조직병리학 및 Histomorphometry 기법으로 관찰한 종양 유발 마우스의 주요 장기에 인진쑥 Methanol 추출물이 미치는 영향 (Effects of Artemisia capillaris Methanol Extract on Organs in Tumor Cells Inoculated Mice Observed the Histopathology and Histomorphometry)

  • This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillaris extracts against Hepa-lc1c7 and Sarcoma 180 cancer cells. In in vivo experimental tests using 210 ICR mice, on the $28^{th}$ day and the $42^{nd}$ day, all animals in vehicle controled HP (Hepa-lclc7 tumor cell inoculated vehicle control) and SP (Sarcoma 180 tumor cell inoculated vehicle control) showed tumor cells in the liver and spleen based on the histopathology. However, the incidences and the percentages of regions occupied by tumor cells were dramatically and dose-dependently decreased by mACH (Artemisia capillaris methanol extracts) treatment on the histomorphometry. Although the exact mechanism of inhibition of the incidences of tumor cells in the parenchyma whether inhibition of metastasis or proliferation is unclear, mACH dramatically reduce the percentages of regions occupied by tumor cells in the liver and spleen apart from the inoculation sites of Hepa-lclc7 and Sarcoma 180. In addition, they also effectively inhibit the abnormal changes on the kidney detected in the present study. The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-lclc7 and Sarcoma 180 m mice.

  • The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillaris extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells in in vivo experimental tests. In in vivo experimental tests using 210 ICR mice, based on flow cytometry, CD3+, CD4+, CD8+ and TNF-${\alpha}+$ splenocytes were significantly (p<0.05) reduced in the Hepa-1c1c7 and Sarcoma 180 inoculated vehicle controls, HP and SP, compared to those of the intact vehicle control on both the $28^{th}$ day and the $42^{nd}$ day, respectively. These decreases of CD3+, CD4+, CD8+ and TNF-${\alpha}+$ cells induced by tumor inoculations were significantly (p<0.05) inhibited by mACH treatment regardless of the type of experiments and tumor cells inoculated. The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.

야생쑥 추출물을 이용한 견직물의 천연염색성에 관한 연구 (The study on Natural Dyeability of silk with Artemisia Extract)

  • The purpose of this study was to investigate the natural dyeability of silk on extract of Artemisia princeps, The experimental items were divided into the mordanting method. comonent of fabric, kind of mordant. The experimental study was done by laundering fastness, abrasion(dry/wet) fastness, perspiration(acid/alkali) fastness, light fastness test and color difference by C.C.M system. The summarized finding resulted from experiments and investigation are suggested as follows; First, in the C. C. M test on mordanting methods, color difference was significantly improved when mordants were treatmented. And the premordanting method showed the highest color difference, color was most yellow-greenish, Second, in the C.C.M test on component of fabrics, color difference of silk was higher than cotton. It is considered that silk has -$NH_2$ , -COOH, -OH than more than cotton. Third, in dyeing-fastness on mordants, laundering fastness showed that color-change was 2~3 grade, the contamination on attached fabric was 4~5 grade. perspiration fastness(acid/alkali) showed 4~5 grade nearly and those of acid was higher than alkali. abrasion fastness(dry/wet) was 4~5 grade and in Fe(3~4 grade) was lower than the other mordants. Forth, in color difference analysis on mordants, Fe(50.0) showed the highest and the order of color difference was alum(16.0), Cu(7.2), Sn(3.5), Al(3.1), Cr(2.3), The Hue was turned into yellow-greenish in alum mordant treatment, the luminocity of color was most dark in Fe(-48.9) and Cu(-7.2), chroma was the highest in alum (15.7) method.

Anticonvulsant Effect of Artemisia capillaris Herba in Mice

  • In the present study, the anticonvulsant effects of Artemisia capillaris Herba (AC) and its major constituent, esculetin (ECT), were tested and the mechanism studied. Locomotion, Myorelaxation, motor coordination and electroshock seizure experiment were conducted in mice. To identify the anticonvulsant mechanism effect of this drug, chemical-induced seizure in mice and the ionic movement in neuroblastoma cells were also observed. The ethanol extract of AC was orally administered to mice 30 min. prior to testing and ECT was intraperitoneally injected. AC and ECT treatment did not change locomotor activities as well as activities on the rota-rod, which indicates that they did not cause a sedative and myorelaxation effect. AC and ECT treatment increased threshold of convulsion induced by electroshock. AC treatment also inhibited convulsion induced by pentylenetetrazole. In the case of strychnine however, only high dose of AC treatment inhibited convulsion. AC and ECT treatment increased the $Cl^-$ influx into the intracellular area in a dose-dependent manner. On the other hand, bicuculline, a GABA antagonist, inhibited the $Cl^-$ influx induced by AC and ECT. These results indicate that ECT induces the anticonvulsive effect of AC extract through the GABAergic neuron.

