만형자 추출물의 항산화 및 주름개선 효과 (Antioxidant and Anti-wrinkling Effects of Extracts from Vitex trifolia L.)

  • 본 연구에서는 만형자 분획물의 항산화 및 주름개선 효과에 대하여 알아보고자 하였다. 만형자 ethyl acetate 분획물은 DPPH, ABTS에서 우수한 항산화 활성을 나타내었으며, 주름개선 효과를 검증하기 위하여 elastase 저해활성을 측정한 결과 $1,000{\mu}g/mL$에서 74% 저해활성을 나타내었다. 만형자 ethyl acetate 분획물의 세포생존율을 확인하기 위하여 섬유아세포로 MTT assay에 의해 확인 한 결과 5, 10, 15, 20, $25{\mu}g/mL$의 농도구간에서 100%에 가까운 세포 생존율을 확인하였다. 만형자 ethyl acetate 분획물의 MMP-1 저해활성을 측정 한 결과 $25{\mu}g/mL$에서 14%의 저해효과를 나타내었으며, 만형자 ethyl acetate 분획물의 MMP-1의 단백질 발현 억제 효과를 확인한 결과 $25{\mu}g/mL$의 농도에서 50% 감소하였다. 이러한 결과로 보아 만형자 ethyl acetate 분획물의 주름개선 효과를 확인할 수 있으며, 천연 기능성 화장품 소재로서의 가능성을 확인하였다.

압출 성형 인삼의 항산화 활성 증강 효과 (Effect of the Extruded Ginseng on Antioxidant Activity)

  • 최근 식품 및 천연 물질로부터 항산화 작용이 있는 생리 활성 물질을 찾고자 하는 연구가 활발하게 진행되고 있다. 본 연구에서는 우리나라를 비롯하여 동양권에서 전통적으로 자양강장을 목적으로 많이 사용해온 인삼을 압출 성형 기술에 의해 가공한 후 80% 에탄올로 추출하고 감압 농축한 에탄올엑기스와 이를 에테르를 이용하여 지방 성분을 제거한 후 수포화 부탄올 처리한 부탄올 분획과 물 분획을 얻었다. 인삼과 압출 성형 인삼의 부탄올 층을 각각 고속 액체 크로마토그라피에 의해 인삼 사포닌 함량을 측정하였다. 실험 결과 압출성형 인삼이 대조 인삼에 비해 ginsenoside Rg1의 경우 약 2.2배, Re의 경우 약 2.3배, Rc의 경우 약 1.2배, Rb2의 경우 약 1.3배, Rd의 경우 약 2.2배의 사포닌 함량이 증가하는 결과를 보였다. 또한, ORAC 방법에 의한 항산화 활성 실험 결과, 압출 성형 인삼의 경우 인삼 중의 여러 유효 성분이 많이 함유되어 있는 80% 에탄올 추출분 획과 일반적으로 사포닌 성분을 많이 함유한 부탄올 분획은 물론이고, 사포닌 함량이 거의 없는 수층 분획 모두에서 대조 인삼의 분획들에 비해 유의성 있는 증가 (0.05

  • In this work the antioxidant effects of browning reaction products prepared by xylose-tryrtophan reaction system were discussed. The antioxygenic brown pigments were separated by solvent extraction, and column chromatography and isolated by gel filteration. The functional groups of the brown pigments which had antioxidant activity were examined. The brown pigments extracted with methanol showed antioxidant effect and were fractionated in 5portions on DEAE-cellulose column. The elutes with methanol: acetic acid(10:30 v/v sol n(A), methanol: chloroform(95:5 v/v) sol n(C), and chloroform: acetic acid(10:30 v/v) sol n(E) only showed antioxidant activity and their compositions were 22.43, 21.51 and $34.43\%$ respectively. When each fraction on DEAE-cellulose column was reseparated on Sephadex LH-20 column, 2 fractions were obtained from portion A and C respectively. Molecular weights of A, C and E fraction of brown pigments were from 2,600 to 3,700. By elucidation of IR spectra, the pigment fractions which showed a strong antioxidant activity were tearing the indole group. It is suggested that the antioxidant function of the brown pigment is due to hydroxy and amino group. A higher activity of the brown pigment fraction E might be attributed to carboxylic acid or carboxylic ester compounds.

