Effects of cultivated wild ginseng pharmacopuncture at Gansoo (BL18) and Gokji (LI11) on lipid composition, cytokine level, liver function and antioxidative capacity in diet-induced obese rats (간유(肝兪)(BL18) 및 곡지(曲池)(LI11) 산삼약침 처치가 비만쥐의 지질구성, cytokine, 간장기능, 항산화능에 미치는 영향)

  • Objevtives & Methods : Effects of cultivated wild ginseng pharmacopuncture at BL18 and LI11 on lipid composition, cytokine level, liver function, anti-oxidative capacity and histological characters were investigated in diet-induced obese rats. Forty male Sprague-Dawley rats weighing about 400g were divided into 4 groups of control, BL18, LI11 and BL18 plus LI11 pharmacopuncture groups and raised for 4 weeks. Results : 1. Plasma ${\beta}$-lipoprotein, free fatty acids level and TNF-${\alpha}$ levels significantly decreased in the pharmacopuncture groups compared to those of no treatment group. Plasma and liver total cholesterol, triglyceride, glucose and thiobarbituric acid reactive substance (TBARS) levels were also significantly lower than those of no treatment group. There was, however, no difference in TBARS level among pharmacopuncture groups. Liver total cholesterol level of BL18 pharmacopuncture group was lower than those of the other two pharmacopuncture groups. In LDL-cholesterol level, BL18 pharmacopuncture and BL18 plus LI11 pharmacopuncture groups only had significantly lower levels than that of no treatment group. 2. There was no significant difference between cultivated wild ginseng pharmacopuncture groups and no treatment group in IL-6, alanine transaminase (ALT), aspartic acid transaminase (AST) levels. 3. Compared with \ those of no treatment group, pharmacopuncture groups had significantly higher levels of HDL-cholesterol, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase activities. There was, however, no significant difference among pharmacopuncture groups. 4. Histological characters of heart, kidney and liver of BL18 pharmacopuncture group were similar to those of normal rats. Conclusions : These results indicate that cultivated wild ginseng pharmacopuncture at BL18 and LI11 may suppress adipose tissue mass and lipid peroxidation and activate antioxidant system.

The Relationship between Some Blood Parameters and Antioxidant Enzyme Activity in Korean Postmenopausal Women (일부 폐경 후 여성에서 영양상태 및 골대사 관련 일부 혈액 지표와 항산화효소 활성과의 관련성 분석)

  • To elucidate the relationship between blood parameters related bone metabolism and antioxidant enzyme activity in postmenopausal period 60 women residing in Iksan area were recruited. Food and nutrient intake of each individual subject were estimated by 24-hour recalls of 3 non-consecutive days. The biochemical markers including total protein, albumin, osteocalcin (intact bone gla protein; BOP), calcium, phosphorus and hemoglobin were measured in fasting blood. In addition, parameters of antioxidative capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) , catalase (CAT) and total antioxidant capacity (TA) were monitored in blood, also. The mean age, height, weight, and BM! of subjects were 64.8 years, 151.1 em, 59.5 kg $26.0\;kg/m^2$, respectively. The mean SOD, GPx, and CAT activities were 138.5 U/ml, 1,273.8 U/ml and 314.3 kU/l respectively, and TA was 1.16 mmol/l without significant difference among different age groups. BMI was positively correlated with SOD activity (p < 0.01). SOD activity and CAT activity showed positive correlation with serum albumin (p < 0.05) and hemoglobin (p < 0.01). In conclusion, this study revealed that antioxidant enzyme activity holds a significant relationship with the blood parameters like as serum albumin and hemoglobin in postmenopausal women and further systematic research is needed to investigate the their relation mechanism.

