Streptomyces acidiscabies에 의한 감자 더뎅이병 (Potato Scab Caused by Streptomyces acidiscabies)

  • Bacterial species isolated from common scab lesion on potato (Solanum tuberosum L. cv. Dejima) was identified as Streptomyces acidiscabies. This organism had flexuous spore chains and white spore mass color, produced melanin pigment on tyrosine agar medium but did not produce on peptone agar medium. S. acidiscabies grew on agar medium at pH 4.0, used L-arabinose, D-fructose, D-glucose, D-mannitol, rhamnose, sucrose, D-xylose and meso-inositol except reffinose as carbon sources. It was also susceptible to thallium acetate (10 $\mu\textrm{g}$/ml, 100$\mu\textrm{g}$/ml), phenol (0.1%, wt/vol), streptomycin (20 $\mu\textrm{g}$/ml), and was resistant to 7% NaCl, crystal violet (0.5 $\mu\textrm{g}$/ml), penicillin (10 IU/ml) and oleandomycin (25 $\mu\textrm{g}$/ml, 100 $\mu\textrm{g}$/ml).

  • The udders of 28 culled dairy cows were removed at slaughter and bacteria isolated. Isolations of major known mammary pathogens comparised Staphylococcus sp. (40.7%), non-identified Gram's negative rods (33.2%) and E. coli (7.4%). In the mixed infection, Staphylococcus sp. was most frequently involved. Drug sensitivity tests were performed on several bacterial species by disc method. These tests indicated that the microorganisms isolated from clinical mastitis of dairy cows were more resistant to antimicrobial agents than the organisrms isolated from milk samples.

  • The treatment of alkylated nitro derivatives of indazole with 2-(4-chlorophenyl)acetonitrile under basic conditions gave the new 8-chloro-3-alkyl-3H-pyrazolo[4,3-a]acridine-11-carbonitriles via the nucleophilic substitution of hydrogen which proceeds at room temperature with concomitant cyclisation in fairly good yields. The structures of all newly synthesized compounds were confirmed by IR, $^1H$ NMR, $^{13}C$ NMR and mass spectral data. Fluorescence experimental results of all newly synthesized compounds revealed remarkable photoluminescence properties and strong green fluorescence properties. Also, the new compounds exhibited potent antibacterial activity and their antibacterial activity (MIC) against Gram positive (Staphylococcuse aureus methicillin resistant S. aureus and Bacillus subtilis) and negative bacterial (Pseudomonas aeruginosa and Escherichia coli) species were determined.

Improved Transformation of the Filamentous Fungus Aspergillus niger Using Agrobacterium tumefaciens

  • Since it is known that Agrobacterium tumefaciens, which has long been used to transform plants, can transfer the T-DNA to yeast Saccharomyces cerevisiae during tumourigenesis, a variety of fungi were subjected to transformation to improve their transformation frequency. In this study, I report the A. tumefaciens-mediated transformation of filamentous fungus Aspergillus niger. Transfer of the binary vector pBIN9-Hg, containing the bacterial hygromycin B phosphotransferase gene under the control of the Aspergillus nidulans trpC promoter and terminator as a selectable marker, led to the selection of $50{\sim}100$ hygromycin B-resistant transformants per $1{\times}10^7$ conidia of A. niger. This efficiency is improved $10{\sim}20$ fold more than reported elsewhere. In order to avoid the difficulties in selection transformant from the over-growing non-transformant, I used top agar containing 900 ${\mu}g/ml$ of hygromycin. Genomic PCR and Southern analysis showed that all transformants contained single T-DNA insert per fungal genome. This technique offers an easier and more efficient method than that of using protoplast.

Phenol 분해 균주의 분리 및 특성 (Isolation and Characterization of a Phenol-Degrading Bacteria)

  • Twelve bacterial strains capable of growing on phenol minimal medium were isolated from iron foundry activated sludge by enrichment culture, and amount them, one isolate which was the best in cell growth and phenol degradation was selected and identified as Acinetobacter junii POH. The optimal temperature, initial pH and phenol concentration in the above medium were 3$0^{\circ}C$, 7.5 and 1000 ppm, respectively. Cell growth of Acinetobacter junii POH dramatically increased 20 hrs cultivation-time and reached a almost stationary phsae 40 hrs cultivation-time then phenol was degraded about 98%. Cell growth was inhibited y phenol at concentrations over 1500 ppm. The isolate was resistant to several antibiotics as well as various heavy metal ions. The growth-limiting log P value of Acinetobacter junii POH on organic solvents was 2.9 in the LB medium. Therefore, it is suggested that Acinetobacter junii POH could be effectively used for the biological treatment of wastewater containing the presence of heavy metal ions and organic solvents.

