To investigate antioxidant effects of tea tree root extracts using various extraction methods, cytotoxicity, DPPH and ABTS radical scavenging, SOD, nitrite scavenging activity and inhibitory activity of lipid peroxidation, reducing power, ferrous ion chelating activity were measured. Cytotoxicity for RAW 264.7 cells was not observed at concentrations treated with below 90 ㎍/mL in all extracts. The maximum DPPH radical, nitrite scavenging, SOD activity and inhibitory activity of lipid peroxidation were obtained at the ethylacetate and 70% ethanol extract. The maximum ABTS radical scavenging activity was obtained at the ethylacetate and hot water extract. However, in the case of reducing power and ferrous ion chelating activity, they were obtained at 70% ethanol and hexane extract, respectively. Nitrate scavenging activity showed the most excellent scavenging ability of 59.6% at 90 ㎍/mL of ethylacetate. The hexane extract had the highest ferrous ion chelating activity, showing 61.05% at 50 ㎍/mL, 66.07% at 70 ㎍/mL and 76.81% at 90 ㎍/mL, respectively. The results of this research show that the ethylacetate and 70% ethanol extracts of tea tree root can be used as a natural material for scavenging the radicals. However, future study is necessary to understand the mechanism of antioxidant activity by identification of substances.
Recently, as a natural substance has been emphasized interest in research to enhance the immune function. Green lettuce (Lactuca sativa L.) is a popular vegetable used fresh and it contains various phytochemicals and antioxidant compounds, and has been reported to have various physiological activities such as antibacterial, antioxidant, antitumor and anti-mutagenic. However, only a few studies have investigated on the mechanism of action of immune-enhancing activity of lettuce. Therefore, in this study, the immunomodulatory activities and potential mechanism of action of Green lettuce extracts (GLE) were evaluated in the murine macrophage cell line RAW264.7. GLE significantly increased NO levels by RAW264.7 cells, as well as expressions of immunomodulators such as iNOS, COX-2, IL-1β, IL-6, IL-12, TNF-α and MCP-1. Although GLE activated ERK1/2, p38, JNK and NF-κB, GLE-mediated expressions of immunomodulators was dependent on p38, JNK and NF-κB. In addition, TLR4 inhibition blocked GLE-mediated expressions of immunomodulators and activation of p38, JNK and NF-κB. Taken together, these results demonstrated that TLR4-MAPK/NF-κB signalling pathways participated in GLE-induced macrophage activation and GLE could be developed as a potential immunomodulating functional food.
P. glehnii, an evergreen conifer found in northern areas, is known as a cold-resistant species. In this experiment, we measured the water content, PSⅡ efficiency, chlorophyll fluorescence, pigments of the xanthophyll-cycle and activity of enzymes of the ascorbate-glutathione cycle during cold acclimation and at subsequent low-temperature conditions to examine the importance of acclimation to cold tolerance. P. glehnii showed a decrease in PSⅡ efficiency (especially in Fv) during cold acclimation and at subsequent low temperatures. However, cold-acclimated needles showed higher PSⅡ efficiency at low temperatures than nonacclimated needles. In addition, 0-YON (first-year needles) showed an increase in $\beta$-carotene and lutein, while 1-YON (one-year-old needles) immediately developed an antioxidant mechanism in the ascorbate-gluthathione cycle as soon as they were exposed to low temperature and both 0-YON and 1-YON showed increased zeaxanthin and de-epoxidation ratios at continuous low temperature. Based on our results, we suggest that P. glehnii maintain PSⅡ efficiency at low temperature by effectively protecting the photosynthetic apparatus from photo-damage by rapid induction of an antioxidant mechanism in 1-YON and dissipation of excess energy by $\beta$-carotene and lutein in 0-YON.

Osteoarthritis is thought to be induced by the ageing-related loss of homeostatic balance between degeneration and repair mechanism around cartilage tissue in which inflammatory mediators such as reactive oxygen species, cytokines and prostaglandins are prone to overproduction under undesirable physiological conditions. Phlorotannins are unique polyphenolic compounds bearing dibenzo-1,4-dioxin skeleton which are not found in terrestrial plants but found only in some brown algal species such as Ecklonia and Eisenia families. Phlorotanninrich extracts of Ecklonia cava including LAD103 showed significant antioxidant activities such as DPPH radical scavenging, ferric ion reduction, peroxynitrite scavenging, and inhibition of LDL oxidation, indicating their possible antioxidative interference both in onset and downstream consequences of osteoarthritis. LAD103 also showed significant down regulation of $PGE_2$ generation in LPS-treated RAW 246.7 cells, and significant inhibition of human recombinant interleukin-$1{\alpha}$-induced proteoglycan degradation, indicating its beneficial involvement in pathophysiological consequences of osteoarthritis, the mechanism of which needs further investigation. Since LAD103 showed strong therapeutic potentials in arthritic treatment through several in vitro experiments, it is highly encouraged to perform further mechanistic and efficacy studies.

