  • This experiment was conducted to examine estimates of heterosis, degree of dominance, heritability and phenotypic and genotypic correlation for several mutant characters in flue-cured tobacco variety. Significant heterosis was noted, recording 2.73 and 6.16% for leaf width, -6.86 and -4.72 for leaf shape index, 19.8 and 23.4% for bacterial wilt disease index in Fl an4 F2 generation, respectively, and -1.44 for days to flower in F2 generation. And also leaf width was appeared partial dominance in Fl, and leaf shape index in Fl and F2, Leaf width in F2 was appeared overdominance. Estimated heritabilities in the broad sense ranged from 0.77 to 0.87 for plant and stalk height and days to flower, while those of leaf number, leaf length and width, leaf shape index and bacterial wilt disease index ranged from 0.50 to 0.65. And the heritabilities in the narrow sense were appeared 0.64 and 0.72 for stalk and plant height, respectively, and the others were ranged from 0.32 to 0.47 Positive genotpic correlations appeared among plant height, stalk height, leaf length and leaf width, and between leaf shape index ands day to flower. And negative genotypic correlations appeared between leaf shape index and plant height and stalk height and leaf width, and between bacterial wilt disease index and leaf length.

황색종 연초 돌연변이 계통 KF8832-85의 특성 (Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Isolation and Characterization of Bacteriophages Infecting Ralstonia solanacearum from Potato Fields

  • Bacterial wilt caused by Ralstonia solanacearum is one of the most devastating diseases in major Solanaceae crops. The pathogen is easily disseminated and survives for many years in plant farming system. Although chemicals are applied to control the disease, they are of limited efficacy and cause several problems. Therefore, the use of phage therapy has been suggested to control the disease as a biological agent. In this study, we discovered bacteriophages lysing diverse Ralstonia isolates from plant and soil samples obtained from the potato cultivated field in Jeju. Three times repeated pickings of plaques resulted in obtaining 173 single phages showing diverse spectrum of host-specificity. With the results, 12 core phages were selected and dendrogram was generated. Genetic diversity of the selected phages was also confirmed by AFLP (Amplified Fragment of Length Polymorphism) fingerprinting. The stability of the phages was investigated in various temperatures and various conditions of pH in vitro. The phages were stable at $16^{\circ}C-44^{\circ}C$ and pH 6-10. Morphological characterization of the phages revealed they were all classified into the Podoviridae, but had diverse head sizes. The results of this research will contribute to control the disease and further researches regarding genetic and molecular aspects will facilitate understanding phage and bacteria interaction.

Biological Potential of Bioorganic Fertilizer Fortified with Bacterial Antagonist for the Control of Tomato Bacterial Wilt and the Promotion of Crop Yields

  • The application of Bacillus sp. in the biological control of plant soilborne diseases has been shown to be an environmentally friendly alternative to the use of chemical fungicides. In this study, the effects of bioorganic fertilizer (BOF) fortified with Bacillus amyloliquefaciens SQY 162 on the suppression of tomato bacterial wilt were investigated in pot experiments. The disease incidence of tomato wilt after the application of BOF was 65.18% and 41.62% lower at 10 and 20 days after transplantation, respectively, than in the control condition. BOF also promoted the plant growth. The SQY 162 populations efficiently colonized the tomato rhizosphere, which directly suppressed the number of Ralstonia solanacearum in the tomato rhizosphere soil. In the presence of BOF, the activities of defense-related enzymes in tomato were lower than in the presence of the control treatment, but the expression levels of the defense-related genes of the plants in the salicylic acid and jasmonic acid pathways were enhanced. It was also found that strain SQY 162 could secrete antibiotic surfactin, but not volatile organic compounds, to suppress Ralstonia. The strain could also produce plant growth promotion compounds such as siderophores and indole-3-acetic acid. Thus, owing to its innate multiple-functional traits and its broad biocontrol activities, we found that this antagonistic strain isolated from the tobacco rhizosphere could establish itself successfully in the tomato rhizosphere to control soilborne diseases.

Field Performance of Resistant Potato Genotypes Transformed with the EFR Receptor from Arabidopsis thaliana in the Absence of Bacterial Wilt (Ralstonia solanacearum)

  • Bacterial wilt caused by the pathogen Ralstonia solanacearum is a devastating disease of potato crops. Harmonizing immunity to pathogens and crop yield is a balance between productive, economic, and environmental interests. In this work, the agronomic performance of two events of potato cultivar INIA Iporá expressing the Arabidopsis thaliana EFR gene (Iporá EFR 3 and Iporá EFR 12) previously selected for their high resistance to bacterial wilt was evaluated under pathogen-free conditions. During two cultivation cycles, the evaluated phenotypic characteristics were emergence, beginning of flowering, vigor, growth, leaf morphology, yield, number and size of tubers, analyzed under biosecurity standards. The phenotypic characteristics evaluated did not show differences, except in the morphology of the leaf with a more globose appearance and a shortening of the rachis in the transformation events with respect to untransformed Iporá. The Iporá EFR 3 genotype showed a ~40% yield decrease in reference to untransformed Iporá in the two trials, while Iporá EFR 12 did not differ statistically from untransformed Iporá. Iporá EFR 12 shows performance stability in the absence of the pathogen, compared to the untransformed control, positioning it as an interesting candidate for regions where the presence of the pathogen is endemic and bacterial wilt has a high economic impact.