Antibiotics and Probiotics Prophylaxis for Recurrent Urinary Tract Infection in Children

  • Since many years, continuous low dose antibiotic prophylaxis (CAP) has been used for children at a risk for recurrent urinary tract infection (UTI), especially those with vesicoureteral reflux (VUR). The incidence of recurrent UTI has been shown to be higher in children with VUR with bladder and bowel dysfunction (BBD) than in those with VUR without BBD. Therefore, CAP has been recommended for children with BBD and VUR because of the increased risk of UTI. However, the use of CAP has become highly controversial because of bacterial resistance developed due to antibiotic over-usage. The preventive effects of probiotics have been proved in various adult urogenital infections, and the antimicrobial activities of lactobacilli against uropathogens have been demonstrated in previous in vitro studies. However, a critical assessment of their efficacy in children with UTI is lacking. The importance of the use of urogenital probiotics is that it is a natural approach that replenishes the depleted normal flora to create a better environment to fight off uropathogens. Probiotics have a great potential, particularly today with the increasing threat of antibiotic-resistant microorganisms.

Comparative Proteomic Analysis for a Putative Pyridoxal Phosphate-Dependent Aminotransferase Required for Virulence in Acidovorax citrulli

  • Acidovorax citrulli (Ac) is the causative agent of bacterial fruit blotch disease in watermelon. Since resistant cultivars have not yet been developed, the virulence factors/mechanisms of Ac need to be characterized. This study reports the functions of a putative pyridoxal phosphate-dependent aminotransferase (PpdaAc) that transfers amino groups to its substrates and uses pyridoxal phosphate as a coenzyme. It was observed that a ppdaAc knockout mutant had a significantly reduced virulence in watermelon when introduced via germinated-seed inoculation as well as leaf infiltration. Comparative proteomic analysis predicted the cellular mechanisms related to PpdaAc. Apart from causing virulence, the PpdaAc may have significant roles in energy production, cell membrane, motility, chemotaxis, post-translational modifications, and iron-related mechanisms. Therefore, it is postulated that PpdaAc may possess pleiotropic effects. These results provide new insights into the functions of a previously unidentified PpdaAc in Ac.

Debridement, antibiotics, and implant retention in infected shoulder arthroplasty caused by Serratia marcescens: a case report

  • Periprosthetic joint infection (PJI) is one of the most devastating complications that can occur after shoulder arthroplasty. Although staged revision arthroplasty is the standard treatment in many cases, surgical intervention with debridement, antibiotics, and implant retention (DAIR) can be an effective option for acute PJI. We report a complex case of infected reverse shoulder arthroplasty (RSA) in a 73-year-old male. The patient had been previously treated for infected nonunion of a proximal humerus fracture caused by methicillin-resistant Staphylococcus epidermidis. He presented with a sinus tract 16 days after the implantation of RSA and was diagnosed with PJI caused by Serratia marcescens. The patient was successfully treated with DAIR and was free of infection at the last follow-up visit at 4 years postoperatively.

Service life of concrete culverts repaired with biological sulfate-resisting mortars

  • The purpose of this study is to examine the effectiveness of biological repairing mortars on restoring the structural performance of a sewage culvert deteriorated by sulfate attack. The biological mortars were developed for protecting concrete structures exposed to sulfate attack based on the block membrane action of the bacterial glycocalyx. The diffusion coefficient of sulfate ions in the biological mortars was determined from the natural diffusion cell tests. The effect of sulfate-attack-induced concrete deterioration on the structural performance of culverts was examined by using the moment-curvature relationship predicted based on the nonlinear section lamina approach considering the sulfuric-acid-induced degradation of the structure. Typical analytical assessments showed that biological mortars were quite effective in increasing the sulfate-resistant service life of sewage culverts.