This study was performed to investigate the antioxidant mechanism of tomato wine with varying lycopene content in rats fed a high fat diet (HFD). Male Sprague-Dawley rats were randomly divided into five groups (n=10 per group) and fed an HFD (35% of total energy from fat) plus ethanol (7.2% of total energy from alcohol), tomato wine with varying lycopene content (0.425 mg%, 1.140 mg% or 2.045 mg% lycopene) or an isocaloric control diet for 6 weeks. Mice fed HFD plus ethanol significantly increased erythrocyte hydrogen peroxide and thiobarbituric acid reactive substances (TBARS) levels with increases in activities of erythrocyte antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and glutathione reductase (GR) compared to pair-fed rats. Supplementation of tomato wine with varying lycopene content decreased ethanol-mediated increases of erythrocyte lipid peroxidation and antioxidant enzyme activities in HFD-fed rats, and tomato wine with higher lycopene appeared to be more effective. Tomato wine also dose-dependently lowered TBARS levels with decreased pro-oxidant enzyme, xanthine oxidase (XOD) activity in plasma of HFD-fed rats. In contrast to erythrocytes, the inhibitory effects of tomato wine on hepatic lipid peroxidation were linked to increased hepatic antioxidant enzymes (SOD and CAT) and alcohol metabolizing enzyme (alcohol dehydrogenase and aldehyde dehydrogenase) activities. There were no significant differences in hepatic XOD and cytochrome P450-2E1 activities among the groups. Together, our data suggest that tomato wine fortified with lycopene has the potential to protect against ethanol-induced oxidative stress via regulation of antioxidant or pro-oxidant enzymes and alcohol metabolizing enzyme activities in plasma, erythrocyte and liver.
Purpose: Ligustri Lucidi Fructus and Ecliptae Herba has long been used for clinical therapy associated especially with menopausal symptoms in Korea. To provide a scientific rationale for such use, we have investigated the antioxidant and vasorelaxant effects of Ligustri Lucidi Fructus, Ecliptae Herba and its mixture. Methods: The antioxidant activity of the extracts from Ligustri Lucidi Fructus, Ecliptae Herba and its mixture were evaluated and compared with that of BHA (butylated hydroxyanisole), BHT (butylated hydroxytoluene), vitamin C and vitamin E, using the ${\alpha}$,${\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH) radical scavenging method. Results: Antioxidant activity of all extracts using the DPPH radical scavenging method decreased in the order vitamin C>BHA>vitamin E>Ligustri Lucidi Fructus>Ligustri Lucidi Fructus:Ecliptae Herba(1:1)>Ecliptae Herba>BHT. The vasorelaxant effects of extracts were investigated on the vasomotor tone of the rat thoracic aorta in an organ bath. All of the extracts elicited along-term relaxing response in the endothelium-intact as well as endothelium-denuded rat aorta contracted with norepinephrine. This relaxant effect was abolished by Precontraction with 72 mM KCI. Thus, it is suggested that the mechanism of vasorelaxant effect of extracts not involve voltage-operated $Ca^{2+}$ channel blocking but receptor-mediated route. Conclusion: These antioxidant and vasorelaxant effecs of the extracts may contribute to the beneficial effects in postmenopausal women.

Antioxidants are involved in the defense mechanism against the attack of free radicals. This study was carried out to determine the antioxidant activity of new variety of Glycyrrhiza cultivar radix, Wongam and Sinwongam. Dissolved freeze dried Wongam and Sinwongam extracts were filtered by $0.2{\mu}m$ filter and serially diluted at the concentrations of $10{\mu}g/mL$, $50{\mu}g/mL$, $100{\mu}g/mL$, $500{\mu}g/mL$, and $1000{\mu}g/mL$. The antioxidant potential was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, ABTS (2,2-azino-bis (3-rthylbenzthiazoline-6-sulfonic acid) diammonium salt) radical cation decolorization assay, nitrite radical scavenging assay, and ferric reducing antioxidant power (FRAP) assay. DPPH radical scavenging activities (i.e. the highest value $50.9{\pm}0.8%$ by Wongam and $82.6{\pm}1.1%$ by Sinwongam), ABTS radical scavenging activities (i.e. the highest value $88.1{\pm}1.8%$ by Wongam and $98.6{\pm}0.1%$ by Sinwongam), and nitrite radical scavenging activities (i.e. the highest value $87.3{\pm}1.6%$ by Wongam and $89.8{\pm}0.8%$ by Sinwongam) increased in a dose-dependent manner. In addition, ferric reducing power activities also increased in a dose-dependent manner. The FRAP value of Wongam and Sinwongam extracts were $0.72{\pm}0.03$ and $0.99{\pm}0.06$ compared to ascorbic acid, as a positive control, was $1.32{\pm}0.02$. These results suggested that Wongam and Sinwongam have beneficial effects as a potent antioxidant.

