Effect of Gibberellic Acid on Seed Germination and Early Growth of Kentucky Bluegrass and Tall Fescue Turfgrass (Gibberellic acid 처리가 Kentucky bluegrass와 Tall fescue 잔디종자의 발아와 초기생육에 미치는 영향)

  • This study was carried out to find the optimum treatment condition for the improving germination using the plant growth regulators with $GA_3$ and BAP, to Kentucky bluegrass and Tall fescue showing low germination. $GA_3$ treatment was more effective on the germination of Kentucky bluegrass than BAP single treatment or the combination treatment of $GA_3$ and BAP, regardless of their treatment concentration. Seed germinability was 6 days treatment was significantly better than that of 3 days treatment in the experiment for treatment period of the plant growth regulators. When seeds were treated with $1000{\mu}M$ of $GA_3$ for 6 days, on the percent germination was improved as much as between 10% and 38% comparing with the control. In Tall fescue seed, the percent germination was not significantly improved, although it was slightly increased when the plant growth regulators were treated. For the optimum condition, on percent germination was improved as much as 10% comparing with the control of Tall fescue treated with $1000{\mu}M$ of $GA_3$ for 3 days. Percent emergence fur the seeds of Kentucky bluegrass and Tall fescue treated $GA_3$ in the field was improved as much as $8{\sim}9%$ comparing with the control, and mean emergence time $(E_{50})$ was also faster for $1{\sim}2$ days than that of the control. However, dry weight, plant height and root length showed no significant effects, although those were slightly improved more than those of the control.

Effects of Aerobic Exercise and Black Garlic Intake on Blood Lipids, Lipid Peroxidation and BAP in Rats (유산소 운동과 흑마늘 섭취가 흰쥐의 혈중지질, 지질과산화 및 항산화 능력에 미치는 영향)

  • The purpose of this study was to analyze the effects of aerobic exercise and black garlic intake on blood lipids, lipid peroxidation and BAP in rats. The subjects for the study were 25 male Sprague-Dawley rats composed of the control group (CG, n=6), exercise group (EG, n=6), which trained for 4 weeks (30-60 min/day, speed at 15 m/min), black garlic intake group (BGG, n=7), and exercise with black garlic intake group (EBGG, n=6). Blood lipids, lipid peroxidation, and BAP were measured in all the subjects after the end of the 4 week treadmill exercise period. The findings of this study were as follows; TC and TG were significantly lower in the EBGG compared to other groups, while there were no significant findings for HDL-C and LDL-C levels. TBARS was significantly lower in the EBGG compared to the CG, and the BAP from the EBGG and EG were significantly higher than in the CG and BGG after 4 weeks of treadmill exercise. Therefore, aerobic exercise with black garlic intake was effective in improving TC, TG, TBARS, and BAP. Consequently, aerobic exercise with black garlic intake will be effective for activating antioxidant defenses and decreasing cardiovascular diseases.

Adventitious Shoot Formation and Plant Regeneration from Explants of Solanum nigrum L. (까마중(Solanum nigrum L.)의 유식물 절편체에서 부정아 형성 및 식물체 재분화)

  • In the present study, the effects of plant growth regulators on adventitious shoot and root formation of various explants of $in$ $vitro$ seedlings of $Solanum$ $nigrum$ L. were investigated to determine the optimum conditions for the high-efficiency plant regeneration of this species. The formation of adventitious shoots was higher in leaf explants than in cotyledon, hypocotyl, or epicotyl explants at low concentrations (0.5~2.0 mg $L^{-1}$ ) of 6-benzylaminopurine (BAP). The number of adventitious shoots and the shoot length were also higher in both leaf and cotyledon explants. In particular, 2.0 mg $L^{-1}$ BAP was most effective for stimulating the induction and multiplication of adventitious shoots. In terms of root formation and root development from shoots that were separated from multiple shoots, indole butyric acid (IBA) and indole acetic acid (IAA) were more effective than ${\alpha}$-naphthalene acetic acid (NAA). The percentage of rooting as well as the number of roots per shoot (4.0), root length (7.82 cm), and shoot length (8.76 cm) was highest on MS media supplemented with 0.05 mg $L^{-1}$ IAA. Furthermore, 100% of the regenerated plantlets survived when transplanted to compost soil. These results suggest that leaf explants are the best source for the high-efficiency regeneration of $S.$ $nigrum$ L., and that 2.0 mg $L^{-1}$ BAP and 0.05 mg $L^{-1}$ IAA are the best conditions for shoot and root induction, respectively.

