Bacterial Biomass and Secondary Productivity in Naktong River Estuary (낙동강 하구생태계의 세균 생물량과 이차생산성)

  • To investigate the bacterial potentials for utilizing dissolved organic matter in highly eutrophic estuary, the annual fluctuations of microbiological and physicochemical environmental parameters were analyzed in Naktong River Estuary. Total bacterial number ranged from 0.33 to $2.09{\times}10^7$ cells/ml, and correlated with the heterotrophic bacterial numbers in more eutrophic sites, especially. Bacterial biovolume and biomass varied between 0.064 and 0.156 2.09${\mu}m^3$/cell, 0.163 and 1.036 ${\mu}g$-C/ml, respectively. Bacterial secondary productivity ranged from 0.24 to 60.86 ${\mu}g$-C/l/h, and showed high correlations with the environmental parameters of pollution indicator. The seasonal variation pattern of bacterial productivity in freshwater sites was high in winter and low in summer, which was interpreted as the results of pollution loads varied with the amount of rainfall. In seawater site, the pattern was different from those of freshwater sites; high in summer and low in winter. In this site, the values of bacterial productivity showed positive correaltions with chlorophyll a, heterotrophic bacterial number, and temperature (r>0.5, p<0.05). These results suggested that the main source of organic matter which influences the bacterial productivity may be allochthonous materials in the upper freshwater zone of Naktong River Barrage, and autochthonous algal excretory products in the lower seawater zone of Naktong River Barrage.

The Relationship between Phytoplankton Productivity and Water Quality Changes in Downstream of Nakdong River (낙동강 하류에서 식물플랑크톤 생산력과 수질 변화와의 관계)

  • The relationship between primary productivity and changes in water quality was investigated at Mulgum station, a site downstream of the Nakdong River, Korea. Phytoplankton production was characterized by blooms of Microcystis aeruginosa during the summer and Stephanodiscus hantzschii during the winter. Primary production and secondary production by bacterioplankton ranged from 1.5~53.5 mg-C/ι day and 0.1~0.3 mg-C/ι day, respectively. Distribution of total organic carbon appeared to be highly correlated with phytoplankton biomass, especially during blooms of M. aeruginosa, when particulate organic carbon was 81% of total organic carbon and the main source of organic materials supplied into the water. The correlation coefficient between chlorophyll-a and BOD was 0.86. Thus it was concluded that autochthonous phytoplankton mostly affected the BOD level. Total bacterial numbers were also highly correlated with chlorophyll-a ($r^2$= 0.84) and the bacterial community appears to be regulated by phytoplankton biomass in this area.

Heterotrophic Bacterial Secondary Productivity and Effect of Environmental Parameters is Naktong Embayment Korea (낙동강하구 연안수역의 세균 생산성과 환경요인의 영향)

  • The ecology of estuarine bacteria in terms of bactenal production and biomass was investigated in Naktong embayment. Intrusion of eutrophic freshwater was one of the major factors affecting on the ecosystem of Naktong embayment. Total bacterial number varied from $2.2{\times}10^5 cells/ml to 9.8{\times}10^5 $ cellslml, and the variation ranges of the bacterial biovolume and biomass were 0.023-0.201TEX>$\mu$$m^3$/cell and 0.010-0.140 TEX>$\mu$g-Clml, respectively, and there was a reciprocal relationship between bacterial number and biomass. Pool size of thymidine varied from 12.93 nM to 44.56 nM. The pool during summer was supposed to be composed of easily utilizable form than the typical one of winter, which suggests thal bacterial productivity measured in summer may be underestimated. Bactenal production varied from 0.12 TEX>$\mu$g-Cllh to 22.38 TEX>$\mu$g-Clllh, and the values were low in winter and increased from spring and reached the highest in summer. The variations of bacterial production showed high correlations with temperature, chlorophyll a, and bacterial biomass. These results suggested that the main source of organic matters which influence the bacterial production in Naktong embayment may be the photosynthetic excretory products of phytoplanktons.

  • This study was performed to improve the yield of bacterial cellulose(BC) by UV and NTG mutagenesis of strain KJ-1 which produced largely BC. some mutants showed high BC productivity with twice elevation compared to that the wild strain KJ-1. A difference was found in production and bioconversion phase of synthesized organic acid, such as gluconic acid, 2-keto gluconic acid, and 5-keto gluconic acid between mutants and strain KJ-1 in the static culture. The organic acid produced in secondary metabolism phase, were more rapidly consumed in the culture with the mutants than that the parent strain after glucose in the broth was conversed to a limiting substrate. Therefore, we suggested the reason for increasing of BC production that the mutant strain consumed more efficiently synthesized acids as substrates than that of the parent strain.

Mass Cultivation and Secondary Metabolite Analysis of Rhodobacter capsulatus PS-2 (광합성세균 Rhodobater capsulatus PS-2의 대량배양 최적화 및 대사산물 분석)

  • Plant growth promoting (PGP) hormones, which are produced in a small quantity by bacteria, affect in plant growth and development. PGPs play an important role on the crop productivity in agricultural field. In this study, a photosynthetic bacterial strain producing the PGP was isolated from paddy soil. Bacterial isolate was gram negative, rod-shaped and motility positive. From the 16s rRNA gene sequence analysis, the isolate was identified as Rhodobacter capsulatus PS-2. The mass cultivation of R. capsulatus PS-2 was optimized by considering of the carbon, nitrogen and inorganic salt sources. Optimal medium composition was determined as Na-succinate 4.5 g, yeast extract 5 g, $K_2HPO_4$ 1 g, $MgSO_4$ 5 g, per liter. From the result of 500 L fermentation for 2 days using the optimal medium, the viable cells were $8.7{\times}10^9cfu/mL$. R. capsulatus PS-2 strain produced the carotenoid and indole-3-acetic acid (IAA). The carotenoid extraction and quantitative analysis were performed by HCl-assisting method. Total carotenoid contents from R. capsulatus PS-2 culture broth were measured as $7.02{\pm}0.04$ and $6.93{\pm}0.05mg/L$ under photoheterotrophic and chemoheterotrophic conditions, respectively. To measure the productivity of IAA, colorimetric method was employed using Salkowski reagent at optical density 535 nm. The results showed that the highest content of IAA was $197.44{\pm}5.92mg/L$ in the optimal medium supplemented with 0.3% tryptophan.
