Neurospora crassa ars-1 프로모터의 발현율 조사 (Transcription level of the ars-1 promoter of Neurospora crassa)

  • Arylsulfatase를 암호화하는 Neurospora crassa의 ars-1 유전자는 황의 결핍에 의해서 발현이 조절된다. lacZ 유전자에 연결된 ars-1 프로모터 (Pars)가 N. crassa RLM 35-35 a his-3 inl 균주에 형질전환되고, 유사 서열 재조합에 의한 단일 교차에 의해서 염색체의 his-3 부위로 도입이 되었다. 황이 결여된 Vogel의 배지에서 자란 균사로부터 $\beta$-galactosidase 활성이 측정되었다. $\beta$-galactosidase 활성은 derepression 조건으로 변환한지 14 시간 후에 최고치를 기록하였다. Microconidiation에 의해서 생산된 homokaryon에서 측정된 활성은 heterokaryon보다 17% 증가가 되었음을 보여주었다. 이 실험은 ars-1 프로모터의 발현율을 측정할 수 있는 한 방법을 제시한 것으로, 보다 수월한 ars-1 발현 조절 연구가 기능할 것이다.

Isolation and ars Detoxification of Arsenite-Oxidizing Bacteria from Abandoned Arsenic-Contaminated Mines

  • The ecosystems of certain abandoned mines contain arsenic-resistant bacteria capable of performing detoxification when an ars gene is present in the bacterial genome. The ars gene has already been isolated from Pseudomonas putida and identified as a member of the membrane transport regulatory deoxyribonucleic acid family. The arsenite-oxidizing bacterial strains isolated in the present study were found to grow in the presence of 66.7 mM sodium arsenate($V;\;Na_2HAsO_4{\cdot}7H_2O$), yet experienced inhibited growth when the sodium arsenite($III;\;NaAsO_2$) concentration was higher than 26 mM. Batch experiment results showed that Pseudomonas putida strain OS-5 completely oxidized 1 mM of As(III) to As(V) within 35 h. An arsB gene encoding a membrane transport regulatory protein was observed in arsenite-oxidizing Pseudomonas putida strain OS-5, whereas arsB, arsH, and arrA were detected in strain OS-19, arsD and arsB were isolated from strain RW-18, and arsR, arsD, and arsB were found in E. coli strain OS-80. The leader gene of arsR, -arsD, was observed in a weak acid position. Thus, for bacteria exposed to weak acidity, the ars system may cause changes to the ecosystems of As-contaminated mines. Accordingly, the present results suggest that arsR, arsD, arsAB, arsA, arsB, arsC, arsH, arrA, arrB, aoxA, aoxB, aoxC, aoxD, aroA, and aroB may be useful for arsenite-oxidizing bacteria in abandoned arsenic-contaminated mines.

한국어판 주의력결핍 과잉행동장애 평가척도의 부모용 규준연구 (Normative Study of the K-ARS(Korean ADHD Rating Scale) for Parents)

  • Objectives : The K-ARS (Korean ADHD Rating Scale) is one of the most important assessment tool of attention-deficit hyperactivity disorder (ADHD) in Korea. in this study, we presented detailed normative data on the K-ARS for school-aged children in Seoul metropolitan area to put it to practical use. Methods : The subjects were 2,397 students(1,223 boys and 1,174 girls, aged 6-12) from 4 elementary schools in Seoul, and one caretaker of each child completed the K-ARS for parents. Children who showed high scores of the K-ARS for parents were screened, and 2 child psychiatrists interviewed them to make a clinical diagnosis. We compared the mean scores of the K-ARS for parents between ADHD and normal group, and examined the percentage of correct classification. Results : There were some differences in score of the K-ARS for parents according to sex and age, so we presented continuous normative data with T score and subdivided cut-off points for ADHD screening. Interviews with child psychiatrists using DSM-IV criteria were performed to test diagnostic validity, and the difference in every the K-ARS for parents index between ADHD and normal group was significant(p<.001). Using 3 different cut-of points(80th, 90th, 93rd percentage), the accuracies of ADHD correct classification were 67.9, 72.2, 71.1% and all 3 canonical discriminants were significant (p<.05) between ADHD and normal group. Conclusion : The normative data and cut-off points on the K-ARS for parents are useful in screening ADHD children in Seoul metropolitan area.