Plant Regeneration via Adventitious Shoot Formation from Hypocotyl Explants of Groundcherry (Physalis angulata L.) (땅꽈리(Physalis angulata L.) 하배축 절편으로부터 신초 형성을 통한 식물체 재분화)

  • In the present study, plant regeneration through adventitious shoot formation from hypocotyl segments of in vitro seedlings of groundcherry (Physalis angulata L.) was investigated to determine the optimum culture conditions for highly efficient regeneration of the species. Adventitious shoots in hypocotyl segments were efficiently induced on MS media with low concentrations of BAP, specifically, with 0.5-1.0 mg/L BAP singly or in combination with 0.1-0.5 mg/L NAA. The 1.0 mg/L BAP single treatment was most effective for forming multiple adventitious shoots. When the induced shoots were transferred to the root induction media, low concentrations of NAA, IBA, and IAA enhanced the development of adventitious roots from adventitious shoots, suggesting that low concentrations of auxins were optimal for producing regenerated plantlets. The number of roots per shoot was large (> 2.0), and the root length exceeded 8.0 cm. In particular, the development and the overall shape of the roots were ideal. Furthermore, the number and length of shoots exceeded 2 and 6.0 cm, respectively. When the regenerated plantlets were transferred to compost soil, the root and shoot systems had developed well to the point that all of the regenerated plantlets acclimated successfully, resulting in normal morphology and growth characteristics, similar to those of the mother plant. Therefore, plant regeneration via adventitious shoot formation is expected to be one of the main methods for producing groundcherry on a large scale for a stable supply of the raw materials.

  • Leaf discs and apical meristems were cultured in Murashige and Skoog (MS) medium supplemented with cytokinin and auxin at different concentrations. Callus production was observed in all tested media after six days of incubation. Callus produced in the presence of high concentration of NAA (2.0mg/1) was fragile in texture and yellow in colour. Highest callus formation was observed from leaf discs in the medium supplemented with 1.0mg/1 NAA and 0.5 mg/l BAP in dark at $25{\pm}1{\circ}C$. Percentage of callus formation was 95% and mean callus fresh weight was 654.88 43.53 mg. Shoots were induced from the callus after 4 weeks in 1/2MS medium supplemented with BAP and kinetin both at 0.5mg/1. When elongated shoots were separated and transferred into multiplication medium (MS+0.5mg/1 BAP+0.5mg/1 kinetin) multiplication rate was 6.4 after 6 weeks. Higher concentrations of BAP caused callus production at the base. Direct shoot induction was observed from apical meristems in MS medium in the presence of 0.175 mg/1 IAA + 2.25mg/1 BAP and 0.175 mg/1 IAA + 3.0 mg/1 BAP in 16 hour day at $25{\pm}1{\circ}C$. Explants (apical meristems) elongated to form a single shoot forming a callus at the base. Adventitious buds were sprouted out from the base. Percentage explants which producing shoots was 28.57 and 65.5 respectively. Multiple shoot induction was also observed in the same media. Highest multiple shoot production was observed in the presence of 0.175 mg/l IAA and 3.0mg/l BAP, Mean number of shoots per explant was 5.36 and the mean shoot length was $16.66{\pm}4.15$mm. Shoots (20 30m length) were tested for root induction. Excised shoots were transferred into rooting media, which contains different concentrations of NAA and IAA. Best rooting performance was observed in 1/2MS medium supplemented with 0.1mg/1 NAA after 10 days of incubation in 16 hr photoperiod at $25{\pm}1{\circ}C$. Mean number of roots per shoot was 6 and the mean root length was 252mm. Rooted plantlets were transferred into sterile coir dust:sand (1:4) mixture and maintained in a humid chamber for two weeks, They were gradually exposed to the natural environment. After three weeks they were transferred to pots containing coir dust:sand (1:2) mixture for further development where the 90% survival was observed.

Influence of Hormones and Selection of Stable Cell Lines of Plumbago rosea for Accumulation of Plumbagin

  • Callus and suspension cultures derived from leaf explants of Plumbago rosea were established on Murashige and Skoog's medium containing 1 mg/L IAA, 0.5 mg/L NAA and 0.3 mg/L BAP. Callus cultures were tested for their growth and accumulation of plumbagin, a naphthoquinone and was identified by $^1H$ NMR and electron ionization mass spectroscopy. While auxins (not 2,4-D) influenced growth and plumbagin accumulation, cytokinins did not influence them much. Increasing concentrations of IAA in presence of NAA and BAP increased plumbagin in suspensions only up to 1 mg/L. Growth of callus was optimum (8.3 g DCW/I) at a hormonal combination of 1.5 mg/L IAA, 0.5 mg/L NAA and 0.3 mg/L BAP, but high plumbagin accumulation (4.9 mg/g DCW) was recorded at 1.0 mg/L IAA plus 0.3 mg/L BAP. Since instability in growth and secondary metabolite accumulation was noticed, several cell lines/clumps of callus were screened for plumbagin accumulation by visual and analytical methods. Biomass and accumulation of plumbagin showed a negative correlation in several cell lines. But one cell line showed stability both in terms of biomass and plumbagin accumulation over a period of 6 months.

