Prediction of Continuous Reactors Performance Based on Batch Reactor Deactivation Kinetics Data of Immobilized Lipase

  • Experiments on deactivation kinetics of immobilized lipase enzyme from Candida cyl-indracea were performed in stirred bath reactor using rice bran oil as the substrate and temperature as the deactivation parameter. The data were fitted In first order deactivation model. The effect of temperature on deactivation rate was represented by Arrhenius equation. Theoretical equations were developed based on pseudo-steady state approximation and Michaelis -Menten rate expression to predict the time course of conversion due to enzyme deactivation and apparent half-life of the immobilized enzyme activity in PFR and CSTH under constant feed rate polity for no diffusion limitation and diffusion limitation of first order. Stability of enzyme in these continuous reactors was predicted and factors affecting the stability were analyzed.

Pharmacokinetic Study of Decursinol Following Oral Administration in Rat (데커시놀추출물의 경구투여후 흰쥐에 있어서의 약물속도론적 연구)

  • The objective of this study is to investigate the pharamacokinetic parameters of decursinol following oral administration in Sprague-Dawley rats. The plasma concentration of decursinol was determined by LC/MS with APCI positive mode. The m/z value of decursinol was observed at 247. Following oral administration of decursinol extract, the apparent clearance was $5.3{\pm}2.7\;ml/hr/rat$, the absorption half life was $2.5{\pm}0.41\;hr$, the elimination half life was $3.05{\pm}1.57\;hr$, and the apparent volume of distribution was $21{\pm}12\;ml/rat$. The LC/MS method was successfully applied to the pharmacokinetic study of decursinol.

Measurements of $^{224}Ra\;and\;^{223}Ra$ in the Nakdong River estuary and the South Sea of Korea

  • The horizontal distributions of $^{224}Ra$ (half life=3.4 days) and $^{223}Ra$ (half life=11.3 days) were measured in surface seawaters of the Nakdong River estuary and the South Sea of Korea in April and October 2002. In order to determine these short-lived Ra isotopes, we used a delayed coincidence counter which is much more rapid and accurate than traditional methods. In an estuarine mixing zone (salinity, ~l6 ppt), the activities of $^{224}Ra\;and\;^{223}Ra$ were much greater than what would be expected from a mixture of freshwater and seawater in the Nakdong River estuary. This excess Ra may be a result of Ra desorption from fresh sediments originating from the river upstream. However, in the more open areas of the Nakdong River estuary (salinity>30 ppt) and the South Sea of Korea, $^{224}Ra\;and\;^{^{223}Ra$ activities decreased exponentially as a function of distance offshore. Using the decrease of $^{223}Ra$ with distance offshore in the South Sea of Korea, we estimated the apparent horizontal eddy diffusivities. The apparent eddy diffusion coefficients in South Sea of Korea are calculated to be approximately $3500-8000\;\textrm{m}^2/sec$.

  • First lactation records (174) of Jersey${\times}$Sahiwal and Holstein Friesian${\times}$Sahiwal half breds under 9 sires maintained at Chandra Shekher Azad University of Agriculture and Technology, Kanpur, Uttar Pradesh, India from 1975-1983, were used to estimate the genetic parameters and to predict herd life milk yield and average milk yield per day of herd life from first lactation traits. The traits included were: age at first calving, first service period, first lactation period, first calving interval, first lactation milk yield, milk yield per day of first calving interval, herd life milk yield, herd life and average milk yield per day of herd life. Most of the production and reproduction traits were found to have positive and significant correlations between them on genetic as well as phenotypic scales. Total twelve regression equations were fitted. The prediction equation of herd life milk yield in both the genetic groups showed linear relationship with AFC, FSP, FLP, FLMY and MY/DCI and was apparent and significant. Similarly, polynomials for milk yield per day of herd life for J${\times}$S and HF${\times}$S half breds also showed linear trend, which was found highly significant. The highest and lowest $R^2$ values were found for FCI and AFC, respectively.

