soil microbial activities and diversities in a newly reclaimed soil. Soil chemical properties, population of microbe, microbial biomass, and properties of microbial community were investigated under 4 different treatment (animal manure compost+green manure, chemical fertilizer, and without fertilizer). The experiment was conducted for 3 years from 2012 to 2014. The most of chemical properties in the animal manure compost+green manure treatment were increased continually compare to chemical fertilizer and without fertilizer. The population of bacteria and fungi were higher in the animal manure compost+green manure treatment, however, there was no difference on actinomyces. Soil microbial biomass C content was higher in the animal manure compost+green manure treatment than in chemical fertilizer and without fertilizer. Biolog examination showed that catabolic diversities of bacterial communities were higher in the treatment of animal manure compost+green manure. It was showed that principle component analysis of the Biolog data differentiated the organic matter amended soils from NPK and control. These results indicated that application of animal manure compost+green manure had a beneficial effect on soil microbial properties.

Little study has been published specifically addressing the dynamics of nitrate reducing bacteria (NBR) during the bioremediation of nitrate-contaminated aquifer. In our previous study we successfully quantified fumarate-enhanced microbial nitrate reduction rate in a nitrate-contaminated aquifer by using a series of single-well push-pull tests (PPTs). In this study we analyzed the suspended population during PPTs. To monitor changes in the microbial community, PCR amplification of 16S rDNA genes and denaturing gradient gel electrophoresis (DGGE) were used to study the dynamics of the bacterial community in detail. Before the stimulation of NBR, the dominant DGGE bands obtained by PCR were affiliated with V-Proteobacteria consisting of Acinetobacter spp. and Pseudomonas fluorescens. However, as NBR biostimulation proceeded, the dominant patterns of DGGE bands changed, and they were affiliated with Azoarcus denitrificans Td-3 and Flavobacterium xanthum. Azoarcus denitrificans Td-3 is known to completely reduce nitrate to nitrogen gas. The series of single-well push-pull tests in this study should prove useful for conducting rapid, low-cost feasibility assessments for in situ denitrification and provide important information about which microorganisms play a key role in bioremediation of a nitrate contaminated aquifer.

The microbial activity and bacterial community structure of activated sludge reactors, which treated free cyanide (FC), zinc-complexed cyanide (ZC), or nickel-complexed cyanide (NC), were studied. The three reactors (designated as re-FC, re-ZC, and re-NC) were operated for 50 days with a stepwise decrease of hydraulic retention time. In the re-FC and re-ZC reactors, FC or ZC was almost completely removed, whereas approximately 80-87% of NC was removed in re-NC. This result might be attributed to the high toxicity of nickel released after degradation of NC. In the batch test, the sludges taken from re-FC and re-ZC completely degraded FC, ZC, and NC, whereas the sludge from re-NC degraded only NC. Although re-FC and re-ZC showed similar properties in regard to cyanide degradation, denaturing gradient gel electrophoresis (DGGE) analysis of the 16S rRNA gene of the bacterial communities in the three reactors showed that bacterial community was specifically acclimated to each reactor. We found several bacterial sequences in DGGE bands that showed high similarity to known cyanide-degrading bacteria such as Klebsiella spp., Acidovorax spp., and Achromobacter xylosoxidans. Flocforming microorganism might also be one of the major microorganisms, since many sequences related to Zoogloea, Microbacterium, and phylum TM7 were detected in all the reactors.
This study aims at characterizing the bacteriophages isolated from activated sludge performing enhanced biological phosphorous removal (EBPR) to understand the interactions between the phage-host system and bacterial community. Sixteen bacterial isolates (E1-E16) were isolated as host bacterial strains from EBPR activated sludge for phage isolation. Forty bacteriophages based on their plaque sizes (2 plaques on E4, 4 on E8, 11 on E10, 5 on E14, 18 on E16) were obtained from filtered supernatant of the EBPR activated sludge. Each bacteriophage did not make any plaque on bacterial strains tested in this study except on its own host bacterial strain, respectively, indicating that the bacteriophages are with narrow host specificity. However, fourteen of the forty bacteriophages obtained in this study lost their virulent ability even on their own host bacteria. All of the lytic phages showed similar one-step growth patterns and had long latent period (about 9 hours) to reproduce their phage particles in their host bacterial cells. On the other hand, their probable burst sizes (6 to 48 per host cell) were large enough to actively lyse their host bacterial cells. Therefore, it could be implied that bacteriophages are also important members of the microbial community in EBPR activated sludge, and lytic phages directly decrease the population size of their host bacterial groups in EBPR activated sludge by lysis.

