목향 추출물의 항산화 및 항암활성 (Antioxidative and Antitumor Activity of Extracts from Saussurea lappa)

  • This study was carried out to investigate the antioxidative and antitumor activities of Saussurea lappa for the purpose of developing a functional food. The methanol extract of Saussurea lappa was fractionated with five solvents (hexane, chloroform, EtOAc, BuOH, water) and examined for antioxidative activities and xanthine oxidase inhibitory activity in addition to growth inhibitory activity of human cancer cell (HT-29, SNU-1, HeLa). Total phenol compound contents were higher in EtOAC fraction than other fractions. Also, electron donating ability was over 90% at $500{\mu}g/ml$ (93.1 %) and $1000{\mu}g/ml$ (94.3%). The hexane fraction revealed stronger nitrite scavenging ability than the positive control (ascorbic acid) and its abilities were 22.4% and 42.8% at $500{\mu}g/ml$ and $1000{\mu}g/ml$, respectively. Xanthine oxidase inhibitory activity had similar tendency to electron donating ability. The EtOAc fraction showed high inhibition to xanthien oxidase activities at $500{\mu}g/ml$(81.9%) and $1000{\mu}g/ml$(90.4%). In the antitumor activity test, the hexane fraction exhibited potent growth inhibition activity against HT-29, SNU-1 and HeLa cells. Therefore, we believe that Saussurea lappa can be developed into a functional food with antioxidant activity. Additional studies are required for the hexane and chloroform fractions of Saussurea lappa to develop them into therapeutic supplements for stomach cancer, colon cancer, and cervical cancer.

오미자의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidative Activities of Omija(Schizandra chinensis Bailon))

  • To develop physiological functionality of Omija extracted with water was evaluated on antioxidative activity. Omija, high acid material with pH 3.6, contain $57.5{\pm}1.03%$ of moisture, and $18.8{\pm}0.12%$ of crude fat. This material have $12.6{\pm}0.04%$ of carbohydrate and $11.1{\pm}0.07%$ of crude protein as well, but ash and crude protein contents were found less than 10%. 10 mineral contents were also found, too; K and Ca showed the highest level, then Al, Mg, Na and Mn were followed. In composition amino acid contents, glutamic acid took the largest portion, $131.7{\pm}1.3$ mg/100 g, aspartic acid $51.5{\pm}0.6%$, and other composition amino acid under 50%. In case of free sugar contents, 7 types were found. Most of them were glucose and fructose. Total phenolic compounds showed the highest level, $2,862.6{\pm}31.7$ mg/100 g. $197.8{\pm}14.6$ mg/100 g of flavonoid and $225.6{\pm}18.2$ mg/100 g were included. In terms of electron donating ability, radical scavenging ability activated as the amount of Omija extract increased. In particular, Omija extract in 1,000 ${\mu}g/m{\ell}$ demonstrated almost similar electron donating ability, $72.4{\pm}0.21%$, to BHT. It was also found that antioxidant activities of electron donating ability, SOD-like ability, hydroxyl radical scavenging ability and nitrite scavenging ability were highly promoted as Omija extract concentration increased. The nitrite scavenging ability was significant when the extract belonged to strong acid region and doping concentrations increased.

홍미삼 에탄올 추출분획의 항산화 활성 (Antioxidative Activity of Ethanol Extraction Fraction from the Korean Red Tail Ginseng)

  • 본 연구의 목적은 홍미삼의 기능성 연구의 일환으로 총 페놀성 화합물의 추출방법과 DPPH에 대한 수소공여능, linoleic acid의 산화방지활성 및 LDL에 의한 항산화활성 등을 중심으로 연구한 결과이다. 홍미삼에 60% 에탄올를 첨가하여 추출회수별 총 페놀성 화합물을 조사한 결과 3회까지 추출했을 때 대부분이 추출되었기 때문에 추출횟수는 3회가 적당한 것으로 판단되었다. 60% 에탄올 추출액의 수율은 3회까지 추출했을 때 약 37.35%가 추출되었다. 추출 용매에 따른 총 페놀성 화합물의 추출 효율은 물 추출구를 100%로 했을 때, 60% 에탄올 용액으로 추출한 뒤 농축하여 물로 부피를 재조정한 시험구는 122%로 조사되었다. 항산화활성 조사에서 DPPH에 대한 수소공여능에서 시간이 경과함에 따라 항산화 활성이 약간씩 증가하나 활성은 약한 편이었다. Linoliec acid 산화에 대한 산화억제 효과는 1,500 ppm 농도에서 약 72.23%의 저해율을 나타났으며, LDL에 대한 산화방지 효과는 250 ppm 농도에서 그 저해율이 약 22.52%로 나타났다.