오죽 잎 용매분획물의 항산화 및 항균 활성 (Antioxidant and antimicrobial activity of solvent fractions from black bamboo leaves)

  • 오죽 잎의 메탄올 조추출물을 극성에 따라 용매분획하고 분획물의 항산화 활성과 항산화 활성, S. aureus 균주에 대한 항균활성을 조사하고 활성성분을 HPLC를 이용하여 정량하였다. 용매분획물 중에서 ethyl acetate 분획물의 DPPH radical 소거 활성, FRAP활성이 모두 가장 강하였으며, 총페놀성 화합물의 함량도 가장 높았다. S. aureus 균주에 대한 항균활성 역시 ethyl acetate 분획물이 모든 시험 농도에서 가장 높은 활성을 보였다. Ethyl acetate 분획물 중에서 caffeic acid, ferulic acid, quercetin, kaempferol 성분을 분리하여 정량하였으며 이 중 kaempferol의 함량이 가장 높았다. 오죽 잎의 ethyl acetate 분획물은 천연 식품보존료로서 활용이 가능할 것으로 보인다.

천궁(Cnidium officinale Makino) 지상부 추출물의 항산화 활성 평가 (Evaluation of Antioxidant Activity of the Extracts from the Aerial parts of Cnidium officinale Makino)

  • In order to obtain basic data for utilization of the aerial parts of Cnidium officinale Makino (APCO), the antioxidant properties of the aerial parts and rhizomes of C. officinale were measured using DPPH and ABTS radicals, and nitrite scavenging assays. The ethyl acetate (EA) fraction prepared from the aerial parts of APCO showed the strongest antioxidant activities, and contained high level of total phenolic compounds (325.81 mgTE/g) and flavonoids (259.16 mgRE/g). The concentrations for 50% reductions ($RC_{50}$) values of the DPPH and ABTS radicals, and nitrite by the EA fraction of APCO were $11.27\;{\mu}g/m{\ell}$, $14.34\;{\mu}g/m{\ell}$, and $10.26\;{\mu}g/m{\ell}$, respectively. APCO exhibited approximately 3-9 times higher antioxidant activity than rhizomes of C. officinale. The antioxidant capacities of APCO were positively correlated with its total phenolic contents. Therefore, it was concluded that the aerial parts of C. officinale can be a useful and cost-effective source of natural antioxidant for food or cosmetics.

Antioxidant Flavonoids and Chlorogenic Acid from the Leaves of Erobotrya japonica

  • The antioxidant activity of Eriobotrya japonica was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical and lipid peroxidation produced when mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA). The methanol extract and its factions of Eriobotrya japonica leaves showed strong antioxidant activity. The antioxidant activity of EtOAc and n-BuOH soluble fractions were stronger than the others, and were further purified by repeated silica gel, MCl gel CHP-20P, and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid, quercetin-3-sambubioside from n-BuOH fraction and methyl chlorogenate, kaempferol- and quercetin-3-rhamnosides, together with the inactive ursolic acid and$ 2{\alpha}$-hydroxyursolic acid from EtOAc fraction were isolated. Antioxidant flavonoids and chlorogenic acid also showed prominent inhibitory activity against free radical generation in dichlorofluorescein (DCF) method.

Antioxidant Activity of Cholesterol Derived from Silkworm Pupae

  • To search of more selective vasculogenic relaxation activity, the antioxidant activity of silkworm male pupae was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, and anticoagulant activity of them was measured clotting time in both activated partial thromboplastin time (aPTT). Because, most of cGMP-enhancing agent such as, sildenafil, promotes thrombin-induced platelet aggregation, developed unexplained thrombic conditions including heart attack. To search more suitable and safe drug for vasculogenic relaxation, we purified silkworm pupae male extract. The ethyl acetate extract of silkworm male pupae showed strong scavenging activity in both DPPH and aPTT anticoagulant activity. The antioxidant activity potential of the individual fraction was in order of ethyl acetate > n-butanol > chloroform > n-hexane. The ethyl acetate soluble fraction exhibiting strong anti-oxidant and anticoagulant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Cholesterol was isolated as one of the active principles from ethyl acetate fraction, together with, minor portion, ${\beta}-sitosterol$.

