연근 추출물에서 주름개선 소재의 개발 (Development of Anti-Wrinkle Agent from Nelumbo nucifera Root Extract)

  • 본 연구에서는 연근의 추출물에서 기능성화장품 소재를 개발하고자 하였다. 용매는 70~100%의 에탄올을 이용하였으며, 연근의 유효성분으로 nuciferine을 확인하였다. 연근 추출물의 화장품소재로서의 응용가능성을 보기 위해 안전성시험(MTT assay), 주름개선 효과시험(Elastase 활성저해 시험) 및 항산화효과시험(DPPH free radical scavenging assay)을 실시하였다. 100% 에탄올로 연근을 추출할 경우 $100{\mu}g/ml$의 농도에서 세포생존율이 80% 이상이어서 연근 추출물은 화장품소재로서 비교적 안전함을 알 수 있었다. 또한 연근 추출물은 $100{\mu}g/ml$의 농도에서 elastase 활성억제가 40~50%를 보여주어서 비교적 우수한 주름개선 효과를 나타내었다. 50% 항산화 저해능을 보이는 농도($FSC_{50}$)는 $5.0{\sim}38{\mu}g/ml$로 연근 추출물의 항산화효과가 매우 우수하였다. 따라서 연근 에탄올추출물은 주름개선 기능성화장품 소재로서 그 가능성이 높음을 알 수 있었다.

강지통맥음(降脂通脈飮)이 항산화 활성과 고지혈(高脂血) 유발(誘發) 백서(白鼠)에 미치는 영향(影響) (The Effects of Gangzitongmaekeum on Antioxidation Activity and Dietary Hyperlipidemia-induced Mice)

  • This experiments was performed to determine the effects of Gangzitongmaekeum(降脂通脈飮 : GTE) on antioxidation activity and hyperlipidemia induced by hypercholesterolemic diet in mice. The results obtained were as follows : 1. GTE showed a safety in cytotoxicity and toxicity of human fibroblast cells and liver. 2. GTE showed DPPH scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory effect on reactive oxygen species (ROS). 3. GTE treated mice showed body and liver weight decrease, compared to the hyperlipidemia-induced control group. 4. GTE decreased total cholesterol and LDL cholesterol levels significantly, but HDL cholesterol levels not significantly. 5. GTE decreased triglyceride levels significantly. 6. Glucose levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 7. Albumin levels in GTE treated mice were similar with the hyperlipidemia-induced control group. 8. The lipophagy in liver compared with the control group tended to be decreased in GTE treated mice. In the change of aorta, the cell was regular and boundary of vessel wall was clear, compared to the hyperlipidemia-induced control group. 9. TBARS levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 10. The change of SOD and catalase activity significantly increased compared to the hyperlipidemia-induced control group. 11. ACAT mRNA level and HMG-CoA reductase mRNA levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. Conclusion : These results suggest that Gangzitongmaekeum is effective in antioxidation activity and dietary hyperlipidemia-induced mice.

Retinol에 대한 항산화 연구 (The Study on Antioxidation of Retinal)

  • Retinol에 대한 합성물질과 천연물질의 항산화력을 비교하였다. 수용성 합성 항산화제 tertiary butylhydroquulone(TBHQ)와 수용성 천연 항산화제 $\alpha$-glycosyl rutin($\alpha$-G rutin), licorice, pycnogenol, 지용성 합성 항산화제 butylated hydroxytoluene(BHT)와 지용성 천연 항산화제 $\alpha$-lipoic acid, ferulic acid, natural concentrated tocopherol(no-tocopherol)를 각각 0.0, 0.0l, 0.02, 0.05, 0.l0wt% 사용하여 리포좀에 봉입하였으며, 4$^{\circ}C$, $25^{\circ}C$, 5$0^{\circ}C$에서 8주 동안 retinol의 잔류량 변화를 HPLC로 측정하였다. 사용된 항산화제는 retinol에 대하여 모두 산화억제효과를 나타내었으며, 수용성 항산화제는 licorice>pycnogenol>TBHQ>$\alpha$-G rutin 순으로, 지용성 항산화제는 $\alpha$-lipoic acid>BHT>no-tocopherol>ferulic acid 순으로 나타났다. 또한 $\alpha$-lipolc acid, BHT, licorice, pycnogenol을 흔합하여 retinol에 대한 항산화력 상승효과를 알아보았다. $\alpha$-lipoic acid와 BHT를 혼합하여 사용하였을 때 retinol에 대한 항산화력이 가장 좋게 나타났다.

감초 추출물의 약리적 특성 및 분석 (A Study on the Pharmacetical Characteristics & Analysis of Glycyrrhizin Extract)

  • From experiment results on pharmacetical characteristics and analysis of Glycyrrhizin extract, some conclusions are obtained as follows. From results on extract experiment of Glycyrrhizin, it appeared about 8%-extraction ratio as semi-solid state, and after dried in freezing from Glycyrrhizin extract of semi-solid state, it obtained about 70%-Glycyrrhizin extract as solid state of yellow gold color. From results on antimicrobial experiment of Glycyrrhizin extract, number of S-typhimurium and Fungus in microbe decreased more and more according to time passage. This phenomenon shows that Glycyrrhizin extract keeps antimicrobial effect. From results on antioxidation experiment of Glycyrrhizin extract, DPPH scavenging activity of free radical shows that Glycyrrhizin extract appears more remarkable reduction ability than reference samples. This phenomenon means that antioxidation of Glycyrrhizin extract appears higher than Vitamin-C and BHA. From results on instrument analysis, the fatty and aromatic components of 2-pentanone, cyclohexasiloxane, tetrasiloxane, benzoquinoline-2-carboxylic acid etcs from Glycyrrhizin extract was detected with GC/MS and inorganic components of Ca, Mg, Ti, Zn, Fe etcs from Glycyrrhizin extract was detected with ICP/OES.

