Saccharomyces cerevisiae TSA1의 보존된 아스파트산 잔기 및 세린 잔기의 변이가 과산화효소 활성 및 샤페론 활성에 미치는 영향 (Effects of Mutation at Two Conserved Aspartate Residues and a Serine Residue on Functions of Yeast TSA 1)

  • 퍼옥시레독신은 티오레독신, 티오레독신 환원효소, NADPH로 이루어진 티오레독신 시스템의 환원력을 이용하여 과산화물을 제거하는 티오레독신 과산화효소 활성과 다른 단백질의 열변성에 의한 응집을 막아주는 샤페론 활성을 갖는 효소이다. 정형 2-Cys Prx군에 속하는 퍼옥시레독신 참고서열 1,024개 중 부분적인 서열 등을 제외한 967개 서열을 정렬하였을 때 75번과 103번 아스파트산 잔기는 99% 보존되었고, 73번 세린 잔기는 97% 보존되었음에도 불구하고 잘 보존된 아스파트산 잔기와 세린 잔기에 대해 알려지지 않았다. 이 잔기가 TSA1의 두가지 효소 활성에 미치는 영향을 알아보기 위해 재조합 단백질을 이용하여 활성도를 알아보았다. in vitro 실험을 통하여 잘 보존된 잔기인 103번 아스파트산은 75번 아스파트산보다 티오레독신 퍼옥시레독신 활성 및 분자 샤페론 활성에 더 영향을 미치고, 103번의 음전하는 분자 샤페론 활성에 중요한 역할을 하며 과산화효소활성에는 75번과 103번의 음전하가 관여함을 알 수 있었다. 또한 73의 세린 잔기 역시 과산화효소에 영향을 미치는 잔기임을 알 수 있었다. 최근 출아 효모 퍼옥시레독신인 TSA2의 79번과 109번의 세린 잔기를 시스테인 잔기로 변이시킨 경우 두 변이 단백질 모두 과산화효소 활성과 샤페론 활성이 증가되었는데 이는 ${\beta}$-sheet 구조의 증가와 관련되는 것으로 보고하였다[28]. 이들 두 세린 잔기는 TSA1 구조에 의하면 모두 ${\alpha}$-나선 구조에 위치하였다. 반면에 73번의 세린 잔기는 ${\beta}$-sheet의 C-말단에 위치하는 잔기로 과산화효소 활성에 대한 영향이 다르게 나타나는 것으로 추정된다. 추후 생체 내 실험을 통하여 아스파트산 잔기의 변이가 과산화물 저항성이 미치는 영향 및 열 저항성(thermal stress)에 미치는 역할을 살펴볼 필요가 있다. 또한 아스파트산 잔기와 과산화물과의 반응 및 분자 샤페론과의 반응에 장애가 되는 요인이 무엇인지에 대한 추가 연구가 필요할 것이다.

Neuroprotective Effect of Acanthopanax sessiliflorus against Toxicity Induced by N-Methyl-D-Aspartate in Rat Organotypic Hippocampal Slice Culture

  • We investigated that water extract of Acanthopanax sessiliflorus roots rescued the N-methyl-D-aspartate (NMDA), agonist of glutamate receptor, -induced toxicity in rat organotypic hippocampal slice culture. When the cell death in NMDA only-treated hippocampal slices was set 100%, A. sessiliflorus decreased the cell death to 75.4, 51.6, 48.9, and 40.6% at 1, 10, 50, and $100\;{\mu}g/ml$ treatment, respectively. On the basis of these results, the water extract of A. sessiliflorus roots may be a preventive agent against NMDA-induced cytotoxicity.

Study of Alanine-73 and Aspartate-9 of HLA-C Locus in Saudi Psoriasis Patients, Using Sequence-specific Primers (PCR-SSP)

  • Alanine at residue 73 (Ala-73) and aspartate at residue 9 (Asp-9) are characteristic to both Cw6 and Cw7 alleles of HLA-C gene and have been suggested as possible markers for psoriasis vulgaris (PsV). However, the results from various ethnic groups/populations are contradictory and inconclusive. In this study, an attempt has been made to examine the association between HLA-C (Ala-73 and Asp-9) and susceptibility to PsV among Saudi patients. Genomic DNA was extracted from 25 Saudi PsV patients and 75 control subjects. Polymerase chain reaction (PCR) was performed to amplify HLA-C sequences using earlier reported primers, C133P and C243PR. Sequence-specific primers were used to specifically detect nucleotide coding for Ala-73 and Asp-9 in all the subjects. The results showed significantly higher frequency of Asp-9 (84.0% versus 61.3%) in PsV patients as compared to controls (p < 0.05, 2-tailed Fisher's exact test). The frequencies of Ala-73 among PsV patients (92%) and controls (88%) did not differ significantly.