야콘, 선학초, 자초 추출물 분획의 항산화 활성에 관한 연구 (Study on Antioxidant Activity of Smallanthus sonchifolius, Agrimonia pilosa, and Lithospermum erythrorhizon Extract Fractions)

  • In this work, the antioxidant activity and total phenolic compound content of 6 fractions of Smallanthus sonchifolius, Agrimonia pilosa, and Lithospermum erythrorhizon extract were investigated. The highest total phenolic compound contents of each plant extracts were obtained from n-butanol ($13.75{\pm}0.21%$) and methylene chloride ($12.89{\pm}1.10%$) fractions (S. sonchifolius), ethyl acetate ($19.69{\pm}1.02%$) and water ($18.72{\pm}0.76%$) fractions (A. pilosa), and n-butanol ($36.26{\pm}1.26%$) and ethyl acetate ($17.66{\pm}0.94%$) fractions (L. erythrorhizon), respectively. As a result of DPPH radical scavenging activity in 10 mg/mL condition, the highest activity were obtained from n-butanol fraction of S. sonchifolius (81.06%), ethyl acetate fraction of A. pilosa (86.32%), and n-butanol fraction of L. erythrorhizon (82.6%), respectively. Also, the highest reducing power was obtained same fractions as well as DPPH adical scavenging activity. Overall, antioxidant activity has relatively closely connected with contents of total phenolic compounds in S. sonchifolius and L. erythrorhizon extracts.

빌레나무 추출물의 항산화, α-글루코시다아제 억제 및 항미생물 활성 (Antioxidant, α-Glucosidase Inhibitory and Antimicrobial Activities of Extracts from Maesa japonica (Thunb.))

  • Maesa japonica (Thunb.) is an evergreen shrub belonged to the Myrsinaceae family, which was discovered in 2006 in South Korea. And, its biological functions have not been well studied. In this study, we determined the antioxidant activities, ${\alpha}$-glucosidase inhibitory effects and antimicrobial activities of methanol extract and the solvent fractions of M. japonica leaves and twigs. The highest antioxidant activity obtained by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay and reducing power assay was found in the ethylacetate fraction of twigs methanol extract, which contained the highest level of total phenolic compounds compared to the other fractions. In addition, ethylacetate fraction of twigs extract exhibited higher inhibitory activities against ${\alpha}$-glucosidase ($IC_{50}=0.8{\mu}g/m{\ell}$) compared to the $IC_{50}$ of the buthanol fraction ($IC_{50}=16{\mu}g/m{\ell}$) of leaves extract. It showed antimicrobial activities against Bacillus atrophaeus and Bacillus subtilis subsp. Spizizenii. Although the data is too limited, the current study is the first report on biological functions of M. japonica.

Antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis from Jeju Island, Korea

  • In this study, we assessed the antioxidant properties of tidal pool microalgae, Halochlorococcum porphyrae and Oltamannsiellopsis unicellularis, from Jeju Island, Korea. Specifically, the antioxidant activity of fractions isolated from 80% methanol extract, and digests produced from five proteases and carbohydrases, were investigated. Almost all the fractions and the 80% methanol extract exhibited higher effects on 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging. The ethyl acetate fraction showed the highest superoxide anion scavenging activity, while both n-hexane and chloroform fractions exhibited higher $H_2O_2$ scavenging activity. Among the enzymatic digests from H. porphyrae and O. unicellularis, all the digests exhibited remarkable DPPH scavenging activities. In nitric oxide inhibition, all the digests recorded significantly higher effects than those of the commercial antioxidants (p < 0.05). Flavozyme and Neutrase digests from H. porphyrae, and Termamyl and Alcalase digests from O. unicellularis, showed significant effects in metal chelating. Lipid peroxidation was significantly inhibited in the ethyl acetate fraction, in the Celluclast and Protamex digests from H. porphyrae, and in the chloroform fraction from O. unicellularis. These findings suggest that the two tidal pool microalgae tested in this study are rich in potential antioxidative compounds, the specific properties of which can be considered for use in the food and pharmaceutical industries.