Suppression of Fusarium Wilt Caused by Fusarium oxysporum f. sp. lactucae and Growth Promotion on Lettuce Using Bacterial Isolates

  • This study was carried out to explore a non-chemical strategy for enhancing productivity by employing some antagonistic rhizobacteria. One hundred eighteen bacterial isolates were obtained from the rhizospheric zone of various crop fields of Gangwon-do, Korea, and screened for antifungal activity against Fusarium wilt (Fusarium oxysporum f. sp. lactucae) in lettuce crop under in vitro and in vivo conditions. In broth-based dual culture assay, fourteen bacterial isolates showed significant inhibition of mycelial growth of F. oxysporium f. sp. lactucae. All of the antagonistic isolates were further characterized for the antagonistic traits under in vitro conditions. The isolates were identified on the basis of biochemical characteristics and confirmed at their species level by 16S rRNA gene sequencing analysis. Arthrobacter sulfonivorans, Bacillus siamensis, Bacillus amyloliquefaciens, Pseudomonas proteolytica, four Paenibacillus peoriae strains, and Bacillus subtilis were identified from the biochemical characterization and 16S rRNA gene sequencing analysis. The isolates EN21 and EN23 showed significant decrease in disease severity on lettuce compared to infected control and other bacterial treatments under greenhouse conditions. Two bacterial isolates, EN4 and EN21, were evaluated to assess their disease reduction and growth promotion in lettuce in field conditions. The consortium of EN4 and EN21 showed significant enhancement of growth on lettuce by suppressing disease caused by F. oxysporum f. sp. lactucae respectively. This study clearly indicates that the promising isolates, EN4 (P. proteolytica) and EN21 (Bacillus siamensis), can be commercialized and used as biofertilizer and/or biopesticide for sustainable crop production.

가지대목 EG203을 이용한 토마토 풋마름병 경감효과 (Reduction of Bacterial Wilt Diseases with Eggplant Rootstock EG203-Grafted Tomatoes in the Field Trials)

  • Ralstonia solanacearum에 의한 시들음 피해는 고온기 재배가 증가함에 따라 피해가 증가하고 있다. 이에 재배적 방법으로 토마토대목을 이용하고 있으나 병이 다 발생하는 지역에서는 효과가 기대에 미치지 못한다. 이에 따라서 AVRDC에서 수집한 풋마름병 저항성 가지 대목인 EG203을 재료로 하여 풋마름병 다 발생포장인 부여 토마토시험장 비닐하우스에서 2003-2005년, 구미시 비닐하우스에서 2009-2011년까지 두 곳에서 각각 3년간 실시하였다. EG203(가지)는 광발아 종자로 파종 시 육묘 트레이에 직접 파종하여야 하며 EG-203(가지)의 파종 적기는 토마토 접수보다 3주전에 파종해야 접수와 비슷한 경경(2.5-3.0 mm)에 도달하여 접목하기에 알맞았다. 접목방법은 맞접과 삽접이 93-96%의 활착률을 보여 대목으로 사용가능한 것으로 확인되었다. 접목 후 포장에 정식한 후의 풋마름병 발생을 2003년부터 2005년까지 3년 동안의 평균은 EG203(가지) 접목 시 4.3%로 실생의 58.0%보다 매우 낮았으며 대목용 품종의 25.0-36.7%보다도 낮게 나타났다. 구미에서 2009년부터 2011년까지 3년간 대목 효과에 대하여 시험한 결과는 2009년 5농가에서 EG203과 실생묘를 비교한 결과 EG203에서는 풋마름병 발생률이 2-5%였으나 실생은 20-80%를 보였다. 2010년에는 토마토대목('B-blocking', '청강')과 가지대목(EG203)을 갖고 비교 한 결과 풋마름병 발생률의 차이는 보이지 않았으나 가지대목(EG203)에서 복화방 출현이 늦어 수확이 감소되는 경향을 보였다. 2011년 토마토대목('청강')과 가지대목(EG203)으로 3농가에서 시험한 결과 토마토대목('청강')에서 풋마름병 발생률은 60-85%, 가지대목(EG203)은 0-1%로 가지대목에서 병발생률이 매우 낮았다. 가지대목(EG203)을 이용할 경우 토마토대목과 비교 복화방이 늦게 발생하여 토마토 생산량이 줄었다. 따라서 병 발생이 낮은 곳에서는 토마토대목을 활용하여 접목을 하고 병이 다 발생하는 지역에서는 가지대목(EG203)을 활용하는 것이 재배에 유리하다고 판단되어진다.