Rat Liver $\beta$-Glucuronidase; Its Purification and Inhibition Studies

  • ${\beta}$-Glucuronidase (EC 3.2.1.31) which hydrolizes D-glucuronate from ${\beta}$-D-glucuronide was purified from rat liver, using ammonium sulfate fractionation, DEAE-cellulose chromatography, Concanavalin-A Sepharose 4B chromatography and gel filtration on Sephadex G-200. This enzyme has the molecular weight of 280,000 daltons by gel filtration and 75,000 daltons by SDS-polyacrylamide gel electrophoresis. As its funtion is reverse of detoxification in the liver, the inhibition of the enzyme was tested with extracts of several food products and medicinal herbs, some are known as anti-cancer agents. Among them, Panax ginseng and Cortnellus shiiake inhibited the enzyme competitively and the $K_1$ values were $9.22 {\times}\;10^{-2}$ and 0.102 mg/ml, respectively. These inhibitors strongly bound to DEAE-cellulose. The negatively charged amino acids, L-aspartate and L-glutamate, inhibited the enzyme, and $K_1$ value of L-aspartate was 0.80 mM. The interaction between ${\beta}$-glucuronidase and p-nitrophenyl-${\beta}$-D-glucuronide was found to involve ionic forces by the effect of ionic strength on the kinetic constant, Vmax/Km. It was inferred from these findings that cationic group at the active center of the enzyme is probably involved in attacking the substrate.

Effect of Feeding High Glucosinolate Rapeseed Meal to Laying Japanese Quail

  • The laying performance of Japanese quails fed graded levels of high glucosinolate ($92.5{\mu}mole/g$) rapeseed meal (RSM) was assessed. One hundred and twenty Japanese quails aged 1 day-old were assigned at random to four dietary treatments consisting of 0, 50, 75 or 100 g/kg RSM in the diet replacing part of the soybean meal and de-oiled rice bran in a standard quail ration. 12 female representative quails from each diet were selected at random and housed in individual cages from 7-20 wk of age. The egg production, feed intake and FCR was comparable among the different dietary groups. The egg quality characteristics, organoleptic evaluation of boiled eggs as well as the haematological (haemoglobin, total erythrocyte count, total leucocyte count) and biochemical (glucose, protein, cholesterol, aspartate amino transferase, alanine amino transferase and alkaline phosphatase) constituents did not differ significantly among the groups. The gross and histopathological studies of vital organs did not reveal any appreciable changes. The feed cost was reduced by the incorporation of RSM in the diet, but only the production cost of quails fed the 75 g/kg RSM was lower in comparison to other groups. In the present study, the laying potential of Japanese quail was well-maintained up to the 100g/kg dietary level of rapeseed meal.

Predictors of nonresponse to intravenous immunoglobulin therapy in Kawasaki disease

  • Purpose: It has been reported that 10% to 20% of children with Kawasaki disease (KD) will not respond to intravenous immunoglobulin (IVIG) treatment. In this study, we aimed to identify useful predictors of therapeutic failure in children with KD. Methods: We examined 309 children diagnosed with KD at the Kyungpook National University Hospital and the Inje University Busan Paik Hospital between January 2005 and June 2011. We retrospectively reviewed their medical records and analyzed multiple parameters in responders and nonresponders to IVIG. Results: Among the 309 children, 30 (9.7%) did not respond to IVIG. They had significantly higher proportion of neutrophils, and higher levels of aspartate aminotransferase, alanine aminotransferase (ALT), total bilirubin, and N-terminal fragment of B-type natriuretic peptide than did responders. IVIG-nonresponders had a significantly longer duration of hospitalization, and more frequently experienced coronary artery lesion, and sterile pyuria. No differences in the duration of fever at initial treatment or, clinical features were noted. Conclusion: Two independent predictors (ALT${\geq}$84 IU/L, total bilirubin${\geq}$0.9 mg/dL) for nonresponse were confirmed through multivariate logistic regression analysis. Thus elevated ALT and total bilirubin levels might be useful in predicting nonresponse to IVIG therapy in children with KD.

A Proposal of Dietary Supplement from Choto-san, a Kampo Medicine

  • Therapeutic effect of a Kampo medicine, Choto-san, in patients with vascular dementia was demonstrated by a double-blind and placebo-controlled clinical trial. To clarify the therapeutic efficacy of Choto-san, anti-ischemic effect in mice, hypotensive effect in spontaneously hypertensive rats (SHR), anti-oxidative effects in vitro, and N-methyl-D-aspartate (NMDA) receptor-blocking activity using Xenopus oocytes were studied. (1) Pretreatment with Choto-san (0.75-6.O g/kg, P.O.) or a component herb Chotoko (Uncaria genus: 75 - 600 mg/kg, P.O.) prevented ischemia-induced impairment of spatial learning behaviour in mice. Indole alkaloids- and phenolic fractions extracted from Chotoko also improved significantly the learning deficit. (2) Subchronic administration of Choto-san (0.5 g/kg, p.o.) caused a significant hypotensive effects in SHR. (3) Choto-san, Chotoko, and the phenolic constituent, (-) epicatechin, significantly protected the NG108-15 cell injury induced by $H_20_2$ exposure in vitro and also inhibited lipid peroxidation in the brain homogenate. (4) Indole alkaloids, rhynchophylline and isorhynchophylline (1-100 uM), reversibly reduced NMDA-induced current in the receptor-expressed Xenopus oocytes. These results suggest that anti-vascular dementia effects of Choto-san are mainly due to the effect of Chotoko. From these results, it is possible to make a novel dietary supplement through several extraction steps from Chotoko.

