Comparison of Acid and Bile Tolerances, Cholesterol Assimilation, and CLA Production in Probiotic Lactobacillus acidophilus Strains

  • This study aimed to compare the probiotic characteristics of twelve strains of Lactobacillus acidophilus including cholesterol assimilation and conjugated linoleic acid (CLA) production. Cholesterol assimilation exhibited some variation among L. acidophilus strains, which could be classified into three groups based on their assimilation levels (p<0.05). The high cholesterol assimilation group exhibited a significantly higher tolerance to 0.3 and 0.5% bile acid than the low cholesterol assimilation group (p<0.05). Cholesterol assimilation showed positive correlation with 0.5% bile tolerance, and a negative correlation with acid tolerance (p<0.01). Glycocholate deconjugation activity showed no relationship with cholesterol assimilation, whereas taurocholate deconjugation activity was shown to have negative correlation with cholesterol assimilation (p<0.05). CLA production by L. acidophilus strains exhibited a wide variation, ranging from 2.69 to 5.04 mg/g fat. CLA production of L. acidophilus GP1B was the highest among the tested strains, but there was no evidence for differences in CLA production in strain specificity. Based on these results, the cholesterol assimilation of L. acidophilus strains may not be related to deconjugation activity, but may in-fact be attributed to their bile-tolerance.

  • Crassulacean acid metaholism (CAM) was investigated in leaves and stems of the succulent $C_4$dicot Portulaca oleracea L. Under 14-hour days, stem tissues showed much greater fluctuation of acidity than leaf tissues. But leaf and stem tissues showed almost same CAM-like pattern of acid fluctuation under 8-hour days. Stem tissues of R oleracea grown under the naturai environment showed high CAM activity, but no CAM activity was seen in leaves of those plants. In the naturally growing plants, the rapid acidification was seen in intact stems at dawn, but defoliated stems showed only a gradual increase. RuBP carlboxylase activity was very high at 2:00 P.M. in both leaves and stems. However, its activity at 1:00 A.M. and 5:30 AM. was hardly detected. particularly, activity of PEP carboxylase in leaves was very high in the early morning, though that in stem tissues was little. These results indicate that $CO_2$ passed through open stomata at dawn may be assimilated by PEP carboxylase in leaves, and then $C_4$ products move to stems. The levels of nitrate concentration and of nitrate reductase were higher in stems than in leaves. The levels were also higher in the light than in the dark. It would be suggested that considerable amount of nitrate absorbed from roots ho assimilated in stems, and nitrate transferred to leaves via stem tissues be reduced there. Molybdate Alters Sulfate Assimilation and Induces Oxidative Stress in White Clover (Trifolium repens L.)

  • Molybdenum (Mo) in rhizosphere influences sulfate assimilation as well as a number of other physiological aspects. In this study, the activity of key enzymes in sulfate assimilatory pathways, such as ATP sulfurylase (ATPs), adenosine 5'-phosphosulphate reductase (APR), as well as the responses of reactive oxygen species (ROS), were analyzed to elucidate the metabolic and physiological effects of external Mo supply to detached leaves of Trifolium repens L. Mo supply with a range from 1 mM to 40 mM depressed the activity of ATPs throughout the entire time course. In the leaves exposed to 1 mM Mo, a continuous decrease in the activity of ATPs was confirmed by Native-PAGE. The APR activity was also declined by Mo treatment. The accumulation of $H_2O_2$ and ${O_2}^{{\cdot}-}$ were not significant up to 10 mM Mo, whereas a remarked accumulation was detected under 40 mM Mo supply. The data suggest that an external supply of Mo has an inhibitory effect on sulfate assimilation, and induces oxidative stress only at an extremely high concentration.

  • Recently, many companies are interested in adopting cloud computing as their IT strategy. However, no distinct results have appeared in the substantial implementation of this technology. The main reason for such result is from the absence of research models leading to high impact studies on cloud computing. Thus, this study attempts to find a possible answer for the following research question : what factors influence an organizational assimilation of cloud computing? This study investigates Technology-Push (TP)/Need-Pull (NP) theory as a main factor affecting cloud computing assimilation. Also, the study examines the moderating role of organizational readiness. TP includes of perceived benefits, vendor pressure, cost savings, and IT activity intensity while NP includes competitor orientation, information technology policy, technological turbulence, and performance gaps. In addition, organizational readiness suggests two variables, financial resources and technological knowledge. Result from 217 adopting organizations showed that all of these factors with exception of competitor orientation and vendor pressure, have statistically significant impact on assimilation of cloud computing. The implications of the findings propose a theoretical framework for the foundation of studies on cloud computing assimilation, which can server as important practical guidelines for technology development.

생물 과제의 자기조절 활동에서 나타나는 중등학생의 연령별 두뇌 활성 -fNIRS 연구 (Age-Specific Brain Activation in Secondary School Students' Self-Regulating Activities on Biological Tasks -fNIRS Study)

  • 이 연구의 목적은 중등학생의 자기조절 과정에서 동화, 갈등, 조절의 세부 과정에 대한 뇌 활성을 비교하고 분석하는 것이다. 자기조절 과제는 생물학적 계통발생 과제로 제시되었고, 뇌활성은 fNIRS로 측정 및 분석되었다. 그 결과 동화 과정과 비교하여 갈등 과정에서 좌측 DLPFC, OFC, FP 영역에서 유의미한 활성이 발견되었고, 조절 과정에서는 DLPFC, VLPFC에서 유의미한 활성화가 발견되었다. 중등학생의 학년이 높아질수록 갈등 과정에서도 DLPFC가 증가하고 동화 과정에서도 VLPFC가 증가한다. 또한, 갈등과 조절 과정을 비교한 결과, 7학년 학생들은 오른쪽 VLPFC에서 유의미한 뇌 활동을 보였고, 9학년 학생들은 조절 과정에서 왼쪽 FP와 DLPFC 영역에서 유의미한 뇌 활동을 보였지만, 11학년 학생들은 이 과정에서 유의미한 뇌 활동을 보이지 않았다. 이러한 결과는 신경학적 연구 방법이 인지 활동과 강의실 교육 상황과 관련된 교육 연구에 적용될 수 있음을 보여준다.