Fusion Peptide Improves Stability and Bioactivity of Single Chain Antibody against Rabies Virus

  • The combination of rabies immunoglobulin (RIG) with a vaccine is currently effective against rabies infections, but improvements are needed. Genetic engineering antibody technology is an attractive approach for developing novel antibodies to replace RIG. In our previous study, a single-chain variable fragment, scFv57R, against rabies virus glycoprotein was constructed. However, its inherent weak stability and short half-life compared with the parent RIG may limit its diagnostic and therapeutic application. Therefore, an acidic tail of synuclein (ATS) derived from the C-terminal acidic tail of human alpha-synuclein protein was fused to the C-terminus of scFv57R in order to help it resist adverse stress and improve the stability and half-life. The tail showed no apparent effect on the preparation procedure and affinity of the protein, nor did it change the neutralizing potency in vitro. In the ELISA test of molecular stability, the ATS fusion form of the protein, scFv57R-ATS, showed an increase in thermal stability and longer half-life in serum than scFv57R. The protection against fatal rabies virus challenge improved after fusing the tail to the scFv, which may be attributed to the improved stability. Thus, the ATS fusion approach presented here is easily implemented and can be used as a new strategy to improve the stability and half-life of engineered antibody proteins for practical applications.

Studies on Ferrokinetics and Copper Metabolism in Various Malignant Tumors (각종악성종양환자(各種惡性腫瘍患者)의 Ferrokinetics 및 동대사(銅代謝)에 관(關)한 연구(硏究))

  • Anemia is a usual finding in advanced malignant diseases. Various mechanisms were reported as to be involved in the development of anemia of this kind, and they may differ in individual cases. Tumor anemias may be due, for instance, to chronic blood loss, shortened life span of the red blood cells or a decreased hemopoiesis in the bone marrow. The serum iron and copper levels, total iron binding capacity, apparent half survival of $^{51}Cr$-labelled red blood cells were measured along with the ferrokinetic studies using $^{59}Fe$ in 64 patients with various malignant tumors. Following were the results: 1. The serum iron levels were decreased in all cases. There existed no correlation between the serum iron levels and the severity of the diseases. 2. The serum copper levels were increased, particularly in lung cancer, rectal cancer, hepatoma and various sarcomas. There was also no correlation between the serum copper levels and the severity of the diseases. 3. The serum iron levels appeared to be inversely proportional to the serum copper levels. 4. The total iron binding capacities were within normal limits in all cases. There were also no correlations between the total iron binding capacities, serum iron levels and the severity of the diseases. 5. The patients could be classified according to the ferrokinetic patterns, namely, that of iron deficiency anemia in 10 cases, that of refractory anemia in 6 cases, normal in 1 case and that of atypical abnormal in 9 cases. 6. Apparent half survival time of $^{51}Cr$-labelled red blood cells were definitely shortened in half of the cases.

Toxicokinetics of paraquat in Korean patients with acute poisoning

  • To conduct a kinetic study of paraquat (PQ), we investigated 9 patients with acute PQ intoxication. All of them ingested more than 20 ml of undiluted PQ herbicide to commit suicide and arrived at our hospital early, not later than 7 h after PQ ingestion. The urine dithionite test for PQ in all of the nine patients was strongly positive at emergency room. Blood samples were obtained every 30 min for the first 2~3 h and then every 1 or 2 h, as long as the clinical progression was stable among the patients for 30 h after PQ ingestion. The area under the plasma concentration-time curve ($AUC_{inf}$), which was extrapolated to infinity, was calculated using the trapezoidal rule. Toxicokinetic parameters, such as the terminal elimination half-life, apparent oral clearance, and apparent volume of distribution ($V_d/F$) were calculated. The maximum PQ concentration ($C_{max}$) and the time to reach maximum PQ concentration ($T_{max}$) were also obtained. Plasma PQ concentrations in nine patients were well described by a bi-exponential curve with a mean terminal elimination half-life of $13.1{\pm}6.8h$. $C_{max}$ and $AUC_{inf}$ were $20.8{\pm}25.7mg/l$ and $172.5{\pm}160.3h{\cdot}mg/l$, respectively. Apparent volume of distribution and apparent oral clearance were $50.9{\pm}61.3l/kg$ and $173.4{\pm}111.2l/h$, respectively. There were a significant correlation (r=0.84; p<0.05) between the PQ amount ingested and $C_{max}$. $AUC_{inf}$ also showed a significant correlation (r=0.83; p<0.05) with the PQ amount ingested. These correlations provide evidence that PQ has dose-linear toxicokinetic characteristics.