The pathogens and community structure of the fishes in Taehwa river were investigated from March 2007 to January 2009. During the study period, 3,504 individuals belonging to 35 species, 17 families and 9 orders were collected. The numerically dominant and subdominat species were Opsarichthys uncirostris (relative abundance 39.7%) and Hemibarbus labeo (relative abundance 30.9%). There were five Korean endemic species (20.8%) including Squalidus chankaensis tsuchigae, Zacco koreanus, Cobitis hankugensis, Coreoperca herzi, Odontobutis platycephala. The large fishes like Hemibarbus labeo or Opsarichthys uncirostris were gathered around the Samho bridge, sampling site 2 according to a season. The reaction to which two kinds of fish pathogenic virus is all negative and no fish pathogenic bacteria was isolated from 220 individuals. The fish pathogenic parasite not present variously with 7 species. Especially, Trichodina sp. was detected monthly and the infective density was high. But it is cosidered that temporary overcrowding of fish is not influenced mass mortality causing diseases in the specific site of river.

Alcaligenes sp. JMP228 carrying 2,4dichlorophenoxyacetic acid (2,4-D) degradative plasmid pJP4 was inoculated into natural soil, and transfer of the plasmid pJP4 to indigenous soil bacteria was investigated with and without 2,4-D amendment. Plasmid pJP4 transfer was enhanced in the soils treated with 2,4-D, compared to the soils not amended with 2,4-D. Several different transconjugants were isolated from the soils treated with 2,4-D, while no indigenous transconjugants were obtained from the unamended soils. Inoculation of the soils with both the donor Alcaligenes sp. JMP228/pJP4 and a recipient Burkholderia cepacia DBO 1 produced less diverse transconjugants than the soils inoculated with the donor alone. Repetitive extragenic palindromic-polymerase chain reaction (REP-PCR) analysis of the transconjugants exhibited seven distinct genomic DNA fingerprints. Analysis of 16S rDNA sequences indicated that the transconjugants were related to members of the genera Burkholderia and Pandoraea. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that inoculation of the donor caused clear changes in the bacterial community structure of the 2,4-Damended soils. The new 16S rRNA gene bands in the DGGE profile corresponded with the 16S rRNA genes of 2,4-Ddegrading transconjugants isolated from the soil. The results indicate that introduction of the 2,4-D degradative plasmid as Alcaligenes sp. JMP228/pJP4 has a substantial impact on the bacterial community structure in the 2,4-D-amended soil.