식방풍의 성분분리 및 생리활성 (Biolosical Activities of Isolated Compounds from Peucedani Radix)

  • In this study, isolation of antioxidative compounds was performed for development of anti-oxidizing agent. $CHCl_3$, $H_2O$, 30%, 60% MeOH, MeOH fractions were examined antioxidative activity by DPPH, test of inhibition on NO production. It was revealed that 30%, 60% MeOH and $CHCl_3$ fractions had significant antioxidative activity. In 30% MeOH and 60% MeOH, $CHCl_3$ fraction, six compounds were isolated and elucidated as adenosine(I), guanosine(II), peucedanol 7-O-$\beta$-D-apiofuranosyl(1$\rightarrow$6)-$\beta$-glucopyranoside(III), peucedanol 7-O-$\beta$-D-glucopyranoside(IV), peucedanol(V) and scopoletin(VI) by physicochemical data and spectroscopic methods. (Negative FAB-MS, $^{1}H-NMR$, $^{13}C-NMR$). The results from antioxidative activity screening for the each compound showed that compound IV was relatively superior antioxidant ability. In anti-inflammatory activation assay, compound III, IV, VI had concentration-dependent-activity and compound IV had superior anti-inflammatory ability. These results suggest that Peucedani Radix might be developed as a potent anti-oxidative, anti-inflammatory agents and ingredients for related functional foods.

느릅나무 추출물의 항산화 효과 및 아질산염 소거능 (Antioxidative Activities and Nitrite Scavenging Abilities of Extracts from Ulmus devidiana)

  • Oil emulsion에 느릅나무 근피(한국산, 중국산), 수피(한국산) 각 추출물 0.05% 첨가에 의한 항산화 작용의 실험 결과는 다음과 같다. 활성 산소종, 금속이온 참가에 따른 지방 산화 억제능은 근피, 수피 각 용매 추출물의 $H_2O_2,-OH,\;KO_2$에 대한 포집능은 모두 우수하여 높은 항산화 효과를 나타내었으며, 각 추출물과 $Fe^{2+}$ ion과 $Cu^{2+}$ ion도 강한 활성 유리 라디칼 포집능으로 우수한 항산화 효과를 보였다. 지방 산화를 촉진하는 nonheme iron 함량에서 $Fe^{2+}$ 함량과 total iron 함량은 근피(중국산) 에탄올, 메탄올 추출물이 가장 높은 함량을 나타내었고, 한국산 근피, 수피의 에탄올, 메탄올 그리고 부탄올 추출물군에서 80% 이상의 높은 활성을 보였고, 물 추출물군의 경우 20~55%의 범위로 낮은 효과를 보였다. SOD 유사활성은 근피(중국산) 부탄올 추출물, 수피(한국산) 물 추출물에서 가장 높은 활성능을 보였다. 아질산염 소거능의 경우도 근피, 수피 에탄올, 메탄올 그리고 부탄올 추출물군 모두 우수한 소거능을 보였고, 특히 pH 1.2에서는 가장 높은 아질산염 소거능을 보였으며, pH 6.0으로 갈수록 급격히 감소하였다.

The Antioxidative Effect of Eclipta prostrata L. Extract on Cultured NIH3T3 Fibroblasts Injured by Manganese-Induced Cytotoxicity

  • Manganese (Mn) is used as main materials in various chemical processes of industry, but it suggested that Mn brings about its toxicant by fume or dust through respiratory system and skin barrier. Mn toxicant induces the loss of mental health and life quality by cerebrovascular and skin diseases. Nevertheless, it lefts much unknown on the mechanism and the effectively therapeutic methods about Mn toxicant. Therefore, this study was evaluated the cytotoxicity induced by manganese dioxide ($MnO_2$) in cultured NIH3T3 fibroblasts, and also, the correlation between $MnO_2$-induced cytotoxicity and oxidative stress was examined. While, the effect of Eclipta prostrata L. (EP) extract belong to Compositae was assessed against $MnO_2$-induced cytotoxicity in the view of antioxidative effect for searching the natural resources mitigating or preventing the $MnO_2$-induced cytotoxicity. In this study, $MnO_2$-induced cytotoxicity was revealed as mid-toxic by Borenfreud and Puerner's toxic criteria, and catalase (CAT), an antioxidant prevented $MnO_2$-induced cytotoxicity by the remarkable increase of cell viability in these cultures. While, in the protective effect of EP extract on $MnO_2$-induced cytotoxicity, EP extract effectively prevented the cytotoxicity induced by $MnO_2$ via antioxidative effects such as xanthine oxidase (XO) inhibitory ability and DPPH-radical scavenging ability. From the above results, EP extract showed the effective prevention against $MnO_2$-induced cytotoxicity correlated with oxidative stress by antioxidative effects. Conclusively, this study may be useful to research or development the alternatively therapeutic agent from natural resources like EP extract for the treatment of diseases resulted in oxidative stress.

