Cultivation of the Hyperthermophilic Archaeon Sulfolobus solfataricus in Low-Salt Media

  • Two low-salt complex media, bactopeptone and desalted yeast extract, were used for high density cultivation of the hyperthermophilic archaeon Sulfolobus solfataricus (DSM 1617). Bactopeptone, which has low mineral ion content among various complex media, was good for cell growth in batch cultures; the maximal cell density in bactopeptone was comparable to that in yeast extract. However, cell growth was rather poor when bactopeptone was added by the fed-batch procedure. Since several vitamins are deficient in abctopeptone, the effect of vitamins on cell growth was examined. Among the vitamins tested, pyridoxine was found to improve the growth rate of S. solfataricus. To reduce the growth inhibition caused by mineral ions, yeast extract was dialyzed against distilled water and then fed-batch cultures were carried out using a fed medium containing desalted yeast extract. Although the concentrations of mineral ions in yeast extract were significantly lowered by the dialysis whether low molecular weight solutes in yest extract are crucial for cell growth, we investigated the effect of trehalose, a most abundant compatible solute in yeast extract, on the growth pattern. Cell densities were increased and the length of the lag phase was markedly shortened by the presence of trehalose, indicating that trehalose plays an important role in the growth of S. solfataricus.

  • A strain of Saccharomyces cerevisiae BY-366 was found to produce a strong sucrose-hydrolyzing enzyme Using this strain, the optimal culture conditions for the production of invertase were investigated. The results are as follows : 1. For enzyme production, optimal temperature, initial pH and critical concentrations of sucrose and raffinose were 3$0^{\circ}C$, 5.0 and 3.0%, respectively. 2. Enzyme production was reached maximum by organic nitrogen source, 0.3% yeast extract plus 0.5% bactopeptone. 3. It was appeared the presence of 0.1 M Mn2+ and Fe2+ ion was essential factors, on the other hand, 0.1 M Ag+ and Hg2+ ion almost block in yeast growth and enzyme production. 4. Invertase productivity was reached maximum within 3 days on stationary culture with medium-composed of sucrose 3%, bactopeptone 0.5%, yeast extract 0.3%, KEHPO. 0,1%, MgSO4.7H2O 0.05%.

  • 김치로부터 분리 동정된 Lactobacillus plantarum KLAB21 균주에 있어서 항돌연변이 활성물질 생산을 위한 최적 조건을 조사하였다. 탄소원으로서 glucose 첨가시 가장 높은 항돌연변이 활성을 나타내었으며, 질소원으로서는 yeast extract와 bactopeptone 첨가시 활성이 우수하였다. 탄소원인 glucose의 농도는 2%에서, 질소원인 yeast extract와 bactopeptone의 농도는 1%에서 가장 우수한 항돌연변이 활성을 나타내었다. 항돌연변이 활성의 최적 배양 조건은 초기 pH 배양온도, 배양속도 각각 7.0, 37$^{\circ}C$, 150rpm이었다. 상기의 최적 조건에서 36시간 배양시 가장 높은 항돌연변이 활성을 나타내었는데 S. typhimurium TA100과 S. typhimurium TA98을 이용한 조사결과 항돌연변이 활성이 각각 73.95%, 59.47%이었다.

Isolation and Characterization of Aniline-Degrading Bacteria

  • Six isolated strains degrading aniline were selected, identified and designated as pseudomonas putida K6, Pseudomonas acidovorans K82, Achromobacter gr. D. V. K24, Achromobacter xylosocidans K4, Moraxella sp. K21 and Moraxella sp. K22. All of them degraded 1000 ppm aniline completely within 30 to 36 hours. Most of these strains are resistant to antibiotics more than one, but Moraxella sp. has not any antibiotic marker tested. Most strains except for P. acidovorans K82 were shown to have resistance to the heavy metal ions such as Ni, Cu, Li, Ba, Co, etc. but not to Hg to which only P. putida K6 was resistant. M. sp. K21 was capable of degrading aniline to a maximum concentration of 2500 ppm without any repression. The incubation of the cell in limited pH ranges (4-8) had no great effect on aniline degradation. The addition of bactopeptone to the minimal media promoted the speed of aniline degradation, but the addition of glucose rather repressed the rate of aniline degradation. Through enzyme assay, A. gr. D. V. K 24 was shown to degrade aniline through artho-pathway and formed .betha.-ketoadipate as intermediate metabolite.

