Bifidobacterium longum HY8001의 섭취가 사람의 장내세균층 및 장내세균 효소에 미치는 영향 (Effect of Bifidobacterium longum HY8001 Administration on Human Fecal Bacterial Enzymes and Microflora)

  • The effects of Bifidobacterium longum HY8001 supplement intake on the fecal microflora and fecal bacterial enzyme activity were studied in ten healthy human volunteers, before, during and after intake (respectively for 3 weeks). During intake of B. longum HY8001 supplement, fecal, $\beta$-glucuronidase and nitroreductase activities significantly decreased 44.6%(p<0.005) and 32.3%(p<0.01), respectively. Although numbers of major bacterial groups of fecal microflora were not affected by B. longum HY8001 intake for 3 weeks, the number of Bifidobacterium was significantly increased (p<0.05). This result indicates that intake of B. longum HY8001 might be potentially beneficial for the prevention and inhibition of colon cancer and improvement of human intestinal microflora composition.

미나리발효액이 장내 유해세균 및 유익균의 In Vitro 생육 및 효소활성에 미치는 영향 (Effect of Extract of Fermented Dropwort on Intestinal Bacteria and Enzymes In Vitro)

  • 미나리(Oenanthe stolonifera) 발효액이 장내 병원성 미생물과 유익균의 생육, 그리고 장내 세균효소에 미치는 영향을 in vitro 에서 조사하였다. 고상 배지 (Agar plate) 에서의 clear zone 형성에 의한 생육저해를 측정한 결과, Vibro, Salmonella 등의 유해 미생물에 대해서 강한 생육저해 효과를 보였으며 Bifidobacterium longum 에 대해서는 생육저해 효과가 크지 않았다. 액체배지에서의 최소저해농도(MIC) 측정에서도 고장배지에서와 같은 경향을 보여 상대적으로 B. longum 에 대한 생육저해가 가장 적었다. 발효 기간에 따른 영향을 보면 발효 기간이 길수록 적은 양으로도 유해한 균들의 생육을 잘 저해할 수 있는 것으로 나타났다. 장내 세균 효소인 ${\beta}$-glucuronidase와 tryptophanase의 활성에 대해 미나리 발효액은 발효하지 않은 액에 비해 저해효과가 컸으며 발효기간이 길수록 저해효과도 증가하였다. 이상의 실험 결과로서 미나리 발효액은 유해세균에 대한 생육저해능이 크고 상대적으로 유익균인 B. longum에 대한 저해는 적어, 음용 시 정장 효과가 있을 것으로 추정된다.

  • 건강한 개의 혈액세포와 골수세포의 세포화학적 특성을 조사하였다. 이 실험에서 혈액과 골수 시료에 항응고제를 일체사용하지 않았으며 시료채취후 즉시 도말표본을 작성하여 30분 이내에 반응을 실시하였다. 이 실험의 결과는 아래와 같다. 1. alkaline phosphatase의 활성은 호산구계통과 간혹 전골수구에서 양성반응을 나타내었다. 2. acid phosphatase의 활성은 대부분의 계통의 세포에서 양성반응을 나타내지만 tartrate로 억제하면 호산구계만 양성반응을 나타내었다. 3. peroxidase 활성은 골수구계통의 모든 세포에서 양성반응을 나타내며 단구에서는 미약한 양성의 미세과립을 나타내었다. 4. naphthyl-AS-D-chloroacetate esterase활성은 호중구계 세포에서만 양성을 나타낸다. 5. $\alpha$-naphthyl acetate esterase활성은 단구와 일부의 임파구에서 양성을 나타낸다 6. Sudan black B 염색은 골수구계 세포와 단구계 세포에서 양성을 나타내었다. 7. $\beta$-glucuronidase 활성은 적혈구계를 제외한 모든 세포에서 양성반응을 나타내었다.