  • Carcadian rhythm dependence of vancomycin pharmacokinetics was evaluated in 5 normal volunteers receiving a single intravenous 1.0 g dose of vancomycin at 8 o'clock in the morning and another occasion at 8 o'clock in the evening in a crossover manner. The serum data were subjected to simultaneous computer nonlinear least squares regression analysis using a two-compartment pbarmacokinetic model. The mean half-life of vancomycin was $4.78\pm0.81$ hr in the morning and $4.25\pm0.51$ hr in the evening. The mean total body clearance of vancomycin was $1.29\pm0.58$ hr in the morning and $5.58\pm0.48$ hr in the evening. No circadian rhythm was found to be apparent in normal volunteers. The mean in intrasubject difference in the half-life between 8 A.M. and 8 P.M. was $15.4\%$ with fluctuations ranging from $10.4\sim33.8\%$, It is reasonable to consider individual circadian rhythm for effective dosage regimen of vancomycin in clinical chronotherapeutics.

Effects of High Dose Irradiation on The Leukocyte Life Span (고선양(高線量) 방사선(放射線)이 백혈구(白血球) 수명(壽命)에 미치는 영향(影響))

  • As a part of studies on acute effects of high dose irradiation the present report was carried out to evaluate the changes of the leukocyte life span in the Newzealand white male rabbits by a single whole body exposure to gamma rays from $^{60}Co$ teleirradiation unit. The exposure was done in dose levels of 100, 300, 550 and 1,000 rad to each experimental group of 10 rabbits. The life span and apparent half survival time of leukocytes, and the elution rate of leukocytes in the circulating blood were measured by McMillan method using $^{51}Cr$. 1. As a critical indicator of radiation hazards of the Newzealand male rabbits, the LD 50/30 and LD 100/30 after whole body exposure was estimated as 550 and 1,000 rads respectively. 2. The life span of leukocyte in the circulation after irradiation was slightly shortened in the 100 rad irradiated group, as compared with the unirradiated control group, but markedly shortened in the 300, 550 and 1,000 rads irradiated group. 3. After irradiation, decrease of leukocyte half survival time in the circulation showed the same pattern as that of leukocytes life span. 4. As the irradiation doses increased, the elution rate of $^{51}Cr$ loss from $^{51}Cr$ tagged leukocytes in the circulation were markedly increased gradually. 5. The life span shortening of leukocytes in the circulation after irradiation seems to occure by two processes of senescence acceleration and early destruction.

Pharmacokinetic-Pharmacodynamic Modeling for the Relationship between Glucose-Lowering Effect and Plasma Concentration of Metformin in Volunteers

  • Metformin is a biguanide antihyperglycemic agent often used for the treatment of non-insulin dependent diabetics (NIDDM). In this study, the pharmacokinetics and pharmacodynamics of metformin were investigated in Korean healthy volunteers during a fasting state for over 10 h. In order to evaluate the amount of glucose-lowering effect of metformin, the plasma concentrations of glucose were measured for a period of 10 h followed by the administration of metformin (oral 500 mg) or placebo. In addition, the concentration of metformin in blood samples was determined by HPLC assay for the drug. All volunteers were consumed with 12 g of white sugar 10 minutes after drug intake to maintain initial plasma glucose concentration. The time courses of the plasma concentration of metformin and the glucose-lowering effect were analyzed by nonlinear regression analysis. The estimated $C_{max}$, $T_{max}$, $CL_{t}$/F (apparent clearance), V/F(apparent volume of distribution), and half-life of metformin were 1.42$\{pm}$0.07 $\mu\textrm{g}$/mL, 2.59$\{pm}$0.18h, 66.12$\{pm}$4.6 L/h, 26.63 L, and 1.54 h respectively. Since a significant counterclock-wise hysteresis was found for the metformin concentration in the plasma-effect relationship, indirect response model was used to evaluate pharmacodynamic parameters for metformin. The mean concentration at half-maximum inhibition $IC_{50}$, $k_{in}$, $k_{out}$ were 2.26 $\mu\textrm{g}$/mL, 83.26 $H^{-1}$, and 0.68 $H^{-1}$, respectively. Therefore, the pharmacokinetic-pharmacodynamic model may be useful in the description for the relationship between plasma concentration of metformin and its glucose-lowering effect.
