This experiment was carried out to study the availability of Korean peat as a main carrier material of rhizobial inoculant, using the alfalfa strain Rhizobium meliloti M 14 which was isolated in the previous report. Modification of powdered peat with calcium carbonate and other materials was studied; inoculation of the peat with culture broth, maturation of the mixture under different conditions, and survival of the strain in the peat culture was examined. The results obtained were as follows. 1. Peat produced in Pyongtak was highly acidic, pH 3.8, and addition of calcium carbonate by 14% was required for pH adjustment to 6.4. However the amount of calcium carbonate could be reduced by 4 to 8% when carbon or charcoal was mixed with the peat. 2. Viable number of the strain reached to $7-9{\times}10^9cells/g$ after 3 days, when inoculated with the culture broth of early stationary growth phase and matured in unsteriled peat of open trays; and the number in steriled peat was $1.1-6.2{\times}10^{10}cells/g$ after 5 days, when matured in closed bottles. 3. Survival of the strain was affected markedly by storage temperature, and positive effect of D-sorbitol on the viability was recognized at elevated temperatures, when added as an additional carbon source and moistening agent. Glycerol, sorbitol, or sodium lactate was utilized by the strain as a sole source of carbon, and the decimal reduction time of viable number in the peat culture was was found to be 8 to 9 weeks at $25^{\circ}C$ when these agents were added by 0.5%.
Metabolic trial with 3 fistulated sheep was conducted in a 3 $\times$ 3 Latin square design and feeding trial with 24 Hanwoo steers in 12 month of age for 20 months was conducted to investigate the replacing effect of rice straw with fermented spent mushroom (Flammuliua velutipes) compost (FSMC) on fermentation characteristics, ruminal effective degradabilty and whole tract digestibility of nutrients in sheep, and to examine the growth performance of Hanwoo steers. Experimental diets for the metabolic trial with sheep were commercial concentrates and rice straw in the ratio of 70 : 30 (CON, DM basis). Same concentrate with 30% FSMC and 70% rice straw (FSMC-30) and 60% FSMC and 40% rice straw(FSMC-60). Diets for Hanwoo steers in three treatments were same as for metabolic trial in replacing ratio of rice straw with FSMC. pH of rumen fluid in sheep was not affected by FSMC. Ammonia-N content in the rumen fluid was highest in the sheep fed FSMC-60 at 3h (P<0.045). The CON diet increased (P<0.001) acetate proportion at 1h and 3h post feeding compared to FSMC-60 diet while propionate proportion was highest in the sheep fed FSMC-60 diet for all the sampling times (P<0.027~P<0.002). Increased proportion of butyrate was observed at 30 min prior to feeding (P<0.031), and 1h (P<0.011) and 6h(P<0.039) post feeding from sheep fed FSMC-30 diet compared to those from sheep fed other diets. Effective degradability in the rumen was not influenced by experimental diets. Whole tract digestibility of crude protein (P<0.031) and neutral detergent fiber (P<0.006) tended to be increased in the sheep fed CON diet while corresponding values were lowest in the sheep fed FSMC-60 diet. Total body weight gain of Hanwoo steers for 8 months was not different among diets, thus daily body gain was not influenced by the experimental diets.
This experiment was conducted in the field to investigate the effects of mulched condition on nutrient's mineralization and its mobility as affected by environmental changes (soil moisture and soil temperature) in the plow layer. 1. Nitrogen mineralization was faster owing to the prevention of evaporation and the increase of soil temperature in the poly ethylene mulched plot. As a result, nitrate form of nitrogen was much earlier available for plant in mulched plot than in non mulched plot. 2. Available $P_2O_5$ content in the plow layer was kept highest in the fertilized zone (10-20 cm depth) regardless of difference between mulched and non mulched condition, and was nearly constant without showing difference between planting and non planting of tobacco due to the lower availability of phosphorus by tobacco. 3. Potassium applied in the plow layer was remained a large quantity until the latter growth stage resulting in the lower recovery of potassium by tobacco. 4. Mineralized nutrients such as $NH_4-N$, $NO_3-N$, and K in the plow layer of mulched plot were transported from the fertilized layer (10-20 cm depth) and accumulated to the surface layer (0-10 cm depth) as the growing time goes by, but those in non mulched plot were distributed to the deeper layer. 5. Mobility of available nutrients in the plow layer was If order as $NO_3-N$$$\geq_-$$$NH_4-N$ > K > available $P_2O_5$. 6. Leaching degree of mineralized nutrients in the mulched plot was much lower than in the non mulched plot.
