녹황색채소류 및 대두에서 분리한 불용성 식이섬유의 항돌연변이 효과 (Antimutagenic Effect of Insoluble Dietary Fibers from Some Green Yellow Vegatagles and Soybean by Binding the Carcinogens)

  • The antimutagenic effects of insoluble dietary fibers(IDF) extracted from some green-yellow vegetables(kale, carrot, spinach, broccoli and soybean sprout) and soybean by binding the carcinogens of MeIQ (2-amino-3,4- dimethyl-imidazo(4,5-f) quinoline) and Trp-P-2(3-amino-1-methyl-5H-pyrido[4,3-b] indole) in Salmonella tylhimirium TA100 and TA98 were studied. All of the insoluble dietary fiber samples which binded MeIQ exhibited high antimutagenic effects by removing the mutagen. Among the samples, IDFs from kale and soybean showed strong binding capacity fo the carcinogen and revealed about 90% of the antimutagenic activity. the IDF samples showed somewhat lower binding capacity to the Trp-p-2. The lignin which extracted from kale, soybean and carrot, and the cellulose strongly removed the mutagenicity of MeIQ by the binding. Among the samples, the level of lignin in kale revealed the highest(about 10%), and it seemed that the higher content of lignin in kale is one of the reasons to increase its antimutagenic effect.

  • 한천올리고당을 식품으로 활용하기 위한 전제 조건인 안전성 실험결과, 한천올리고당 LD$_{50}$ 은 1359mg/kg으로 GRAS(Generally Recognized As Safe)등급에 해당하는 무해첨가물에 해당되었다. 또한 5% 한천올리고당의 항돌연변이 효과는 TA 98균주에 88.3%, TA 100균주에 54%의 저해효과를 나타내어 강력한 항돌연변원성 물질임을 알 수 있었다. 한천올리고당의 면역활성을 평가하기 위하여 비장세포에 한천올리고당을 200${\mu}\ell$/$m\ell$ 첨가하여 배양한 결과, 대조군과 비교시 배양 20일 이후에도 비장세포수가 감소하지 않아 한천올리고당의 면역증진 효과를 알 수 있었다.

  • The ethanol extracts from Angelica gigas Nakai and Angelica acutiloba Kitagawa were fractionated to diethyl ether and aqueous partitions. Both partitions had strong antimutagenic effect on the MNNG (N-methyl-N-nitro-N-nitrosoguanidine) by Ames mutagenicity test. Diethyl ether fractions exhibited the greatest antimutagenic effect suppressing the mutagenicity of MNNG with inhibition of 78-80%. The ethanol extracts from both species showed the inhibitory effect on the growth of several human cancer cell lines. Especially, the diethyl ether fraction from ethanol extracts was most effective on human hepatocellular carcinoma cells, inhibiting 90-95% of cell growth. However, the aqueous fractions had least inhibition activity on many cancer cells. There was little cytotoxicity on human normal liver cell by ethanol extracts. Diethyl ether fraction from Angelica gigas Nakai ethanol extract had cytotoxicity less than 20% on human normal liver cells, compared with that from Angelica acutiloba Kitagawa ethanol exract. The adding of 0.5 (g/l) of diethyl ether fractions of Angelica gigas Nakai or Angelica acutiloba Kitagawa increased immune activity by enhacing human B and T cells up to three to four times. It was proven that diethyl ether fraction (0.7 g/1) from Angelica gigas Nakai could control blood pressure by suppressing angiotensin converting enzyme activity up to 98%. From TLC, it was appeared that both of diethyl ether partitions had umbelliferon, known to one of active substances from Angelica gigas Nakai and Angelica acutiloba Kitagawa.

마늘의 쿨로로포름 분획에서 동정된 항돌연변이 물질 (Antimutagenic Compounds Identified from the Chloroform Fraction of Garlic (Allium sativum))

  • 마늘의 메타놀 추출물을 클로로포름층과 수층으로 분획하여 각 추출물의 aflatoxin $B_1(AFB_1)$에 대한 항돌연변이 효과를 실험한 결과, Salmonella typhimurium TA98과 TA100에서 클로로포름층이 가장 효과가 크게 나타났다. 클로로포름층을 다시 silica gel column 및 thin layer 크로마토그라피를 사용하여 분리한 결과, 4개의 분획(ASC F1, 2, 3, 4)를 얻었는데, 이중 ASC F2가 $AFB_1$에 대해 항돌연변이 효과가 가장 컸었다. ASC F2로부터 18가지의 화합물이 GC-MS, NMR, FT-IR을 이용하여 잠정적으로 분리, 동정되었으며, 이중 methyl linoleate는 가장 다량으로 함유되어 있는 화합물이었다. Methyl linoleate 표준품의 항돌연변이 효과를 조사한 결과 효과가 매우 큰 것으로 나타났다.

