Effects of the Feeding Mixed Oils of the Butter, Sardine and Safflower Oils on Fatty Acid Metabolism of Serum and Liver in Rats (버터, 정어리유 및 홍화유를 혼합 급이한 흰쥐의 혈청 및 간장의 지방산 대사에 미치는 영향)

  • This study was designed to observe the effects of the feeding mixed oils of the sardine oil containing n-3 EPA(eicosapentaenoic acid) and DHA (docosahexaenoic acid) and the safflower oil in which were plenty of n-6 linoleic acid, on the fatty acid metabolism of serum and liver in the dietary hyperlipidemic rats. Experimental oils mixed with 16% butter(control group), 8% butter + 8% olive oil and 8% butter with various level of sardine and safflower oils were administered to the male rats of the Sprague Dawley for 4 weeks. In the fatty acid compositions of serum phospholipid, triglyceride and cholesteryl ester, according as the contents of mixed safflower oil increased, n-3 PUFA(polyunsaturated fatty acid) contents and ratio of EPA/AA(arachidonic acid) tended to be decreased, but n-6 PUFA contents and ratio of AA/PUFA tended to be increased. In the fatty acid composition of triglyceride component in liver lipid, DHA contents were most in the liver lipid. And components of cholesteryl ester, n-6 linoleic acid contents were high percentage. Fatty acid compositions of serum and liver lipids were influenced from the fatty acid composition of the test lipids.

Effects of Dietary Fatty Acids on fatty Acid Pattern in Development Rat Brain Phospholipids - Effects on P/M/S and $\omega$3/$\omega$6 Fatty Acid Ratios -