To utilize composts more efficiently, combining composts with fertilizer to meet crop requirements is an appealing alternative. A pot experiment was conducted to study the effect of application rate of composted pig manure blended with fertilizer on the availability and loss of fertilizer-N. Chinese cabbage (Brassica campestris L. cv. Samjin) plants were cultivated for 30 and 60 days. 15N-Labeled urea ($5.24\;^{15}N\;atom\;%$) was added to soil at $450mg\;N\;kg^{-1}$, and unlabeled compost ($0.37\;^{15}N\;atom\;%$) was added at 0, 200, 400, and $600mg\;N\;kg^{-1}$. The amount of plant-N derived from urea was not affected by compost application at rate of $200mg\;N\;kg^{-1}$. However, compost application at 400 and $600mg\;N\;kg^{-1}$ significantly (P<0.05) increased plant assimilation of N from urea irrespective of sampling time, probably because of physicochemical changes in the soil properties allowing urea-N to be assimilated more efficiently. The amount of immobilized urea-N increased with increasing rate of compost application at both growth periods, as the results of increased microbial activities using organic C in the compost. Total recovery of urea-N (as percentage of added N) by Chinese cabbage and soil also increased with increasing rate of compost from 71.5 to 95.6% and from 67.0 to 88.2% at the 30- and 60-days of growth, respectively. These results suggest that increasing rate of compost blending increases plant uptake of fertilizer-N and enhances immobilization of fertilizer-N, which leads to decrease in loss of fertilizer-N. However, information about the fate of immobilized N during future crop cultivation is necessary to verify long-term effect of compost blending.

This study was aimed to improve the productivity and income of mushroom farming by developing a new casing material as a substitute for clay loam casing soil, which is becoming more difficult to acquire. When the new casing materials were used for the stable production of button mushroom (Agaricus bisporus), a 1:1 mixture of clay loam and button mushroom media obtained after harvest supported 13% greater mycelial growth ($32.0kg/3.3m^2$). This material was better than clay loam soil in preventing contamination with environmental compounds and pests. The use of an inexpensive 1:1 mixture of peat moss and coco peat resulted superior mycelial growth with 4% better yield ($32.9kg/3.3m^2$) compared with conventional clay loam soil. Advantages of these casing materials included ready availability and improved productivity. Mixtures of peat moss + coco peat + zeolite (50%:30%:20%) and coco peat + coal ash (75%:25%) could substitute for conventional casing soil. Additionally, the novel mixtures containing material obtained after cultivation might be used to produce organic fertilizer.