  • The antimutagenic activity of a methanol extract of roasted Cassia tora seed against aflatoxin $B_1(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using Salmonella typhimurium TA100 and or TA98. The MeOH extract was then sequentially partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. The $CH_2Cl_2$ and n-BuOH fractions possessed antimutagenic activity, but the EtOAc and $H_2O$ fractions were inactive. Both the MeOH extract and its fractions were capable of inhibiting the indirect-acting mutagen $AFB_1$, suggesting that these fractions may prevent the metabolic activation of $AFB_1$, or scavenge the electrophilic intermediate capable of inducing mutations. Column chromatography using silica gel yielded pure alaternin from the $CH_2Cl_2$ a fraction, and adenosine and isorubrofusarin gentiobioside from the n-BuOH fraction. Alaternin and isorubrofusarin gentiobioside demonstrated significant antimutagenic activities.

Melanoidin의 항산화성 및 항돌연변이원성 (Antioxidative and Antimutagenic Characteristics of Melanoidin Related Products)

  • Melanoidins, as brown colored polymers, are formed through the diversified reaction systems of Maillard type and other reactions. Melanoidins are important components in relation to food quality and also are known to have antioxidative, mutagenic and antimutagenic activities. Since these aspects have been extensively reviwed elsewhere, only the recent studies regarding to their antioxidative and antimutagenic activities are discussed in this review. Even though their mechanisms are not clearly identified, melanoidins or specific fractions isolated from their mixtures have shown varied antioxidative activities depending on the reaction systems and reaction conditions. Those activities presumely are derived from the complex functional properties of hydrogen / electron donors and metal chelating power, which are originated from their reductone structure and others. It is considered that pyrolysate and other mutagens are formed by the given conditions in some cases during browning reaction, whereas melanoidins and their fractions have antimutagenic effects on chemical and other mutagens. There are positive correlationship among the color intensity, antioxidative activity and antimutagenicity of melanoidins or their fractions. These suggest that the antimutagenicity of melanoidins could be attributed to their antioxidative properties, however, it might also be due to other factors, because the relevant responses for antimutagenicity are very complicate and not clear. Accordingly, further studies are required to determine the actual acitivities and mechanisms involved in antioxidation and (anti)mutagenicity of melanoidins by reaction systems / conditions and by the isolated fractions. And also, additional studies are needed to evaluate the applications of melanoidins and their relevant effects to food and human health.

멸치액젓으로부터 분리한 Bacillus subtilis JM-3의 생리활성기능에 관한 연구 (Studies on Physiological Activity of Bacillus subtilis JM-3 Isolated from Anchovy Sauce)

  • 속성 발효 및 기능성 멸치액젓의 제조에 사용할 수 있는 미생물 starter의 개발을 목적으로 전보에서 분리한 단백질 분해활성 및 혈전용해활성이 가장 우수했던 B. subtilis JM-3는 저온성 병원성 세균인 Listeria monocytogenes에 대한 항균활성이 가장 강하였으며, linoleic acid의 자동산화를 80%이상 억제하는 항산화활성을 나타내었다. 그리고 N-nitroso compound인 NDMA 및 NMO에 대하여서는 약 50%의 변이억제활성을 나타내었으며, SNU-1(사람의 위암세포)에 대하여 배양 원액 $20\;{\mu}L$를 첨가한 경우 88.9%의 높은 성장 억제율을 나타내어 기능성 멸치액젓 제조용 starter로서의 가능성이 확인되었으며, 앞으로 본 균주의 기능성 멸치액젓 starter로서의 이용을 극대화하기 위하여서는 돌연변이에 의한 호염성 변이주의 유전적 육종이나 형질 전환 방법 등을 응용한 호염성 미생물과의 접목을 통한 내염성 균주의 개발도 필요할 것으로 여겨진다.