Effect of Iron Availability on Induction of Systemic Resistance to Fusarium Wilt of Chickpea by Pseudomonas spp.

  • Selected isolates of Pseudomonas fluorescens (Pf4-92 and PfRsC5) and P. aeruginosa (PaRsG18 and PaRsG27) were examined for growth promotion and induced systemic resistance against Fusarium wilt of chickpea. Significant increase in plant height was observed in Pseudomonas treated plants. However, plant growth was inhibited when isolates of Pseudomonas were used in combination with Fusarium oxysporum f. sp. ciceri (FocRs1). It was also observed that the Pseudomonas spp. was colonized in root of chickpea and significantly suppressed the disease in greenhouse condition. Rock wool bioassay technique was used to study the effect of iron availability on the induction of systemic resistance to Fusarium wilt of chickpea mediated by the Pseudomonas spp. All the isolates of Pseudomonas spp. showed greater disease control in the induced systemic resistance (ISR) bioassay when iron availability in the nutrient solution was low. High performance liquid chromatography (HPLC) analysis indicated that an the bacterial isolates produced more salicylic acid (SA) at low iron ($10\;{\mu}M$ EDDHA) than high iron availability ($10\;{\mu}Fe^{3+}$ EDDHA). Except PaRsG27, all the three isolates produced more pseudobactin at low iron than high iron availability.

잎담배 세균성마름병(立枯病)의 방제체계에 관한 연구 제1보 저항성품종, 경종적 및 토양훈증제처리가 잎담배 세균성 마름병 방제효과에 미치는 영향 (Study on Control System of Bacterial Wilt in Flue-cured Tobacco 1. Control Effect of Bacterial Wilt in Tobacco Line NC82 by the Cultural Practice and Fumigation)

  • [ $1982\~1983$ ]년 사이에 한국인삼연초연구소 음성시험장의 세균성마름병 상습발병포지에서 이 병의 방제체계 확립을 위해 시험한 결과 1. 세균성마름병은 세균증식에 적합한 온도와 토양습도가 주어진 7월하순${\cdot}$8월하순에 가장 큰 피해를 나타냈으며 토양훈증제처리구 및 저항성품종재배구에서는 병발생이 적고 느리게 나타낮다. 2. 세균성마름병의 방제효과는 감수성품종인 NC23 26에 비하여 저항성품종인 NC82의 무처리재배의 경우 $44.7\%\~55.8\%$의 방제효과를 보았으며 토양훈증제처리에 저항성품종인 NC82를 재배 할 경우 $95\~99\%$의 방제효과를 보았다. 경종적처리에서는 무처리재배에 비하여 $0.8\~20\%$로 낮게 나타났으며 저항성품종이나 토양훈증제를 함께 사용 할 경우 그 상승효과는 거의 인정되지 않았다. 3. 세균성마름병의 종합적인 방제체계확립은 경종적처리(춘추경, 잔간근제거, 조기이식, 피복물제거)에 토양훈증(Cylon)를 10a당 40l 토양관주한 후 저항성 품종(NC82)을 재배할 때 $95\%$ 이상의 방제효과를 거둘 수 있었다.

  • This experiment were conducted to investigate heterosis and combining ability for several mutant characters by analyzing dialled crosses of flue-cured tobacco. In a dialled cross of 3 flue-cured varieties and the mutant line 83H -5, the heterosis was somewhat higher in Fl than in F2. For growth character, the heterosis was 0.28-6.03% in plant height, leaf number, leaf shape index and yield, and was 43.2% for bacterial wilt disease index. The mutant line 83H-5 showed significantly negative GCA effect for plant height, leaf width and bacterial wilt disease index in Fl and F2, leaf length in F2, and positive GCA effect for total alkaloids, total nitrogen in Fl and days to flower in F2, respectively. Specific combining ability(SCA) in 83H-5 x Hicks was significant in negative effect for leaf length(F2), number of leaves(F2), leaf shape(F1, F2), bacterial wilt(F2) and alkaloids(F1), and in 83H-5 x NC 2326 in positive effect for leaf length(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in positive effect for plant height(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in Positive effect for Plant height(F1, F2), leaf length(F2) and yield(F1, F2).
