Verification of aecial host ranges of four Gymnosporangium species based on artificial inoculation.

  • Aecial host ranges of four Gymnosporangium species causing cedar-apple rust diseases, G. asiaticum, G. cornutum, 5. japonicum and G. yamadae, were investigated through artificial inoculation. Thirteen species of nine genera among Rosaceous plants, which have been reported as social hosts in Korea, were inoculated with fresh teliospores spores in early days of May of 2000 and of 2001, respectively. In the results, we re-confirmed that there was highly specific relationship between the rust species and aecial hosts and report new aecial hosts of four Gymnosporangium species. Teliospores of G. cornutum collected from Juniperus rigida successively produced spermogonia and aecia only on Sorbus alntifolia, the first report on host alteration of G. cornutum in Korea. Positive responses by teliospores of G. japonicum from J. chinenis of Suwon and from J. chinenis var. horizontalis of Jeju island were obtained only on P. villosa. Crataegus pinnatifida was confirmed as a new aecial host of G. viatium. Until this time, G. ymadae was believed to have Malus as the aecial host. However, teliospores of G. yamadae collected from J. chinensis var. kaizuka successively formed spermogonia and aecia on the leaves of Chaenomeles lagenaria, C. sinensis, Pyrus pyrtifolia var, culta, P. ussuriensis, Malus pumila and M. sileboldii. The date for maturation of spermogonia and aecia, and symptom development varied according to the rust fungi and aecial host plants, respectively.

  • 멕시코와 네팔에서 도입된 고추 유전자원 50점과 대조품종 등을 포함한 총 130여점에 대하여 풋마름병과 역병에 대한 저항성을 검정하였다. 풋마름병에는 KC897, KC939, KC936가 KC126, KC350, KC351, KC353에 더하여 새로운 저항성 재료로 나타났다. 역병에는 저항성이 발견되지 않았다.

Occurrence of Dry Rot on Cymbidium Orchids Caused by Fusarium spry. in Korea

  • Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.

Occurrence of Sclerotinia Rot on Composite Vegetable Crops and the Causal Sclerotinia spp.

  • Composite vegetable crops grown in greenhouses and open fields in Korea were surveyed from 1995 to 1999. Occurrence of Sclerotinia rot was observed in 123 of 277 lettuce fields, in 11 of 18 head lettuce fields, in 12 of 14 endive fields, and in 4 of 38 garland chrysanthemum fields surveyed during the growing seasons. The disease most severely occurred up to 80% on lettuce. Incidence of the disease was as high as 20% at its maximum on endive and garland chrysanthemum but relatively low on head lettuce. Symptoms of the disease developed on leaves of all the composites, crowns of lettuce and head lettuce, and stems of garland chrysanthemum. Out of 240 isolates of Sclerotinia species obtained from the diseased composites, 169 isolates were identified as Sclerotinia sclerotiorum, and the others as S. minor based on their morphological and cultural characteristics. S. sclerotiorum was isolated from all the composites, and S. minor only from lettuce and endive. Eight isolates of S. sclerotiorum and four isolates of S. minor were tested for their pathogenicity to the composites by artificial inoculation. All the isolates of the two Sclerotinia spp. induced rot symptoms on the plants of the composites by artificial inoculation, which were similar to those observed in the fields. The pathogenicity tests revealed that there is no significant difference in virulence of the isolates to the composites and in susceptibility of the composites to the isolates.

The Impact of Environmental and Host Specificity in Seed Germination and Survival of Korean Mistletoe [Viscum album var. coloratum (Kom.) Ohwi]

  • Humankind has been searching for medicinal materials from various plant sources in an attempt to treat disease. Mistletoe is one indubitable plant source for these materials due to its effectiveness in treating various diseases, but it has almost disappeared from the mountainous areas of Korea due to excessive harvesting. In this study, in order to select host tree species for Korean mistletoe [Viscum album var. coloratum (Kom.) Ohwi] by seed inoculation and to clarify the effect of host specificity among various tree species were conducted for the purpose of gaining basic information for the artificial cultivation of Korean mistletoe. Almost all the seeds of Korean mistletoe germinated in vitro at the temperature of 15℃. Among host trees used in this study, Prunus mume showed the highest parasitic affinity with inoculated Korean mistletoe, compared with any other host plants. However, treatment of hormones could not increase the low survival rate of Korean mistletoe on the host trees.