Storage temperature profoundly influences the storability of paprika (Capsicum annuum L.). However, the impact of storage temperature on storability and its association with the antioxidant activity of paprika are poorly understood. In this study, we evaluated the storage attributes, activity, and gene expression levels of antioxidant enzymes in paprika stored at 4, 10, and 20℃ for 14 d and then at 20℃ for an additional 5 d (14+5 d; retail conditions). Storage at 10℃ effectively mitigated pitting, stalk browning, shriveling, and decay while significantly enhancing the marketability of paprika. The fruits stored at 4℃ were prone to pitting, whereas those stored at 20℃ were sensitive to stalk browning and decay. Moreover, paprika stored at 10℃ exhibited higher 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) activity and total phenolic content than those stored at 4 and 20℃, indicating improved antioxidant activity. Additionally, storage at 10℃ upregulated the expression levels of the antioxidant genes, catalase and peroxidase, suggesting the mechanism underlying the quality enhancement of paprika. Our findings suggest that paprika storage at 10℃ alleviates chilling injuries, preserves the quality and marketability, and enhances the antioxidant potential of paprika. These findings provide insights into how temperature influences the quality and minimizes post-harvest losses during the storage and distribution of paprika.
Ginger (Zingiber officinale Roscoe) is traditionally used as appetite enhancer, improver of the digestive system, antithusive, anti-cold, antipyretic, analgesic, and antiinflammation. In vitro evaluation using human lymphocyte cultures showed almost similar indication with those in in vivo mouse study, NK cell lysing activity was improved significantly. Proliferation activity of B and T cells, and CD3$^{+}$ and CD3$^{+}$CD4$^{+}$T cell subset were better observed using oleoresin or gingerol and shogaol fractions. Although there were higher activities in gingerol, the improvement was almost equal to that by oleoresin. Shogaol did not show better improvement except at higher concentration. It could be concluded that treatment with single bioactive compound, such as gingerol, did not show significant effects compared to oleoresin, the crude extract. In human study, involving healthy male adult, the improvement of NK cell lysing activity was again demonstrated and even more apparent. The mechanism involved in the protection seemed to be through the antioxidant activity of gingerol. However, other mechanism underlying the improvement of NK cell lysing activity must be involved since this improvement seemed to be specifically toward NK cell activity. Since NK cells ave specific for the elimination of virus-infected cell and mutated cells, this positive effect on the immune system are very interesting. This work has also scientifically proved that the traditional beliefs that ginger had preventive effects on common cold appeared to be reasonable.
Beneficial effects of dehydroepiandrosterone (DHEA) supplement on age-associated chronic diseases such as cancer, cardiovascular disease, insulin resistance and diabetes, have been reported. However, its mechanism of action in hepatocellular carcinoma in vivo has not been investigated in detail. We have previously shown that during hepatocellular carcinogenesis, DHEA treatment decreases formation of preneoplastic glutathione S-transferase placental form-positive foci in the liver and has antioxidant effects. Here we aimed to determine the mechanism of actions of DHEA, in comparison to vitamin E, in a chemically-induced hepatocellular carcinoma model in rats. Sprague-Dawley rats were administered with control diet without a carcinogen, diets with 1.5% vitamin E, 0.5% DHEA and both of the compounds with a carcinogen for 6 weeks. The doses were previously reported to have anti-cancer effects in animals without known toxicities. With DHEA treatment, cytosolic malate dehydrogenase activities were significantly increased by ${\sim}5$ fold and glucose 6-phosphate dehydrogenase activities were decreased by ${\sim}25%$ compared to carcinogen treated group. Activities of Se-glutathione peroxidase in the cytotol was decreased siguificantly with DHEA treatment, confirming its antioxidative effect. However, liver microsomal cytochrome P-450 content and NADPH-dependent cytochrome P-450 reductase activities were not altered with DHEA treatment. Vitamin E treatment decreased cytosolic Se-glutathione peroxidase activities in accordance with our previous reports. However, vitamin E did not alter glucose 6-phosphate dehydrogenase or malate dehydrogenase activities. Our results suggest that DHEA may have decreased tumor nodule formation and reduced lipid peroxidation as previously reported, possibly by increasing the production of NADPH, a reducing equivalent for NADPH-dependent antioxidant enzymes. DHEA treatment tended to reduce glucose 6-phosphate dehydrogenase activities, which may have resulted in limited supply for de novo synthesis of DNA via inhibiting the hexose monophophaste pathway. Although both DHEA and vitamin E effectively reduced preneoplastic foci in this model, they seemed to fimction in different mechanisms. In conclusion, DHEA may be used to reduce hepatocellular carcinoma growth by targeting NADPH synthesis, cell proliferation and anti-oxidant enzyme activities during tumor growth.
