다양한 환경 조건의 하수처리시설 반응조 내 세균 및 고세균 군집 (Bacterial- and Archaeal Communities in Variously Environmental Conditioned Basins of Several Wastewater Treatment Plants)

  • 하수의 종류(생활하수, 축산폐수) 및 다양한 처리 공정에 따른 미생물군집구조를 비교하기 위해 A2O공법으로 운영되는 생활하수처리시설(안심·서부·신천)의 10개 생물학적 반응조와 축산폐수처리시설의 활성슬러지를 채취해 DNA 유전체를 추출한 후 세균은 프라이머 27F/518R, 고세균의 경우, 프라이머 Arch519F/A958R를 이용해 유전체를 증폭시켰고 그 염기서열을 Roche 454 GS-FLX Titanium을 이용한 pyrosequencing 법으로 분석하였다. 그 결과, 생활하수와 축산폐수에 따른 미생물 군집구조의 차이는 컸지만 A2O공법에 따른 산소 유무 등의 환경 변화와 관련된 군집구조의 변화는 크지 않았다. 혐기조 및 무산소조 반응조들에서만 분석한 고세균 군집 결과에서는 동일 하수처리시설의 반응조들의 고세군군집들끼리만 모이는 하수처리시설별 집괴현상을 나타냈다. 세균다양성 및 종 풍부도가 높은 서부처리시설이 다른 시설보다 더 높은 질소 및 인 제거율을 나타냈다.

  • The 5-stage biological nutrient removal (BNR) process with step feed system showed a very stable organic carbon and nutrient removal efficiency ($87\%\;COD\,;79\%\;nitrogen,\;and\;87\%$ phosphorus) for an operation period of 2 years. In each stage at the pilot plant, microbial communities, which are important in removing nitrogen and phosphorus, were investigated using fluorescence in-situ hybridization (FISH) and 165 rDNA characterization. All tanks of 5-stage sludge had a similar composition of bacterial communities. The totat cell numbers of each reactor were found to be around $2.36-2.83{\times}10^9$ cells/ml. About $56.5-62.0\%$ of total 4,6-diamidino-2-phenylindol (DAPI) cells were hybridized to the bacterial-specific probe EUB388. Members of ${\beta}$-proteobacteria were the most abundant proteobacterial group, accounting for up to $20.6-26.7\%$. The high G+C Gram-positive bacterial group and Cytophaga-Flexibacter cluster counts were also found to be relatively high. The beta subclass proteobacteria did not accumulate a large amount of polyphosphate. The proportion of phosphorus-accumulating organisms (PAOs) in the total population of the sludge was almost $50\%$ in anoxic-1 tank. The high G+C Gram-positive bacteria and Cytophaga-Flexibacter cluster indicate a key role of denitrifying phosphorus-accumulating organisms (dPAOs). Both groups might be correlated with some other subclass of proteobacteria for enhancing nitrogen and phosphorus removal in this process.

Effects of Soil-Plant Interactive System on Response to Exposure to ZnO Nanoparticles

  • The ecotoxicological effects of nanomaterials on animal, plant, and soil microorganisms have been widely investigated; however, the nanotoxic effects of plant-soil interactive systems are still largely unknown. In the present study, the effects of ZnO nanoparticles (NPs) on the soil-plant interactive system were estimated. The growth of plant seedlings in the presence of different concentrations of ZnO NPs within microcosm soil (M) and natural soil (NS) was compared. Changes in dehydrogenase activity (DHA) and soil bacterial community diversity were estimated based on the microcosm with plants (M+P) and microcosm without plants (M-P) in different concentrations of ZnO NPs treatment. The shoot growth of M+P and NS+P was significantly inhibited by 24% and 31.5% relative to the control at a ZnO NPs concentration of 1,000 mg/kg. The DHA levels decreased following increased ZnO NPs concentration. Specifically, these levels were significantly reduced from 100 mg/kg in M-P and only 1,000 mg/kg in M+P. Different clustering groups of M+P and M-P were observed in the principal component analysis (PCA). Therefore, the M-P's soil bacterial population may have more toxic effects at a high dose of ZnO NPs than M+P's. The plant and activation of soil bacteria in the M+P may have a less toxic interactive effect on each of the soil bacterial populations and plant growth by the ZnO NPs attachment or absorption of plant roots surface. The soil-plant interactive system might help decrease the toxic effects of ZnO NPs on the rhizobacteria population.

