Relationship between Endogenous Polyamines and Floral Bud Differentiation in Chrysanthemum morifolium under Short-day Conditions

  • This study was carried out to investigate the relationship between endogenous polyamines (PAs) and floral bud differentiation in chrysanthemum (Chrysanthemum morifolium). In this study, PA content (free, bound, and conjugated) in apical buds, leaves, and roots changed appreciably during floral bud differentiation. PAs accumulated during series of processes such as floral induction, differentiation of floret primordia, and crown formation in apical buds; changes in PAs in apical buds may have a relationship with those in leaves and roots. The levels of free PAs and conjugated PAs [putrescine (Put) and spermine (Spm)] in apical buds rapidly increased during the initiation stage of floral bud differentiation, while free and conjugated spermidine (Spd) reached their highest levels at the stage of floret primordium differentiation. In the free, conjugated, and bound PA fractions, the changes in Spm content were negligible compared to those of Put and Spd throughout the experiment. These findings indicate that PAs participate in regulating the process of flower bud differentiation in chrysanthemum.

A STUDY ON THE PATTERN OF THE ROOT RESORPTION FOLLOWING MAXILLARY INCISOR RETRATION (상악 전치부 견인시 치근 흡수 발생 양상에 관한 연구)

  • This study was carried out in order to find out the pattern of the root resorption following maxillary incisor retraction after maxillary 1st bicuspid extration in maxillary protrusion patients. For this study, thirty two patient who received orthodontic treatment were chosen. The results were as follows; 1. Of the total 192 teeth, 61 teeth(31.77%) showed no apical root loss, 64 teeth(33.33%) fell into the class "slight", 46 teeth(23.96%) fell into the class "moderate". Only 21 teeth(10.94%) were classified as "excessive". 2. No correlation was noted between the amount of apical root loss and the types of tooth movement of the maxillary central incisors. 3. The patients who were treated with standard brackets had more changes in tooth axis and less movement of root apexes, but the patients with straight brackets had less changes in tooth axis and more movement of root apexes. 4. Comparing the degree of root resorption between bracket types, patients who had used standard brackets showed more apical root loss than patients who had used straight brackets. 5. The most frequent degree of root resorption observed in standard bracket patients was second degree, followed by first degree and third degree. The most frequent degree of root resorption observed in straight bracket patients was zero degree, followed by first, second degree and third degree.

Bone Density Relationship of Mandible and Cervical Vertebrae in Panoramic Radiography

  • Purpose: Upper cervical vertebrae are commonly imaged together with the jaw bones in panoramic radiography. There have been many studies investigating the possible role of mandible as an indicator of osteoporosis. But the result doesn't show unanimity. This study measured bone densities of mandible and second and third cervical vertebrae to find out any relationship between these two areas. These results may contribute in panorama being used as a screening method in detecting possible osteoporotic patient. Materials and Methods: Randomly selected 226 digitized panoramic images with cervical vertebrae shadows from 156 dental patients between 5 to 80 years of age were used. And the bone densities of second and third cervical vertebrae, apical areas of first and second mandibular molars and interdental areas were measured. The bone density measurements were restricted to the cancellous bone and the average and standard deviations and paired t-tests were done to each measurements. Results: All the measurements were statistically significantly related. The best relationship was found between the third cervical vertebrae and first and second mandibular apical areas. The average and standard deviations of the measured bone density ratios of these areas were 1.20±0.45 and 1.34±0.48 each. Conclusion: Patients whose panoramic bone density of the third cervical vertebrae are much below those of mandibular first or second molar apical areas may have osteoporosis.

New Record of the Freshwater Dinoflagellate Peridinium umbonatum Stein (Dinophyceae) from Togyo Reservoir, Korea

  • Cells of the dinoflagellate Peridinium were frequently observed in water samples of Togyo reservoir, and some species were responsible for dense blooms. Recently, we could identify them as P. bipes f. occultatum Lindem. and P. aciculiferum Lemm., considering morphology (Ki et al. 2005a; Ki and Han 2005b): However, some unidentified Peridinium cells with different shapes and body sizes were found among the samples collected during early spring. Here we describe their morphological characteristics such as thecal plate and body size to characterize its taxonomic identity by morphological characters. The formula of epithecal plates was recorded as 4 apical, 2 intercalary and 7 precingular plates (i.e. 4’', 2a, 7’'’') and the epicone in an apical view was symmetric. An apical pore was easy to make out under a light microscope. No cingular displacement was observed. The average body size was 33 $\mu$m in length with a range of 26-36 $\mu$m, and average 26 $\mu$m in width with a range of 21-31 $\mu$m, respectively; the cell was, therefore, shown slightly elongated. This way we identified Peridinium umbonatum Stein, 1883 for the first time from Korean freshwaters.