Background: Suppressive soil inhibits soil-borne diseases if pathogens are present, and ginseng does not show injury even if replanted in the same field. Methods and Results: Soil chemical properties and microbial community of soil were investigated in soil suppressive and conducive to ginseng root rot. Root rot disease in 2-year-old ginseng was tested by mixing conducive soil, with suppressive or sterilized suppressive soil. The root rot ratio in suppressive soil was 43.3% compared to 96.7% in conducive soil. Biological factors acted to inhibit the root rot because disease ratio was increased in the sterilized suppressive soil compared to that in non-suppressive soil. The suppressive soil had lower pH, nitrate nitrogen and sodium than the conducive soil. Dominat bacteria and fungi (more than 1.0%) were 3 and 17 species in conducive soil and 7 and 23 species in suppressive soil, respectively. The most predominant fungi were Pseudaleuria sp. HG936843 (28.70%) in conducive soil and Pseudogymnoascus roseus (7.52%) in suppressive soil. Conclusion: Microbial diversity was more abundant in the suppressive soil than in the conducive soil, and the proportion of pathogens (Nectriaceae sp.) causing root rot was significantly lower in the suppressive soil than in the conducive soil.
Cyperus rotundus L. is a perennial herb that was found to be dominating an area in northeast Brazil previously contaminated with petroleum. In order to increase our knowledge of microorganism-plant interactions in phytoremediation, the bacterial community present in the rhizosphere and roots of C. rotundus was evaluated by culture-dependent and molecular approaches. PCR-DGGE analysis based on the 16S rRNA gene showed that the bacterial community in bulk soil, rhizosphere, and root samples had a high degree of similarity. A complex population of alkane-utilizing bacteria and a variable nitrogen-fixing population were observed via PCR-DGGE analysis of alkB and nifH genes, respectively. In addition, two clone libraries were generated from alkB fragments obtained by PCR of bulk and rhizosphere soil DNA samples. Statistical analyses of these libraries showed that the compositions of their respective populations were different in terms of alkB gene sequences. Using culturedependent techniques, 209 bacterial strains were isolated from the rhizosphere and rhizoplane/roots of C. rotundus. Dot-blot analysis showed that 17 strains contained both alkB and nifH gene sequences. Partial 16S rRNA gene sequencing revealed that these strains are affiliated with the genera Bosea, Cupriavidus, Enterobacter, Gordonia, Mycoplana, Pandoraea, Pseudomonas, Rhizobium, and Rhodococcus. These isolates can be considered to have great potential for the phytoremediation of soil with C. rotundus in this tropical soil area.

Objective: As laying hens become aged, laying performance and egg quality are generally impaired. One of the practical methods to rejuvenate production and egg quality of aged laying hens with decreasing productivity is a forced molting. However, the changes in intestinal microbiota after forced molting of aged hens are not clearly known. The aim of the present study was to analyze the changes in excreta bacterial communities after forced molting of aged laying hens. Methods: A total of one hundred 66-wk-old Hy-Line Brown laying hens were induced to molt by a 2-d water removal and an 11-d fasting until egg production completely ceased. The excreta samples of 16 hens with similar body weight were collected before and immediately after molting. Excreta bacterial communities were analyzed by high-throughput sequencing of bacterial 16S rRNA genes. Results: Bacteroidetes, Firmicutes, and Proteobacteria were the three major bacterial phyla in pre-molting and immediate post-molting hens, accounting for more than 98.0%. Lactobacillus genus had relatively high abundance in both group, but decreased by molting (62.3% in premolting and 24.9% in post-molting hens). Moreover, pathogenic bacteria such as Enterococcus cecorum and Escherichia coli were more abundant in immediate post-molting hens than in pre-molting hens. Forced molting influenced the alpha diversity, with higher Chao1 (p = 0.012), phylogenetic diversity whole tree (p = 0.014), observed operational taxonomic unit indices (p = 0.006), and Simpson indices (p<0.001), which indicated that forced molting increased excreta bacterial richness of aged laying hens. Conclusion: This study improves the current knowledge of bacterial community alterations in the excreta by forced molting in aged laying hens, which can provide increasing opportunity to develop novel dietary and management skills for improving the gastrointestinal health of aged laying hens after molting.

The scientific information between microbial community and chemical properties of reclaimed tidal soil is not enough to understand the land reclamation process. This study was conducted to investigate the relation between chemical properties and microbial activities of soils from reclaimed tidal lands located at south-western coastal area (42 samples from Goheuong, Samsan, Bojun, Kunnae, Hwaong and Yeongsangang sites). Most of the reclaimed soils showed chemical characteristics as salinity soil based on EC. Only $Na^+$ in exchangeable cation was dependent on EC of reclaimed soil, whereas other cations such as $K^+$, $Ca^{2+}$, and $Mg^{2+}$ were independent on EC. The mesophilic bacteria decreased with an increase in EC of soil. Microbial population increased with soil organic content in the range of $0{\sim}10g\;kg^{-1}$ and dehydrogenase activity less than $100{\mu}g-TPF\;g^{-1}h^{-1}$. Microbial population of soils from reclaimed tidal lands was closely related to the microbial community containing hydrolytic enzyme activities of cellulase, amylase, protease, and lipase.