Chemopreventive Activity of Turmeric Essential Oil and Possible Mechanisms of Action

  • This study aimed to evaluate the antimutagenic and anticarcinogenic activity of turmeric essential oil as well as to establish biochemical mechanisms of action. Antimutagenicity testing was accomplished using strains and known mutagens with and without microsomal activation. Anticarcinogenic activity was assessed by topical application of 7, 12 - dimethylbenz[a]anthracene (DMBA) as initiator and 1% croton oil as promoter for the induction of skin papillomas in mice. Inhibition of p450 enzymes by TEO was studied using various resorufins and aminopyrene as substrate. Turmeric essential oil (TEO) showed significant antimutagenic activity (p<0.001) against direct acting mutagens such as sodium azide ($NaN_3$), 4-nitro-O-phenylenediamine (NPD) and N-methyl-N-nitro N'nitrosoguanine (MNNG). TEO was found to have significant antimutagenic effect (>90%) against mutagen needing metabolic activation such as 2-acetamidoflourene (2-AAF). The study also revealed that TEO significantly inhibited (p<0.001) the mutagenicity induced by tobacco extract to Salmonella TA 102 strain. DMBA and croton oil induced papilloma development in mice was found to be delayed and prevented significantly by TEO application. Moreover TEO significantly (P<0.001) inhibited isoforms of cytochrome p450 (CYP1A1, CYP1A2, CYP2B1/2, CYP2A, CYP2B and CYP3A) enzymes in vitro, which are involved in the activation of carcinogens. Results indicated that TEO is antimutagenic and anticarcinogenic and inhibition of enzymes (p450) involved in the activation of carcinogen is one of its mechanisms of action.

Salmonella typhimurium Strain TA98, 100에서 감잎차, 녹차, 우롱차 추출물의 돌연변이 억제 효과 (Antimutagenic Effects of Water Extracts of Persimmon Leaf Tea, Green Tea and Oolong Tea on Reversion and Survival of Selected Salmonella Tester Strains)

  • Water extracts of persimmon leaf tea(PLTE), green tea(GTE) and oolong tea(OTE), at the con centration used for human consumption, were examined for inhibitory effects on the mutagenicity of major classes of dietary and environmental mutagens including indirect acting mutagens, B[ ]P (benzo[ ]pyrene), IQ(2 amino 3 methylimidazo[4,5 f]quinoline), 2 AA(2 aminoanthracene) in the presence of S9 mix and direct acting mutagen, 4 NQO(4 nitroquinoline 1 oxide) without S9 mix, using the modified Ames Salmonella/microsome assay. PLTE, GTE and OTE showed very potent and concentration dependent antimutagenic effects against indirect acting mutagens B[ ]P and IQ. At the maximum concentration(16,200 g/plate) of each tea extract, number of colonies decreased in a dose dependent manner up to 82~100%. Similar inhibition of PLTE, GTE and OTE were seen at higher concentration in the mutagenicity of the 2 AA following an initial increase in the activity at lower concentration. However, the mutagenicity of the direct acting mutagen 4 NQO were not suppressed at lower concentration of the three tea extracts, and higher concentration of the tea extracts enhanced mutagenic activity of the mutagen. There were no differences in the mode of antimutagenesis between PLTE, GTE, and OTE, in both Salmonella typhimurium TA98 and TA100 strains against the same mutagen. In conclusion, the water extracts of persimmon leaf tea, green tea and oolong tea possess marked antimutagenic potential against a variety of important dietary and environmental indirect acting mutagens, but the activity was not observed against the direct acting mutagens. These results suggest that the mode of inhibitory action may not have resulted from direct interaction between tea extracts and the mutagens, but rather from indirect metabolic inactivation of mutagens by tea extracts.

  • The extracts of aerial parts of Artemisia princeps var. orientalis, A. argyi, A. capillaris, A. rubripes and A. laciniata were investigated on the antimutagenic effect againt aflatoxin $B_1$ $(AFB_1)$. The methanolic extracts of 5 Artemisia species reduced the mutagenicity of $AFB_1$. And also 4,5-O-dicaffeoylquinic acid isolated from A. princeps var. orientalis revealed the antimutagenic activity.
