경북지역의 사과생산 및 유통에 관한 연구 (Studies on the production and marketing of apple of Kyungpook region and strategies of its improvement)

  • Apple is most favorable fruit in Korea, and apple farmland has been increased before and after the agreements of Uruguay Round and apple is considered as one of strategic agricultural products. Especially expansion of apple farmland is concentrated in Kyungpook region because of the suitable climate and its market share is about 70 percents in 1992. The marketing channels of apples of Kyungpook region are widely classified by merchant, agricultural or horticultural co-op and large scale farmer's. Among them market share of merchant's occupy over 65 percent. In marketing margins, commercial profit is higher than cost in total marketing margins and, assembler and retailer's margin is not less than wholesaler's. The fluctuation of the price of apple is high in year. The marketing problems of apples are several, first, complex marketing channels, secondly, the high percentage of market share by growing district assembler, thirdly, low rate of package and quality standardization, finally, concentration of shipment of apple because of the shortage of apple processing, storage and marketing facilities, of newly produced apples. In conclusion, to increase apple grower's income with the stabilization of supply and quality upgrade, the improvement measures of marketing system are as follows, first, government level's support in marketing facilities and mechanism, secondly, the increment of supply by grower's cooperatives, thirdly, the establishment of a serious of marketing system to increase the efficiency, fourthly, the establishment of cold-chain system and quality standardization of apple, finally, production of various kinds of apple processing goods.

국내 유통 주요 사과 품종 왜성대목 및 접수의 바이러스 검정 (Virus Detection of Dwarfing Rootstock and Scion in Major Commercial Apple Cultivars)

  • Apple (Malus domestica) is one of the most economically important fruits in Korea. But virus infection has decreased sustainable production of apple and caused the serious problems such as yield loss and poor fruit quality. Virus or viroid infection including Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), Apple mosaic virus (ApMV) and Apple scar skin viroid (ASSVd) has been also reported in Korea, furthermore, its damages and economic losses have increased constantly. In our research, we tried to survey virus infection for commercial nursery trees of major apple cultivars, especially dwarfing rootstocks 'M.9' and 'M.26' as well as scions. Trees were collected from 11 locations which have produced a great amount of apple nursery stocks in Korea. Infection degree was investigated in apple cultivars, 'Hongro' and 'Fuji' using RT-PCR method. In the scion of cultivar 'Hongro', infection ratio of ACLSV, ASPV and ASGV were 100%, 81.8% and 100% respectively. In the rootstock of cultivar 'Hongro', infection ratio of ACLSV, ASPV, ASGV and ApMV were 90.9%, 81.8%, 100% and 9.1% respectively. In the scion of cultivar 'Fuji', infection ratio of ACLSV, ASPV and ASGV were 81.8%, 90.9% and 100% respectively. In the rootstock of cultivar 'Fuji', infection ratio of ACLSV, ASPV, ASGV and ApMV were 81.8%, 90.9%, 100% and 9.1% respectively. Infection of ASSVd was not detected in both cultivars. From our results, it was found that most of apple rootstocks and scions had multiple infections by apple viruses which have caused economic damage in fruit production.

사과원 잡초관리 방법에 따른 사과해충 및 천적의 군집구조 (Community Structure of Phytophagous Arthropods and Their Natural Enemies at Different Weed Management Systems in Apple Orchards)

  • 사과원 잡초군락이 해충 및 천적발생에 미치는 영향을 알아보기 위하여 제초제를 살포하여 잡초를 제거한 구(제초구)와 수관하부까지 잡초를 키우면서 인력으로예초한 구(예초구)에서 수상의 해충 및 천적과 잡초상의 곤충류를 조사하여 다음과 같은 결과를 얻었다. 응애류와 진딧물의 발생은 처리간에 유의성있는 차이는 없었으나, 응애류의 경우는 예초구에서 발생이 적은 경향이었다. 사과굴나방의 경우 예초구에서 1993년에는 피해엽률이 10% 정도 낮았고, 1994년은 4% 정도 낮은 발생을 보였다. 응애류와 진딧물의 천적류 발생은 쳬초구에서 높은 경향이었으나 큰 차이는 없었으며 사과굴나방의 기생청 천적류는 예초구에서 1993년에는 6~10%, 1994년에는 20~25% 높은 발생을 보였다. 사과원 잡초상에서 포충망을 이용 곤충류를 채집 조사한 결과, 사과해충은 발견되지 않았으며, 사과해충 천적류로서 좀벌류, 고치벌류, 애꽃노리재, 무당벌류, 풀잠자리류 등이 발견되었다. 사과생육기간중 잡초상 및 수상에서 군충군집변화는 상이하였다. 수상에서의 곤충군집형성에 약제살포가 큰 영향을 주었으나, 잡초상에서는 약제살포에 상대적으로 안정된 군집이 유지되었다. 결론적으로 사과원 잡초는 수상의 사과해충 및 천적발생에 영향을 주고 있었으며, 사과굴나방 기생벌같은 기주특이적인 천적들은 수상이동에 의하여 밀도억제작용을 하였고, 잡초상에 먹이가 풍부하거나 또는 광식성인 천적들은 잡초상 곤충군집에 연관되어 수상의 해충밀도에 큰 영향을 주지 못했다.