  • Docosahexaenoic acid(DHA), a $\omega$3 series fatty acid and arachidonic acid(AA). a $\omega$6 series fatty acid were found in relatively high concentrations in the phospholipids(PLs) of cell membranes of nerve tissues, and they can be affected by various factors. The present study examined the effects of dietary $\omega$6 and $\omega$3 fatty acid composition on P/M/S and on $\omega$3/$\omega$6 fatty acid ratios in brain PLs of 2nd generation rats. The expeimental diets consisted of 10% fat(by wt), which were computer- searched mixed oil('M') with P/M/S ratio, 1 : 1.4 : 1 and $\omega$6/$\omega$3 ratio, 6 : 1 and safflower oil('S') poor in $\omega$3 fatty acids. The experimental diets were started 3-4 wks prior to conception. During the lactation period, the feeding mothers were switched 1 wk after birth and provided the pups for 2 wks with milk which had compositions different from that of their natural mother. The same diet as their mothers was provided from weaning to 9 wks of age. The 'M'and 'S' rats were again subdivided into MM, MS, SS, SM rats according to diet which their lactating mothers were fed from the begining of the experiment. The relative percentage of P/M/S fatty acids in brain PLs in all experimental groups converged to a very similar value at 9 wks of age, indicating the existence of a control mechanism for the degree of fatty acids, unsaturation. The $\omega$3/$\omega$6 fatty acid ratios of brain PLs converged to about 1.0 in MM & SM groups and to 0.7 in SS & MS groups, suggesting also the existence of some balance between $\omega$3 and $\omega$6 fatty acids in developing rat brain. The concentrations of $\omega$3 fatty acids, especially DHA, in the SM group were increased and became similar to those in MM group at 9 wks of age. The increase in DHA of brain PLs was counterbalanced b)r a decrease in 22 5$\omega$6. Therefore, the ratios of 22 : 6$\omega$6/22 : 5$\omega$6 were higher in both MM & SM groups than those of SS & MS groups at 9 wak of age. Although dietary $\omega$3 and $\omega$6 fatty acids affected 22 : 6$\omega$S and 22 : 5$\omega$6 contained in rat brain PLs reciprocally, the relative percentage of AA did not appear to be significantly influenced by the diet in all groups at 9 wks of age, suggesting that a mechanism for the maintenance of a certain level of AA in brain PLs exists. In conclusion, the $\omega$3/$\omega$6 fatty acid and 22 : 6$\omega$3/22 : 5$\omega$6 ratios, but not P/M/S ratio, of rat brain PLs were affected by the postnatal dietary changes. Futher studies are required to clarify the mechanism(S) of ensuring a certain level of DHA and of maintaining a similar level of AA in rat brain PLs after. weaning(9 wk) regardless of prenatal and postnatal dietary changes.   • 자연계에 존재하는 수은중 유기수은은 생태계 먹이사슬을 통하여 체내의 여러장기에 축적되어 조직손상을 일으키는 것으로 잘 알려져 있다. 그러나 이러한 세포독성에 대한 정확한 생화학적 기전에 대해서는 자세히 알려진 바가 없다. 포스포리파아제 $A_2$(PLA$_2$)는 세포막의 인지질로부터 Arachidonic acid (AA)와 Lysophospholipid를 유리시키는 효소로 최근 세포손상과 관련하여 그 역할이 주목되고 있으며, 극히 최근, 일차배양 소뇌신경세포를 이용한 연구에서 메칠수은처리에 의해 세포독성의 지표인 Lactate dehydrogenase (LDH)의 유리와 함께 AA 유리가 증가되는 것이 관찰되었으나 여러형태의 PLA$_2$중 어느형태의 효소가 관련되어 있는지, 또한, 그 자세한 기전에 대해서는 불분명한 점이 많다. 본 연구에서는 신장세포의 일종인 MDCK세포를 이용하여 메칠수은의 처리에 의한 PLA$_2$의 활성화 및 그 생화학적인 기전을 구명하고자 하였다. [$^3$H]AA를 MDCK세포의 배양액에 첨가하여 라벨링한 후 메칠수은을 처리하였을때 [$^3$H]AA가 대조군에 비해 농도의존적 및 경시적으로 현저하게 증가하였으며 동시에 LDH의 유리도 함께 관찰되었다. 이러한 [$^3$H]AA의 유리 증가는 세포질 PLA$_2$에 특이적인 저해제로 알려진 AACOCF$_3$의 전처리에 의해 거의 완전히 억제되었으나 LDH의 유리는 오히려 증가하였다. 또한, 글루타치온(GSH)의 전구체인 NAC (N-Acetyl Cysteine)에 의해 [$^3$H]AA의 유리는 부분적으로 감소하였으나, LDH의 유리는 변함이 없었다. 돼지비장이나 MDCK 세포에서 얻어진 세포질 PLA$_2$에 메칠수은을 직접 처리하였을때는 오히려 PLA$_2$의 활성은 감소되었다. 위의 결과들로부터 메칠수은에 의한 [$^3$H]AA의 유리 증가는 세포질 PLA$_2$효소에 대한 직접적인 작용이 아니라 세포내 -SH기의 차단이나 Oxidative Stress에 의해 간접적으로 활성화되는 것으로 예상되며, 세포질 PLA$_2$에 의해 유리된 AA의 세포독설과 관련된 세포내의 역할에 대해 의문이 제기되었다.

Fatty Acid Concentration of Serum Phospholipids in Normal and Glaucoma Patients (정상인과 녹내장 환자의 혈청 인지질 지방산 농도)

  • Glaucoma is one of the leading causes of blindness worldwide and is characterized by degeneration of the optic nerve. Elevated intraocular pressure (IOP) is the major contributing factor to optic nerve damage. However, some patients develop glaucoma even with normal IOP. Other factors, including age, race, myopia, and nutrition, can affect glaucoma risk. Dietary fat intake and serum fatty acid composition are closely related with the fatty acid profile of the retina and thus can modulate glaucoma risk. In this study, we collected serum samples from 34 glaucoma patients (26 primary open angle glaucoma (POAG) and 8 normal tension glaucoma (NTG)) and 45 healthy controls and analyzed their serum phospholipid fatty acid concentrations. The results show that the ratio of oleic acid to stearic acid (OA/SA), which is an indicator of stearoyl-CoA desaturase activity, was higher in POAG patients when compared to normal controls. The ratio of eicosapentaenoic acid to arachidonic acid (EPA/AA), which is an indirect marker of eicosanoid biosynthesis, was also higher in glaucoma patients (p=0.048). These results imply that dietary fatty acid intake and serum fatty acid profile can influence glaucoma risk. Additional study is necessary to identify the relationship between fatty acids and glaucoma.