Anticytotoxic and Radical Scavenging Activities of Acer tegmentosum Maxim Stem Extracts (산겨릅나무 줄기 추출물의 세포독성 억제 및 라디칼 소거 활성)

  • The objective of this study was to investigate anticytotoxic and antioxidatative capacities of ethanol extracts from Acer tegmentosum Maxim (A. tegmentosum) stem in vitro. The extract at concentration of 200 ug/mL inhibited 10 and 20 ug/mL arsenic trioxide-induced cytotoxicity of HepG2 cells by 79.3 and 57.5%, respectively. The extract at concentration of 200 ug/mL inhibited 0.2 and 0.5 mM t-BHP-induced cytotoxicity of HepG2 cells by 66.3 and 35.7%, respectively. Antioxidative effects of the extract were examined via measurement of ABTS, superoxide, and peroxyl radical scavenging activities. ABTS radical scavenging activity of the extract was higher than that of ${\alpha}$-tocopherol. Superoxide scavenging activity of the extract was higher than that of catechin. Oxygen radical absorbance capacity of the extract was higher than that of ascorbic acid. Cupric reducing antioxidant capacity of the extract was higher than that of ${\alpha}$-tocopherol. The extract at concentrations of 100 and $500{\mu}g/mL$ inhibited 10 mM t-BHP-induced lipid peroxidation of HepG2 cells by 38.2 and 80.7%, respectively. The extract prevented supercoiled DNA strand breakage induced by hydroxyl or peroxyl radical. Total phenolic and flavonoid contents of the extract at concentration of $100{\mu}g/mL$ were 71.3 nmol/mL gallic acid and 18.8 nmol/mL catechin equivalents, respectively. Thus, strong cytoprotective and antioxidant effects of A. tegmentosum stem extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in polyphenolic contents.

The impact of Caesalpinia Sappan L. on Oxidative Damage and Inflammatory Relevant Factor in RAW 264.7 Cells and HUVEC (소목(蘇木)이 산화적 손상 및 Raw 264.7 cell과 HUVEC에서의 염증 유관 인자에 미치는 영향)

  • Objectives : This study investigated the impact of Caesalpinia sappan L. on oxidative damage and inflammatory relevant factor in RAW 264.7 cells and human umbilical vein endothelial cells (HUVEC). Methods : We determined whether fractionated EtOH extracts of Caesalpinia sappan L. (CSL) inhibit free radical generation such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), reactive oxygen species (ROS) and nitric oxide (NO) and pro-inflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 cells and HUVEC. Result : 1. DPPH removal capacity was increased by CSL. 2. LPS-induced ROS, and NO inhibitory capacity were increased by CSL. 3. LPS-induced cell death of Raw 264.7 cells was decreased by CSL. 4. The amount of cytokine generation in Raw 264.7 cell was decreased significantly by CSL. 5. The amount of cytokine generation in HUVEC was decreased significantly by CSL. Conclusions : These results suggest that CSL supplement may attenuate oxidative stress by elevated antioxidative processes, and suppress inflammatory mediator activation.

Effect of Ojayeonjonghwan(五子衍宗丸) on Antioxidant Capacity in D-galactose Induced Aging Rats (오자연종환(五子衍宗丸)이 노화유발(老化誘發) 흰주의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives : Ojayeonjonghwan(五子衍宗丸) is composed of Polygonum multiflorum THUNB, and some medical herbs that are known as formula of senescence delay effects. The purpose of this study is to investigate th effect of Ojayeonjonghwan on antioxidant enzyme activities, such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione preoxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and Ojayeonjonghwan administered group (D-galactose 50mg/kg and Ojayeonjonghwan extracts 245.0mg/200g 1time/day for 6 weeks, OJY Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, Plasma total lipid, Plasma triglyceride and cholesterol were measured in rat erythrocytes and blood plasma. Results : TBARS in plasma concentration of OJY group was significantly lower than that of control group. Red blood cell(RBC) SOD activity of OJY group was significantly higher than that of control group(F=16.057, p=0.0001, ANOVA test), RBC GSH-px activity of OJY group was increased(F=4.271, p=0.034, ANOVA test). RBC catalase activities of all experimental group were not significantly different. Total lipid and triglyceride concentration in plasma of all experimental groups were not significantly different. Total cholesterol concentrations in plasma of OJY group were significantly lower than those of control group(F=4.387, p=0.032, ANOVA test). Conclusions : According to the above results, it is considered that Ojayeonjonghwan is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