열처리와 경정배양을 이용한 바이러스 무병 사과 생산 시스템 (Production System of Virus-free Apple Plants Using Heat Treatment and Shoot Tip Culture)

  • 국내외적으로 ACLSV, ASGV, ApMV, ASSVd와 같은 바이러스 및 바이로이드 병의 발생으로 사과 과실의 생산량 감소와 기형적인 외형 등 많은 문제점들이 보고되었다. 하지만 사과 바이러스의 감염에 대한 방제 대책은 거의 알려진 바가 없는 실정이다. 따라서 본 논문에서는 사과 신품종인 '단홍', '홍안', '새나라', '썸머드림'을 분양하기에 앞서 바이러스 무병묘를 생산하는 시스템을 확립하고자 하였다. $37^{\circ}C$가 유지되는 항온 항습장치에서 4주간 열처리를 하였으며 기내에서 경정 배양을 하였다. 열처리된 각각의 사과 신품종들은 바이러스 진단 프라이머를 통해 RT-PCR을 수행하여 바이러스 진단을 수행하였다. 결과적으로 '단홍'은 28%의 바이러스 무병묘를 확보할 수 있었으며 '홍안'은 16%, '새나라'와 '썸머드림'은 12%의 확률로 바이러스 무병 사과를 확보할 수 있었다. 본 연구결과는 열처리 및 경정배양을 통해 사과 신품종에서 바이러스 무병묘 생산 시스템 구축이 가능함을 보여주었다.

Ecological Characteristics and Unique Diagnostic Techniques of Apple Blotch Disease Caused by Marssonina coronaria in Korea

  • Apple blotch, caused by Marssonina coronaria, induce early defoliation in apple and leading to critical economic losses in apple orchards in Korea. Since M. coronaria is difficult to culture, we developed isolation and cultural method. We collected M. coronaria isolates from Gyeongbuk Province and then constructed phylogentic tree based on ITS regions. As the results, phylogenetic relationship indicated that all Korean isolates formed a same cluster and closely related to Chinese isolates [1]. Ecological characteristic of M. coronaria have been observed in apple orchards which located in Gyeongbuk Province from 2011 to present. As the results, the typical apple blotch symptoms were observed from July, and then the infected leaves were discolored and formed acervuli on the leaves. After rainfall, severe infection of symptoms such as discoloration and early defoliation were continuously observed until October. Also overwintered conidia were observed in next March on the fallen diseased leaves [2]. In the last 5 years, ascopores of M. coronaria were not observed in apple orchards which were severely infected by M. coronaria in Korea. Thus, it is assumed that overwintered conidia could be a primary inoculum of M. coronaria. Meanwhile, apple blotch has long latent periods compare to other apple disease. During the latent period, early diagnosis of apple blotch is the most important to control the disease by spray fungicide. In this reason, we developed novel diagnostic method to detect M. coronaria during latent period using optical coherence tomography (OCT) and Loop-mediated isothermal amplification (LAMP) method [2, 3]. In this presentation, it will introduce ecological characterization of M. coronaria in Korea and unique detection technique of M. coronaria in apple. It will be helpful to develop new strategies to control apple blotch in Korea.

Rapid Screening of Apple mosaic virus in Cultivated Apples by RT-PCR

  • The coat protein (CP) gene of Apple mosaic virus (ApMV), a member of the genus Ilarvirus, was selected for the design of virus-specific primers for amplification and molecular detection of the virus in cultivated apple. A combined assay of reverse transcription and polymerase chain reaction (RT-PCR) was performed with a single pair of ApMV-specific primers and crude nucleic acid extracts from virus-infected apple for rapid detection of the virus. The PCR product was verified by restriction mapping analysis and by sequence determination. The lowest concentration of template viral RNA required for detection was 100 fg. This indicates that the RT-PCR for detection of the virus is a 10$^3$times more sensitive, reproducible and time-saving method than the enzyme-linked immunosorbent assay. The specificity of the primers was verified using other unrelated viral RNAs. No PCR product was observed when Cucumber mosaic virus (Cucumovirus) or a crude extract of healthy apple was used as a template in RT-PCR with the same primers. The PCR product (669 bp) of the CP gene of the virus was cloned into the plasmid vector and result-ant recombinant (pAPCP1) was selected for molecule of apple transformation to breed virus-resistant transgenic apple plants as the next step. This method can be useful for early stage screening of in vitro plantlet and genetic resources of resistant cultivar of apple plants.