Induction on in vitro Plant Regeneration the Apple Rootstocks of Fire Blight Resistance by Plant Growth Regulators (생장조절제 처리에 따른 과수화상벙 저항성 사과대목의 기내 식물체 유도)

  • Apple (Malus×domestica Borkh.; Rosaceae) is an important fruit crop grown mainly in temperate regions of the world. Tissue culture in vitro is a biotechnological technique that has been used to genetically improve cultivars (scions) and rootstocks. This could be important in the production of genetically uniform scions and rootstocks for commercial apple production. In nurseries, apple plants are produced by grafting scions onto rootstocks. The Cornell-Geneva (Geneva® series) breeding program has bred several dwarf rootstocks that are resistant to diseases and pests and are also cold hardy. This study was conducted to determine the optimal medium strength to improve sprouting shoot rate of apical meristem of the apple rootstocks of fire blight resistance. The apple rootstocks apical meristem at size (0.2 mm to 0.3 mm) with axillary buds were cultured on the MS(Murashige & Skoog) medium supplemented with plant growth regulators. The sprouting ratio and growth characteristics was evaluated after eight weeks in vitro culture. The highest rate of bud differentiation and shoot formation were 23.8% and 55.6%, respectively. After 6 weeks, shoots were regenerated from apical meristem, and their growth characteristics was significantly varied on the respective basal medium with different plant growth regulators. Our studies showed that the apple rootstocks the apple rootstocks of fire blight resistance plantlets could be successfully produced from apical meristem differentiated out of young twigs via organogenic regeneration.

The Variation of Winter Buds among 10 Selected Populations of Kalopanax septemlobus Koidz. in Korea

  • The objective of this study was to understand the conservation of gene resources and provide information for mass selection' of winter bud characters among the selected populations of Kalopanax septemlobus Koidz using analysis of variance(ANOVA) tests. The obtained results are shown below; 1. Ten populations of K. septemlobus were selected for the study of the variation of winter bud characters in Korea. The results of the analysis of variance(ANOVA) tests shows that there were statistically significant differences in all of the winter bud characters among those populations. 2. Correlation analysis shows that width between Height and DBH(Diameter at breast height) characters have negative relationship with all of the characters, as ABL(Apical branch length), ABW(Apical branch width), AWBL(Apical branch winter bud length), AWBW(Apical branch winter bud width), ABT(Apical branch No. of thorns), ABLB(Apical branch No. of lateral bud) and LBL(Lateral branch length), LBW(Lateral branch width), LBT(Lateral branch No. of thorns), LBLB(Lateral branch No. of lateral bud). 3. The result of principal component analysis(PCA) for winter buds showed that the first principal components(PC' s) to the fourth principal component explains about 78% of the total variation. The first principal component(PC) was correlated with AWBW, LWBW, and LBL and the ratio of ABL/ABW and LBL/LBW out of 16 winter bud characters. The second principal component correlated with ABL, ABW, ABLB, LWBL(Lateral branch winter bud length), and LBW and the ratio of AWBL/AWBW. The third principal component correlated with ABL, ABW, LWBL, LBL, and the ratio of LBL/LBW. The fourth principal component correlated with LBL and the ratio of LWBL/LWBW(Lateral branch winter bud width), LBL/LBW. Therefore, these characters were important to analysis of the variation for winter bud characters among selected populations of K. septemlobus in Korea. 4. Cluster analysis using the average linkage method based on 10 selected populations for the 16 winter bud characters of K. septemlobus in Korea showed a clustering into two groups by level of distance 1.1(Fig. 3). As can be seen in Fig. 3, Group I consisted of three areas(Mt. Sori, Mt. Balwang and Mt. Worak) and Group Ⅱ contisted of seven areas(Suwon, Mt. Chuwang, Mt. Kyeryong, Mt. Kaji, Mt. Jiri, Muan, and Mt. Halla). The result of cluster analysis for winter bud characters corresponded well with principal component analysis, as is shown in Fig. 2.

COMPARISON OF CANAL SHAPING METHODS WITH GTTM ROTARY FILE AND CONDENSATION METHODS (GT rotary file을 이용한 근관성형법과 충전방법의 비교)

  • The purpose of this study was to compare the shaping time of two shaping methods and the leakage of three different obturation techniques. Ninty three canaled human molar teeth were used, which were randomly divided into two groups of forty teeth each and ten control teeth. After working length determination, the one group was prepared crown-down technique using rotary root canal instruments of GT rotary files .12/20, .10/20, .08/20 and .06/20 taper(Maillefer Instrument SA. Switzerland). The other group was instrumented with Gates Glidden burs(#1, #2, and #3) to coronal preparation and GT rotary files .08/20 and .06/30 taper to apical preparation. Shaping time was measured. After root canals were instrumented, they were divided to three subgroups and obturated as follows : Subgroup 1, obturated with single cone method Subgroup 2, obturated with lateral condensation : Subgroup 3, obturated with continuous wave technique. Three subgroups were obturated using non-standardized gutta-percha cone(Diadent, Korea, .06 or .08 taper) and AH-26(Dentsply DeTrey, Germany) as a root canal cement. Ten unobturated teeth served as positive and negative controls. After immersion in 2% methylene blue solution for 1 month, the teeth were washed during 24h. The teeth were demineralized in 10% nitric acid and dehydrated by immersion in 80, 90 and 100% ethyl alcohol. The teeth were finally cleared and stored in 100% methylsalicylate, and apical dye penetration was evaluated under stereomicroscope(Leica M420, LC, U.S.A)at $\times$8.75 magnification. Liner measurement of dye penetration was assessed with the use of digitalized image analysing system (analySIS, GmbH, Germany) The data were analysed statistically using independent T-test and Two-way ANOVA and Tukey test. The result were as follows 1. In canal prepared with GT$^{TM}$ rotary file, shaphing time taked more than the group of using Gates Glidden drill to coronal preparation without statistical significance (p>0.05) 2. The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files showed significantly more apical leakage than those of lateral condensation and continuous wave technique regardless of shaping method (p<0.05). 3 The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files and Gates Glidden drill showed significantly more apical leakage than those of continuous wave technique regardless of shaping method (p<0.05). 4. Regardless of shaping method, The group of continuous wave obturation showed less apical leakage than those of lateral condensation without statistical significance (p>0.05). 5. The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files and Gates Glidden drill showed more apical leakage than the group of lateral condensation using same shaping method with-out statistical significance (p>0.05).