Cytotoxic and Apoptotic Effects of Extracts of Artemisia ciniformis Krasch. & Popov ex Poljakov on K562 and HL-60 Cell Lines

  • Artemisia, as one of the largest genera in the tribe Anthemideae of the Asteraceae comprises an important part of Iranian flora. While cytotoxic and apoptotic properties have already been reported for some species of the genus there is not any report on cytotoxic effects of A. ciniformis. Petroleum ether (40-60), dichloromethane, ethyl acetate, ethanol and ethanol-water (50:50) extracts of the aerial parts of A. cinformis were subjected to cytotoxic and apoptotic evaluations on two cancer human cell lines (K562 and HL-60) and on J774 normal cells. Among multiple extracts evaluated for cytotoxicity, dichloromethane ($CH_2Cl_2$) and petroleum ether (PE) extracts were shown to possess the highest anti-proliferative effects on HL-60 and K562 cells with $IC_{50}$ values of 31.3 and $25.5{\mu}g/ml$ respectively. Apoptosis induction verified by sub-G1 peaks was seen in flow cytometry histograms. Increase in the amount of Bax protein, formation of DNA fragments, and cleavage of PARP to 24 and 89kDa sub units all confirmed induction of apoptosis by A. cinformis extracts. Taken together according to the result of the present study some extracts of A. cinformis could be considered as sources for natural cytotoxic compounds and further mechanistic and phytochemical studies are recommended to fully understand the underlying mechanisms of cnacer cell death as well as identification of responsible phytochemicals.

사철쑥 (Artemisia Capillaris Thunberg)이 흰쥐 혈액 및 간지질 대사에 미치는 영향 (Effects of Artemisia Capillaris Thunberg on the Plasma and Liver Lipid Metabolism in Rats)

  • 사철쑥 (Artemisia capitlaris Thunberg)으로 성인병의 예방목적으로 흰쥐 (Sprague-Dawley rats)의 혈액 및 간지질 대사에 미치는 영향에 대한 효과를 알아보았다. 1) 사료이용효율은 L, LM, LP군에서 높았다 (p < 0.01). 혈청의 콜레스테롤 농도, 동 맥 경화지수 (p <0.01), 중성지질 (p < 0.01)은 L, LM, LP군에서 높았지만, 사철쑥 분말 첨가에 의해 HDL-콜레스테롤과 HTR를 증가시켰다 (p < 0.01). 2) 혈청 및 간 조직의 과산화지질은 C군에 비하여 L군이 유의적으로 많았으며, 사철쑥 첨가시 감소하였다 (p < 0.05). 3) 간 조직의 지방침착은 L군에서 지방양성구의 크기와 분포가 두드러지게 높았고. LM군, LP길군에서는 감소하였다. 이상의 결과는 사철쑥은 혈액 및 간의 지질조성 및 지질 대사에 바람직한 효과를 갖는 것으로 보인다. 이는 사철쑥이 성인병예방에 유용한 기능성 식품으로서 가치가 있을 것으로 사료된다.

엉겅퀴, 울금, 개똥쑥 복합 추출물의 지방간 개선효과 (Ameliorative Effects of Cirsium jaonicum, Artemisia annua and Curcuma longa on Non-alcoholic Fatty Liver Disease)

  • In this study, the effect of complex hot water extracts of Cirsium jaonicum, Artemisia annua and Curcuma longa (CAC) on the improvement of non-alcoholic fatty liver disease (NAFLD) was investigated. CAC inhibited fatty acid synthesis and lipid accumulation in HepG2 cells cultured with free fatty acid (FFA). In the NAFLD animal model, CAC extract suppressed the increase in body weight, liver, and epididymis fat weight, and suppressed the increase in hepatocyte fat and blood triglyceride. In addition, by blocking the Nrf2/HO-1 signaling pathway, cells were protected from oxidative stress in hepatocytes. Moreover, CAC inhibited the expression of COX-2, iNOS, TNF-α and IL-17 in hepatocytes. These results suggest the possibility that CAC extract can be applied in the field of health functional foods and pharmaceuticals for improvement and prevention of NAFLD.