Effects of Unsaturated Fatty Acid Diets Feeding PEriods on the Antithrombosis the Hematological Changes in the Blood and Fatty Acid Compositions of Platelets in Rats (불포화 지방산의 종류와 사육기간이 흰쥐의 항혈전 작용, 혈액구성 및 혈소판의 지방산 조성 변화에 미치는 영향)

  • This study was performed to investigate the effect on polyunsaturate fatty acid diets and feeding periods on the antithrombosis. the hematological changes in the blood and fatty acid compositions of platelets in rats. Each group of rats was fed a diet containing 20%(W/W) corn oil beef tallow sardine oil and the general stock diet for 10, 20. 40 and 80 days. Rats fed sardine oil diet showed significantly longer bleeding time than any other diet groups after 20 days feeding The whole blood clotting time of sardine oil group fed for 80 days was increased significantly. The number of platelet and the concentration of hemoglobin showed no significant difference among all groups. The number of white blood cell was decreased continously in sardine oil group after 10 days feeding. The level of malondialdehyde generation during thrombin-induced aggregation of platelets was decreased continously in sardine oil grou after 20 days feeding. With regard to the composition of platelet fatty acid the ratio of eicosapentaenoic acid(EPA 20: 5 $\omega$-3) to arachidonic acid(AA 20:4 $\omega$-6) was increased in sardine oil group but decreased in corn oil groups and beef tallow groups with days. In conclusion the rats fed sardine oil diet for more than 20 days showed the fact that EPA induced the antithrombosis. the changes in number of white blood cell and the fatty acid composition of platelets.

The Anti-Inflammatory Effect of Pegmatite by in Vivo and in Vitro Study (In vivo 및 in vitro 시험을 통한 페그마타이트의 항염증 효과)

  • Purpose: This work aimed to elucidate the anti-inflammatory effect of pegmatite in vitro and in vivo. Methods: Author evaluated the suppressive effects of pegmatite on lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production, TNF-${\alpha}$ and IL-6 release in the RAW 264.7 murinemacrophages. Results: Treatment of RAW 264.7 cells with pegmatite significantly reduced LPS-stimulated NO production and inflammatory cytokine such as TNF-${\alpha}$ and IL-6 secretion in a concentration-dependent manner. Also pegmatite showed topical anti-inflammatory activity in the arachidonic acid (AA)-induced ear edema and acetic acid-induced increase in capillary permeability assessment in mice. It was also found that pegmatite (10 mg per ear in DW) inhibited arachidonic acid induced edema at 24 h more profoundly than 1 h by topical application. Furthermore, the vascular permeability increase induced by acetic acid was significantly reduced in mice that received pegmatite in 50 mg per mouse. Conclusion: Therefore the results of the present study suggest that pegmatite is a potent inhibitor of the LPS-induced NO and inflammatory cytokine in RAW 264.7 macrophages and showed anti-inflammatory activities in vivo animal model.

Effect of Dietary Docosahexaenoic Acid on Maze-learning Ability in Aged Mice Fed N-3 Fatty Acid Deficient Diet

  • The effect of docosahexaenoic acid (DHA) on maze-learning ability in aged mice was investigated. Aged mice fed a diet deficient in n-3 fatty acid received a semi-purified diet of 0.5, 1 and 2% DHA-ethyl ester (DHA-EE) for 4 months. Maze-learning ability was assessed at 3 months after the start of the experiment. The time required to reach the maze exit and the number of times that a mouse strayed into blind alleys in the maze were measured in 3 trials conducted at 4-day intervals. The time was significantly less in the DHA-EE 0.5% and DHA-EE 2% groups than in the control group (p<0.05). The number of times mice strayed into blind alleys in the maze was significantly lower in the DHA-EE 2% group than in the control group (p<0.05). Mice fed DHA showed increased DHA and decreased level of arachidonic acid (AA) in the brain. These results suggest that the intake of a 2% DHA diet improves learning ability in aged mice, which is associated with a higher DHA content in the brain.