Optimization of Culture Medium for Lactosucrose ($^4G-{\beta}$-D-Galactosylsucrose) Production by Sterigmatomyces elviae Mutant Using Statistical Analysis

  • In this study, the optimization of culture medium using a Sterigmatomyces elviae mutant was investigated using statistical analysis to increase the cell mass and lactosucrose ($^4G-{\beta}$-D-galactosylsucrose) production. In basal medium, the cell mass and lactosucrose production were 4.12 g/l and 140.91 g/l, respectively. However, because of the low cell mass and lactosucrose production, optimization of culture medium was carried out to increase the cell mass and lactosucrose production. Culture media were optimized by the S. elviae mutant using analysis of variance (ANOVA) and response surface methodology (RSM). Central composite designs using RSM were utilized in this investigation. Quadratic models were obtained for cell mass and lactosucrose production. In the case of cell mass, optimal components of the medium were as follows: sucrose 1.13%, yeast extract 0.99%, bactopeptone 2.96%, and ammonium sulfate 0.40%. The predicted maximum value of cell mass was about 5.20 g/l and its experimental value was 5.08 g/l. In the case of lactosucrose production, optimal components of the medium were as follows: sucrose 0.96%, yeast extract 1.2%, bactopeptone 3.0%, and ammonium sulfate 0.48%. Then, the predicted maximum value of lactosucrose production was about 194.12 g/l and the corresponding experimental value was about 183.78 g/l. Therefore, by culturing using predicted conditions, the real cell mass and lactosucrose production increased to 23.3% and 30.42%, respectively.

  • 미생물을 이용하여 pyridine계 보효소인 NAD를 생산하기 위하여, 12종의 효모로부터 NAD 함량이 높은 Saccharomyces sake KBA No. 6을 선별하였으며, NAD 생산에 미치는 여러 가지 효과를 검토하였다. NAD 생산에 있어서 탄소원으로는 4% glucose, 질소원으로는 2% bactopeptone이 가장 좋았으며 최적 PM와 온도는 각각 5.0과 $30^{\circ}$C$이었다. 한편 NAD 전구물질로서 nicotinamide와 adenine을 동시에 사용한 경우 NAD 생산이 가장 좋았으며 이때의 농도는 각각 4 mg/ml과 3 mg/ml이었다. 또한 NAD 생산을 증가시키기 위하여 배양시 2가 금속이온들을 사용하였으며 이들 중 $Zn^{2+}$가 매우 효과적이었다. 계면활성제의 영향은 음이온 계면활성제인 sodium dodecyl sulfate(SDS)를 사용하였을 때 효과적이었다. 상기의 최적 조건하에서, 144시간 배양 후에 최대 NAD 생산량은 배지 100 ml당 35 mg이었으며 전체량의 89%인 31mg의 NAD가 배지 중에 유리되었다.

  • A bacterium producing pullulanase was from soil, and was identified Bacillus cereus and named as Bacillus cereus JK36. The optimal culture conditions for the efficident production of pullulanase from B. cereus JK36 was obtained by cultivating with the medium composed of 1% pullulan, 1% teast extract, 1% bactopeptone, 0.1% NaH$_{2}$PO$_{4}$, 2H$_{2}$O, 0.02% MgSO$_{4}$\ulcorner7H$_{2}$O at 40$\circ$C, initial pH 6.5 for 70 hours. Using the culture supernatant as crude enzyme, the optimal pH and temperature of the pullulanase of this strain were 6.5 and 50$\circ$C. In effect of pH and temperature on the stability of the enzyme, the enzyme was stable in the range of pH6.0$\sim$9.5 and up to 40$\circ$C, respectively. The hydrolysis product on pullulan was mainly maltotriose.