Assimilation of Peptides and Amino Acids and Dissimilation of Lactate During Submerged Pure Cultures of Penicillium camembertii and Geotrichum candidum

  • The behavior of Penicillium camembertii and Geotrichum candidum growing in submerged pure cultures on simple (glutamate) or complex (peptones) substrates as nitrogen and carbon sources and lactate as a second carbon source was examined. Similar to the behavior previously recorded on a simple substrate (glutamate), a clear differentiation between the carbon source and the energy source was also shown on peptones and lactate during P. camembertii growth, since throughout growth, lactate was only dissimilated, viz., used for energy supply by oxidation into $CO_2$, whereas peptides and amino acids from peptones were used for carbon (and nitrogen) assimilation. Because of its deaminating activity, G candidum preferred peptides and amino acids to lactate as energy sources, in addition to being assimilated as carbon and nitrogen sources. From this, on peptones and lactate, G candidum grew faster than P. camembertii (0.19 and 0.08 g/l/h, respectively) by assimilating the most readily utilizable peptides and amino acids; however, owing to its lower proteolytic activity, the maximum biomass was lower than that of P. camembertii (3.7 and 5.5 g/l, respectively), for which continuous proteolysis and assimilation of peptides were shown.

  • Twelve strains of Lactobacillus acidophilus isolated from feces of human or animal sources were tested for probiotic properties such as cholesterol assimilation, bile and acid tolerances, and CLA production. Although the cultures showed some variation with respect to each test, the 12 strains could be classified into 3 groups based on their ability to assimilate cholesterol. The cholesterol assimilation showed positive correlation with bile tolerance and negative correlation with acid tolerance. The cholesterol assimilation of L. acidophilus strains may not be related to the deconjugation activity, but may in fact be attributed to its bile tolerance. CLA production by lactic acid bacteria (LAB) exhibited a wide variation that ranged from 2.69 to 7.64 mg/g fat. CLA production of Bifidobacterium longum ATCC 15707 was the highest among the LAB tested, but there was no evidence for differences in CLA production between genus and species.

Plasmid-Mediated Aniline Assimilation by Pseudomonas sp. B10

  • An aniline-utilizing microorganism identified as a species of Pseudomonas was isolated from soil contaminated highly with aniline and urea-herbicide. This strain was able to utilize aniline as the sole source of carbon and energy, and was shown to harbor a single large plasmid mediating the aniline assimilation. Subsequent plasmid-curing of this bacterium resulted in the abolishment of the aniline utilizing phenotype and the loss of catechol-C2,3O-oxygenase. The reestablishment of the plasmid, denoted pB10, in cured Pseudomonas sp. via filter surface mating, resulted in restoration of the aniline assimilation abilities and enzyme activity.

Characteristics of Peptide Assimilation by Helicobacter pylori: Evidence for Involvement of Cell Surface Peptidase

  • Peptide assimilation by Helicobacter pylori was investigated using L-phenylalanyl-3-thia-phenylalanine (PSP) as a detector peptide; the release of thiophenol upon enzymatic hydrolysis of PSP was spectrophotometrically detected with the aid of 5,5'-dithiobis[2-nitrobenzoic acid] (DTNB). By adding PSP to whole-cell suspension, thiophenol was produced progressively, resembling that found in Esherichia coli or Staphylococcus aureus. Interestingly, the rate of thiophenol production by H pylori in particular was markedly reduced when cells were pretreated with trypsin, indicating surface exhibition of peptidase. According to the competitive spectrophotometry using alanyl-peptides, H pylori did not appear to assimilate PSP through the peptide transport system. No discernible PSP assimilation could be ascertained in H pylori cells, unless provided with some additives necessary for peptidase activity, such as $Ni^{2+}\;or\;Mg^{2+}$ and an appropriate concentration of potassium or ammonium salts. These observations strongly suggest that, regardless of a presumptive peptide transport system, peptide assimilation of H. plori appears to be highly dependent upon milieu conditions, due to unique peptidase exhibition on the cell surface.

Isolation of the Cholesterol-Assimilating Strain Pediococcus acidilactici LRCC5307 and Production of Low-Cholesterol Butter

  • The objective of the present study was to evaluate the cholesterol-assimilation ability of lactic acid bacteria (LAB), which were isolated from kimchi, a Korean traditional fermented cabbage. The isolated strain, using modified MRS medium, showed 30.5% cholesterol assimilation activity and was named Pediococcus acidilactici LRCC5307. Types and concentrations of bile were investigated for their effects on increasing the cholesterol assimilation ability of the LRCC5307 strain, a 74.5% decrease in cholesterol was observed when 0.2% bile salts were added. In addition, the manufacture of low-cholesterol butter using LRCC5307 was examined. After fermentation, LRCC5307 with butter showed 8.74 Log CFU/g viable cells, pH 5.43, and a 11% decrease in cholesterol. These results suggest that LRCC5307 could help in the production of healthier butter by decreasing cholesterol and including living LAB.
