -)

  • This study was conducted to estimate the biofunctional characteristics, namely nitrite scavenging effect of barley leaves (BL) and their mixture (AM) with other antioxidants. Aqueous or methanol extracts were obtained from BL and AM. Aqueous and methanol extracts from barley leaves had relatively higher nitrite scavenging effects and its activities and contents increased in a concentration-dependent manner. The activities and contents of methanol extracts obtained from BL and AM were higher than those of aqueous extracts. Especially, AM containing BL and other antioxidant mixture had the highest activities and contents increased in a concentration-dependent manner. BL or AM were added to macrophage cells of RAW 264.7. Survival rates of the cells treated with BL and AM were measured to be different. $IC_{50}$ value decreased in a concentration-dependent manner by the addition of aqueous or methanol extracts from BL and AM. Especially, methanol extracts of AM had the highest nitrite scavenging effects. Thus, BL and AM may have protective effect against carcinogen and immune ability against reactive molecules through NO (nitric oxide) signal pathway. From the above results, barley leaves appear to contain natural anticancer and immune-related agents and may have potentiality to be used as functional food ingredients.

The Effects of Acidic Electrolytic Water on the Development of Barley Chloroplast (산성 전해수가 보리(Hordeum vulgae L.) 엽록체의 발달에 미치는 영향)

  • To investigate the effects of strong acidic electrolytic water on the chloroplast, barley leaves were treated with strong acidic electrolytic water(pH 2.5). And to investigate the effects of weak acidic electrolytic water on the chloroplast development, etiolated barley leaves were treated with weak acidic electrolytic water(pH 6.5) during greening period. Chl contents, Fo, Fv, and Chl fluorescence quenching coefficient in barley leaves were measured during and after treatment of acidic electrolytic water. The following results were obtained. Chl a, b, and carotenoid were decreased with treatment of strong acidic electrolytic water. Chl contents were significantly decreased than that of the control after 5 min. These results provide evidence that the strong acidic electrolytic water dissimilate the Chl and so that the value of Fo was slightly increased. The strong acidic electrolytic water damaged PS II because Fo was increased and Fv, Fm, and Fv/Fm ratio were decreased. qP, qNP and qE were decreased. On the other hand qI was increased than that of the control. But Chl content and Chl fluorescence patterns were a little changed as the pH increase over 4.0 Chl a, b, and carotenoid were increased with treatment of weak acidic electrolytic water during greening period. Chl contents were significantly increased than that of control after 12 hours greening. These results provide evidence that the weak acidic electrolytic water accelerated the chlorophyll synthesis. And the weak acidic electrolytic water accelerated PS II development because Fv, Fm, qP and Fv/Fm ratio were increased than that of the control.

Genetic Analysis of Photoinhibition in Barley

  • Winter cereals are acclimated during wintering, and thereafter their freezing resistance is increased. In order to analyze inheritance and heritabilities for photoinhibition of photosynthesis by high light intensity under low temperature, and to evaluate the relationship between low temperature-induced photoinhibition and winter survival, 4 parental half diallel crosses were used. The detached leaves of 7-8cm long from plants grown for 35 and 55 days were placed on wet filter paper and placed in trays at 5$^{\circ}C$ cold room with 1,200 $\mu$mol $m^{-2}$ $s^{-1}$ PPFD. Chlorophyll fluorescence was measured with a chlorophyll fluorescence system after dark adaptation for 30 min. The Fv/Fm of 35day old plants was reduced from 0.714 in the control leaves to 0.409 and 0.368 following photoinhibitory treatment of 6h and 8h and the CVs were increased from 0.8% to 22.2-22.3%. The Fv/Fm of 55-day old plants was reduced from 0.775 in the control leaves to 0.485 and 0.439 following photoinhibitory treatment of 10h and 12h, respectively. According to half diallel cross analysis, Reno and Dongbori 1 (highly resistant to photoinhibition) was dominant, but Oweolbori (susceptible to photoinhibition) was recessive, and photoinhibition showed partial dominance with highly additive gene action. Dongbori 1 showed the greatest GCA effects for photoinhibition, and GCA/SCA ratios (8.7-22.3 times) indicated that the additive variance for the character was more important. Winter survival in barley crosses was positively correlated with resistance to photoinhibition and significantly fitted by linear regression ($R^2$=0.751$^{**}$-0.779$^{**}$). The chlorophyll fluorescence measured by Fv/Fm has been found to be highly inheritable and very useful in evaluating relative levels of freezing resistance in barley.ley.