Foot Rot of Bok Choy and Kale Caused by Rhizoctonia solani AG-2-1 in Korea

  • Foot rot symptoms were occasionally observed on young bok choy (Brassica rapa subsp. chinensis) and kale (B. oleracea var. viridis) plants grown in vinyl greenhouses located in Icheon and Yangpyeong, Gyeonggi Province, Korea. These observations were made during disease surveys in April 2020. The incidence of diseased plants in the vinyl greenhouses investigated was 0.5-1.0% in bok choy and 0.5-5.0% in kale. Five isolates of Rhizoctonia sp. were obtained from diseased roots of bok choy and three isolates of Rhizoctonia sp. were taken from diseased stems of kale. All the Rhizoctonia sp. isolates were identified as Rhizoctonia solani AG-2-1 based on the morphological characteristics and anastomosis test. Three isolates each of R. solani AG-2-1 from bok choy and kale were tested for pathogenicity in their host plants by artificial inoculation. The tested isolates induced foot rot symptoms on the inoculated bok choy and kale plants. The symptoms on the bok choy and kale, induced by the artificial inoculation, were similar to those observed on plants from the vinyl greenhouses that were investigated. This is the first report of R. solani AG-2-1 causing foot rot in bok choy and kale in Korea.

Damages caused by infection with viruses in cut-flower production of Lilium $\chi$. fomolongi

  • Viral disease symptoms were investigated in the field grown Longiflorum hybrid cultivars, and the damages caused by infection with Lily mottle virus (LMoV) and Cucumber mosaic virus (CMV) were assessed by comparing growth of plants produced from seeds of Longiflorum hybrid cultivar both infected by artificial inoculation and free from infection with theses viruses. Dominant symptom caused by spotaneous infection with LMoV and CMV in the field was mottle combined with chlorotic stripe on leaves. LMoV developed brownish necrotic lesion on floral leaves. The incidence of viral disease by mixed infection with LMoV, CMV or Lily symptomless virus (LSV) in the filed grown Longiflorum hybrid cultivar, cultivated for more than 6 years, was 80 to 84 percent. In comparison with virus-free plants, plants doubly infected with CMV and LMoV by artificial inoculation decreased stem length by 14 percent and fresh weight by 38 percent. In conclusion, flower quality and the stem length of Longiflorum hybrid cultivar were affected by LMoV and CMV infection.

Comparison of Resistance Level to Cotton leaf curl virus(CLCuV) Among Newly Developed Cotton Mutants and Commercial Cultivars

  • Four newly developed cotton mutants (M-111, M-7662, M-358 and M-218) were compared for their resistance against Cotton leaf curl virus(CLCuV) together with commercial resistant (CIM-443, CIM-482, CIM-473, FH-900 and FH-901) and susceptible (5-12) varieties by artificial inoculation through grafting and under natural field conditions. Infectivity and success of grafting were 100% in all cases. None of the grafted plants were found immune or asymptomatic. All the grafted mutants and most of their single plant progeny rows (SPPRs) showed highly resistant responses as the symptoms displayed by these mutants were milder than the commercial cultivars. Grafted mutants also had delayed disease reactions as they took more time (25-30 days) to produce disease symptoms, as compared with resistant commercial varieties that produced disease 18-22 days after inoculation. Growth of the grafted SPPRs of tested mutants was normal, which is an indication that there will be no production losses. Observations under natural infestation of whitefly showed that two SPPRs of M-ll/CE and M-7662-1/2 and one resistant variety CIM-443 exhibited slight incidence of disease, while one SPPR of M-l1/59 and S-12 were moderately susceptible and highly susceptible with 21% and 97.l% disease incidence, respectively. This study also showed that plants displaying more disease symptoms through grafting were easily infected under natural conditions. These results suggest that preference should be given to those plants that exhibited highly resistant responses after artificial inoculation.

