Pharmacological Evaluation of Proprietary Preparation from Bacterial Metabolites with Special Reference to its Immunomodulatory Actions

  • A mixture of several bacterial metabolites $(Sterodin{\circledR})$ was used to study its effect on major immunocytes, in vivo and in vitro. This mixture of bacterial metabolites increased number of macrophages and neutrophils and their phagocytic activity in experimental animals for a transient period. BSA induced antibody production was found to be higher in the drug treated group. These results indicated that the bacterial metabolites probably acted through non-specific defence mechanism against invading organisms and the chance of reinfection was reduced.

유백피 추출물의 항세균 작용 (Antibacterial Activity of Ulmus pumila L. Extract)

  • 유백피 추출물의 항세균 효과를 측정하기 위해 각각 다른 농도의 추출물의 함유한 배지에서 MIC를 측정하고, 증식에 미치는 영향을 조사한 결과는 다음과 같았다.이 추출물은 실험에서 사용된 세균 중 S.aureus, S.epidermidis, S.faecalis, B.subtilis, M.luteus와 같은 그람 양성균 및 E.coli, S.typhi, P.vulgaris, P.aeruginosa와 같은 그람 음성균에서 높았고, K.pneumoniae에 대해서는 항균력을 나타내지 않았다. 이 추출물 중의 항세균 효과를 가진 물질은 $121^{\circ}C$에서 20분간 멸균한 후에도 항세균 효과에는 별다른 변화가 없었다.

포도상구균(Staphylococcus aureus)에 대한 키토산의 항균성 -키토산의 수용성, 탈아세틸화도 및 분자량이 항균성에 미치는 효과- (Antibacterial Activity of Chitosan against Staphylococcus aureus -The Effect of Watersolubility, Degree of Deacetylation and Molecular Weight of Chitosan on Antibacterial Activity-)

  • The antibacterial activities of several types of chitosan were measured against Staphylococcus aureus and evaluated for their application to antibacterial textile finishing. The % reduction of bacteria of the chitosans prepared in our laboratory were between 72 and 87%. The two water-soluble chitosans with molecular weights 1,000 and 3,000 did not show antibacterial activities. The deacetylation of chitosan was appeared to increase antibacterial activity. The % reduction in bacterial density of the 86%-deacetylated chitosan solution was 56% where that of the 76%-deacetylated chitosan solution was only 17% at 0.1% chitosan concentration. Molecular weights of the chitosans seemed not to affect antibacterial activities of chitosans. The antibacterial activity of the acid-soluble, 86%-deacetylated chitosan with 4 cps showed 98% of the % reduction at the level of 0.2% chitosan. The % reduction of bacteria of this chitosan was higher at the higher concentration of acetic acid in the chitosan-bacterial mixture. The antibacterial activity was increased with the pH change over the range of 4.0 to 6.5. The 100% of the % reduction of bacteria was achieved within 4 hour incubation of the chitosan-bacterial mixture. According to the data obtained from the above experiments, the four chitosans among the six prepared in our laboratory were proved to be valuable for antibacterial textile finishing.

The Effect of Polyphenols Isolated from Cynanchi wilfordii Radix with Anti-inflammatory, Antioxidant, and Anti-bacterial Activity

  • Recently, Cynanchi wilfordii Radix has gained wide use in Asian countries as a functional food effective for relieving fatigue, osteoporosis, and constipation, particularly in menopausal disorders. However, its anti-inflammatory and anti-microbial activities have not been explored in detail to date. The anti-inflammatory, antioxidant, and anti-bacterial properties of the Cynanchi wilfordii Radix extracts obtained with water, methanol, ethanol, and acetone were compared. All 4 polyphenol-containing extracts exhibited anti-inflammatory and antioxidant effects. The ethanol extract was found to elicit the most potent reduction of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and cytokine (IL-$1{\beta}$, IL-6, IL-10, and TNF-${\alpha}$) levels, as well as inhibit the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in a concentration-dependent manner. The evaluation of antioxidant activity also revealed the ethanol extract to have the highest free radical scavenging activity, measured as $85.3{\pm}0.4%$, which is equivalent to 99.9% of the activity of ${\alpha}$ -tocopherol. In the assessment of anti-bacterial activity, only ethanol extract was found to inhibit the growth of the Bacillus species Bacillus cereus and Bacillus anthracis. These results show that polyphenols of Cynanchi wilfordii Radix have anti-inflammatory, antioxidant, and anti-bacterial properties that can be exploited and further improved for use as a supplementary functional food, in cosmetics, and for pharmaceutical purposes.