황산카드뮴독성의 산화적 손상에 대한 부들 추출물의 항산화 효과 (Antioxidative Effect of Typha orientalis L. Extract on the Oxidative Stress Induced by Cytotoxicity of Cadmium Sulfate)

  • Objectives: This study was carried out to analyze the cytotoxicity of cadmium sulfate ($CdSO_4$) and the antioxidative effect of Typha orientalis L. (TO) extract on the oxidative stress induced by cytotoxicity of $CdSO_4$ in the cultured NIH3T3 fibroblasts. Methods: For this study, the cell viability and the antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide dismutase (SOD)-like activity and xanthine oxidase (XO)-inhibitory activity were assessed. Results: The cadmium sulfate significantly decreased cell viability in dose-dependently, and $XTT_{50}$ value was measured at $47.4{\mu}M$ of $CdSO_4$. The cytotoxicity of $CdSO_4$ was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The butylated hydroxytoluene (BHT) as antioxidant significantly increased cell viability injured by $CdSO_4$-induced cytotoxicity in these cultures. In the protective effect of TO extract on $CdSO_4$-induced cytotoxicity, TO extract remarkably increased the inhibitory ability of LP and XO as well as SOD-like ability. Conclusions: From the above results, it is suggested that the oxidative stress is involved in the cytotoxicity of $CdSO_4$, and TO extract effectively protected $CdSO_4$-induced cytotoxicity by antioxidative effects. The natural component like TO extract may be a putative therapeutic agent for treatment of the toxicity induced by heavy metallic compound like $CdSO_4$ correlated with the oxidative stress.

맨드라미, 비트, 홍갓 색소 추출물의 항산화 및 항균 효과 (Antioxidant and Antibacterial Activity of Extracts from Brassica juncea czerniak et coss., Celosia cristata L., and Beta vulgaris L.)

  • We sought to study the qualities and scientific benefits of Dongchimi, a traditional Korean food. We compared and analyzed ingredients used for the appearance and storability of dongchimi - honggot (Brassica juncea czerniak et coss), cockscomb (Celosia cristata L.), and beet Beta vulgaris L.). We specifically examined the antioxidative and antibacterial activity of pigments from extracts of these ingredients. Distilled water ($H_2O$) and 1% citric acid were used to safely extract pigments. The antioxidative activity of the pigments was then measured for total phenolic compounds, SOD (Super Oxide Dismutase), and EDA (Electron Donation Ability) by DPPH. The antibacterial activity of was also assessed by a Paper disc solution. Our results show that the pigments had sufficient antioxidative activity and had antibacterial properties against Gram positive and negative bacteria. In particular, Cockscomb (used for enhancing color) contained the highest amount of polyphenol compounds and had the most efficient antioxidative activity.

Screening of Antioxidative Activity of Bifidobacterium Species Isolated from Korean Infant Feces and Their Identification

  • Among 59 Korean isolated, 20 were confirmed as members of the genus Bifidobaferium species based on gram staining, microscopic examination of cell morphology and the TLC method, The oxygen tolerance and antioxidative activities of these 20 Bifidobacterium strains and 5 standard Bifidobaferium strains were tested. All the strains demonstrated antioxidative activities as regards inhibiting linoleic acid peroxidation. The antioxidative activities of isolated and standard strains were found to range from 10.7-46.4% and from 10.7-22.2%, respectively. In addition, all tested strains exhibited a Scavenging ability on DPPH free radicals, range from 15-41% for the isolated strains and 8.3-22% for the standard strain. Accordingly. the isolated Bifidobarterium strains demonstrated higher antioxidative artivities than the 5 standa rd Bifidobarterium strains. On the base of grades for each test, HJL 7511 was identified 35 the best strain, followed by HJL 7501. 2 strains were identified with Polymerase Chain Reaction (PCR) assay using group-specific primers designed from the nucleotide Sequences of the 16S rRNA and internal transcribed spacer (ITS) regions of the Bifidobacteria. Based on the Sequencing results, HJL 7511 and HJL 7501 were identified as Bifidobacterium infantis.

매염제인 ZnSO4의 피부독성에 대한 멍석딸기 추출물의 항산화 및 미백효과 (Antioxidative and Whitening Effects of Rubus parvifolius L. Extract on Dermal Cytotoxicity of ZnSO4, Mordant)

  • 본 연구는 매염제인 황산아연(ZnSO4)의 피부독성을 배양 피부세포주인 SK-MEL-3 세포를 재료로 산화적 손상측면에서의 조사와 함께 ZnSO4의 독성에 대한 멍석딸기(RP)의 영향을 항산화와 미백효과 측면에서 알아보았다. 본 실험을 위하여, 세포생존율, DPPH-라디칼 소거능 및 melanin합성 저해능을 분석하였다. 본 연구에서 ZnSO4는 농도 의존적으로 세포생존율을 유의하게 감소시켰으며, XTT50값이 173.3 uM로 중간 독성으로 나타났다. 또한, 항산화제의 일종인 ascorbic acid는 ZnSO4에 의하여 손상된 세포생존율을 유의하게 증가시켰다. 한편, ZnSO4의 독성에 대한 RP 추출물의 영향에서, RP 추출물 처리는 유의한 세포생존율의 증가와 함께 DPPH-라디칼 소거능과 melanin합성 저해능을 통하여 항산화와 미백효과를 나타냈다. 결론적으로, RP 추출물과 같은 천연성분은 향후 항산화제와 미백제로서의 대체물질 개발에 있어 활용적 가치가 클 것으로 생각된다.