마늘의 첨가가 사과잼의 품질특성에 미치는 영향 (The Effects on Quality Characteristics Resulting from the Use of Varying Amounts of Garlic as Additives in Apple Jams)

  • Influence of garlic on the quality characteristics of apple jam was investigated. Apple jams with various amounts of sugar and garlic were prepared, and their sweetness, color difference, and instrumental texture were evaluated. The results were as follows: 1. When equal amount of sugar was added, the sweetness of apple jam decreased as the garlic content increased. 2. As the garlic content increased, the lightness and yellowness tended to be high and the redness and ΔE tended to be low, resulting that the color of apple jam was improved. 3. As the sugar content increased, the hardness and adhesiveness of the jam tended to increase when garlic was added at 10%. 4. Apple jam with 60% sugar and 10% garlic was the best in adhesiveness, glossiness, and overall quality by sensory evaluation. As the addition of garlic did not cause any significant difference in flavor, cooked garlic could be applied in apple jams.

  • Studies on the changes of methanol content in the manufacturing process of apple wine and apple brandy. The results from the studies of transition and changes of methanol content in the fermentation of wine and brandy from Korean apple, Kugkwang and Iwai are as follows. 1) Pectin, the source of methanol, can be extracted as dregs more than 85% of its in the process of pressing to get juice. 2) In the process of fermenting wine, the occurence of methanol depends on the condition of the apple itself (i.e. species, freshness, change in quality, or corruption). It seems that the insoluble pectin in the fresh apples changes into the soluble pectin as time goes by. 3) The heating treatment of fresh apples produced more methanol compared with nonheating treatment. 4) The content of methanol in apple brandy can influence free methanol content in mash pulp.

The Rapid Apple Decline Phenomenon: Current Status and Expected Associated Factors in Korea

  • Rapid apple decline (RAD) is a complex phenomenon affecting cultivated apple trees and particularly dwarf rootstocks on grafted young apple trees. Since its first appearance in the United States, RAD has been reported worldwide, for example in Canada, South America, Africa, and Asia. The phenomenon has also been observed in apple orchards in Korea, and it presented similar symptoms regardless of apple cultivar and cultivation period. Most previous reports have suggested that RAD may be associated with multiple factors, including plant pathogenic infections, abiotic stresses, environmental conditions, and the susceptibility of trees to cold injury during winter. However, RAD was observed to be more severe and affect more frequently apple trees on the Malling series dwarf rootstock. In this study, we reviewed the current status of RAD worldwide and surveyed biotic and abiotic factors that are potentially closely related to it in Korea.

기외 열처리와 경정접목을 이용한 사과 폿트묘에서의 바이러스 제거 (Combining ex vitro thermotherapy with shoot-tip grafting for elimination of virus from potted apple plants)

  • 사과는 국내 과수산업에서 가장 많이 재배되고 있는 과종이다. 하지만 apple mosaic virus (ApMV), apple stem grooving capillovirus (ASGV), apple stem pitting virus (ASPV), apple chlorotic leaf spot virus (ACLSV), apple scar skin viroid (ASSVd)와 같은 바이러스 및 바이로이드에 감염되면 과실의 수확량 감소 및 품질 저하를 야기시킨다. 본 연구에서는 국내 사과 농가에서 가장 많이 감염되어 있는 ASGV 바이러스를 제거하기 위한 효율적인 무병화 시스템을 확립하고자 하였다. ASGV에 감염된 폿트묘를 36℃, 38℃, 40℃가 유지되는 항온·항습장치에서 4주간 열처리를 수행하였으며, 신초 생장율과 바이러스 제거율을 조사하였다. 신초 생장률은 36℃ 처리구에서 가장 높았으며 신초의 중간부와 상단부는 바이러스가 제거되었으나 하단부는 바이러스가 제거되지 않았다. 38℃, 40℃ 처리구는 신초의 모든 구간에서 바이러스가 제거되지 않았으며, 40℃ 처리구는 신초의 생장 없이 열처리 3주 후 고사되었다. 36℃ 온도에서 열처리된 폿트묘의 경정을 절취하여 기외에서 접목하였으며 94%의 생존율과 20%의 바이러스 제거율을 보였다. 따라서 열처리 및 경정접목을 통해 무병묘 생산이 가능할 것으로 판단되었다.
