A blue pigment-producing bacterium, Vogesella indigofera, was isolated and quantified for the relationship between its synthesis of a blue pigment and exposure concentrations of $Cr^{6+}$. The concentration of $Cr^{6+}$ and the percentage of blue colonies on agar plates was negatively correlated ($r^{2}$ =-0.8683). Critical concentrations inhibiting bacterial pigment production were found to be between 100-150 $\mu\textrm{g}$$Cr^{6+}$/ml on agar plates and 200-300 $\mu\textrm{g}$$Cr^{6+}$/ml in liquid culture. As the blue color is characteristic and easily observable, the bacterium Vogesella indigofera may have potential applications in the detection and monitoring of environmental pollution.
This present study has been carried out to examine the correlation between the distribution of the coliform bacterial plasmids and the viability test against heavy metals in the upper stream of Han river(Gapyung; clean water) and the its basin (Anyang Cheon; polluted water). And the distribution of plasmids were examined to be used as trophic indicator for analysis between the clean and polluted waters. 1. A total of 110 isolates were analyzed for the presence of plasmids by means of the boiling method and agarose gel techniques. Plasmids were significantly more frequent in the strains which had been isolated from the clean water (14.3%). Also, there were much higher multiplicity of plasmids at the polluted water(41.0%), compared with the clean water (33.0%). By the comparision between molecular weight of bacterial plasmids in the clean water and those of polluted water, there were no significant differences of the clean water from the polluted water, to such extent as 30.0%, 28.6%, respectively, in frequency for occurrence of high molecular weight plasmids iver 35.8% Mdal. 2. Each isolate was carried out the resistance test for mercury(10-5M), nickel(10-3M) and arsenic (0.1%). At the polluted site, the survival ability of the plasmid-carrying straings(Hg, 31.0%l Ni, 5.7%; As, 65.7%) was higher than that of the non plasmid-carrying strains(Hg, 12.1%; Ni, 3.0%; As, 54.6%). This trend was more remarkable in the clean site (plasmid-carrying strains: 16.7%,-,-). As a result it is suggested that plasmids could be used as an indicator of a certain types of water pollution. In addition, heavy metals might have inflyenced, some extent, to the distribution of plasmids in the environment which has been surveyed in the present study.
부영양화된 낙동강 하구역에서 세균에 의한 용존 유기물의 이용능을 조사하고 이에 미치는 환경요인의 영향을 파악하기 위하여 미생물학적 요인과 물리화학적 환경요인의 연중 변화를 분석하였다. 총세균수는 0.33 ~ $2.09{\times}10^7$ cells/ml의 변화폭을 나타냈으며, 오염부하량이 큰 정점에서 종속영양세균의 개체수와 비례하였다. 세균이 체적과 생물량은 각각 0.064~0.156 ${\mu}m^3$/cell, 0.163~1.036 ${\mu}g$-C/ml을 나타냈다. 세균의 이차생산성은 0.24~60.86 ${\mu}g$-C/l/h의 범위로 측정되었으며, 부영양화 지표로 사용되는 환경인자와 밀접한 관계를 보였다. 담수역에 속하는 정점에서의 세균생산성의 계절별 변화는 동계에 최고치를, 하계에 최저치를 나타내어 갈수기의 오염부하량 증가와 밀접한 상관성을 보였다. 해수역에 속하는 다대의 경우 동계에 낮은 값을, 하계에 높은 값을 나타내고, 환경요인 중 엽록소 a, 수은 등과 밀접한 상관성을 보였다 (r>0.5, p<0.05). 이로 미루어 낙동강 하구의 하구언 상류역에서는 주위에서 유입되는 유기물이, 하구언 하류역에서는 식물성 플랑크톤의 광합성 산무링 세균의 이차생산성에 미치는 주 영양물질임을 암시하고 있다.
In 2005, the 5.84-Km length of Cheonggyecheon stream, previously covered with concrete road, was uncovered in the middle of Seoul, Korea. We investigated microbial water quality in various sites in Cheonggyecheon stream. We took water samples on three different days. The sampling sites included inflow water from upper stream (Mojeongyo), midstream (Ogansugyo), and downstream (Muhakgyo). Fecal pollution indicator microorganisms were measured by both IDEXX $Colilert^{(R)}$ and $Enterolert^{(R)}$. Microbial community from these sampling sites was also characterized based on 16S rRNA gene sequences. The average concentrations of total coliform are 5 CFU/100 mL, 1474 CFU/100 mL, and 1776 CFU/100 mL at Mojeongyo, Ogansugyo, and Muhakgyo, respectively. The average concentrations of fecal coliform were 28 CFU/100 mL, 47 CFU/100 mL in Ogansugyo, and Muhakgyo, respectively. The concentrations of other fecal indicator microorganisms including E. coli and Enterococcus sp. increased in downstream. When we characterized the microbial community, unique microbial community were discovered at different sampling sites. This study suggests that Cheonggyechoen stream is likely affected by non-point fecal sources and has unique microbial environment as the river flows downstream.
