Sequence-tagged site (STS) markers tightly linked to the bacterial leaf blight (BLB) resistance genes, xa5, xa13 and Xa21, were used in this study. A survey was conducted to find polymorphisms between the resistant and susceptible germplasm in rice. 500 of Korean varieties and 100 of landraces were evaluated in this study. STS marker, RG207 was used to having xa5 resistance gene of rice germplasm. 27 varieties of Korean germplasm showed resistant for xa5 gene. The RG136 an xa-13 marker resulted in a single band of approximately 1kb in all the rice accessions studied. In order to detect polymorphism, digestion of the polymerase chain reaction (PCR) product was performed using a restriction enzyme Hinf Ⅰ. The resistant lines resulted in two bands 0.5kb on digestion with Hinf Ⅰ, while the same enzyme did not digest the PCR product of susceptible lines. No polymorphism was detected in Korean varieties and landraces, indicating that they probably do not contain xa13 gene. pTA248 an Xa-21 marker detected a band of 1kb in the resistant lines and bands of either 750bp or 700bp in the susceptible lines. Among germplasm tested, there are no varieties and landraces with Xa21 resistant gene. The results of the germplasm survey will be useful for the selection of parents in breeding programs aimed at transferring these bacterial blight resistance genes from one varietal background to another.

Agriculture is the most primitive civilized Activities of mankind but also the propellant of civilization development. Because it is the most basic material goods source of mankind. Among these materials rice is one of the most important part of these, we call them the substance of survival. From the beginning of the agricultural activities to the present we have experienced three industrial revolutions and are experiencing the Fourth Industrial Revolution. With the development of science and technology makes the efficiency of agricultural production is higher and higher, but compared with the original we are facing the same problem: natural disasters; pests and diseases; now also face the depletion of resources, environmental degradation and other issues. Therefore, improve and cultivate new crop varieties to make it better resistance and more production for better develop modern agriculture. It's very helpful for human social development. And also it is the responsibility and task of modern molecular breeding. In this study, I used bacterial leaf blight to find a better resistance gene to improve the resistance of rice. Frist Cultivate k3 of bacterial leaf blight, than inoculation by leaf clipping method (Kauffman,1973) in CNDH and SNDH population at 40days after rice transplanting. Check the lesion length by inoculation plants at 14days after inoculation, and record data for QTL analysis program. Than I get 4 intervals in 3 different chromosomal regions. I found these defense genes in the 4 intervals. So I used NCBI Justbio, Rapdb, etc. to finding these genes in physical map, than design primer for map base cloning. At last these defense genes will be employed in further research for introduction of the gene to the parental plant and rice breeding for solving food crisis.
This study was conducted to test on the resistance of varieties bred by crossing with Asominori to bacterial leaf blight. Nakdongbyeo and Dongjinbyeo which were susceptible to HB 9011, 8 resistant varieties including Ilmibyeo derived from Asominori and Asominori, Hwangok, 13 varieties including Chukoku 45 which were resistance to HB 9011, HB 9022 and HB 9033 were used to screen their res ponce depending on the various screening methods such as the true resistance, the secondary infection resistance and the disease common field test methods, and the results are as follows: Among 13 varieties tested, 11 varieties including llmibyeo showed tme resistance to HB 9011. Less than 1.0cm of disease lesion were developed on these varieties. Disease lesion was not developed on most of the Asominori lines including Daechongbyeo against IIB9011 and 1lmibyeo was also resistance to HB 9011, on this variety disease lesion area rate was 1.2%, and 7 varieties including Hwajinbyeo showed field resistance to HB 9022. Disease lesion area rate were 19.6% on Nakdongbyeo. 15.6% on Dongjinbyeo, from 3.0% to 2.4% on Asominori lines, and 0.5% on the Asominori when screened at disease common area. Disease was not developed on Keumnambyeo. Significant correlation coefficiences were found between the results from the test methods of the true resistance, the field resistance ancl the field resistance at disease common field tests on Asominori lines, but in some cases, even the varieties on which disease lesions developed, showed field resistance to HB 9022 and HB 9033.
The influence of rice bacterial leaf blight (BLB) disease incidence on yield and quality of Nampyung, Gang-baek and Iksan493 was investigated in three areas, Gimje, Yeongam and Jangheung, frequently found BLB infested rice. The infection rate of Nampyung, susceptible to BLB disease, was higher ($23{\sim}93%$) than Gang-baek ($2{\sim}15%$) and Iksan493 ($2{\sim}6%$), have resistant gene Xa7 and xa5, respectively. BLB disease incidence was severely found in Gimj and then yield of Nampyung was reduced 65% (352 kg/10a) compared to Iksan493 (540 kg/10a) due to the decrease in the ripen grain filling, brown/rough ratio and 1,000 grain weight. There was no difference of rice yield among Nampyung, Gangbaek and Iksan493 in Yeongam and Jangheung. The occurrence time of disease in Yeongam and Jangheung was later than Gimje and BLB disease was less infected in these areas. BLB infected rice grain showed inferior grain appearance and taste index to sound grain. independent of rice with resistant gene. In Yeongam and Jangheung, Gangbaek and Iksan493 cultivated showed lower ratio of white color to belly than Nampyung. Iksan493 showed better grain appearance and lower ratio of white core to belly than Gangbaek. In protein content of rice grain, the range of Gangbaek and Iksan493 was $6.9{\sim}9.8%$, but Nampyung showed unstable ranged from undetermined to 9.0%.

