Effect of Temperature on Growth and Related Gene Expression in Alternative Type Wheat Cultivars (양절형 밀 생장에 대한 온도의 영향과 유전자 발현 양상)

  • We have investigated the effects of ambient temperature on the growth of wheat in Korea. The differences in the growth phase of wheat were compared according to the temperature treatment. The productive tiller number and dry weight were decreased in a plot under a higher temperature treatment. We found that the growth of Jinpum was different from that of the alternative wheat cultivars, which were bred in Korea, at 50 days after treatment. While the Jinpum wheat grown at 17℃ showed vegetative stage growth, that grown in the 23℃ growth chamber entered the heading and flowering stage. The differences in the expression of 16 genes known to be involved in high-temperature responses were checked by using Jinpum wheat 50 days after two temperature treatments (17℃ and 23℃), which showed apparent differences in expression between the higher and lower temperatures during the growth phase. In the 23℃ treatment samples, the genes with increased expression were HSP70, HSP101, VRN2, ERF1, TAA1, YUCCA2, GolS, MYB73, and Histone H2A, while the genes with decreased expression were VRN-A1, DREB2A, HsfA3, PIF4, PhyB, HSP17.6CII, rbcL, and MYB73. YUCCA2, HSP101, ERF1, and VRN-A1 showed a significant difference in gene expression between lower- and higher-temperature conditions. Overall, combining the means of the expression of various genes involved in thermosensing, vernalization, and abiotic stresses, it is possible to conclude that different sets of genes are involved in vernalization and summer depression of wheat under long term, high ambient temperature conditions.

Evaluation of the Removal Characteristics of Pollutants in Storm Runoff Depending on the Media Properties (여재 특성에 따른 강우 유출수 내 오염물질 제거특성 평가)

  • The aims of this study were to evaluate the removal efficiency for various pollutants in urban storm runoff by a filtration device, and to determine design parameters depending on filter media properties. Appropriate selection of filter media will affect the size and life time of the filtration device. Sets of column tests were performed in order to evaluate the removal efficiency by perlite and a synthetic resin. An investigation of surface properties including CEC (cation exchange capacity) and zeta-potential suggested that the perlite had a superior adsorption capability for cationic pollutants. TCODcr and turbidity were analyzed to investigate the removal characteristic of particulate pollutant. In both columns, the particles in the collected storm runoff was almost completely capture with a small EBCT (empty bed contact time) of 2.5 minutes. Complete clogging at the EBCT of 2.5 minutes occurred after 630 minutes in the perlite column and 810 minutes in the resin column. The removal efficiency of TCODcr and turbidity at the EBCT of 2.5 minutes decreased to below 70% due to an wall effect. The removal efficiency for dissolved pollutant (SCODcr) was negligible due to the insufficient contact time for adsorption. The removal of heavy metals (Cu, Zn, Pb) was mostly ascribed to the filtration of particles containing metals, since the relationship between CEC and the removal efficiency was not apparent. The result of this study would be valuable for the application of filtration device to control of urban storm runoff.

A Study of Water Quality of Lake Daeho (대호(大湖) 수질변화(水質變化)에 관한 연구(硏究))

  • This study was designed to find out whether the water quality of Lake Daeho would be suitable for Sgricultural purposes during 1984-1988 perird. Thr results were as follows ; 1. There was an apparent seasonal temperature fluctuation$(6-25^{\circ}C)$ above the entrance of the culvert at the depth of 14m, but the temperature below remained constant at $11,9^{\circ}C$. 2. The yearly water qualities observed at the deepest site by the seadike were 7.4-7.5 pH, 16,800-1,472 ${\mu}mhos/cm$ EC, 9.2-10.8ppm DO, 1.3-2.5ppm DOD, 2.4-5.3ppm COD, 0.22-2.29ppm T-N, 0.01-0.10ppm T-P. 3. The average values of water qualities at the epilimnion in 1988 were 7.6 pH, 1,745umhos/cm EC, 10.8ppm DO, 1.8ppm DOD, 2.4ppm COD, 0.52ppm T-N, 0.05ppm T-P. 4. The salinity of the epilimnion at a 6m depth was 29,000ppm before the final closure of the seadike. It was 11,000 ppm in March 1984, 4,300ppm in March 1985, 2,000ppm in March 1986 and 1987, and 900ppm in March 1988. The salinity of the whole water column decreased from 29.000ppm to 1,200 ppm in March 1988. The average salinity above and below the culvert in 1987 was about 1,300ppm, and 30,000ppm respectively reaching that of seawater. 5. The highest salinity was observed at the epilimnion by the seadike, showing about 5,835ppm in 1984. 6. The seasonal salinity fluctuation was 2,000ppm in May, 800ppm in October, and 485ppm in September 1987. 7. The halocline was observed at the depth of 14m where the entrance of the culvert was located. Therefore, the epilimnion water is suitable for agricultural purposes, and the intake of water from Lake Sapkyo seems to be unnecessary.