Antioxidative Capacity and Quality Characteristics of Yanggaeng added with Beaknyuncho (Opuntia ficus-indica var. saboten) Powder (백년초 분말을 첨가 제조한 양갱의 항산화활성과 품질 특성)

  • This study investigated the physicochemical and sensory characteristics of yanggaeng prepared with various amounts of Opuntia ficus-indica var. saboten powder (as ratios of 1%, 3%, 5%, 7% to the total materials). According to the results, the pH of yanggaeng decreased significantly with the addition of Opuntia ficus-indica var. saboten powder (p<0.001). Increasing the amount of Opuntia ficus-indica var. saboten powder in the yanggaeng tended to increased $^{\circ}Brix$ value (p<0.01). For color values, lightness (L) values were reduced by increasing the amount of Opuntia ficus-indica var. saboten powder, redness (a) and yellowness (b) while values were increased significantly (p<0.001). Texture measurement scores in terms of hardness, springness, and Adhesiveness for yanggaeng showed that sample groups were lower than those of the control group. Cohesivene and chewiness were higher in the additive group than in the control group. Antioxidant activities, evaluated by DPPH radical scavenging capacity, Nitrites cavenging activity, and total phenolic substances content increased when increased the amount of Opuntia ficus-indica var. saboten powder (p<0.001). Results of descriptive analysis showed that by increasing the amount of Opuntia ficus-indica var. saboten powder, some attributes such as the intensity of the color, sweet, hardness and chewiness increased (p<0.01). It concluded that yanggaeng containing Opuntia ficus-indica var. saboten can be prepared with good sensory properties and antioxidant activities, and the addition of 3% Opuntia ficus-indica var. saboten powder was found to be ideal. Sensory evaluation scores in terms of color, odor, taste, texture, and overall preference of groups with 3% of Opuntia ficus-indica var. saboten powder did not show any significant differences when compared to the control group. Based on the above results, this study suggests that the addition of 3% Opuntia ficus-indica var. saboten powder may be the best substitution ratio for yanggaeng.

Antioxidative capacity of hydrolyzed rapeseed cake extract and oxidative stability of fish oil-in-water emulsion added with the extract

  • Rapeseed cake was extracted with 80% ethanol and then fractionated with $H_2O$ (fraction I) as well as with 30% (II), 50% (III), 70% (IV), and 100% ethanol (V). Total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric-reducing antioxidant potential, and Trolox equivalent antioxidant capacity were in the order of fractions II > III > I > IV > V. The three fractions with high antioxidant activities and TPC (I, II, and III) were pooled and hydrolyzed by NaOH solution, resulting in 18.97 mg sinapic acid/g hydrolyzed extract and 21- and 2.2-fold increases in TPC and DPPH radical scavenging activity, respectively. Hydrolyzed rapeseed cake extracts (200, 500, and 1,000 ppm) and catechin (200 ppm) as a comparison were added to 10% fish oil-in-water emulsion, and their effects on oxidative stability were investigated by measuring hydroperoxide values (PV) during refrigerated storage. PVs were significantly lower in the emulsions with added hydrolyzed extract as compared to the control (p<0.05) and significantly decreased with increasing extract concentration (p<0.05) over a period of 29 days. The emulsion added with hydrolyzed extract showed higher PV than that added catechin at the same concentration (200 ppm) during 13-22 days (p<0.05), but after then, the PV was not significantly different (p>0.05). This study indicates that hydrolyzed rapeseed cake extract rich in sinapic acid may inhibit oxidation in a fish oil-in-water emulsion in a concentration-dependent manner.