In-Vitro, Anti-Bacterial Activities of Aqueous Extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo Against Gram Positive and Gram Negative Bacteria

  • Objective: Evaluations of the in-vitro anti-bacterial activities of aqueous extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and Shilajita mumiyo against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) and gram-negative bacteria (Escherichia coli, klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa) are reasonable since these ethnomedicinal plants have been used in Persian folk medicine for treating skin diseases, venereal diseases, respiratory problems and nervous disorders for ages. Methods: The well diffusion method (KB testing) with a concentration of $250{\mu}g/disc$ was used for evaluating the minimal inhibitory concentrations (MIC). Maximum synergistic effects of different combinations of components were also observed. Results: A particular combination of Acacia catechu (L.F.) Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo extracts possesses an outstanding anti-bacterial activity. It's inhibiting effect on microorganisms is significant when compared to the control group (P<0.05). Staphylococcus aureus was the most sensitive microorganism. The highest anti-bacterial activity against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) or gram-negative bacteria (Escherichia coli, Klebsiella pneumonia, Proteus mirabilis, and Pseudomonas aeruginosa) was exerted by formula number 2 (table 1). Conclusion: The results reveal the presence of anti-bacterial activities of Acacia catechu, Castanea sativa husk, Ephedra sp. and Mumiyo against gram-positive and gram-negative bacteria. Synergistic effects in a combined formula, especially in formula number 2 (ASLAN$^{(R)}$) can lead to potential sources of new antiseptic agents for treatment of acute or chronic skin ulcers. These results considering the significant anti-bacterial effect of the present formulation, support ethnopharmacological uses against diarrheal and venereal diseases and demonstrate use of these plants to treat infectious diseases.

동전기 생물학적 복원에서 전기분해반응이 미생물 활성에 미치는 영향 (Effect of Electrolysis on Bacterial Activity in Electrokinetic Bioremediation)

  • 미생물을 접종하지 않은 동전기 공정에서 전기분해에 의한 양극조에서 산소의 발생은 전류밀도에 비례하였으며 전해질을 순환시킴에 따라 생물반응기내 전해질과 함께 용존산소농도가 증가하였다. 전류의 공급과 함께 미생물 농도는 급격히 증가하였으며 이때 미생물의 산소소비량이 증가되어 용존산소농도가 감소되었다. Pentadecane-오염토양에 대한 동전기 생물학적 복원의 결과에서 높은 전류밀도 1.88 $mA/cm^2$에서 비록 산소의 발생량은 많았지만 오히려 증가된 유기산이 전해질 pH와 미생물 활성을 감소시키므로 미생물 농도와 제거효율이 0.63 $mA/cm^2$보다 낮게 나타났다. 0.63 $mA/cm^2$에서 적절한 산소의 공급과 동시에 전해액 pH의 감소가 작았으므로 최적의 미생물 농도와 제거효율을 얻을 수 있었다.

Image Analysis of Bacterial Cell Size by Diurnal Changes in Lake Soyang, Korea

  • To define the effects of zooplankton and phytoplankton to bacteria, bacterial numbers, frequency of dividing cells (FDC) and size distribution were performed with image analysis in the surface layer of Lake Soyang. In August 1992, when Anabaena was blooming, the bacterial number increased at daytime. Bacterial numbers and FDC value had a negative correlation (r = 0.83, P < 0.01). Bacterial size spectrums were dynamically changed during the day and night, especially the small bacteria less than $0.5\;{\mu}m^3$. Meanwhile, in October, after the bloom, the bacterial number was only one third of that in August, even though the FDC was higher than that in August. The bacterial numbers of small size class dropped at 13:00. But the size spectrums were relatively constant during the night time. These results suggest that the bacterial growth was tightly coupled with phytoplankton during Anabaena bloom. And after the bloom, the bacterial number was controlled grazing activity of zooplankton at daytime.

Propolis의 어류 병원성 세균에 대한 in vitro 항균 효과 (In vitro antimicrobial activity of Korean propolis against fish pathogenic bacteria)

  • The present study was to investigate in vitro antimicrobial effects of propolis against six different fish bacterial pathogens, Aeromonas hydrophila, Edwardsiella tarda, Vibrio vulnificus, V. parahaemolyticus, A. salmonicida subsp. masoucida, A. salmonicida subsp. salmonicida) using minimum inhibitory concentration (MIC) and minimum bacteriocidal concentration (MBC) tests. In the results, propolis exhibited antimicrobial activity against all bacteria used in the present study, but there was no marked difference in bacterial species except Vibrio species. Collectively, propolis was thought to be an usefulness antimicrobial substance for controlling bacterial diseases in the fish industry.

온실재배 토마토에서 발생하는 위조병의 미생물학적 제어 (Biological Control of Fusarium Wilt of Tomato Plants by Antagonistic Microorganism in Greenhouse)

  • This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it’s identification. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

Anti-inflammatory Activity of Detoxified Bacterial Strains in Wistar Rats

  • A mixture of several detoxified bacterial strains ($Sterodin^{(R)}$) has been studied for anti-inflammatory effect in Wistar rats on carrageenin, dextran and prostaglandin $E_1$ ($PGE_1$) induced edema in acute model and cotton pellet and carrageenin induced sub-acute model, while, Freund's adjuvant induced chronic model. The bacterial strains showed strong inhibitory activity in acute, sub-acute and chronic models of inflammation. Further, it reduced ${\alpha}1$ acid glycoprotein and ${\alpha}2$ macroglobulin levels in serum and prostaglandin $E_2$ in inflamed paw. These results indicated that the bacterial strains probably act through prostaglandin mediatory pathways and may be useful in treatment of inflammation.