비파식초의 사염화탄소에 의한 간독성 보호 효과 (Protective Effect of Eriobotrya japonica Lindl. on Hepatotoxicity by Carbon Tetrachloride)

  • 비파 식초가 사염화탄소에 의한 간독성에 대한 보호효과를 흰 쥐를 이용하여 확인하고자 연구하였다. 4주간 SD rat 암컷을 정상군, 음성대조군, 양성대조군, 비파 식초군으로 나누어 샘플을 처리하였고, 29일째 되는 날 사염화탄소를 처리하였다. 혈청 AST 및 ALT 효소 활성 수치 변화에서 비파 식초가 사염화탄소를 단독으로 투여한 군보다 유의한 수치로 각각 49.77%, 75.88%로 감소하였다(p < 0.001). 간 조직에서는 CAT 및 SOD 효소 활성은 비파 식초군이 사염화탄소를 단독으로 투여한 음성대조군보다 유의한 차이로 높은 활성 수치를 보였다. 조직학적 관찰에서는 비파식초군이 사염화탄소를 단독으로 투여한 음성대조군에 비해 지방변성이 적게 분포하고, 조직의 섬유화가 진행되지 않았으며, 정상군과 비슷한 간세포의 구조가 관찰 되었다. 이와 같은 결과를 보아 비파 식초는 항산화 효소의 증가로 활성산소를 제거하여 간 손상을 억제할 뿐아니라 혈청의 AST 및 ALT 효소 활성을 감소시켜 간 보호 효과가 있는 것으로 건강기능식품으로써 긍정적인 가능성이 있다고 사료된다.

Effects of Intraperitoneal N-methyl-D-aspartate (NMDA) Administration on Nociceptive/Repetitive Behaviors in Juvenile Mice

  • Dysregulation of excitatory neurotransmission has been implicated in the pathogenesis of neuropsychiatric disorders. Pharmacological inhibition of N-methyl-D-aspartate (NMDA) receptors is widely used to model neurobehavioral pathologies and underlying mechanisms. There is ample evidence that overstimulation of NMDA-dependent neurotransmission may induce neurobehavioral abnormalities, such as repetitive behaviors and hypersensitization to nociception and cognitive disruption, pharmacological modeling using NMDA has been limited due to the induction of neurotoxicity and blood brain barrier breakdown, especially in young animals. In this study, we examined the effects of intraperitoneal NMDA-administration on nociceptive and repetitive behaviors in ICR mice. Intraperitoneal injection of NMDA induced repetitive grooming and tail biting/licking behaviors in a dose- and age-dependent manner. Nociceptive and repetitive behaviors were more prominent in juvenile mice than adult mice. We did not observe extensive blood brain barrier breakdown or neuronal cell death after peritoneal injection of NMDA, indicating limited neurotoxic effects despite a significant increase in NMDA concentration in the cerebrospinal fluid. These findings suggest that the observed behavioral changes were not mediated by general NMDA toxicity. In the hot plate test, we found that the latency of paw licking and jumping decreased in the NMDA-exposed mice especially in the 75 mg/kg group, suggesting increased nociceptive sensitivity in NMDA-treated animals. Repetitive behaviors and increased pain sensitivity are often comorbid in psychiatric disorders (e.g., autism spectrum disorder). Therefore, the behavioral characteristics of intraperitoneal NMDA-administered mice described herein may be valuable for studying the mechanisms underlying relevant disorders and screening candidate therapeutic molecules.

알콜유도 지질과산화에 미치는 어성초 영향 (Effect of Houttuynia cordata on EtOH-induced Lipidperoxidation in Rat)

  • The antilipidperoxidative and hepatopreventive effects of Houttuynia cordata extract(HCE) were investigated at the level of liver homogenates and serum of SD rats intoxicated EtOH(3g/kg for 3days). We measured MDA(malondialdehyde) in the live. homogenate, AST(1-aspartate-2-oxoglutarate aminotransferase) and ALT( 1-alanine-2-oxoglutarate aminotransferse) in the serum. The analysis of the measurement indicated that HCE reduced MDA, ALT and AST significantly and their reduction was in relation to dose dependence. In rat liver homogenate intoxicated with EtOH, HCE treatment group inhibited lipidperoxidation by 50-75%. In rat serum intoxicated EtOH, HCE inhibited AST and ALT by 40-70%.
