HepG2 간암세포에서 아라키돈산에 의한 세포사멸기전에 미치는 NADPH 산화효소의 역할 (Role of NADPH Oxidase in the Mechanism of Arachidonic Acid-induced Apoptosis in HepG2 Human Hepatoblastoma Cells)

  • Previously, we have reported that arachidonic acid (AA) appears to be involved in the induction of apoptosis in HepG2 human hepatoblastoma cells. In this study we investigated the possible role of the NADPH oxidase, a membranebound enzyme generating reactive oxygen species (ROS), in the mechanism of AA-induced apoptosis in HepG2 cells. Apoptotic cell death induced by AA was significantly suppressed by various inhibitors of the NADPH oxidase, diphenylene iodonium (DPI), apocynin (Apo) and neopterine (NP). In addition, these inhibitors of the NADPH oxidase completely blunted the AA-induced ROS elevation. Next, we investigated the implication of metabolic pathway of AA in these AA actions. Both apoptosis and ROS production induced by AA were not significantly altered by treatment with indomethacin (Indo) or nordihydroguaiaretic acid (NDGA), selective inhibitors of cyclooxygenase (COX) and lipoxygenase (LOX), respectively, suggesting that AA metabolites produced by COX or LOX may not have an essential role in the AA-induced apoptosis and ROS generation. Collectively, these results suggest that the NADPH oxidase may be a key player in the mechanism of AA-induced apoptosis in HepG2 cells. These results further suggest that NADPH oxidase may be a good target for the management of human hepatomas.

Inhibition of Red Ginseng on 5-Hydroxyeicosatetraenoic Acid (5-HETE) Biosynthesis from Arachidonic Acid in Helicobacter Pylori-infected Gastric Cells

  • Helicobacter pylori (H. pylori) infection rapidly stimulated either COX-2 or 5-LOX and released arachidonic acid metabolites that have been considered as pivotal mediators in H. pylori-induced inflammatory responses. To determine whether red ginseng extract (RGE) can suppress the biosynthesis of 5(S)-hydroxyeicosatetraenoic acids (HETE), a precursor metabolite of leukotrienes B4 (LTB4) in H. pylori-provoked inflammatory responses in gastric epithelial cells, the biosynthesis of monohydroxy fatty acids was measured using radioactive arachidonic acid and validated by RP-HPLC using non-radioactive AA as substrate in AGS cells cocultured with H. pylori (ATCC 43504) with or without pretreatment of RGE. Among three known major HETEs, H. pylori infection specifically induced the biosynthesis of $^{14}C-5(S)-HETE$ rather than the complex of $^{14}C-15S-/^{14}C-12(S)-HETE$ from $^{14}C-AA$, concomitantly obtained by HPLC(p<0.01). RGE, 1 to $100{\mu}g/ml$, selectively suppressed H. pylori-stimulated $^{14}C-5(S)-HETE$ production implying the attenuation of 5-lipoxygenase activity, of which was similar to known LOX inhibitor NDGA $(10{\mu}M)$ (p<0.01). However, the amount of 5(S)-HETE was significantly reduced by higher dose of RGE $(100{\mu}g/ml)$ (p<0.05). These results indicated that LOX pathway might be one of principle pathogenic mechanisms of H. pylori and red ginseng could be a nutraceutical against H. pylori infection through inhibiting action of LOX activity.

일반란 및 기능란의 난황속의 지방산 조성 및 함량 비교 (Comparison of Composition and Content of Fatty Acid in Egg Yolk Oil among General and Functional Eggs)

  • This study was designed to compare content and composition of fatty acid in egg yolk oil among general eggs from chicken, quail, duck. We also compared those of general and functional chicken egg. Fatty acids were determined by GC method and the results were as follows: Palmitic acid, palmitoleic acid, stearic acid, oleic acid, linoleic acid, linolenic acid and timnodonic acid were identified in egg of chicken, quail and duck. The major fatty acid was oleic acid and palmitic acid in three kinds of eggs. Arachidonic acid and docosahexaenoic acid (DHA) were analyzed in egg of quail , but no in chicken. Monounsaturaterd fatty acid (MUFA) was higher in egg yolk oil of chicken and quail. Polyunsaturated fatty acid (PUFA) was higher in duck egg. Ginseng egg had significantly higher palmitic acid and oleic acid lower than general chicken egg. Gamgoal egg had lower palmitic acid and oleic acid, and higher palmitoleic acid and stearic acid than general chiekcn egg. The content of oleic acid was lower in DHA egg than in general chiecken egg, but arachidonic acid was detected only in DHA egg. Ginseng egg had the highest content of saturated fatty acid among chicken eggs. The content of MUFA acid was the highest in gamgoal egg and general chicken egg. DHA egg had the most amount of PUFA among all chicken egg.