Rice bacterial leaf blight (BLB) by Xanthomonas oryzae pv. oryzae (Xoo) is considered to be one of the major rice diseases steadily occurring around the rice-producing countries. In this study, we developed a recombinase polymerase amplification (RPA) assay for the rapid, convenient and specific diagnosis of Xoo by targeting Xoo-specific transposase A gene. As the target gene can be amplified in 10 min without DNA extraction process and special equipment for temperature control, RPA for BLB can be useful and practical component for on-site diagnosis.
Gamma-ray irradiation is known to induce various mutations in plants caused by chromosome alterations. This study investigated disease responses of selected gamma-ray induced rice mutants generated from seven Japonica-type rice cultivars against three plant diseases. Among the tested 22 mutants, three gain-of-function mutants and six loss-of-function mutants against rice blast were obtained, as well as three loss-of-function mutants against bacterial leaf blight (BLB). Two of the loss-of-function mutants were susceptible to both rice blast and BLB. Gain-of-function mutation has not been frequently observed in rice plants, thus, the mutants can be used to identify loci of novel genes for the regulation of disease resistant response.
Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) in rice (Oryza sativa L.). In this study, we investigated the effect of a mutation in opsX (XOO1056), which encodes a saccharide biosynthesis regulatory protein, on the virulence and bacterial chemotaxis of Xoo. We performed DNA micro-array analysis, which showed that 63 of 2,678 genes, including genes related to bacterial motility (flagellar and chemotaxis proteins) were significantly downregulated ($<\;-2\;log_2$ fold changes) by the mutation in opsX. Indeed, motility assays showed that the mutant strain was nonmotile on semisolid agar swarm plates. In addition, a mutant strain (opsX::Tn5) showed decreased virulence against the susceptible rice cultivar, IR24. Quantitative real-time RT-PCR reaction was performed to confirm the expression levels of these genes, including those related to flagella and chemotaxis, in the opsX mutant. Our findings revealed that mutation of opsX affects both virulence and bacterial motility. These results will help to improve our understanding of Xoo and provide insight into Xoo-rice interactions.
Six hundred and twenty five isolates of Xanthomonas campestris pv. oryzae, cauing bacterial leaf blight (BLB) of rice, were classified into five pathotypes during $1979\~1983$ in Korea. Among them, 483 isolates$(77.3\%)$ were classified as pathotype I, 83 $(13.3\%)$ as pathotype II, 54$(8.6\%)$ as pathotype III, 3 $(0.5\%)$ as pathotype IV and 2$(0.3\%)$ as pathotype V. Pathotype I was the most prevalent throughout the country. Pathotype III was widely distributed in the western or southern parts of Korea, particularly in Jeonnam, Chungnam and Gyeongnam provinces. One isolate of pathotype IV was found in Gyeongnam in 1980, and two isolates were found in Jeonnam in 1981. Most of the isolates were obtained from Milyang 23 varietal group, while the isolates of pathotype III were collected from Yushin and Tongil varietal group.
Xanthomonas oryzae pv. oryzae, causal agent of bacterial leaf blight (BLB) of rice, had been collected and identified using Biolog and fatty acid analysis. Epidemics of BLB had been occurred all the times at several rice cultivating areas in Korea in 1999-2004. Most X. oryzae pv. oryzae isolated in 1999 and 2002 belonged to Korean race K1, but more than 50% of the pathogen isolated in 2003 belonged to Korean race K3. Especially, most pathogens isolated in Jeonnam and Joenbuk provinces belonged to Korean race K3. Inoculation test of near isogenic lines (NIL) of rice carrying single resistance genes against BLB showed that many isolates belonging to Korean race 1 reacted differently to diverse resistant monogenic lines of rice. Southern blot analysis also showed that the bacterial pathogens belonged to the same race had different numbers of avirulence genes. This results suggested that each Korean race type may respond to many resistance genes of rice. All the K3 races isolated in Jeonnam and Joenbuk provinces were able to cause disease on Xa3 monogenic lines of rice. Since most rice cultivars cultivated in Jeonnam and Jeonbuk were carrying Xa3 resistance genes, the bacterial pathogens isolated in Jeonnam and Jeonbuk were likely to develop to adapt to Xa3 resistance gene. Together with avirulence gene patterns of the bacterial isolates and the results of disease reaction of monogenic lines of rice to them, Korean X. oryzae pv. oryzae was classified into 19 pathotypes. This newly classified pathotypes should help the breeding of new resistance rice cultivars in Korea.