  • Patient with skeletal class II relationship was treated with L-ARS. L-ARS is fixed functional appliance that could be effective in children and adolescent patient who don't wear activator. The following results were obtained ; 1) Growth of Mandible was stimulated and overjet was decreased, therefore coupling of anterior teeth was established with L-ARS which is fixed functional appliance. 2) These changes were accomplished with Mandibular skeletal growth with no effect on the Maxilla. 3) L-ARS was especially effective on patient who deny to wear the removable functional appliance.

  • 곤충 병원균을 미생물 살충제로서 이용하기 위한 대량배양 기술을 확립코자 미생물 살충제로 잠재성이 있는 Metarhizium anisopliae var. majus ARS 978의 대량배양을 위해 액체배양 조건을 확립하였다. 최적배양을 위한 온도와 pH는 각각 $28^{\circ}C$와 pH 7.0 이었다. M. anisopliae ARS 978의 균체량, 효소들의 활성 및 포자의 생성에 1.0 %(v/v) molasses, 1.0%(w/v) 주박, 0.3 %(w/v) $CaCO_{3}$가 가장 적합한 최적배지 조건이었다.

딸기재배 온실용 온습도 계측시스템의 성능개선 (Performance Improvement of a Temperature and Humidity Measuring System for Strawberry Cultivation Greenhouse)

  • 본 연구는 기존에 사용하여 왔고, 최근에 온습도의 정확도를 검증하였던 강제 흡출식 복사선 차폐장치(Aspirated Radiation Shield; ARS)를이용하여 모 기업(A 회사)에서 개발한 시스템의 성능을 개선하고, ARS 장치의 풍속이 온습도에 미치는 영향에 대해서도 시험적으로 검토하였다. 그 결과를 요약하면 다음과 같다. A 회사 제품의 시스템을 개선하기 전, A 회사 시스템의 온도는 ARS 장치로 측정한 온도보다 최대 10.2℃정도 높았고, 상대습도는 20.0%정도 낮게 나타났다. 시스템을 개선한 후, 노드 1, 2의 온도 및 상대습도는 거의 일치하는 것으로 나타났다. 개선 후의 노드 2와 ARS 장치로 측정한 온도간의 최고, 평균 및 최저온도를 포함한 온도편차는 각각 0.2~0.7℃정도로써 ARS 장치가 약간 낮거나 높게 나타나는 경향이 있었다. 상대습도의 경우, 일몰 직후 ARS 장치의 상대습도가 약 10.0%정도 높게 나타나는 경향이 있었지만, 그 이외에는 평균적으로 1.9%정도 ARS 장치가 약간 낮게 나타나는 경향이 있었다. 그리고 노드 1을 최소-중간 사이, 중간-최대 사이 및 최대로 설정한 경우, 노드 1, 2의 최고, 평균 및 최저온도를 포함한 편차는 각각 0.1~0.4℃, 0.0~0.2℃ 및 0.0~0.5℃정도였다. 그리고 노드 1의 3개 측점과 ARS 장치의 최고, 평균 및 최저온도를 포함한 편차는 각각 0.2~0.5℃, 0.1~2.2℃ 및 0.1~1.1℃정도의 범위로써 풍속의 크기에 따른 온도편차는 아주 미미한 것으로 나타났다. 또한 선행연구 및 본 연구의 결과를 종합하여 보면, 온도오차를 개선하기 위한 적정 풍속은 1.0~2.0m·s-1 정도의 범위일 것으로 판단되었다.