Characterization of Microbial Communities in Chinese Rice Wine Collected at Yichang City and Suzhou City in China

  • Two typical microbial communities from Chinese rice wine fermentation collected in Yichang city and Suzhou city in China were investigated. Both communities could ferment glutinous rice to rice wine in 2 days. The sugar and ethanol contents were 198.67 and 14.47 mg/g, respectively, for rice wine from Yichang city, and 292.50 and 12.31 mg/g, respectively, for rice wine from Suzhou city. Acetic acid and lactic acid were the most abundant organic acids. Abundant fungi and bacteria were detected in both communities by high-throughput sequencing. Saccharomycopsis fibuligera and Rhizopus oryzae were the dominant fungi in rice wine from Suzhou city, compared with R. oryzae, Wickerhamomyces anomalus, Saccharomyces cerevisiae, Mucor indicus, and Rhizopus microsporus in rice wine from Yichang city. Bacterial diversity was greater than fungal diversity in both communities. Citrobacter was the most abundant genus. Furthermore, Exiguobacterium, Aeromonas, Acinetobacter, Pseudomonas, Enterobacter, Bacillus, and Lactococcus were highly abundant in both communities.

An Overview of Different Techniques on the Microbial Community Structure, and Functional Diversity of Plant Growth Promoting Bacteria

  • Soil is a dynamic biological system, in which it is difficult to determine the composition of microbial communities. Knowledge of microbial diversity and function in soils are limited because of the taxonomic and methodological limitations associated with studying the organisms. In this review, approaches to measure microbial diversity in soil were discussed. Research on soil microbes can be categorized as structural diversity, functional diversity and genetic diversity studies, and these include cultivation based and cultivation independent methods. Cultivation independent technique to evaluate soil structural diversity include different techniques such as Phospholipid Fatty Acids (PLFA) and Fatty Acid Methyl Ester (FAME) analysis. Carbon source utilization pattern of soil microorganisms by Community Level Physiological Profiling (CLPP), catabolic responses by Substrate Induced Respiration technique (SIR) and soil microbial enzyme activities are discussed. Genetic diversity of soil microorganisms using molecular techniques such as 16S rDNA analysis Denaturing Gradient Gel Electrophoresis (DGGE) / Temperature Gradient Gel Electrophoresis (TGGE), Terminal Restriction Fragment Length Polymorphism (T-RFLP), Single Strand Conformation Polymorphism (SSCP), Restriction Fragment Length Polymorphism (RFLP) / Amplified Ribosomal DNA Restriction Analysis (ARDRA) and Ribosomal Intergenic Spacer Analysis (RISA) are also discussed. The chapter ends with a final conclusion on the advantages and disadvantages of different techniques and advances in molecular techniques to study the soil microbial diversity.

녹비 시용이 초기 논 토양 미생물군집에 미치는 영향 (Response of Soil Microbial Communities to Applications of Green Manures in Paddy at an Early Rice-Growing Stage)

  • 논 토양에서 청보리, 자운영과 트리티케일을 시용하고 벼생육초기에 토양 미생물 생태계의 변화에 미치는 영향을 검토하였다. 녹비를 시용한 처리구는 관행에 비해 토양 유기물 함량은 유의적으로 증가하였으나 토양의 나트륨 함량은 유의적인 감소를 나타냈다. 내생균근균의 함량과 구성비율은 녹비 시용으로 관행 보다 유의적으로 높아졌으며 특히 청보리 처리구가 가장 높았다. 또한 토양의 그람음성 세균과 그람양성 세균의 비율과 불포화지방산과 포화지방산 함량의 비율은 청보리 처리구가 다른 처리구에 비해 유의적인 증가를 보였다. 그리고 주성분 분석에서 PC 1에 가장 큰 영향을 미친 미생물 군집은 그람음성 세균인 것으로 나타났으며 청보리 처리구는 관행과 가장 큰 차이를 나타냈다. 이러한 결과를 종합한 결과 청보리는 논 토양의 초기 미생물 다양성을 증대시킬 수 있는 최적의 녹비작물로 판단된다.

한국농촌(韓國農村)의 기생충성인축공통질환(寄生蟲性人畜共通疾患)의 현황(現況)과 문제점(問題點) (Current Problems of Parasitic Zoonoses in Rural Korea)

  • Zoonoses are those diseases and infections that are naturally transmitted between vertebrate animals and man. At present, approximately 175 diseases caused by virus, rickettsia, bacteria, protozoa and helminthes as well as some of arthropods and the number is continually increasing. Although zoonoses are generally recognized as significant health problems, the actual prevalence and incidence of zoonotic infections is difficult to determine. Medical services in a rural area are scarce and ill persons may have little chance to see a physician. Furthermore, a lack of physician awareness and of appropriate diagnostic support is made often misdiagnosis. In rural Korea, parasitic disease is still important subject in the field of zoonotic infections considering its number and morbidity. Recently there was conspicuous reduction of soil-transmitted parasitic diseases (i.e. ascariasis. hookworms, trichuriasis), but clonorchiasis. intestinal trematodiases and taeniasis are a considerable problem. This is often significantly related to human behavioral patterns based on socio-economic and cultural conditions and linked with the local biologic and physical environment. The most of all parasitic zoonoses to man and animals are the food-transmitted parasitic diseases, and domestic animals such as dog, cat, pig, as well as fat and some mammals, ruminants and birds are act as reservoir hosts. In the present topics, current problems of parasitic zoonoses caused by protozoan infections (i. e. toxoplasmosis, pneumocystosis and cryptosporidiosis) which are the most common opportunistic in AIDS or immunocompromised persons, trematode infections (i.e. clonorchiasis, paragonimiasis and some intestinal tremadodiases). cestode infections such as cysticercosis and sparganosis, and some nematode infections of anisakiasis, thelaziasis and strongyloidiasis, are discussed here those on the occurrence in man and animals and the source of transmission as well as prevention and control measures in Korea.