Effects of Dietary Probiotic Complex on Growth Performance, Blood Immunological Parameters and Fecal Malodor Gas Emission in Growing Pigs (복합 생균제 첨가가 육성돈의 생산성, 면역관련 혈액학적 지표 및 분내 유해가스 발생에 미치는 영향)

  • This study was conducted to evaluate effect of probiotic complex(Lactobacillus acidophilus, Bacillus subtilis and Aspergillus oryzae) on growth performance, blood immunological parameters and fecal malodor gas emissions in growing pigs. Forty-eight pigs[(Landrace × Yorkshire) × Duroc, 25.31±1.29kg average initial body weight] were used in 35d growth trial. Dietary treatments included CON(basal diet), PC1(basal diet + 0.1% probiotic complex) and PC2(basal diet + 0.2% probiotic complex). From d 0 to 20, ADFI was significantly increased in PC1 and PC2 compared to CON(Linear effect, P=0.013). From d 21 to 35, ADFI was increased in PC1 compared to CON(Quadratic effect, P=0.024). For the whole period, ADFI was increased PC2 and PC1 compared to CON(Linear effect, P=0.009, Quadratic effect, P=0.004). For the whole period, ADG was increased in PC1 compared to CON(Quadratic effect, P=0.017). G/F was not affected by treatments. Dry matter digestibility in PC2 was higher than PC1 and CON(Linear effect, P=0.001). Nitrogen digestibility was significantly higher in PC2 and PC1 than CON(Linear effect, P=0.005). In blood immunological parameters, Total protein, IgG, red blood cell(RBC) and white blood cell(WBC) were increased in PC2 compared to PC1 and CON(Linear effect, P<0.001, Quadratic effect, P<0.001). In fecal malodor gas emission, ammonia and acetic acid were significantly reduced in PC2 compared to CON(Linear effect, P<0.02). Hydrogen sulfide was significantly reduced in PC2 compared to CON(Linear effect, P=0.0002, Quadratic effect, P=0.018). However, total mercaptans was not affected by treatments. Water content of feces was not significantly different among the treatments. In conclusion, 0.2 % probiotic complex improved ADFI, apparent dry matter and nitrogen digestibility, Total protein, IgG, RBC and WBC. Also, it decreased ammonia, acetic acid and hydrogen sulfide emissions in growing pigs.

Frequency of Meals and Hyperlipogenesis of Rat (쥐의 급식회수(給食回數)와 체지방과잉합성(體脂肪過剩合成))

  • This experiment was performed to investigate the effect of the frequency of meals on the metatolism and the body composition of rats when equal amount of purified diet was ingested. Thirty approximately days old rats weighing 290 g and thirty-two about 40 days old rats weighing 180 g were employed for the period of 34 days. Rats fed ad libitum (10 to 15 meals per day) and two-meal per day were pair-fed and equal amount of diet was fed to each rat in pair. The experimental results obtained are summarized as follows: 1. Frequency of meal did not exert any effect on the body weight gain. However, rats fed two-meal per· day gained significantly (p <0.005) more fat and energy than ad libitum group. The rate of gain of protein in ad libitum group was higher than that of two-meal group. No difference was observed for the mineral deposition of rat body. 2. From the preperation of rat liver it was found that the activity of glucose-6-phosphate dehydrogenase was much higher for the rats fed two-meals per day than those fed ad libitum. Therefore, it is suggested that the metabolic pathway of carbohydrate for two-meal group has been shifted from glycolysis to Hexose Monophosphate Shunt and produced more NADPH which would be the essential cofactor of fatty acids synthesis. 3. The rate of excretion of urinary nitrogen for two-meal group was significantly (p<0.005) higher than that of ad libitum group. It is apparent that considerable amount of over-loaded amino acids by feeding two-big-meal daily· could not be used for the protein biosynthesis all at once and excreted following deamination through urine. The residual carbon chain could be served as a precursor of fatty acids synthesis. 4. The heat production rate of rats fed two-meal group was significantly (p<0.005) lower than that of ad libitum group. It seems possible that the activity of thyroid gland (and consequently BMR) can be depressed by the frequency of meal.