To understand the ecological function of heterotrophic bacterial community in water column of large freshwater lakes in the permafrost zone, we investigated the structure and function of bacterial community in Lake Khuvsgul, Mongolia. Species composition of overall bacterial community was analyzed by denaturing gradient gel electrophoresis (DGGE) of 16S rRNA gene fragments, and bacteria that can be cultured at 10oC were isolated and characterized. Based on the depth profile of environmental parameters, thermocline and chemocline were recognized at the 5~10 m zone of the water column. The stratified DGGE profile indicated that the discontinuity of water properties might influence the structure of bacterial community: band profiles in the 0~5 m zone were diverse with large change by depth, but the profile was relatively stable at the $\geq$10 m zone, with predominance of the band identified as Acidovorax facilis. Bacterial cultures were screened for protease, cellulase, amylase and lipase activity, and 23 isolates were selected for high activity of the hydrolytic enzymes. The isolates were identified based on their 16S rRNA gene sequences. In the surface water (zero meter depth), Acidovorax defluvii and Sphingobacterium faecium with high cellulase activity were present. Flavobacterium succinicans, Mycoplana bullata and A. facilis were stably predominant isolates at 2 m, 5 m, and $\geq$10 m depths, respectively. F. succinicans isolates showed high protease activity while M. bullata isolates showed moderate levels of protease and celluase activity. A. facilis isolates showed either cellulase or lipase activity, exclusively to each other. According to the profile of growth rates of the isolates in the temperature range of $0\sim42^{\circ}C$, the surface-zone (0~5 m) isolates were facultative psychrophiles while isolates from $\geq$10 m depth were typical mesophiles. This stratification is believed to be due to stratified availability of organic materials to the bacterial decomposers. In the water column below the chemoline, the environment is extremely oligotrophic so that the trait of rapid growth in low temperature might not be demanded by deep-lake decomposers. The stratified distribution of community composition and decomposer activity in Lake Khuvsgul implies that ecological functions of bacterial community in lakes of cold region are sharply divided by water column stratification.

Climate and land use changes have impact on availability water resource by hydrologic cycle change. The purpose of this study is to evaluate the hydrologic behavior by the future potential climate and land use changes in Anseongcheon watershed ($371.1km^2$) using SWAT model. For climate change scenario, the HadGEM-RA (the Hadley Centre Global Environment Model version 3-Regional Atmosphere model) RCP (Representative Concentration Pathway) 4.5 and 8.5 emission scenarios from Korea Meteorological Administration (KMA) were used. The mean temperature increased up to $4.2^{\circ}C$ and the precipitation showed maximum 21.2% increase for 2080s RCP 8.5 scenario comparing with the baseline (1990-2010). For the land use change scenario, the Conservation of Land Use its Effects at Small regional extent (CLUE-s) model was applied for 3 scenarios (logarithmic, linear, exponential) according to urban growth. The 2100 urban area of the watershed was predicted by 9.4%, 20.7%, and 35% respectively for each scenario. As the climate change impact, the evapotranspiration (ET) and streamflow (ST) showed maximum change of 20.6% in 2080s RCP 8.5 and 25.7% in 2080s RCP 4.5 respectively. As the land use change impact, the ET and ST showed maximum change of 3.7% in 2080s logarithmic and 2.9% in 2080s linear urban growth respectively. By the both climate and land use change impacts, the ET and ST changed 19.2% in 2040s RCP 8.5 and exponential scenarios and 36.1% in 2080s RCP 4.5 and linear scenarios respectively. The results of the research are expected to understand the changing water resources of watershed quantitatively by hydrological environment condition change in the future.
In order to observe the lasting effect of NK-compound mineral fertilizer and organic compound fertilizer including Myweon organic liquid fertilizer, a pot experiment was conducted with rice (Oriza Sativa) variety: Nong Back. These fertilizers were applied as basal and the same amount of urea form of nitrogen was top dressed about a month after transplanting, July seventh, 1977. Results obtained are as follows; 1. The lasting effect of various fertilizers were laid in following decreasing order; Myweon liquid < Organic compound fertilizer (Myweon coop. made) > NK-compound fertilizer (Chosun coop. made) Single fertilizer. It was considered that organic matter served as microbial feed and lead a temporary fixation of available plant nutrients in the soil, and the reduced surface area of the compound fertilizers slowed down the availability of the fertilizers. 2. The fertilizer showing greater lasting effect produced more panicles per hill and less grains per panicle than the fertilizers showing less lasting effect, and brought low maturity, which resulted in low paddy yield the paddy producing efficiency of nitrogen absorbed by straw was also low in the former fertilizers. Such advanced effect of the former fertilizer was considered to be related with the variety of early maturity and unseasonable topdressing of fertilizer which made at the maximum tillering stage. 3. For the production of Japonica type paddy with heavy fertilization which may required to depress the early growth a little and promote the late growth, it might be necessary to develop slow releasing fertilizers such as single fertilizer formulated to a large grains or compound fertilizer containing organic matter. 4. If the nitrogen content of paddy, Nong Back, far excess 0.64 or 0.65% and reaches 0.68% or above, the yield of the variety seemed to be decreased remarkbly through the low maturity rate and thousand grain weight.