Transepithelial transport and dynamic changes on apical membrane area of turtle bladder (Turtle Bladder 정단세포막(丁端細胞膜)의 역동적(力動的) 변화와 상피수송(上皮輸送)에 관하여)

  • The present study was carried out to analyze the evidence of membrane recycling, and the regulation of cellular transport by dynamic changes in apical membrane area that functionally interacts with the number of cytoplasmic vesicles. Under scanning electron micrographs, turtle bladder mucosa contain three main type of cells; granular cells and carbonic anhydrase (CA)-rich cells, deviding into a and b type of epithelial cell. The granular cell is the majority cell type of the mucosa comprising 80% of the total cell number. The remaining 20% of the cells are characteristically rich in carbonic anhydrase. Uptake of HRP was detected in the most vacuoles or tubulovesicles in both type of CA-rich cells in the turtle bladder, indicating that the part of plasma membrane was internalized in the apical cytoplasmic vacuoles. It seems quite likely that CA-rich cells possess intracellular vesicles carrying proton pumps which are recycling back to the apical plasma membrane. In turtle bladder, the granular cells actively secrete large quantities of mucin and other proteins by an exocytotic mechanism in an apparently constitutive fashion. The possibility that bladder epithelial cells secrete mucin via a regulated secretory pathway has not been rigorously examined and much is still to be determined about these issues from this cell type.

  • Comparative study on dynamic change of canopy structure during ripening period were carried out by using newly bred high yield rice cultivar (IR 667-Suwon 213) and a commercial variety, Jinhung in relation to nitrogen nutrition. The results were as follows. 1. Canopy structure pattern (vertical distribution of dry matter density at heading)was vertical type for Jinhung and horizontal type for IR 667. 2. The vertical distribution pattern of leaf area density (or weight) in the canopy was central dominant type for IR 667 while apical dominant type for Jinhung. 3. Canopy conservation pattern and percent distribution pattern of leaf area density followed the vertical distribution pattern of leaf area density. 4. Canopy persistence was weaker in IR 667, thus they have smaller canopy conservation ratio indicating faster senescence. 5. Slow supply of nitrogen (sulfur coated urea) showed a trend to change the apical dominant pattern into the central dominant pattern by the conservation of central portion, and it-resulted in higher yield though nitrogen nutrition did little affect canopy pattern. 6. The central and apical dominant pattern appeared to be well matched to the upper leaf-dependent type and the lower leaf-dependent type of grain yield, respectively.

Establishment of intestinal organoids from small intestine of growing cattle (12 months old)

  • Recently, we reported the robust in vitro three-dimensional (3D) expansion of intestinal organoids derived from adult bovine (> 24 months) samples. The present study aimed to establish an in vitro 3D system for the cultivation of intestinal organoids derived from growing cattle (12 months old) for practical use as a potential alternative to in vivo systems for various purposes. However, very few studies on the functional characterization and 3D expansion of adult stem cells from livestock species compared to those from other species are available. In this study, intestinal crypts, including intestinal stem cells, from the small intestines (ileum and jejunum) of growing cattle were isolated and long-term 3D cultures were successfully established using a scaffold-based method. Furthermore, we generated an apical-out intestinal organoid derived from growing cattle. Interestingly, intestinal organoids derived from the ileum, but not the jejunum, could be expanded without losing the ability to recapitulate crypts, and these organoids specifically expressed several specific markers of intestinal stem cells and the intestinal epithelium. Furthermore, these organoids exhibited key functionality with regard to high permeability for compounds up to 4 kDa in size (e.g., fluorescein isothiocyanate [FITC]-dextran), indicating that apical-out intestinal organoids are better than other models. Collectively, these results indicate the establishment of growing cattle-derived intestinal organoids and subsequent generation of apical-out intestinal organoids. These organoids may be valuable tools and potential alternatives to in vivo systems for examining host-pathogen interactions involving epithelial cells, such as enteric virus infection and nutrient absorption, and may be used for various purposes.
