산사(山査)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響) (Effect of CRATAEGII FRUCTUS(山査) on Antioxidant Capacity in D-galactose induced Aging Rats)

  • Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.

산화된 low density lipoprotein (LDL)에 의해 유도된 내피세포의 손상에 대한 마늘 대사산물인 allylmercaptan의 보호 효과 (Protection Effects of Allylmercaptan, Metabolite of Garlic on Endothelial Cell Injury Induced by Oxidized Low Density Lipoprotein)

  • Low density lipoprotein (LDL)의 산화는 동맥경화의 유발과 진행에 결정적 역할을 하는 것으로 알려져 있다. 본 연구에서는 마늘의 주요 대사산물인 allylmercaptan의 산화 low density lipoprotein에 의해 손상된 내피세포의 보호효과에 대하여 실험하였다. Allylmercaptan의 항산화 활성은 thiobarbituric acid substance (TBARS)로 측정하였다. Allylmercaptan은 0.1, 1 및 10 mM의 농도에서 $Cu^{2+}$에 의해 유도된 LDL의 산화를 용량의존적으로 억제하였다. 폐동맥 내피세포를 $37^{\circ}C$, 5% $CO_2$ 상태에서 24시간 동안 미리 배양시킨 후 세측한 다음 다시 24시간 동안 0.1 mg/ml oxidized LDL (ox-LDL)을 첨가하여 배양하였다. 이 때 ox-LDL이 Lactate dehydrogenase (LDH)의 방출과 glutathione (GSH)를 감소시키는 원인으로 세포막 손상의 지표로 LDH와 GSH 함량을 조사하였다. 본 실험 결과 allylmercaptan을 일정 농도 별로 endothelial cell에 첨가하여 배양했을 때 LDH의 방출과 GSH의 감소를 현저하게 억제하였다. Peroxide를 형광분석법으로 24 well plate에서 직접 측정한 결과 allylmercaptan이 폐동맥 내피세포 내에서 ox-LDL 유도 peroxide의 방출을 억제하였다. 그리고 allylmercaptan은 과산화수소의 소거능도 있었다. 본 실험결과 allylmercaptan은 ox-LDL 유도 폐동맥 내피세포를 보호할 수 있었으므로 allylmercaptan은 동맥경화의 예방에 유용할 것으로 생각된다.

옥죽(玉竹)의 지질강하 및 항산화효과 (Effect of Polygonatum Odoratum on Lowering Lipid and Antioxidation)

  • Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.

인삼 추출물이 난소를 절제한 백서의 항산화효소 활성에 미치는 영향 (The Effects of Panax Ginseng Extract on Antioxidative Enzyme Activity in Ovariectomized Rats)

  • 노화과정에 산소라디칼이 관여할 가능성을 노화유도된 쥐를 동물모델로 하여 알아보았다. 난소를 절제한 rat와 난소를 절제하지 않은 정상의 rat에서의 생리활성에 미치는 인삼 추출물의 효과를 연구하였다. 난소를 절제한 rat에 각각 GTS (Ovx+GTS 군)와 GW (Ovx+GW 군)를 투여하고, 이것과 비교하기 위해 난소를 절제한 대조군(Ovx군)과 마지막으로 난소를 절제하지 않은 정상군(Sham군)으로 하여 항산화효과에 관한 실험방법으로 진행하였다. 노화유도된 쥐의 각 조직에서 lipid peroxidation이 증가되었고 free radical반응이 더 심하게 일어난 간 조직에서 노화진행이 그만큼 촉진되었음을 알 수 있었다. 간소라디칼반응이 항진된 원인을 규명코자 간 조직에서 미토콘드리아와 세포질분획에서 항산화효소의 활성을 조사한 결과, SOD와 glutathion reductase활성이 감소되었으며 노화나 지질과산화가 심하게 진행되었던 간 조직에서 이들 항산화효소의 활성감소가 심한 경향을 나타내었다. 생체는 free radical을 조절할 수 있는 다양한 항산화계가 존재하여 free radical을 효율적으로 제거함으로써 생체의 항상성을 유지할 수 있다. 이러한 항산화계에는 superoxide dismutase (SOD), glutathion (GSH) peroxidase, GSH reductase, GSH S-transferase, catalase등의 항산화효소가 있다. SOD는 반응성을 가진 산소species인 superoxide anion을 macrophage와 polymorphonuclear leucocyte의 항균작용같은 세포방어기구에서 중요한 역할을 한다. 원칙상 눈, 피부등의 byaus이 공기중의 산소에 노출되면 oxidative stress가 커짐으로서 조직이 훼손되나 SOD같은 효소에 의해 보호되고 손상이 방지되는 것으로 알려지고 있다. 실제로 신체부위에 적용해 보면 쉽게 활성상실이 일어나므로 fatty arid와 공유결합시켜 효과적으로 superoxide radical을 제거할 수 있다. AST의 활성은 Ovx군이 Sham군에 비해 2.11배 높게 나타났으며, ALT 활성은 변화가 나타나지 않았다. AST 활성에서는 각각의 물질을 투여한 군에서는 유의적인 감소 경향을 보였다. 과산화지질함량은 Ovx군이 Sham군에 비해 높게 나타났으며, 각각의 인삼추출물을 투여한 군에서는 62%∼72%의 저해률을 보였다. 항산화 효소들은 Sham군에 비해 Ovx군에서 낮게 나타났는데, 각 물질의 투여 군에서는 유의성있는 증가를 보였다. 이러한 결과에서 보듯이, 난소 절제로 생성된 많은 free radical은 항산화효소에 의해서 감소되었으며, 그리고 인삼 추출물이 항산화 효소 대신에 노화를 저해하는 작용을 하는 것으로 사료되었다.