Effect of Aster scaber extract on the Growth of Bifidobacteria and Clostridium perfringens

  • Growth responses of some intestinal bacteria such as bifidobacteria and Clostridium perfringens to the extracts of certain foodstuffs were investigated in vitro. Among edible mountain herbs, the extracts of several chui-na-muls (Aster tataricus, Ligularia fischeri and Aster scaber) had an inhibitory activity against C. perfringens on the agar plate and the water extract of Aster scaber worked selectively on it among intestinal bacteria. The water extract showed growth-promoting effect toward bifidobacteria such as B. adolescentis, B. animalis, B. bifidum, B. infantis and B. thermophilum in the broth culture. When the faecal inoculum was incubated in the culture with the extract, the population of C. perfringens decreased, whereas that of bifidobacteria increased by $10^3$ scale.$\beta$-glucuronidase activity in the culture with the water extract of Aster scaber digested with pepsin and pancreatin was lower than that in the control culture.

Bioconversion of Flavones During Fermentation in Milk Containing Scutellaria baicalensis Extract by Lactobacillus brevis

  • Scutellaria baicalensis (SB), a traditional herb with high pharmacological value, contains more than 10% flavone by weight. To improve the biological activity of flavones in SB, we aimed to enhance the bioconversion of baicalin (BG) to baicalein (B) and wogonoside (WG) to wogonin (W) in SB during fermentation using beta-glucuronidase produced from Lactobacillus brevis RO1. After activation, L. brevis RO1 was cultured in milk containing SB root extract with various carbon or nitrogen sources at $37^{\circ}C$ for 72 h. During fermentation, the growth patterns of L. brevis RO1 and changes in the flavone content were assessed using thin-layer chromatography and high-performance liquid chromatography. After 72 h of fermentation, the concentrations of B and W in the control group increased by only 0.15 and 0.12 mM, respectively, whereas they increased by 0.57 and 0.24 mM in the fish peptone group. The production of B and W was enhanced by the addition of 0.4% fish peptone, which not only improved the growth of L. brevis RO1 (p < 0.001) but also enhanced the bioconversion of flavones. In conclusion, the bioconversion of flavones in SB may provide a potential application for the enhancement of the functional components in SB.

Studies on the Pharmacological Actions of Cactus:Identification of Its anti-Inflammatory Effect

  • The ethanol extracts of Opuntia ficus-indica fructus (EEOF) and Opuntia ficus-indica stem (EEOS) were prepared and used to evaluate the pharmacological effects of cactus. Both the extracts inhibited the writhing syndrome induced by acetic acid, indicating that they contains analgesic effect. The oral administrations of EEOF and EEOS suppressed carrageenan-induced rat paw edema and also showed potent inhibition in the leukocyte migration of CMC-pouch model in rats. Moreover, the extracts suppressed the release of $\beta$-glucuronidase, a lysosomal enzyme in rat neutrophils. It was also noted that the extracts showed the protective effect on gastric mucosal layers. From the results it is suggested that the cactus extracts contain anti-inflammatory action having protective effect against gastric lesions.

Isolation and Properties of $\beta$-N-Acetyl-D-glucosaminidase B from Rat Uterus

  • ${\beta}$-N-Acetyl-D-glucosaminidase B was highly purified with the following sequence of steps; DEAE-cellulose, CM-cellulose, and Sephadex G-200 gel filtration chromatograpies. The specific activity of the purified ${\beta}$ -N-acetyl-D-glucosaminidase B was 2.2 units/mg protein with 12.9 % yield and 196.2 fold purity. The purified ${\beta}$-N-acetyl-D-glucosaminidase B showed single band on polyacrylamide gel electrophoresis. The final preparation of ${\beta}$ -N-acetyl-D-glucosaminidase B was completely free friom arylsulfatase and ${\beta}$-glucuronidase. ${\beta}$ -N-Acetyl-D-glucosaminidase B had pH optimum of 4.5 in 0.5 M sodium citrate buffer. The molecular weight of ${\beta}$-N-acetyl-D-glucosaminidase B was 133,000 by Sephadex G-200 gel filtration. The Km value of ${\beta}$-N-acetyl-D-glucosaminidase B using p-nitrophenyl-N-acetyl-${\beta}$-D-glucosaminide as substrate was 1.0 mM and $V_{max}$ was 0.014 ${\mu}$ mole/min. ${\beta}$-N-Acetyl-D-glucosaminidase B was stable at $55^{circ}C$ for 70 minutes. The crude ${\beta}$ -N-acetyl-D-glucosamiinidase in 70 % ammonium sulfate retained 93 % activity after 7 months storage at -$55^{circ}C$. Bovine serum albumin, sodium chloride, and phosphate activated ${\beta}$ -N-Acetyl-D-glucosaminidase B. N-Acetyl-D-glucosamine, ${\alpha}$-methyl-D-mannoside, and acetate inhibited ${\beta}$ -N-acetyl-D-glucosaminidase B.