Quality Characteristics of White Bread with Barley Leaves Tea Powder (보리잎차 분말을 대체한 식빵의 품질 특성)

  • In this study, white bread was prepared containing 1.5, 3.0, 4.5 or 6.0% barley leaves tea powder (BLTP). The samples along with a control were then compared regarding their quality characteristics, including pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, moisture content, color, textural characteristics, internal surface appearances and sensory qualities, all to determine the optimal ratio of BLTP. As the BLTP content increased, the pH of the dough and bread and the lightness decreased, whereas the total titratable acidity increased. The fermentation power of dough expansion increased with a longer incubation time. The control group was evidenced by a significantly higher specific volume and baking loss than that observed in the BLTP samples. However, greenness and yellowness evidenced the opposite effect. Hardness was highest at a substitution level of 1.5% and lowest at a level of 4.5%. Fracturability and resilience were not significantly different among the samples. For the internal surface appearance, darkness and greenness increased both increased at higher BLTP content. In the sensory evaluation, color, flavor and overall acceptability were highest in the control bread samples but minimal at a substitution level of 6.0%. Softness was the highest at the 3.0% substitution level and lowest in the control bread samples. Barley leaf flavor, astringency, bitterness and off-flavor increased as the BLTP content increased. Delicious taste was not significantly different among the samples. In conclusion, the results demonstrated that 4.5% BLTP may prove quite useful as a substitute for wheat flour in the production of white bread and may provide favorable nutritional and functional properties.

Distribution and Isolation of Soil borne Wheat Mosaic Virus in Korea

  • This study was conducted to investigate the occurrence of Soil borne wheat mosaic virus(SbWMV) in barley fields in Korea and to examine the host pathogenicity of SbWMV. By using the ELISA test, SbWMV was detected in the six regions : Suwon, Milyang, Jinju, Youngkwang, Iksan, and Chonju. SbWMV was isolated from the two strains, Albori strain from Jinju and Eunpamil strain from Milyang. SbWMV was collected from leaves showing mosaic, yellowing and necrosis stripes. SbWMV was inoculated mechanically on 1∼1.5 leaf stages with leaf-rubbing to identify the host pathogenicity of 36 Korean barley cultivars, a wheat cultivar, two rye cultivars, three Japanese barley cultivars and Chenopodium amaranticola. Viral sympoms of inoculated leaves appeared on moulted loaves about 4 to 6 weeks of inoculation. Baegdong and Tapgolbori, infected from Albori strain and Eunpamil strain infected from Samdobori showed much higher susceptibility than C. amaranticola and C. quinoa which showed ring spots and chlorotic spots respectively. Virus particles were observed by the electron microscope. They were rod-shapes, which are bipartite, of 142 nm or 281 nm in length with 20 nm diameter on infected leaves. Specific detection and identification of SbWMV was set up using the RT-PCR. PCR fragments of SbWMV(0.5kb) were obtained by using the designed primers for SbWMV RNA 2.

Characteristics of Induced Resistance to Erysiphe graminis f. sp. hordei in Near-Isogenic Barley Lines. (근 동질유전자 보리계통에서 보리흰가루 병에 대한 유도저항성의 특성)

  • Some characteristics of the resistance induced by Erysiphe graminis f. sp. hordei on near-isogenic barley leaves were evaluated. Inoculation of heat-killed incompatible inducer conidia did not protect barley leaves against compatible challenger race when the inducer inoculum was removed prior to inoculation of challenger. However, the colony formation of challenger race was greatly reduced by 87.1 to $91.2\%$ when the heat-killed inducer inoculum was not removed from the leaves. Although incompatible inducer conidia were removed before they penetrate the host cell, colony formation of challenger was markedly decreased without change in its infection type. After penetrating the host cell by inducer, however, a change in infection types occurred on the challenged leaves. Irrespective of compatibility of previously inoculated inducer on middle part of leaves, there was no reduction in colony formation of challenger race both on the adjacent acropetal and basipetal parts of the same leaves free of inducer inoculation. The colonies formed on the basipetal part by challenger race showed normal 4 type, whereas the infection type of colonies formed on the acropetal part was somewhat changed, thereby sporulation being reduced. The possibility of translocation of resistance-inducing factors was discussed.