Hepatoprotective effect of Samchulgeonbi-tang via Nrf2 Activation (Nrf2활성화를 통한 삼출건비탕(蔘朮健脾湯)의 간세포보호효과)

  • Objectives : Oxidative stress is an important cause of many diseases including liver injury. Therefore, adequate regulation of oxidative stress plays a pivotal role in maintaining liver function. Until recently, there has been no studies on the hepatoprotective effect of Samchulgeonbi-tang (SCGBT). Therefore, the hepatoprotective effect of SCGBT was investigated in HepG2 cells. In this study, oxidative stress was induced by arachidonic acid (AA) and iron. Methods : To analyze the hepatoprotective effects of SCGBT against oxidative stress induced by AA + iron, the cell viability, apoptosis-related proteins and intracellular ROS, glutathione (GSH), and mitochondrial membrane permeability (MMP) were measured. In addition, nuclear factor erythroid 2-related factor 2 (Nrf2) transcription activation and expressions of Nrf2 target gene were analyzed through immunoblot analysis. Results : SCGBT increased the cell viability from AA + iron - induced cell death and inhibited apoptosis by regulating apoptosis related proteins. SCGBT protected cells by inhibiting ROS production, GSH depletion, and MMP degradation against AA + iron induced oxidative stress. Furthermore, Nrf2 activation was increased by SCGBT, and the Nrf2 target genes were also activated by SCGBT. Conclusions : These results suggest that the SCGBT has a hepatocyte protection effect and antioxidant effect from AA + iron induced oxidative stress.

Effect of Rheum undulatum Linne extract and Glycyrriza uralensis Fischer extract against arachidonic acid and iron-induced oxidative stress in HepG2 cell and CCl4-induced liver injury in mice (대황과 감초 병용의 항산화 및 간보호효과)

  • Objectives : Rheum undulatum Linne and Glycyrriza uralensis Fischer are widely used herbal medicine. In this study, anti-oxidant and liver protective effects of R. undunlatum extract (RUE) and G. uralensis extract (GUE) were investigated in HepG2 cells, respectively. Oxidative stress and liver fibrosis were induced by arachidonic acid (AA) and iron, and CCl4.Methods : MTT assay was assessed for cell viability, and immunoblotting analysis was performed to detect expression of apoptosis related proteins. In addition, reactive oxygen species (ROS) and mitochondrial dysfunction were measured. In vivo, BALB/c mouse were orally administrated with the aqueous extract of 10 mg/kg RUE and 100 mg/kg GUE for 3 days and then, injected with CCl4 0.5 ml/kg body weight to induce acute liver damage. Serum ALT level was measured, and histological change was observed in Harris's hematoxylin and eosin stainResults : RUE and GUE pre-treatment increased relative cell viability in concentration dependent manner and altered the expression levels of apoptosis-related proteins such as procaspase 3, PARP and Bcl-xL. RUE and GUE also inhibited the mitochondrial dysfunction and excessive reactive oxygen species (ROS) production induced by AA and iron. In addition, RUE and GUE activated liver kinase B1 (LKB1), by increasing phosphorylation. Moreover, RUE and GUE treatment decreased liver injuries induced by CCl4, as evidenced by decreases in histological liver damage as well as serum alanine amino transferase (ALT) level.Conclusions : These data suggest that RUE and GUE has anti-oxidant and liver protective effects against AA and iron-induced oxidative stress and CCl4-induced liver injury.

Cytoprotective Effects of Artemisia princeps Extract through Inhibition of Mitochondrial Dysfunction (애엽(艾葉)의 미토콘드리아 보호 효과)

  • Objectives : Artemisia princeps is used as moxa in moxibustion and traditional herbal medicine. And its extracts or compounds is known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. This study was performed to investigate the cytoprotective effect of Artemisia princeps extract (APE) against arachidonic acid (AA)+iron-induced oxidative stress on HepG2 cell. Methods : The effects of APE on cell viability has been assessed using MTT assay. And flow cytometric analysis was performed to estimate APE's effects on mitochondrial function. To investigate its underlying mechanism, related protein was analysed by using immunoblot analysis. Results : Treatment of APE increased relative cell viability, prevented a decline of B-cell lymphoma-extra large (Bcl-xL) and cleavage of poly(ADP-ribose) polymerase (PARP) and procaspase-3, and also protected mitochondrial membrane permeability (MMP) against oxidative stress induced by AA+iron. In addition, APE treatment increased phosphorylation of AMP-activated protein kinase (AMPK) exerts a cytoprotective effect. Conclusions : This results demonstrate that APE has an ability to activation of AMPK which protects cells from AA+iron-induced oxidative stress and restores MMP.