High Frequency Shoot Regeneration from Leaf Explants of Some Chrysanthemum Cultivars

  • This study was conducted to examine differences in shoot regeneration among chrysanthemum cultivars. Leaf explants of chrysanthemum cultivars 'Sulhwa', 'Puma', 'Geummokseo' and 'Sulpoong' were used. Explants cultured on the medium for 2 weeks formed calli at the cut surfaces. Shoots regenerated on MS basal medium supplemented with various concentration combinations of NAA and BAP. Explants were cultured under cool-white fluorescent lamps with a light intensity of $40\mu{Mm}^{-2}$.$s^{-1}$ for 16 $hday^{-1}$, at $25^{\circ}c$ and 70-80% relative humidity. 'Geummokseo' and 'Sulpoong' were the most responsive cultivars in shoot regeneration. Most effective medium for 'Sulhwa' and 'Puma' was MS basal medium supplemented with 10.0 $\mu{M}$ NAA and 5.0 $\mu{M}$ BAP and for 'Geummokseo' MS supplemented with 10.0$\mu{M}$ NAA and 20.0$\mu{M}$ BAP. Regeneration of multiple shoots was observed on MS basal medium supplemented with 1.0$\mu{M}$ or 10.0 $\mu{M}$ NAA and 5.0$\mu{M}$ BAP. High frequency regeneration of adventitious shoots from leaf explants and efficient induction of root from these regenerated shoots were obtained.

Anthocyanin Production from Culture of Alnus hirsuta Callus (물오리나무 캘러스로부터 Anthocyanin 생산)

  • Callus tissues derived from protoplast of Alnus hirsuta were shown to produce red pigments under the continuous light. The R4 cell line with high yield of the pigments was selected. One of the red pigments was identified as anthocyanin; glucose and galactose as sugar moiety and cyandin as aglycone, by spectroscopic analysis, cellulose TLC and GLC. Both callus growth and anthocyanin accumulation were remarkably stimulated by 2,4-D. The highest anthocyanin yield was observed when $1\;\mu\textrm{M}$ 2,4-D in combination with $0.1\;\mu\textrm{M}$ BAP was supplemented to the culture medium. In case of inorganic salts, anthocyanin yield was enhenced by reducing concentration of phosphate from 5.0 mM to 2.5 mM and by increasing the ratio of ${NO_3}^{-}$ type to 80% for total nitrogen source.source.

In vitro Plant Regeneration from Apical Bud and Nodal Segments of Anthocepahalus Cadamba - An important sacred and medicinal tree

  • Multiple shoot induction and plant regeneration using apical bud and nodal explants of 100 year old tree of Anthocephalus cadamba, an important sacred and medicinal tree in India was achieved for the first time. Aseptic explants cultured in Murashige and Skoog (MS) medium augmented with different concentrations of BAP (0.1, 0.5, 1.0, 2.5, 5.0 and 10 mg/l), when maintained for 60 days, healthy shoots were induced in presence of BAP (1 mg/l). Lower concentrations of BAP (0.1 - 0.5 mg/l) induced only one shoot per explant. Increase in number of shoots per explant was observed in presence of higher concentrations of BAP (2.5, 5.0 and 10 mg/l). However, elongation of shoots was completely inhibited. Bud break and shoot regeneration was largely associated with seasonal factors. Apical buds cultured during June to August exhibited early bud break within two weeks of initial culture. In rest of the months, bud break and shoot regeneration was very slow irrespective of the various concentrations of BAP used in the medium. Explants sourced from three different maturity levels of shoots indicated that actively growing shoots from the mother plant with 1 - 2 nodal segments was more suitable for culture initiation than the explants collected from mature shoots at dormant stage. Regenerated shoots with 2 - 3 pairs of leaves when transferred to half strength MS medium fortified with IBA (1 mg/l), 60% of the shoots induced healthy roots, indicating the possibility of large scale micropropagation.

  • Enzyme activities and phenolic compound were compared to investigate the physiological characteristics during shoot formation from diploid and haploid of Nicotiana tabacum w BY4. The Nakata medium with 1.0 mg/L IAA, 0.5 mg/L Kinetin and 3 g/L activie carbon was excellent to induce the haploid plants from the middle size anther within 30 days after culture. The MS medium with 0.5 mg/L 2,4-D was good for callus induction from leaf explants of diploid and haploid, and a lot of plane were regenerated from calli on the MS medium supplemented with 2.0 mg/L BAE Activities of peroxidase for both of diploid and haploid plane were the highest at 2.0mg/L BAP Activities of IAA oxidase and catalase of haploid Plants were high or than those of diploid plants. On the other hand, activity of peroxidase of haploid plants were lower than those of diploid plants.
