Studies on the Antioxidative Effect of Korean Traditional Medicinal Plants as Skin Care Product (피부보호용품제조를 위한 한국재래약초의 항산화성 연구)

  • The total phenolic contents and antioxidative activities of the ethanol and methanol extracts from 9 Korean medicinal herbs were tested for the application of skin care products such as soaf and lotion. For the evaluation of antioxidative activity of the extract the method of electron donating ability with resolution reagent DPPH(1 ,1-diphenyl-2-picryl hydrazyl) and TBARS (Thiobarbituric acid reactive substances) value were used in comparison with α-tocopherol and BHA. As the results, the total phenolic contents of the extracts calculated as chlorogenic acid were ranged from 0.063-0.124 mg/ml. All the medicinal plant extracts showed some appreciable antioxidative effects. Among them, Zanthoxylum piperitum, Cinnamomum cassia and Caesalpinia sappan were shown to have relatively high antioxidative effects comparable with BHA and α-tocopherol..

Varietal Variation in Antioxidative Activity of Rice Grain by DPPH and TBA Methods

  • This study was to investigate antioxidative activity of rice grain using 1, 1-Diphenyl-2-picrylhydrazyl(DPPH) and thiobarbituric acid (TBA) method and germination ability for screening rice varieties with high antioxidative activities on korean native and foreign rice varieties harvested in 1998 and 1999. The average antioxidative activity of foreign rice varieties (DPPH 63.5% and TBA 55.2%) was significantly higher than that of native rice varieties (DPPH 47.2% and TBA 45.6%) on varieties harvested in 1999. The promptness index (PI) of native rice varieties was higher in stored rices for three months (mean PI=160.7) than that of stored rices for a year (mean PI=141.6). On the other hand, the PI of foreign rice varieties was higher in stored rices stored for a year (mean PI=176.7) than that of stored rices for three months (mean PI=157.5). Varieties with high redness of hulled rice (a-value) showed significant lipid peroxidation inhibitory activity to DPPH in a stored rices for a year (r=${0.5744}^**$) and stored rices for three months (r=${0.5630}^**$) . These results indicate that the pigments of hulled rice varieties may play important antioxidative roles and colored rice varieties with higher antioxidative potentials can be developed and also may provide information with rice breeder to breed rice variety with a high antioxidative activity for a rapid screening of a small amounts of a large number of samples using color value.

Evaluation of Antioxidative Effects of Lactobacillus plantarum with Fuzzy Synthetic Models

  • Numerous studies suggest that the effects of lactic acid bacteria (LAB) on oxidative stress in vivo are correlated with their antioxidative activities in vitro; however, the relationship is still unclear and contradictory. The antioxidative activities of 27 Lactobacillus plantarum strains isolated from fermented foods were determined in terms of 2,2-diphenyl-1-picrylhydrazyl, hydroxyl radical, and superoxide radical scavenging abilities, reducing activity, resistance to hydrogen peroxide, and ferrous chelating ability in vitro. Two fuzzy synthetic evaluation models, one with an analytic hierarchy process and one using entropy weight, were then used to evaluate the overall antioxidative abilities of these L. plantarum strains. Although there was some difference between the two models, the highest scoring strain (CCFM10), the middle scoring strain (CCFM242), and the lowest scoring strain (RS15-3) were obtained with both models. Examination of the antioxidative abilities of these three strains in $\text\tiny{D}$-galactose-induced oxidative stress mice demonstrated that their overall antioxidative abilities in vitro could reveal the abilities to alleviate oxidative stress in vivo. The current study suggests that assessment of overall antioxidative abilities with fuzzy synthetic models can guide the evaluation of probiotic antioxidants. It might be a more quick and effective method to evaluate the overall antioxidative abilities of LAB.