세균군집의 분포에 따른 지하수의 세균학적인 안전성을 평가하기 위하여 음용을 목적으로 개발된 4곳의 관정에서 심층피압대수층의 지하수를 채수하여 물리화학적 환경요인, 세균의 분포와 활성, 지표세균의 분포 및 종속영양세균의 종조성을 조사하였다. 조사한 4개의 관정은 모두 빈영양생태계의 특징을 나타냈으나 관정 C-2는 무기이온의 농도, 유기탄소, 종속영양세균의 수 및 활성, 지표세균의 수가 다른 관정에 비해 매우 높았다. C-1을 제외한 모든 관정에서 분변성 오염이 존재하였으며 특히, 관정 C-2는 다른 관정들에 비해 그람양성 세균의 비율이 낮았으며, 총대장균군, 분변성대장균군, fecal streptococcus 가 각각 시료 250ml 에 최고 61, 33, 55 CFU로 존재하여 지표에서 분변성 오염물질이 지하수로 유입되고 있음이 확인되었다. 종속영양세균의 동정결과 55 CFU 로 존재하여 지표에서 분변성 오염물질이 지하수로 유입되고 있음이 확인되었다. 종속영양세균의 동정결과 지하수에 주로 존재하는 세균속은 Acinetobacter, Eikenella, Moraxella, Pseudomonas, Bacillus, Micrococcus 속이었다. 한편, 지하수에 일반적으로 존재하지 않는 Enterobacteriaceae 그룹과 Staphylococcus 속과 Enterococcus 속이 조사과정에서 검출됨에 따라 이들 세균이 지하수 분변성 오염 판정의 지표세균으로 사용할 수 있음이 확인되었다.

Background: To confirm whether shellfish are suitable for consumption, the quality of seawater and shellfish in shellfish-producing areas must be assessed regularly. This study was conducted to evaluate the bacterial quality on the Changseon area, containing a designated shellfish-producing area, in Korea during 2011-2013. Result: Even though many inland pollutants near the area were identified, they showed no significant impact on the designated area and the shellfish therein. The concentrations of fecal bacteria in all the seawater and mussel samples from the designated area during the harvesting season were within the standards of various countries. Pathogenic bacteria were not detected in any of the mussel samples. In our previous study, the hazardous metal levels in all the mussels from the same area were also within the limits of different countries. Conclusion: The mussel products in this area are suitable for consumption based on fecal pollution, pathogenic bacteria, and also heavy metals.
The impact of rainfall events on the sanitary indicator bacteria density of the shellfish-growing waters in Geoje Bay and Jaran Bay in Korea was investigated. The shellfish-growing area in Geoje Bay, which is a nearly closed basin, was not affected significantly, except near the stream mouth after 11.5 mm of rainfall in 1 day. However, most of the shellfish-growing water in the bay was polluted by fecal coliform bacteria after rain as heavy as 43.0 mm, and the levels of fecal indicator bacteria in some of the sea near the coast did not recover completely until 24 hours after the rainfall. By contrast, in Jaran Bay, which has no significant pollution source in the drainage area, although 9.3-490 MPN/100 mL of fecal coliform bacteria were detected near the stream mouth after rainfall of 33.5 and 81.0 mm, a very low level of the indicator bacteria was detected in the designated shellfish-growing area. During the investigation, the correlations between the sanitary indicator bacteria density and physical parameters, such as salinity and turbidity, were evaluated. Both the total coliform and fecal coliform densities were inversely correlated with salinity. Turbidity was positively correlated with the indicator bacteria density. The survey results suggest that for more efficient management of the shellfish-growing areas located in coastal areas, such as shellfish harvesting after rainfall, a detailed investigation of the effects of rainfall on the bacterial water quality in each growing area is needed.