$\beta$-Glucan Suppresses LPS-stimulated NO Production Through the Down-regulation of iNOS Expression and $NF{\kappa}B$ Transactivation in RAW 264.7 Macrophages

  • The antioxidant and anti-inflammatory protective effects of $\beta$-glucan from barley on RAW 264.7 murine macrophage cells induced by lipopolysaccharide (LPS) were examined. The RAW 264.7 murine macrophages were preincubated with various concentrations ($0-200\;{\mu}g/mL$) of $\beta$-glucan and stimulated with LPS to induce oxidative stress and inflammation. The $\beta$-glucan treatments were found to reduce thiobarbituric acid-reactive substance (TBARS) accumulation, and enhance glutathione levels and the activities of antioxidative enzymes, including superoxide dismutase (SOD), catalase, glutathione reductase, and glutathione peroxidase (GSH-px) in the LPS-stimulated macrophages as compared to the LPS-only treated cells. Nitric oxide (NO) production was significantly suppressed in a dose-dependent manner (p<0.05) with an $IC_{50}$ of $104\;{\mu}g/mL$. Further treatment with $\beta$-glucan at $200\;{\mu}g/mL$ suppressed NO production to 2% of the LPS-control, and suppressed the levels of inducible nitric oxide synthase (iNOS) protein and mRNA in a dose-dependent manner. The specific DNA binding activity of nuclear factor ${\kappa}B\;(NF{\kappa}B)$ was significantly suppressed by $\beta$-glucan treatment with an $IC_{50}$ of $220\;{\mu}g/mL$ in a dose-dependent manner. Finally, barley $\beta$-glucan ameliorates NO production and iNOS expression through the down-regulation of $NF{\kappa}B$ activity, which may be mediated by attenuated oxidative stress in RAW 264.7 macrophages.

비타민 C 보충 및 영양 교육이 흡연 남자 청소년의 영양지식, 식행동과 체내 일부 항산화 관련 효소체계에 미치는 영향 (The Effects of Vit C Supplementation and Nutrition Education on Nutrition Knowledge, Food Habits and Antioxidative Enzyme Activity in Male Adolescent Smokers)

  • The study evaluated the effectiveness of intervention for male adolescent smokers by making an assessment in terms of changes in food habits, nutrition knowledge, plasma catalase, superoxide dismutase(SOD), glutathione peroxidase peroxidase(GSH-px) activities and thiobarbituric acid reactive substance(TBARS) after Vit C supplementation and nutrition education. The subjects, male adolescent smokers, were assigned into four groups : Control group(19 students), education(Educ.) group(19 students), Vit C supplementation (suppl) group(19 students), and Educ. + Vit C suppl. group(19 students). The Educ. group and Educ. + Vit C suppl. group received nutrition education once a week for 2 weeks. The Vit C suppl. group Educ. + Vit C suppl. group received 500 mg ascorbic acid for 35 days. All data were collected before intervention and after intervention. Nutrition knowledge of those who received education increased, and the frequency of fruit and yellow-green vegetable consumption also increased. Plasma antioxidant enzyme activities were not different except for the SOD activity in the Educ. + Vit C suppl. group, which was significantly increased. The plasma ceruloplasmin level of groups that received Vit C supplementation was reduced more than any other groups, and the specific ceruloplasmin ferroxidase activity of groups that received Vit C supplementation was elevated more than other groups. These intervention programs had an impact on food habits, nutrition knowledge, plasma antioxidant enzyme activities, and plasma TBARS in male adolescent smokers. Various nutrition education programs must be implemented for adolescent smokers, and further studied are needed regarding sorts and amount of antioxidant nutrients and supplementation duration.