Spontaneous Pneumomediastinum: Clinical Investigation (자연성 종격동 기종의 임상적 고찰)

  • Background: Spontaneous pneumomediastinum is an uncommon, benign, self-limited disorders that usually occurs in young adults without any apparent precipitating factors or disease. The purpose of this study was to review our experience in dealing with this entity and describe a reasonable course of assessment and management. Material and Method: A retrospective case series was conducted to identify adults patients with SPM who were diagnosed and treated in a single institution between 2001 and 2005. Result: Fifteen patients were identified who included 14 men and 1 women with a mean age of 26 years. Presenting symptoms were chest pain in 12 patients ($80\%$), dyspnea in 5 patients ($33\%$), and throat discomfort in 4 patients ($26\%$). Two cases were associated with use of inhalational drugs and 3 cases were associated with exercise. The predisposing factors were asthma, excessive exercise, and vomiting in spontaneous pneumomediastinum. The physical findings were subcutaneous emphysema in 10 patients ($77\%$). Chest radiography and computerized tomography were the diagnostic methods in all cases with CT scan revealing six cases with associated pulmonary abnormalities. Esophagogram and flexible bronchoscopy were selectively used. Fifteen patients ($100\%$) were admitted to the hospital. Their mean hospital stay was 3 days. All patients were conservatively treated. In a follow-up of 3 years no complications or recurrences were observed. Conclusion: Most simple spontaneous pneumomediastinum cases were benign diseases and most of them ($77\%$) had shown typical chest pain, dyspnea and subcutaneous emphysema. Inhalational drug use was not a major cause of SPM; however, increased use of bronchoinhalers was a suspicious cause of SPM.

Antioxidative, Antimicrobial and Cytotoxic activities of Fagopyrum esculentum $M{\ddot{o}}ench$ Extract in Germinated Seeds (발아 메밀 추출물의 항산화.항균활성 및 세포독성)

  • This research was conducted to investigate the possibilities of usage of germinated-buckwheat (Fagopyrum esculentum $M{\ddot{o}}ench$) by examining antioxidative, antimicrobial and cytotoxic effects of extracts from different germinated root length of buckwheat. Antioxidant activity $(RC_{50})$ was shown higher in extracts of non-germinated seed $(50.41\;{\mu}g/mL)$ and root length 10 mm $(80.57\;{\mu}g/mL)$, 2 mm $(93.77\;{\mu}g/mL)$, 5 mm $(107.09\;{\mu}g/mL)$ than BHT $(163.96\;{\mu}g/mL)$ as a synthetic antioxidant. In antimicrobial activity, non-germinated and germinated seeds were formed inhibitory zone against S. aureus $(4{\sim}10\;mm)$, P. aeruginosa $(2{\sim}9\;mm)$ at the concentrations of $10{\sim}40\;mg/mL$ but B. subtilis, E. coli and S. typhimurium were not apparent antimicrobial activity. Extracts of germinated seed also decreased their antimicrobial activity compared to non-germinated seed extract. In addition, the growth of Calu-6 was inhibited of both 5 mm root length germinated and non-germinated seeds $(800\;{\mu}g/mL)$ as 95.12% and 87.15%, respectively, but these did not show any influence on cytotoxic effect against MCF-7 and Caco-2 cell lines. Extracts of 2 mm and 5 mm germinated seeds were also inhibited against Calu-6 and SNU-601 cell lines.

Effect of Soil Characteristics and Fertilizers Application on Fresh Root Yield of Aralia continentalis K. -I.   • These studies were Conducted to survey cultivation methods, and to analyze soil chemical properties for stable production of Aralia continentalis K., a promising medicinal crop, in the main producting districts. Aralia continentalis K. was cultivated with planting budstocks in distances of $90cm{\times}60cm$ for 3~4 years in the same field, and application rates were N 10~31 kg/10a and P, K 8~17/10a using inorganic fertilizer, that is compound fertilizer(21-17-17) for basal dressings and urea for topdressings. Most of all, the soil surveyed was coarse loamy class which was well drained and soil depth is 50~150cm in the valleys. Soil pH was low, and content of organic matters and av. $P_2O_5$ was abundant but that of exchangeable cation such as Ca, Mg, K was deficient. The relationship between growth characteristics and weight of fresh root was positive correlation in the order of No. of root, stem width, No. of node and branch, plant height and root width. On the path coefficiant analysis, the relationship between content of soil K, Ca and root yield was more apparent than other chemical properties.