  • 자란(B. striata)의 미성숙종자로 부터 유도한 embryogenic callus를 현탁배양하고 이와같은 embryogenic cell suspensions로부터 분리한 원형질체에 electroporation 방법을 사용하여 reporter genes이 들어있는 pBI121 plasmid DNA 를 도입하고 그 발현을 확인하였다. 배양된 세포에 있어서 GUS의 활성은 plasmid DNA양의 증가와 함께 높게 나타났으며, 200-300 voltage/1180 uF에서 GUS의 활성 및 원형질체의 생존율(viability)이 가장 좋았다.

사군자탕(四君子湯)이 TNBS로 유발(誘發)된 생쥐의 염증성(炎症性) 장질환(腸疾患)에 미치는 영향(影響) (The effect of Sagunja-tang on TNBS-induced Inflammatory Bowel Disease in Mouse)

  • Objectives : The present study aimed to find out the effect of Sagunja-tang on the prevention and treatment of inflammatory bowel disease using mice with TNBS-induced inflammatory bowel disease. Methods : Mice with TNBS-induced inflammatory bowel disease were medicated with Sagunja-tang, and the weight changes, colon length, lipid peroxidation, and myeloperoxidase activity were observed. Levels of the inflammatory markers interleukin (IL)-$1{\beta}$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}$B (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis were also measured. Finally, the activation of fecal bacterial enzyme, ${\beta}$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG) and hyaluronic acid were observed. Results : We found that oral administration of Sagunja-tang inhibited TNBS-induced colon shortening and also inhibited myeloperoxidase activity in the colon of mice as well as IL-$1{\beta}$ and COX-2 expression. Sagunja-tang also inhibited TNBSinduced lipid peroxidation and pp65 activation in the colon of mice. In addition, Sagunja-tang inhibited ${\beta}$-glucuronidase activation and fecal hyaluronic acid degradation activation. Conclusions : It is supposed that Sagunja-tang has a potential therapeutic effect on inflammatory bowel disease through the inhibition of both NF-${\kappa}$B activation and lipid peroxidation, and the improvement of intestinal conditions.

인삼섭취가 장내세균 및 세균대사에 미치는 영향 (Effect of Panax ginseng Extract on Growth Responses of Human Intestinal Bacteria and Bacterial Metabolism)

  • The growth responses of a variety of human intestinal bacteria to extracts of Panax ginseng and five other oriental medicinal Araliaceae were elraluattd in vitro and in vivo. The extracts enhanced the growth of Brifidobnnerilrm breve and B. longlim in media with or without carbon sources, suggesting that bifidus factors) might be involved in the phenomenon. This effect was most pronounced with water extract of P. ginseng, the growth of 27 bifidobacteria strains belonging to B adolescentis, B. longum, B. brim and B. infantis being greatly stimurated, whereas seven B. bifidum strains and other bacteria such as clostridia and Escherichin soli had little or no ability to utilise it for growth. Methanol extracts of p. ginseng were found to selectively inhibit growth of various clostridia including bifidobacteria. Paraputrificum, but this effect was not observed on other bacteria including bifidobacteria. The effect of ginseng extract intake (600 mg/day for two weeks) on the faecal microflora, pH, volatile fatty acids, ammonia, putrefactive products, and -glucuronidase, -glucosidase and nitroreductase activities, and on the blood components (triglyceride, total cholesterol and ammonia) were investigated using seven healthy human volunteers. The total concentration of faecal microflora including Bifidnkaderiifm app. during the period of ginseng extract intake %twas significantly unaffected from the preceding and subsequent control peroids. However, the frequency of occurrence of subjects having C. perfringens was significantly decreased. The faecal pH value was also significantly decreased, suggesting that the intake might increase the activity of Bifidobncterium spry. Other biochemical properties in faeces did not changed significantly. The levels of ammonia and triglycerid in blood were decreased with ginseng extract intake. These results may be an indication of at least one of the Pharmacological actions of p. ginseng as an adaptogen.