균근연구(菌根硏究)의 농림업(農林業)에의 응용(應用) (Application of Mycorrhizal Research to Agriculture and Forestry)

  • Recently mycorrhizal research has been one of the most fast-growing research areas in modern plant science and microbiology. The application potential of mycorrhizal techniques to agriculture and forestry is enormous in view of the ubiquitous nature of mycorrhizae and known benefits of mycorrhizae to host plants. Unfortunately, very few scientists in Korea are currently involved in mycorrhizal research. When a team of American plant pathologists visited Korea in September 1982 to participate in the Korea-U.S.A. Joint Seminar on Forest Diseases and Insect Pests, they were surprised by the principal author's statement that there was no single research project on mycorrhizae sponsored by Korean government or any scientific institutions. The author initiated a few years ago a research project on the ecology of tree mycorrhizae with a foreign financial support. Major areas of interest were survey of ectomycorrhizae in relation to soil fertility, taxonomic distribution of mycorrhizae among woody plants, identification of ectomycorrhizal fungi, and growth response of woody plants to artificial inoculation. In spite of the enormous application potential of mycorrhizae to agronomic plants, the subject of mycorrhizae has not been recognized by Korean agronomists, foresters or pathologists. The purpose of this review rather written in Korean is to introduce the techniques of mycorrhizal research to Korean scientists and to urge them to participate in challenging new scientific field which might bring us a remarkable increase in crop productivity and tree growth through manipulation of this unique symbiosis. In this review, following topics were discussed in the same order: introduction; brief history of mycorrhizal research; morphology and classification of mycorrhizae; distribution of mycorrhizae in plant kingdom and in soil profile; physiology of mycorrhizae (functions, mineral nutrition, mycorrhizal formation); interaction of mycorrhizae with soil-born plant pathogens. mycorrhizae in nitrogen-fixing plants; application of mycorrhizal techniques to nursery practices (isolation, culture, inoculation, and response); prospect in the future.

균근균(菌根菌)의 인공접종(人工接種)에 의(依)한 소나무류(類)의 생장촉진(生長促進) (Growth Stimulation of Pines by Artificial Inoculation with Mycorrhizal Fungus, Pisolithus tinctorius)

  • 美國에서 Pisolithus tinctorious(P.t.), Thelephora terrestris(T.t.)의 두 가지 외생균근균(外生菌根菌)을 도입(導入)하여 한국(韓國)에 있는 5개(個) 소나무수종(樹種)에 인공접종(人工接種)하여 그들의 생장촉진효과(生長促進効果)를 측정(測定)하였다. P.t.와 T.t.균계(菌系)를 대량증식배양(大量增殖培養)하여, 파종(播種) 직전(直前)에 Methyl bromide로 훈증(薰蒸)된 토양(土壤)에 접종(接種)하였다. 생장(生長) 첫해에, P.t.로 접종(接種)한 Pinus densiflora, P. thunbergii, P. rigida, P. rigida${\times}$taeda묘목(苗木)은 무처리묘목(無處理苗木)보다 수고생장(樹高生長)이 각각(各各) 55%, 36%, 69%, 37% 증가(增加)되었다. P. densiflora, P. rigida${\times}$taeda묘목(苗木)의 건중량(乾重量)에서는 P.t. 접종묘(接種苗)가 무처리묘목(無處理苗木)보다 각각(各各) 143%, 128% 증가(增加)되었다. 그러나 P. koraiensis에서는 생장(生長) 첫해에 균근균접종효과(菌根菌接種効果)가 나타나지 않았고 접종(接種)된 5개수종(個樹種)에서 T.t. 접종효과(接種効果)는 없었다. 우수(優秀)한 균근균(菌根菌)을 선발(選拔)하여 묘포장(苗圃場)에 인공접종(人工接種)할 경우 파종묘(播種苗)의 초기생장(初期生長)이 촉진(促進)되고, 묘목(苗木)의 품질향상(品質向上)을 도모(圖謀)할 수 있다고 결론(結論)지을 수 있다. 맹폐지조림시(荒廢地造林時) P.t.균(菌)의 이용(利用)에 관(関)하여서도 토의(討議)하였다.