Comparison of Tibial Tuberosity Transposition and Anti-rotation Suture for Medial Patellar Luxation in 133 Small-breed Dogs

  • This retrospective study is designed to compare the clinical results of tibial tuberosity transposition (TTT) and anti-rotation suture (ARS) treatments for medial patellar luxation (MPL). Medical records of 133 dogs were reviewed that had undergone surgical correction of MPL between January 2013 and May 2017. MPL correction was performed on 182 stifles, with TTT and ARS being performed on 101 stifles and 81 stifles. The common dog breeds receiving surgical treatment for MPL were Maltese, Pomeranian, Chihuahua and Poodle. Mean age of dogs with MPL was 32.6 months, and their mean body weight was 4.26 kg. Seventy dogs (52.6%) were male and 63 (47.4%) were female. Of the 182 stifles with MPL, grade II, III and IV were 18.7%, 72.0% and 9.3%. Total complications after TTT and ARS were recorded in 16.8% and 29.6%. Major complications after TTT and ARS were recorded in 5.9% and 12.3%, minor complications after TTT and ARS were recorded in 10.9% and 17.3%. The risk of complication and reluxation rate after TTT were significantly lower than that for ARS (p < 0.05). However, the rate of reluxation among dogs treated by ARS (1.2%) for grade II MPL was significantly lower than that for dogs treated by ARS (8.6%) for grade III MPL (p < 0.05). The TTT group had a shorter recovery period after surgical intervention than that in the ARS group (p < 0.001). In conclusion, TTT had a significantly lower incidence of complication and a shorter recovery period than ARS. However, ARS for grade II MPL appears to be a good surgical option for reducing the rate of reluxation after surgery. These results of this study could be used to provide therapeutic guidelines for surgical MPL correction in small-breed dogs.

ARS를 이용한 2바퀴 로봇의 자세 제어에 관한 연구 (A Study on Pose Control for 2 wheel Robot using ARS)

  • In this paper, configuration control for the Horizontal Maintenance of the 2 wheel robot has been studied using ARS(Attitude Refrence System). The 2 wheel robot technique is getting attention and there have been many researches on the seg-way since the US. Using its 2 freedom, a 2 wheel robot can move in various modes and Our robot performs goal reaching ARS.2 wheel robot fall down to the forward or reverse direction to converge to the stable point. Kalman Filter is normally used for the algorithm and numerous research is progressing at the moment. To calculate the attitude in ARS using 2 axis gyro(roll, pitch) and 3 axis accelerometers (x, y, z). In this paper we present a two wheel robot system for an autonomous mobile robot. This paper realized the robot control method which is much simpler but able to get desired performance by using the ARS control.

  • An Integrated Dynamics Control system with four wheel Steering (IDCS) is proposed and analysed in this study. It integrates and controls steer angle of front and rear wheel simultaneously to enhance lateral stability and steerability. An active front steer (AFS) system and an active rear steer (ARS) system are also developed to compare their performances. The systems are evaluated during brake maneuver and several road conditions are used to test the performances. The results showed that IDCS vehicle follows the reference yaw rate and reduces side slip angle very well. AFS and ARS vehicles track the reference yaw rate but they can not reduce side slip angle. On split-${\mu}$ road, IDCS controller forces the vehicle to go straight ahead but AFS and ARS vehicles show lateral deviation from centerline.

$Sm^3+$-ARS착물의 전기화학적 연구 (Electrochemical Studies on $Sm^3+$-ARS Complexes)

  • $Sm^3+$과 Alizarin red S(ARS)가 반응하여 생성한 착물의 전기화학적 성질을 직류폴라로그래피, 펄스차이폴라로그래피 및 순환전압전류법으로 연구하였다. $Sm^3+$과 ARS는 1 : 3 착물을 형성하였다. 0.1M LiCl 지지전해질 용액에서 착물의 환원파는 확산지배적이며 흡착성 착물파임을 확인하였다. 착물파의 환원전위는 ARS의 환원전위보다 음전위에서 나타났으며 $Sm^3+$의 농도를 증가시키면 ARS의 봉우리전류($P_1$)는 감소하고 착물의 봉우리 전류($P_2$)는 증가하였다. ARS의 농도 $2{\times}$$10^-4$ M에서 $Sm^3+$의 농도 $2{\times}$$10^-6$ M~$3.2{\times}$$10^{-5}$ M 범위에서 직선적으로 변하였다.