Antioxidative and Anti-Inflammatory Activities of Salvia plebeia R. Br Extracts (배암차즈기(Salvia plebeia R. Br.)의 항산화 활성 및 항염증효과)

  • This study provides data to explore functional medicinal food materials that can prevent adult diseases, and verified antioxidant and anti-inflammatory of each solvent fraction of the methanol extract of Salvia plebeia R. Br. in Korea. In the analysis of total phenol content, DPPH radical scavenging ability, and FRAP reduction ability as indicators of antioxidant activity, the methanol fraction and ethyl acetate fraction of the Salvia plebeia R. Br. group showed high antioxidant activity. Ethyl acetate fraction of Salvia plebeia R. Br. methanol extract also showed excellent antioxidative activity as compared with BHT. In the mouse macrophage line Raw 264.7 cells, the NO production ability by LPS treatment was significantly increased in the LPS treatment group compared to the untreated group. In inflammatory reactions induced by LPS treatment in Raw 264.7 cells, inflammatory cytokines (TNF-α, PGE2, IL-1β) and NO production were decreased in the EtOAc fraction and MeOH fraction of the methanol extract of Salvia plebeia R. Br. compared to the case of LPS treatment alone. The anti-inflammatory effect was proved by significantly inhibiting the production of inflammatory cytokines. The present results suggest that Salvia plebeia R. Br. supplementation is beneficial for the suppression of antioxidant and anti-inflammation.

Antioxidative activity of some natural products which have been orientally used as ophthalmic drugs (한방에서 안질환에 사용되어 온 천연물들의 항산화 작용)

  • Antioxidative activities of methanol extracts of 7 natural products which have been orientally used or studied as ophthalmic drugs were investigated. All of 7 extracts assayed - Rubus coreanus, Dendrobium monile, Eriocaulon sieboldianum, Ligustrum japonicum, Cassia tora, Cicadae Periostacum and Polygonatum odoratum var pluriflorum had some electron-donating ability to OPPH radicals, in which R. coreanus and D. monile showed strong activity as similar as ascorbic acid or BHT. At the concentration of $200{\mu}g/m{\ell}$, methanol extract of L. japonicum showed superoxide dismutase-like activity the same as 0.1 mM ascorbic acid and methanol extract of R. coreanus inhibited effectively the production of ABTS radical produced by $H_2O_2$. Furthermore, methanol extract of C. tora showed the highest nitrite scavenging ability. The inherent antioxidative activities of these natural products appear to be responsible for its ophthalmic effects.

The Antioxidative, Antimicrobial and Nitrite Scavenging Effects of Schizandra chinensis RUPRECHT(Omija) Seed (오미자종자의 항산화성, 항균성, 아질산염소거능)

  • This study was carried out to examine the antioxidative, antimicrobial and nitrite scavenging effects of various solvent extracts and different solvent fractions from Schizandra chinensis RUPRECHT(Omija) seed. The results were as follows; The antioxidative activities using lard, soybean oil and linoleic acid were the highest in methanol, ethanol and methanol extract from omija seed, respectively. The free radical scavenging activity using DPPH method was the strongest in acetate fraction of methanol extract from seed. The methanol extract from omija seed had the strongest antimicrobial activities to L. plantarum, B. subtilis, E. coli and P. citrinum, while ethyl acetate extract had the strongest against S. aureus and S. typhimurium. The buthanol fraction from methanol extract had the highest antimicrobial activities, followed by B. subtilis, L. plantarum, E. coli, S. aureus and S. typhimurium. The nitrite scavenging ability was pH dependent, highest at pH 1.2 and lowest at pH 6.0. The buthanol fraction of methanol extract from omija seed had more effective nitrite scavenging ability than other fractions of extracts.

Analysis on the Antioxidant Activity of Catechin Concentrations and Green Tea Extract Powder (녹차 분말과 카테킨의 농도에 따른 항산화력 비교 분석)

  • Antioxidative activities of catechin from green tea extracts were examined by the methods of 1,1-Diphenyl-2-picrylhydrazyl (DPPH) electron donating ability, hydroxy scavenging activity and the inhibitory effect on xanthine oxidase activity. The resulted demonstrated the fact that Catechin one (containing Green tea extracts plus catechin 51%) of green tea extracts product showed at 65.5% in electron donating activity on the DPPH. The electron donating ability on the DPPH of Catechin one was increased to 10% than purified catechin. Catechin one showed the activity at 64.5% in scavenging activity using hydroxy radical method. To the Catechin one provided to increase the hydroxy scavenging activity up to 3 fold. Inhibitory effects of the catechin one measured with xanthine oxidase method was 6.5%. Although the antioxidative activity of catechin (98% purified) was lower than that of Catechin one (containing Green tea extracts plus catechin 51%) in same catechin concentrations ($5{\mu}g$, respectively). Therefore, we may suggest that Catechin one can be used as a functional food additive possessing the potent antioxidative activity.