양파 알코올 추출물의 항산화 효과 및 중금속 제거 가능성 연구 (Studies on the Heavy Metals Elimination and Antioxidation of the Onion Ethnolic Extract)

  • In order to study for Onion ethanolic extract on the Heavy Metal Elimination and antioxidation, the peroxide values of oil and eliminated metal were analyzed. The results are follows: It was very effective as a retardant for autoxidation processing of the soybean and olive oil by the Onion ethanolic extract. Quercetin in the Onion ethanolic extract was affected as a ligand for chelating with some metals. 녹차 추출물의 약리적 특성 및 분석 (A Study on the Pharmacetical Characteristics and Analysis of Green-tea Extract)

  • From the result of pharmacetical characteristics and analysis of Green-tea extract, it could obtain some conclusions as follows. The extract experiment of Green-tea appeared about 10%-extraction ratio as semi-solid state, and after dried in freezing from Green-tea extract of semi-solid state, it obtained about 65% Green-tea extract as solid state. In the results on antimicrobial experiment of Green-tea extract, number of S-typhimurium and fungus in microbe decreased more and more according to the time passage. This phenomenon could show that Green-tea extract keeps antimicrobial effect. In the results on antioxidation experiment of Green-tea extract, DPPH scavenging activity of free radical showed that Green-tea extract appears more remarkable reduction ability than reference samples. This phenomenon means that antioxidation of Green-tea extract appears higher than Vitamin-C and BHA sample. In the results on intrument analysis, the fatty and aromatic components of aniline, acetaldehyde, acetic acid, trichloroethylene, caffeine etcs from Green-tea extract was detected with GC/MS analysis and inorganic components of Ca, Mg, Cu, Mo, Sb, Ti etcs from Green-tea extract was detected with ICP/OES analysis.

호박잎의 항산화 효과 연구 (Experimental Study on Effects of Cucurbita moschata Duch. on Antioxidation)

  • In recent year, We are concerned about anti-aging, disease-protection, long-life, many methods are used in solving this problem. And Those are related with antioxidative ability. Recently, We heard that Cucurbita moschata Duch. has anti-hypertensive effect and good for health. So I let made a experiment for this result of the anti-oxidant effect of Cucurbita moschata Duch. used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity and Bovine serum albumin(BSA). The results of this study were as follows: We measured level of DPPH radical scavenging activity. And we obtained results that the ability of DPPH radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $20\;mg/m{\ell}$. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $10\;mg/m{\ell}$. When we inspected antioxidation with Bovine serum albumin(BSA). And we obtained that antioxidative ability was increased after$2.5\;mg/m{\ell}$ of Cucurbita moschata Duch.. So I guess that Cucurbita moschata Duch. has effect of antioxidative ability. Hereafter we need differential experimental methods for evidence of antioxidative effect on Cucurbita moschata Duch.

장기간 다른 강도의 운동이 항산화 효소의 활성도와 지질 과산화에 미치는 영향 (The Effect of Long-term Exercise of Different Intensity on the Activation of Antioxidation Enzyme and Lipid Peroxidation)

  • This study was to investigate the effect of 10th weeks Exercise of Different Intensity on the activation of antioxidation enzyme and lipid peroxidation. 15 subjects were divided into the 65$\%$ running exercise(EG; 5), 85$\%$ running exercise(EG: 5), and the control(CG: 5) groups. The exercise group had 10 week of running exercise 4 times a week with ACSM(1995) protocol, 65$\%$ of HRmax from the beginning to 5th week, 85$\%$ of HRmax from 6th to 10th week. The subjects had 60 min exercise for each day. After extracting the blood sample, the activation of antioxidation enzyme and lipid peroxidation were compared between the groups. The one way ANOVA was conducted about pretest, 5th week, and 10th week of the subject data. The results obtained from this study were as follows; 1. The 65$\%$ exercise group showed the significantly greater increase in the activation of SOD after 10 weeks for the rest and all-out exercise. (p < .05). 2. The 65$\%$ exercise group displayed the significantly higher increase in the activation of CAT after 10th weeks for the rest exercise. (p < .05). 3. The 65$\%$ exercise group had the significantly greater decrease in the activation of MDA after 10 weeks for the all-out exercise. (p < .05).

추출용매에 따른 브로콜리의 항산화와 항염증에 대한 효과 (Effects of Broccoli on Anti-inflammation and Anti-oxidation According to Extraction Solvent)

  • 본 실험에서는 여드름 치료에 사용되던 국소 레티노이드제와 항생제를 대용 할 수 있고 화장품에 사용되는 원료로 추출하여 항여드름 치료제 또는 화장품 원료를 개발하기 위해 실험한 결과 broccoli proplyeneglycol 그룹에서 DPPH, superoxide radical, nitric oxide assay 에서 모두 vitamin C에 가까운 활성을 나타내었고 paper disk diffusion test에서는 broccoli ethanol과 broccoli hexane 그룹에서 P. acne의 저해 활성을 나타낸 것으로 보아 항여드름 치료제 또는 화장품 원료로써의 유용 가치가 있다고 사료 되어진다.