검은별무늬병균 Cladosporium cucumerinum KACC 40576에 대한 길항균주 Bacillus subtilis KMU-13의 선발 및 항진균 활성 (Selection and Antifungal Activity of Antagonistic Bacterium Bacillus subtilis KMU-13 against Cucumber scab, Cladosporium cucumerinum KACC 40576)

  • 오이검은별무늬병균인 Cladosporium cucumerinum KACC 40576에 대하여 높은 항진균 활성을 나타내는 KMU-13은 Norway Lillehammer 근교 토양시료에서 분리하였으며, 형태학적, 배양 및 생화학적인 특성을 조사한 결과와 16S rDNA 염기서열을 분석한 결과, B. subtilis로 판단되었다. B. subtilis KMU-13의 항진균 물질의 생산을 위한 최적조건은 LB broth를 기본배지로 하여 탄소원으로 0.5% maltose와 질소원으로 0.5% bactopeptone을 첨가하였을 때 가장 높은 항진균 활성을 나타내었으며, 배양조건은 $30^{\circ}C$, 180rpm, 48시간이였고, 배양초기 pH는 6.0으로 조사되었다. TLC에서 Rf값이 0.64로 확인된 밴드에서만 항진균 활성을 가지는 것으로 조사되었으며 잿빛곰팡이병, 고추 탄저병, 참외 탄저병, 수박 등의 박과작물의 덩굴마름병, 토마토 시들음병, 글라디올러스 마른썩음병, 토마토 질병, 수박 덩굴쪼김병, 수박 질병, 사과나무 역병, 벼 잎집무늬마름병, 참외 줄기썩음병, 그리고 고추 균핵병에 대하여 양호한 항진균 활성을 나타내는 것으로 조사되었다.

  • 김치에서 분리한 Lactobacillus plantarum KLAB21 균주에 있어서 MNNG와 NQO에 대한 항돌연변이원성 물질 생산을 위한 최적 조건을 조사하였다. 탄소원으로 glucose를 첨가시 가장 높은 항돌연변이 효과를 나타내었으며, 질소원으로서는 yeast extract와 bactopeptone첨가시 항돌연변이 효과가 우수하였다. 탄소원으로 glucose의 농도를 2% 첨가시 MNNG와 NQO에 대한 항돌연변이 효과가 가장 우수하였으며, 질소원으로서 yeast extract는 1%와 bactopeptone의 농도는 MNNG인 경우 1%, NQO인 경우 1.5%에서 가장 우수한 항돌연변이 효과를 나타내었다. 항돌연변이원성 물질 생산을 위한 최적 배양 조건은 초기 pH, 배양 온도, 배양 속도가 각각 7.0, $37^{\circ}C$ 및 150 rpm이었다. 상기의 최적 조건에서 36시간 배양시 가장 높은 항돌연변이 효과를 나타내었는데 S. typhimurium TA100과 TA98을 이용한 경우 항돌연변이 효과가 각각 98.4%와 57.3%이었다.

Streptomyces thermocyaneoviolaceus의 Xylanase 생산조건 및 Xylooligo당의 생산 (Production Conditions of Xylanase from Streptomyces thermocyaneoviolaceus and Production of Xylooligosaccharides)

  • 농산폐자원으로부터 기능성물질인 xylooligo당을 생산하기 위해서 내열성 균주인 S. thermocyaneoviolaceus가 생산하는 xylanase의 생산최적조건을 검토한 결과 0.8% 밀기울, 0.06% yeast extract, 0.06% bactopeptone, 0.05% $MgSO_4{\cdot}7H_2O$, 0.005% $FeSO_4{\cdot}7H_2O$, 0.05% $KH_2PO_4$ 및 0.2% $K_2HPO_4$를 함유한 배지(pH7.0)에서 $50^{\circ}C$, 24시간 배양시 최고효소활성(2.47 unit/ml)의 배양상징액을 얻을 수 있었다. 효소의 최적반응조건은 pH5.5, $65^{\circ}C$였다. 또한 pH안정성을 조사한 결과 pH 4.5~9.5사이에서 $4^{\circ}C$에서 12시간후에도 80% 이상의 효소활성을 유지하였고, 열 안정성은 $60^{\circ}C$에서 1시간 처리후 94%이상의 효소활성을 유지하는 내열성이 있는 효소였다. 생산된 xylanase birchwood xylan 반응생성물을 TLC 및 HPLC로 확인해 본 결과, pH가 낮을수록(pH 5.0~6.0) xylobiose와 xylotriose및 소량의 xylose의 양이 증가하였고, pH가 높을수록(pH8.0~9.0) $X_4$이상의 각종 xylooligo당의 양이 상대적으로 증가하였다. 또한 24 시간후에는 xylan의 상대량이 25% 이하로 감소하면서 주분해산물로 xylobiose가 생산되었으며 xylotriose와 xylose 및 $X_4$ 이상의 각종 xylooligo당이 생산되었다.