Effect of Amino Acids Addition on Stability and Antioxidative Property of Anthocyanins (아미노산의 첨가가 anthocyanins 색소의 안정성과 항산화능력에 미치는 영향)

  • Effects of amino acids (Arg, Lys, Gly, Ile, Glu, Asp, and Met) on the color intensity, stability and antioxidative properties of anthocyanins extracted from grape skins were investigated. Intensity of anthocyanins was significantly increased by the addition of Asp. Except for basic amino acids such as Arg and Lys, stabilities of anthocyanins were significantly improved by the addition of other amino acids including neutral, acidic and sulfur containing amino acids during the storage at $30^{\circ}C$ at pH 3.5. In case of control anthocyanins was remained unchanged the intensity of red color decreased significantly during the storage whereas their antioxidative activity were unchanged. Although effects of amino acids addition on electron donating abilities of anthocyanins were not differentiated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method, the addition of Asp or Met resulted in increased ferric reducing ability which measured by FRAP (ferric reducing ability of plasma) assay.

Antioxidative Activity of Beverage with Water and Ethanol Extracts of Maegmundong (Liriope platyphylla) (맥문동의 물추출액 및 에탄올추출액 이용 음료의 산화방지 활성)

  • The antioxidative activities of beverages with water or ethanol extracts from Maegmundong (Liriope platyphylla) were investigated with free amino acid and mineral analysis, as well as saponin, total phenolics and flavonoid determination. Antioxidative activity was evaluated by electron donating ability and ABTS radical scavenging activity. There was only a small difference between water extract beverages and ethanol-extract beverages with respect to their electron donating ability and ABTS radical scavenging activity, although the total saponin, total phenolics and flavonoid were found to be greater in the ethanol extract beverages than in the water extract beverages. Three major free amino acids of the Magemundong beverages with water- or ethanol extract were asparagine (58.30, 60.68 mg%), methionine (15.10, 13.95 mg%, respectively) and proline (12.31, 14.00 mg%, respectively). The most abundant mineral in the Maegmundong beverage with water and ethanol extract was potassium (238.68, 244.32 mg%, respectively).

Varietal Difference in Antioxidative Activity of Ethanolic Extracts from Colored Rice Bran (유색미 에탄올 추출물의 품종간 항산화 활성 변이)

  • Interrelation between the antioxidative activities of 70% ethanol extracts from bran fraction of 23 kinds of colored rice and chuchung as a control were examined. Antioxidative activities were evaluated by assaying reducing power, electron-donation ability to DPPH free radical, scavenging activity of hydroxy radical $({\cdot}OH)$ generated through Fenton reaction and inhibitory activity on lipid peroxidation using linoleic acid autoxidation system, respectively. Among 24 varieties of colored rice LK 1-3-6-12-1-1 had the strongest reducing power followed by Elwee, DZ 78, Jumlalocal-1 and SC-45 in decreasing order. The electron-donating ability to DPPH radical was higher in order of HP 883-1-1-1-B-1-1, HP 833-1-3-1-1-1, LK 2-7-12-1-1 and DZ 78. The hydroxy radical scavenging activity was higher in order of DK-1, IR 1544-38-2-2-1-2-2, SC-5 and SC-45 but LK 2-7-12-1-1 had oxidative effect. In the liaoleic acid autoxidation model system, RGS No 336, LK 1B-2-1-1, LK 1B-4-12-1-1, LK 1A-2-12-1-1, LK 2-7-12-1-1 and HP 883-1-1-1-B-1-1 exhibited strong antioxidative activities but Elwee, Jumlalocal-l and SC-45 showed to have oxidative effects. The rice variety of highest pigment content was Elwee and the next were RGS-No 336, IR 1544-38-2-2-1-2-2 and SC-5 with the order of higher content. The reducing power was correlated with the quantity of the pigment in the ethanolic extract of rice bran and SC-5 showed relatively high antioxidative activity in every results of antioxidative activity tests.