The Codonopsis pilosula is traditional oriental herb associated with immune-modulatory functions and has anti-inflammatory properties and antioxidative activity. The present study was conducted to investigate the effects of dietary supplementation of Codonopsis pilosula extract powder (CEP) on the immunity and productivity in sows and piglets. A total of 20 pregnant sows were randomly assigned to two dietary treatments, which was given a corn-soybean meal diet with 0% and 0.5% CEP from 40 days prepartum to weaning (25 days postpartum). The immune system of piglets was expected to build up, because the feeding of CEP might increase the colostrum contents in sows. We also predicted that the growth performance of piglets also could be increased by some compensation effects due to the increased immunity of piglets. In results, immunities of sows and piglets as well as growth performance of piglets were not affected by the supplementation of CEP. The correlation between the colostrum and the serum on the IgG contents tended to have a positive correlation, although there was no significant (coefficient, 0.435; P=0.102). This result suggests that the supplementation of CEP may increase IgG contents in sows and piglets and contribute to improve immunity of piglets. In conclusion, it is thought that the physiologically active substances of CEP did not influence to the immune synergic effects in vivo of sows. Thus, the metabolic and action mechanism of the physiologically active substances of CEP should be explored to evaluate the availability of CEP as a feed additive for the immunomodulator.
The effect of water activity ($a_w,\;0.9{\sim}0.995$) and temperature ($18{\sim}30^{\circ}$C) on in vitro growth and interactions between ochratoxin-producing Aspergillus ochraceus and six other fungi (Alternaria alternata, Aspergillus candidus, A. flavus, A. niger, Eurotium amstelodami, E. rubrum) isolated from maize grain were investigated. A. ochraceus and each six other species were paired and their interactions given a numerical score to obtain an index of dominance ($I_D$) for each species. Generally A. ochraceus was very competitive and dominant against other fungi. It was, however, dominanted by Alternaria alternata and A. niger at high $a_w\;(0.995\;a_w)$, and mutually antagonistic when paired with E. amstelodami and E. rubrum at low $a_w\;(0.9\;a_w)$. The growth rates of each species were also calculated under the same range of environmental conditions. They were markedly influenced by aw and temperature. At high temperature ($30^{\circ}C$), A. ochraceus grew most rapidly under slightly drier conditions ($0.95\;a_w$), while A. alternata, A. flavus and A. niger did at high water availability level ($0.995\;a_w$). At $18^{\circ}C\;and\;25^{\circ}C$, and high $a_w$ level ($0.995\;a_w$), A. alternata grew fastest, while A. candidus, E. amstelodami and E. rubrum grew very slowly. Using Biolog plates the effect of $a_w$ and temperature on utilization patterns of carbon sources in maize was evaluated. The niche overlap index (NOI) relative to A. ochraceus was determined and compared with that of each interacting species. Under high water available condition ($0.995\;a_w$). the NOI of A. ochraceus was often >0.9, indicative of the coexistence with other interacting species. However, against E. amstelodami and E. rubrum at $18^{\circ}C$, the species had NOI <0.8, indicative of occupation of different niches. At low $a_w\;(0.95\;a_w)$, NOI for A. ochraceus was <0.8 when paired with A. alternata and A. niger also suggested the occupation of different niches.