Effects of Nitrogen Fertilization on the Yield and Effective Components of Chrysanthemum boreale M. (질소시비가 산국의 수량과 유효성분에 미치는 영향)

  • Chrysanthemum boreale M. (hereafter, C. boreale M.), a perennial flower, has been historically used as a natural medicine in Korea. With increasing concerns for health-improving foods, the demand for C. boreale M. has become higher than ever. Howevr, the amount of wild C. boreale M. collected from mountainous areas is not enough to cover all demands. The cultivation system and fertilization strategy are required to meet increasing demand on C. boreale M. with a good quality. We investigated the effects of nitrogen application on plant growth and effective components of C. boreale M. to suggest optimum rate of nitrogen fertilization. C. boreale M. was cultivated in a pot scale (1/2000a scale), and nitrogen applied with rate of 0(N0), 50(N50), 100(N100), 150(N150), 200(N200), and $250(N250)kg\;ha^{-1}$. Phosphate and potassium were applied at the same level ($P_2O_5-K_2O=80-80kg\;ha^{-1}$) in all treatments. Maximum yield achieved in 246 and $226kg\;ha^{-1}$ N treatment on the whole plant and the flower part, a valuable part as a herbal medicine, respectively. Proline was the most abundant amino acid in the flower of C boreal M. and the contents of amino acids increased with increasing nitrogen application rate in flower. Nitrogen recovery efficiency was high more than 41% in all nitrogen treatments and increased to 61.8% in nitrogen N100 treatment. From the nitrogen content, the high nitrogen uptake, the low residue of mineral N and the reasonably good apparent fertilizer recovery, it can be inferred that C. boreale M. made efficient use of the available nitrogen. In flower, contents of Cumambrin A. which is a sesquiterpene compound and has the effect of blood-pressure reduction, decreased with increasing nitrogen application. However, the amount of Cumambrin A in flower increased as nitrogen rate increased, because of increasing flower yield. Conclusively, nitrogen fertilization could increase yields and enhance quality. The optimum nitrogen application rate might be on the range of $225{\sim}250kg\;ha^{-1}$ in a mountainous soil.

A Seroepidemiologic Study of Hepatitis A Virus in the Healthy Children and Adolescent in Kyonggi-do Province (경기도 지역의 건강한 소아와 청소년에서 A형 간염 바이러스의 항체 보유율에 대한 역학 조사)

  • Purpose: The incidence of hepatitis A virus(HAV) infection has markedly decreased in the last 20 years due to industrialization and improvements in the standard of living and hygiene in Korea. The reduction in seroprevalence rates indicates infection potential for young adult population, and a need for vaccinations in high-risk adults and children groups has been suggested. In this study we evaluated the seroprevalence rates and natural infection rates of hepatitis A in children and adolescent to obtain the basic data for vaccination of hepatitis A. Methods: A total of 334 children and adolescent subjects below 20 years old in Kyonggi-do province were examined for HAV antibody and seroprevalence rates in each age group was investigated. In 584 elementary school students residing in Kyonggi-do province, serum samples collected in 1993 and 1996 from the same subjects for investigation of natural seroconversion rates. Method of testing antibody was enzyme immunoassay. Results: 1) The seropositive rate of HAV in 334 subjects aged below 20 years old was 5.4%. According to age, the seropositive rates were 27.3% in infant group, 0.0% in 1~4 year-old group, 0.0% in 5~9 year-old group, 2.9% in 10~14 year-old group and 15.0% in 15~19 year-old group. 2) In the study of 584 elementary school children, only one subject showed seropositive in 1993, and in 1996 three different subjects showed seropositive results(0.5%); the natural seroconversion rate during 3 years was 0.5%. 3) The seroprevalence rates of below 20 year-old subjects reported in previous studies were 63.8% in 1979 and 47.3% in 1989 while the present study showed the rate at 5.4%. Conclusion: Since natural antibody formation is rarely occurring, there is a high risk for apparent hepatitis A infection in adults. Therefore vaccination in high risk groups is essential at present, and in order to reduce the chance for hepatitis A infection in adults, vaccination in children may be needed.