간유(肝兪)(BL18) 및 곡지(曲池)(LI11) 산삼약침 처치가 비만쥐의 지질구성, cytokine, 간장기능, 항산화능에 미치는 영향 (Effects of cultivated wild ginseng pharmacopuncture at Gansoo (BL18) and Gokji (LI11) on lipid composition, cytokine level, liver function and antioxidative capacity in diet-induced obese rats)

  • Objevtives & Methods : Effects of cultivated wild ginseng pharmacopuncture at BL18 and LI11 on lipid composition, cytokine level, liver function, anti-oxidative capacity and histological characters were investigated in diet-induced obese rats. Forty male Sprague-Dawley rats weighing about 400g were divided into 4 groups of control, BL18, LI11 and BL18 plus LI11 pharmacopuncture groups and raised for 4 weeks. Results : 1. Plasma ${\beta}$-lipoprotein, free fatty acids level and TNF-${\alpha}$ levels significantly decreased in the pharmacopuncture groups compared to those of no treatment group. Plasma and liver total cholesterol, triglyceride, glucose and thiobarbituric acid reactive substance (TBARS) levels were also significantly lower than those of no treatment group. There was, however, no difference in TBARS level among pharmacopuncture groups. Liver total cholesterol level of BL18 pharmacopuncture group was lower than those of the other two pharmacopuncture groups. In LDL-cholesterol level, BL18 pharmacopuncture and BL18 plus LI11 pharmacopuncture groups only had significantly lower levels than that of no treatment group. 2. There was no significant difference between cultivated wild ginseng pharmacopuncture groups and no treatment group in IL-6, alanine transaminase (ALT), aspartic acid transaminase (AST) levels. 3. Compared with \ those of no treatment group, pharmacopuncture groups had significantly higher levels of HDL-cholesterol, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase activities. There was, however, no significant difference among pharmacopuncture groups. 4. Histological characters of heart, kidney and liver of BL18 pharmacopuncture group were similar to those of normal rats. Conclusions : These results indicate that cultivated wild ginseng pharmacopuncture at BL18 and LI11 may suppress adipose tissue mass and lipid peroxidation and activate antioxidant system.

Forsythiaside, a Constituent of the Fruits of Forsythia suspense, Ameliorates Scopolamine-Induced Memory Impairment in Mice

  • Forsythiaside is a polyphenolic constituent of the fruits of Forsythia suspense Vahl which are widely used as anti-inflammatory herbal raw materials in traditional Chinese medicine. In the present study, the authors assessed the effects of forsythiaside on learning and memory impairments induced by scopolamine using a passive avoidance and the Morris water maze tests in mice. Drug-induced amnesia was induced by scopolamine treatment (1 mg/kg, i.p.). Forsythiaside (10 mg/kg, p.o) administration significantly prevented scopolamine-induced step-through latency reduction in the passive avoidance test and scopolamine-induced increased escape latency in the Morris water maze test (p<0.05). Moreover, in an ex-vivo study, forsythiaside treatment (10 mg/kg, p.o) significantly reduced the increase of thiobarbituric acid reactive substance levels induced by scopolamine (p<0.05). Taken together, the present study suggests that forsythiaside could be useful for the treatment of cognitive impairment, and that its beneficial effects are mediated, in part, by its antioxidative properties.

Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과 (Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

가미소건중탕(加味小建中湯)이 D-galactose로 노화(老化)를 유발(誘發)시킨 백서(白鼠)의 항산화능(抗酸化能)에 미치는 영향(影響) (Effect of Gamisogunjung-tang on Antioxidation Activity in Rats Induced Aging by D-Galactose)

  • Objective : This experiment has been done to evaluate the effects of Gamisogunjung-tang(GST) on antioxidant capability and lipidic concentration in blood which are presumed to be related to aging. Method : In this study, we divided 14 weeks old SD rats into normal group, control group and GST group. Control and GST group were induced aging by D-galactose. At the same time GST group were administered extract of Gamisogunjung-tang for 6 weeks. After then we took blood, and measured the activities of SOD, GSH-px, catalase in erythrocytes and measured TBARS values, concentrations of total lipid, tryglyceride, total cholesterol, HDL-cholesterol in plasma. Results : The activities of SOD, GSH-px, catalase in erythrocytes increased significantly in GST group compared with control group. The value of TBARS and the concentration of total lipid, total cholesterol in plasma decreased significantly in GST group compared with control group. The concentration of HDL-cholesterol increased significantly in GST group compared with control group. The concentration of triglyceride were not noticeable. Conclusion : it is considered that Gamisogunjung-tang has an influence on control aging by activation the antioxidative enzyme systems in erythrocytes and decreasing the concentration of lipid in blood plasma.