Wildlife is a bio-indicator of environmental pollution by antimicrobial resistant bacteria or genes, however, there is no information on antimicrobial resistance in wildlife-origin bacteria. This study aimed to investigate the normal microbiota of staphylococci and their antimicrobial resistance in wildlife that did not take any antimicrobials. After sampling and bacterial isolation/identification, antimicrobial resistance profiles were examined by broth microdilution test, Kirby-Bauer disc diffusion test and mecA genetargeted PCR. Of 90 isolates from wildlife, 83 were coagulase-negative staphylococci while only 7 were coagulase-positive staphylococci. Methicillin-resistance was found in 63 (70%) isolates and 35 of 90 (38.9%) isolates were multidrug-resistant staphylococci. When considering that all of the animals did not take any medication or contacted any medical device before the sampling, the results indicate significantly high prevalence of antimicrobial resistance in wild environments. Further study would be necessary to investigate the transmission route of antimicrobial resistance.
캄보디아는 동남아시아 지역에서 대표적인 저개발 국가이다. 수자원의 관점에서 캄보디아는 풍부한 수량을 갖고 있지만, 농촌지역에서는 안전하지 않은 식수와 처리되지 않은 분뇨에 의하여 공중 보건의 문제가 이어지고 있다. 본 연구는 캄보디아 지역에서의 물 원조 프로그램을 위한 신전략수립과 준비를 위해 현지조사, 검토 및 농촌지역에서의 물 오염현황분석, 그리고 해당지역의 물 순환을 분석하였다. 조사 대상지역은 푸르사트주에 있는 쁘로얍과 랭뜰을 조사 대상지로 정하였다. 연구대상 농촌지역의 사람들은 식수전용 웅덩이, 빗물 항아리, 관리되지 않은 우물물 등 지표수에 의존하고 있었다. 분석은 분원성 미생물(TTC)과 일반항목으로 크게 두 가지로 하였으며, 대상지역에서 채수한 시료들을 분석/평가하였다. TTC는 수인성 배설물 오염의 세균 지표인데 26개의 시료중 단 한개만이 WHO가 규정한 TTC 허용한계인 불검출/100 mL의 기준을 충족시켰다.
부산시 영도구의 혁신지구의 인공해수천은 높아진 하상과 조류의 특성으로 인해 물이 순환되지 않고 더구나 주위의 오수가 유입되고 있어서 수질이 나빠지고 악취를 발생하고 있다. 이 문제를 해결하기 위한 방안으로 가장 오염되고 조류이동을 감안한 하천의 지점에 생물증강법을 적용하여 친환경적, 효율적으로 하천을 정화하고자 하였다. 현장에서 활성화된 복합미생물을 가장 오염된 지점(Site 2)에 7~10일 간격으로 투입하여, 수질의 pH, 온도, DO, ORP, SS, COD, T-N, 및 T-P를 측정하였고 또한 하상퇴적토의 COD 및 미생물군집을 분석하였다. 13개월 후 Site 2의 수질 SS, COD, T-N 및 COD (퇴적토)의 처리효율은 각각 51%, 58%, 27% 및 35%으로 나타났으나 T-P는 오히려 47% 증가를 보였다. 대부분의 측정지점에서 황산염환원세균(sulfate reducing bacteria)그룹 (Desulfobacteraceae_uc_s, Desulfobacterales_uc_s, Desulfuromonadaceae_uc_s, Desulfuromonas_g1_uc and Desulfobacter postgatei)과 Anaerolinaeles의 밀도는 대체적으로 생물증강에 의한 정화가 진행될수록 감소하였으며, 반면에 Gamma-proteobacteria (NOR5-6B_s and NOR5-6A_s), Bacteroidales_uc_s, 및 Flavobacteriales_uc_s의 밀도는 증가하는 경향이었다. 상대적으로 COD가 낮고 DO가 높은 청정지점인 St. 4에서는 호기성미생물인 Flavobacteriaceae_uc_s가 우점하였다. 이러한 미생물군은 하천의 정화과정을 추적할 수 있는 지표미생물로 활용될 수 있을 것으로 판단되었다. 생물증강 시행 후의 대표적 시점 퇴적토시료의 미생물군집 alpha diversity 지수(OTUs, Chao1 및 Shannon 지수)는 시행 전에 비해 감소하는 경향을 보였으며, 또한 beta diversity 분석기법(fast unifrac 분석)으로 분석한 결과 정화 전이나 초기에 비해서 정화가 진행될수록 전반적으로 시료에 무관하게 미생물군집의 유사성을 보여 생물증강이 현장 토착 미생물의 군집구조를 변화시키고 있음을 확인하였다. 이러한 사실로 보아 본 복합미생물에 의한 현장 생물증강법은 brine water 및 담수로 이루어진 오염된 하천을 환경친화적, 경제적으로 정화할 수 있는 대안으로 판단이 되었다.