쑥의 추출물 및 Coumaric Acid의 항균활성 (Antimicrobial Activity of Extracts and Coumaric Acid Isolated from Artemisia princeps var. orientalis)

  • 천연자원에서 향균성 물질을 찾고자 수종의 식용 식물을 검색하는 과정에서 쑥(Artemisia princeps var. orientalis)의 추출물이 항균활생이 비교적 높았다. 쑥의 메탄올 추출물을 겨l통분획한 ethyl acetate 분획물(EtOAc)은 agar diffusion법에셔 항균활성이 가장 높았으며, EtOAc로부터 분리된 화합물 중 0­C coumaric acid(200 -600ppm)는 B. subtilis(12.6 ~ -18.0mm)와 s. typmηwrium( 12.6-16.6mm)에 대하 여 우수한 항균활성을 나타내였고, 그 유도체인 p­C coumanc a디d는 약 1.2 -1.7배 정도의 항균활성을 증가시켰다 Coumaric acid의 3가지는 액체배양에 서 B. subtilis 증식응 모두 강하게 억제하였으며, S. typmmuriuη2, P. aeruginosa 및 s. aureus의 증식 억제에는 각각 0-, m- 및 p-coumaric a디d가 효과적 이었다. B. subtilis의 반고체 agar 배양에셔 pc coumaric acid의 MID는 $\100∼200mu\textrm{g}$/disk였다.

Effect of Fermented Lactic Acid Bacteria on Antiallergic Effect of Artemisia princeps Pampanini

  • Artemisia princeps Pampanini, which is named as Sajabalssuk (SJ-1) in Korea, was fermented with lactic acid bacteria (LAB), and their antiallergic activities were investigated. When SJ-l was fermented with some LAB isolated from human feces, the inhibition of NO production in RAW264.7 cells and antioxidant activities of SJ-1 were not affected. However, the inhibitory activity of SJ-1 against degranulation of RBL-2H3 cells induced by IgE was increased by LAB fermentation. Among the LAB tested, Bifidobacterium infantis K-525 provided the most potent inhibitory effect of SJ-1 against degranulation of RBL-2H3 cells. SJ-1 extract fermented with B. infantis K-525 (F-SJ-1) potently inhibited the mouse passive cutaneous anaphylaxis reaction induced by IgE with antigen, skin dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate, and scratching behaviors induced by compound 48/80. These inhibitory activities of F-SJ-1 were more potent than those of SJ-1. These findings suggest that the inhibition of SJ-1. extract against IgE-induced allergic diseases, such as rhinitis and asthma, can be enhanced by LAB fermentation.

Antibacterial compounds against fish pathogenic bacteria from a combined extract of Angelica gigas and Artemisia iwayomogi and their quantitative analyses

  • In the search for antibiotic alternatives from safe and effective medicinal plants against fish pathogenic bacteria, we found that a combined extract (CE) of 1:1 (w/w) ratio of Angelica gigas Nakai roots and aerial parts of Artemisia iwayomogi Kitamura showed antibacterial activity against the fish pathogenic bacteria. By antibacterial activity-guided fractionations and isolations, five compounds were isolated and identified as decursinol angelate (1), decursin (2), xanthotoxin (3), demethylsuberosin (4), and 2,4-dihydroxy-6-methoxyacetophenone (5) through spectroscopic analyses, such as nuclear magnetic resonance (NMR) and mass spectrometry (MS). Among the compounds, 1 and 2 showed the highest antibacterial activities against Streptococcus iniae and Vibrio anguillarum, showing minimum inhibitory concentrations (MICs) of 62.5-250 ㎍/mL. Compounds 3, 4, and 5 were also found to be active, with MICs of 31.25-1,000 ㎍/mL for those strains. Furthermore, active compounds, 1 and 2 in CE were simultaneously quantified using high-performance liquid chromatography-tandem MS (HPLC-MS/MS). The average contents of 1 and 2 in CE was 3.68% and 6.14%, respectively. The established method showed reliable linearity (r2 > 0.99), good precision, accuracy, and specificity with intra- and inter-day variations of < 2 % and recoveries of 90.13%-108.57%. These results may be helpful for establishing the chemical profile of CE for its commercialization as an antibiotic alternative in aquaculture.