형상이 다른 메주로 제조한 재래식 간장 중의 유기산과 지방산 조성 (The Contents of Organic Acid and Fatty Acid in Traditional Soy Sauce Prepared from Meju under Different Formations)

  • Three kinds of soy sauce were prepared using the brick type of conventional menu(A), the brick type of meju of Aspergillus oryzae (B) and the grain type of menu Aspergillus oryzae (C). Organic acid and fatty acid were analyzed In accordance to aging time of those products Citric acid, lactic acid, acetic acid, malonic acid, butyric acid, oxalic acid, and propionic acid were dejected in all kinds of soy sauce. The content of lactic acid was shown higher than those of any other organic acids. The content of lactic acid was much higher at beginning of preparation and at 180 days in soy sauce B than any other conditions. The content of acetic acid was much higher at beginning of preparation, at 120 days in soy sauce C and at 180 days in soy sauce B than any other conditions. The content of citric acid was highest at beginning preparation in soy sauce C, and that was highest in soy sauce B except beginning preparation to 120 days. Myristic, palmitic, stearic, oleic, linoliic, linolenic, arachidonic acid were detected in all kinds of soy sauce after 180 days. The content of oleic acid were shown 32.59∼53.79% in soy sauce B and in soy sauce C. The content of stearic acid was shown 49.7oA In soy sauce A. Linolinec acid and arachidonic acid were detected in only soy sauce C.

  • 한국 서해 상낙월도에서 채집한 연체 동물인 Bullacta exarata로부터 통상적인 9-hexadecenoic acid ethyl ester 이외에 불포화된 all-cis-5,8,11,14-eicosatetraenoic(arachidonic), 7,10,13-hexadecatrienoic, 10,13-octadecadienoic acid ethyl ester 및 상당량의 glyceryl ether인 chimyl alcohol이 검출되었다.

  • Objectives : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide-induced expression phospholipase $A_2$, cyclooxygenase-2 and inducible nitrogen oxide synthase, and the generation of arachidonic acid, prostaglandin D2 and E2 in RAW 264.7 cells, a murine macrophage cell line. Methods : The expression of phospholipase $A_2$, cyclooxygenase and inducible nitrogen oxide synthase was determined by western blotting with corresponding antibodies, and the generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ was assayed by ELISA method in RAW 264.7 cells. The non-toxic concentrations (0.1 to $5\;{\mu}g/ml$) of bee venom determined by MTT assay, were used in this study. Results : 1. Bee venom inhibited lipopolysaccharide-induced expression of phospholipase $A_2$ in a dose dependent manner after 48 hours treatment. 2. Bee venom inhibited lipopolysaccharide-induced expression of cyclooxygenase-2 in a dose dependent manner after 24 and 48 hours treatment. 3. Bee venom inhibited lipopolysaccharide-induced expression of inducible nitrogen oxidesynthase in a dose dependent manner after 48 hours treatment. 4. The generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ was not much affected by the treatment of bee venom on the lipopolysaccharide-induced generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ in RAW 264.7 cells.

  • The purpose of the present study was to measure and compare the arachidonic acid metabolites in diseased periodontal tissue and vital pulp tissue of the tooth, and to investigate the relationship between periodontal and pulp disease. Diseased periodontal tissue of periodontally involved human teeth and vital pulp tissue from the same teeth which were intact with no periapical lesions were obtained. Each periodontal and pulp tissue homogenates from the same tooth were incubated with $^{14}C$ - arachidonic acid. Lipid solvent extracts were separated by thin layer chromatography to be analyzed by autoradiography and TLC analyzer. 1. The conversion into $TXB_2$, 6 - keto - $PGF_{1a}$ and $PGE_2$, and unidentified metabolite in pulp tissue were less than that in diseased periodontal tissue(P<0.05). 2. Biosynthetic levels of $TXB_2$, unidentified metabolite, 6 - keto - $PGF_{1a}$ and HETEs were not satistically significant between diseased periodontal tissue and pulp tissue. $LTB_4$ was measured highly in pulp tissue(P<0.1). 3. The percentage of each metabolite to the total converted metabolites were not statistically significant between diseased periodontal tissue and pulp tissue. But the percentage of $LTB_4$ in pulp tissue was higher than that in diseased periodontal tissue(P<0.05). 4. The relative amounts of the total metabolites formed in lipoxygenase pathway to those formed in cyclo - oxygenase pathway were 6 fold in diseased periodontal tissue and 12 fold in pulp tissue. But there was no statistical significance between diseased periodontal tissue and pulp tissue(P>0.05).

임신 중인 생쥐 자궁에 있어서 아라키돈산에 특이적인 Acyl-CoA Synthetase 4의 발현 (Expression of Arachidonate-Preferring Acyl-CoA Synthetase 4 in the Mouse Uterus during Pregnancy)

  • 본 연구는 생쥐 자궁에 있어서 아라키돈산으로부터 prostaglandin의 생성에 관여하는 것으로 추측되는 acyl-CoA sytnhetase 4 유전자의 임신단계별 발현을 확인하고자 실시하였다. Acyl-CoA sytnhetase 4 유전자는 착상 전에는 발현이 증가하는 경향을 나타내었으며 착상 후에는 감소하였다. 이러한 발현의 양상은 세포막의 인질로부터 아라키돈산을 유리시키는 cPLA2의 발현과 유리된 아라키돈산으로부터 prostaglandin의 생성에 관여하는 COX1과 COX2의 발현 양상과 일치하였다. 이러한 결과는 세포막에서 유리된 아라키돈산이 무한적으로 COX1과 COX2에 의하여 prostaglandin의 생성에 이용되는 것이 아니라 acyl-CoA sytnhetase 4에 의하여 세포막의 인지질로 되돌려져 prostaglandin의 생성을 조절하는 기능을 세포가 수행하고 있는 것으로 추정된다.