미생물 위해성 평가 방법을 이용한 살모넬라 발생수 추정 (Estimation of the Number of Salmonellosis Using Microbial Risk Assessment Methodology)

  • 미생물 위해성 평가 방법론을 이용하여 살모넬라 발생 수를 추정하고, 기존의 연구 결과와 비교하여 그 활용 가능성에 대해 알아보았다. 살모넬라균의 오염수준은 국내에 발표된 각종 문헌(1997∼2000)과 관련 기관의 모니터링 자료(1999∼2001)를 수집하여 추정하였고, 식품 섭취 량은 2001 국민건강영양조사, 용량-반응 모델은 외국에서 발표된 연구 결과를 이용하였다. 각 변수를 시뮬레이션을 통해 추정한 결과 1년 동안 국내에서 발생 가능한 살모넬라 환자 수는 753,368명으로 나타났는데, 이는 보고된 환자수의 약 115배로 WHO의 추정배수보다는 낮은 결과를 보였다. 이상의 결과를 볼 때, 미생물 위해성 평가 방법은 식중독 발생 환자 수를 추정하고 식중독 관리에 있어서의 우선순위를 결정하는 데 이용이 가능한 것으로 보인다. 그러나 아직까지 검증 방법이 확립되어 있지 않고, 식중독균의 오염농도, 식품 섭취량, 용량-반응 관계, 유통ㆍ보관ㆍ조리 등의 실태에 관한 연구가 좀더 필요할 것으로 생각된다.

수원지역 초등학교 고학년 아동의 급식 위생지식과 수행수준의 평가 (Evaluation of Food Hygiene Knowledge and Health Practice for Elementary School Students in Suwon)

  • The purpose of this study was to examine food hygiene knowledge and health practice levels of elementary school students at foodservice in the Suwon area. Of the 500 upper graders from three elementary schools, 486 students (97.2%) participated in the study. The questionnaire was composed of general characteristics including experience of serving food at school, food hygiene knowledge (25 questions), and health practice (18 questions). The results were as follows: The education experiences of food hygiene were below 40% though most students (88.1%) participated in providing food at foodservice. The percentage of correct answers in food hygiene knowledge was over 70% in most questions, but relatively lower in food preservation temperature (44.7%) and food poisoning bacteria (43.2%). When we examined food hygiene behavior of elementary school students in 5scales, the level of personal hygiene management was 4.04, sanitary management in food product was 3.62, environmental hygiene was 3.92, and foodborn disease and food microorganism was 3.81. Each level in each subarea was significantly related in the frequency of hygiene education experiences. Finally, the food hygiene knowledge was positively correlated with its behavior level in elementary school foodservice. These results suggested that the knowledge of food hygiene may affect its behavior, and therefore, regular education of food hygiene at home and school would be needed to improve food safety in foodservice.

Improved Detection of Helicobacter pylori Infection and Premalignant Gastric Mucosa Using Conventional White Light Source Gastroscopy

  • Background: The gold standard diagnosis of H. pylori related gastritis is evidence of bacteria on histopathological examination of gastric mucosa. Our aim was to study the correlation between gastric mucosal morphology and histopathological severity of H. pylori related gastritis. Materials and Methods: Division was made on morphological features into:Type 1, showing regular arrangement of red dots; Type 2, showing cleft-like appearance; Type 3, with a mosaic appearance; and Type 4, having a mosaic appearance with focal or diffuse hyperemia. Results: Types 1 and 2 gastric mucosal morphologies were statistically significant in predicting an H. pylori negative status (137/145, p<0.01), while Types 3 and 4 were significant a positive status (139/155, p<0.01). The sensitivity, specificity, positive and negative predictive values of Type 3 and 4 morphologies for predicting H. pylori positive were 94.6%, 89.5%, 89.7% and 94.5%, respectively, with a good correlation with inflammation grading (p<0.01). Conclusions: Our study suggests that gastric mucosal morphology can be reliably identified using conventional white light source gastroscopy with good correlation between findings and inflammation grading.