Antioxidative and Antimicrobial Activities of Castanopsis cuspidata var. sieboldii Extracts (구실잣밤나무 추출물의 항산화 및 항균 활성)

  • This study was designed to investigate the possible utilization of Castanopsis cuspidata as a source of antiseptic agents. The leaves of C. cuspidata, extracted by 80% ethanol, were sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and n-butanol. In order to effectively screen for a natural preservative agent, we first investigated the antioxidant activities such as DPPH radical scavenging capacity, superoxide radical scavenging capacity, and xanthine oxidase inhibitory activity of the C. cuspidata ethanol extracts and fraction. Using a screening system, we found that the ethylacetate fraction had the strongest antioxidant activity, which followed a dose-dependent manner. The antimicrobial activities were shown in the ethylacetate fraction of C. cuspidata. Among the five fractions, the ethylacetate fraction showed the highest antimicrobial activities against microorganisms tested, which were Bacillus sublitis, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella enteritidis and Salmonella typhimurium. In addition, leaf extracts of C. cuspidata could be suitable for the development of food preservatives.

Evaluating Pre-silicon Treatment to Alleviate Drought Stress and Increases Antioxidative Activity in Zoysia japonica

  • This study was performed to determine the effects of silicon on zoysiagrass after the application of drought stress. The daily amount of water or scilicon solution was 150 ml per a pot. For 14 days, plants were treated with 0.1 and 1.0 mM silicon (Si) and with distilled water for control and the drought only-treatment. Afterward, the plants in Si and drought treatment were exposed to a 21-day under drought stress condition but the plants in control received water. The results indicated that the growth and the moisture and chlorophyll contents decreased in the drought only-treatment and 0.1 mM Si compared to the control. However, 1.0 mM Si showed an increase in the growth with a significant increase of water and chlorophyll contents. The MDA and $H_2O_2$ concentrations and electrolyte leakage decreased, while the radical scavenging capacity increased in 1.0 mM Si. 1.0 mM Si showed little to no differences in the growth and no differences in water and chlorophyll contents, electrolyte leakage, MDA and $H_2O_2$ concentrations and antioxidant capacity compared to the control. These results suggested that application of silicon is useful for drought tolerance improvement of zoysiagrass under drought that is occurring in turf fields.

Vitamin C and antioxidant capacity stability in cherry and romaine during storage at different temperatures (저장 조건에 따른 로메인 및 체리의 비타민 C 함량 및 항산화능의 변화)

  • Purpose: The aim of this work was to study the change in antioxidant activity depending on storage temperature and storage period in romaine and cherry. Method: The plant material was stored at $0.7{\pm}0.6^{\circ}C$, $3.5{\pm}2.8^{\circ}C$, and $4.7{\pm}1.4^{\circ}C$. Cherry and romaine were stored for a period of 9 days and 7 days, respectively. The cherry was taken from each group of samples at regular intervals of days and the romaine was taken from each group of samples at regular intervals of 2days. Vitamin C, total polyphenol, and total flavonoid stability and antioxidant capacity including DPPH, total antioxidant capacity (TAC) were measured. Results: For cherry, the levels of TAC and flavonoid were higher at the $0.7{\pm}0.6^{\circ}C$ condition than other conditions (p < 0.05). The polyphenol and vitamin C levels were not significantly different among storage conditions. In the case of romaine, the level of TAC was highly preserved until 7 days at the $0.7{\pm}0.6^{\circ}C$ condition. Vitamin C level was significantly lower at the $3.5{\pm}2.8^{\circ}C$ condition (p < 0.05). DPPH activity was highest at the $0.7{\pm}0.6^{\circ}C$ condition (p < 0.05). DPPH activity was shown in order of $0.7{\pm}0.6^{\circ}C>4.7{\pm}1.4^{\circ}C>3.5{\pm}2.8^{\circ}C$. Conclusion: The results indicated that the narrow differences and fluctuation in temperature were associated with antioxidant capacity and it might enhance the nutritional shelf life of vegetables and fruits.
