소염진통제에 관한 연구 Arylpropionic acid 류의 합성 및 항부종시험 (Studies on the Synthesis of Arylpropionic Acids and their Antiinflammatory Activity)

  • Eight derivatives of arylpropionic acid, arylcarbonylpropicnic acid, and arylhydroxybutyric acid were synthesized and their structures were elucidated on the basis of elemental analysis and spectral data. Also their antiinflammatory activities were evaluated by the method of the inhibitory effect on the carrageenin-induced rat paw edema and compared with phenylbutazone. Compound (I) exhibited 59% edema inhibition vs control when 100 mg/kg, was orally administered, whereas phenylbutazone showed 57.8% for 50mg/kg.

Antiinflammatory and Analgesic Effects of Higenamine, a Component of Aconiti Tuber

  • The antiinflammatory and analgesic activities of higenamine were evaluated by measuring edema volume and pain threshold in adjuvant arthritic rats and acetic acid-induced writhing test in mice. Higenamine, with consecutive oral administrations at doses of 10 and 50 mg/kg/day, showed significant antiedemic effect and elevation of pain threshold during the secondary lesion of adjuvant arthritis. Higenamine also showed a significant inhibition of acetic acid-induced writhing syndrome with a single oral administration (200 mg/kg). From these results, it is postulated that higenamine might possess both of centrally and peripherally mediated analgesic properties.

  • Polygonum cuspidatum which is widely distributed in Korea has been used as treatments of dermatitis, gonorrhea, favus athlete's foot, hyperlipidemia and inflammation in traditional medicine. We examined anti-oxidant and antiinflammatory activity by measuring DPPH radical scavenging activity and the inhibition of IFN-${\gamma}$ and LPS-induced NO production in RAW 264.7 murine macrophage cells. We isolated and characterized resveratrol (1), trans-resveratrol 3-O-${\beta}$-Dglucopyranoside (2). Compounds 1 and 2 showed potent activities compared with L-NMMA ($N^G$-monomethyl-L-arginine). These results suggested that the stilbene compounds isolated from Polygonum cuspidatum might be used as antiinflammatory agents.

  • As an attempt to search for bioactive natural constituents exerting antinociceptive and antiinflammatory activities, we examined the potency of the extract of R. coreanus fruits by the activity-guided fractionation. The EtOAc- and BuOH fraction and those alkaline hydrolysates showed significant antinociceptive effects as assessed by writhing-, hot plate- and tail flicks tests in mice and rats as well as antiinflammatory effect in rats with carrageenan-induced edema. BuOH extract was subjected to column chromatography to obtain a large amount of niga-ichigoside F$_1$ (1, 23-hydroxytormentic acid 28-O-glc), which was again hydrolyzed in NaOH solution to yield an aglycone 23-hydroxytormentic acid (1a).   • The cycloooxygenase enzymes catalyze the oxidative conversion of arachidonic acid into prostag1andin H$_2$Which mediates both benificial and pathological effects. The COX-1 is constitutively expressed in most tissues and in blood platelets wherease the expression of COX-2 isoform is induced in response to inflmmatory stimuli such as cyctokynes. Thus the identification of a novel COX-2 selective inhibitor should offer excellent antiinflammatory activity with minimal side effects such as gastrointestinal toxicity. Recently, a group of structurally unique and biologically active pimarane diterpenoids has been isolated from indigenous Korean medicinal plants. These new diterpenoids turned out to be potential analgesic and antiinflammatory agent due to their potent inhibitory activities of prostaglandin synthesis. We have also found that the inhibition of PGE$_2$synthesis is attributed to the potent COX inhibition by pimarane diterpenoid in arachidonic acid cascade. In conjunction with development of new analgesic and nonsteroidal antiinflammatory agent, a series of works on these diterpenoids have been extensively carried out in our laboratories. These efforts involve the structure-activity relationship of pimaradienoic acid, molecular modelings and COX inibitory activities as well as actiinflammatory effects of its structural analogues. In addition, the total syntheses of the new natural pimarane diterpenoids, their stereoisomers and other structural variants were intensively investigated.

지실의 생리활성성분 (Biological Active Components of Fruits of Poncirus trifoliata)

  • The biological activities of fruits of Poncirus trifoliata have been studied. 70% Ethanol extract of Ponciri Fructus was fractionated with ether, ethyl acetate and n-butanol by turns. The ether fraction showed antibacterial activity on Staphylococcus aureus and inhibited the contractability of the isolated mice ileum. The ethyl acetate and n-butanol fractions showed the anti-edematous effect on the carrageenin-induced edema of rat hindpaw. From the n-butanol fraction, one of the flavonoid glycoside was isolated and identified as poncirin. Ponciri Fructus contained about 6% poncirin which was 5,7-dihydroxy-4´-methoxy flavanone rhamnoglucoside and poncirin showed antiinflammatory activities. Poncirin was considered as the major effective component of Ponciri Fructus.

  • A new natural product, 3,4,8-trihydroxybenzopyranopyran-6,9-dione was isolated from the aqueous ethanolic gall extract of Tamarix aphylla (Tamaricaceae) along with the known phenolics, monodecarboxyellagic acid and brevifolin carboxylic acid as well. The structures have been established by ESI-MS, $^1H$ and $^{13}C$ NMR spectral analysis. Antiinflammatory, antipyretic and ulcerogenic activities determination for both plant (Tamarix aphylla and Phragmites australis) were carried out on aq. ethanolic of extracts.

Dammarane-type triterpene oligoglycosides from the leaves and stems of Panax notoginseng and their antiinflammatory activities

  • Background: Inflammation is widespread in the clinical pathology and closely associated to the progress of many diseases. Triterpenoid saponins as a key group of active ingredients in Panax notoginseng (Burk.) F.H. Chen were demonstrated to show antiinflammatory effects. However, the chemical structures of saponins in the leaves and stems of Panax notoginseng (PNLS) are still not fully clear. Herein, the isolation, purification and further evaluation of the antiinflammatory activity of dammarane-type triterpenoid saponins from PNLS were conducted. Methods: Silica gel and reversed-phase C8 column chromatography were used. Furthermore, preparative HPLC was used as a final purification technique to obtain minor saponins with high purities. MS, NMR experiments, and chemical methods were used in the structural identifications. The antiinflammatory activities of the isolated saponins were assessed by measuring the nitric oxide production in RAW 264.7 cells stimulated by lipopolysaccharides. Real-time reverse transcription polymerase chain reaction was used to measure the gene expressions of inflammation-related gene. Results: Eight new minor dammarane-type triterpene oligoglycosides, namely notoginsenosides LK1-LK8 (1-8) were obtained from PNLS, along with seven known ones. Among the isolated saponins, gypenoside IX significantly suppressed the nitric oxide production and inflammatory cytokines including tumor necrosis $factor-{\alpha}$, interleukin 10, interferon-inducible protein 10 and $interleukin-1{\beta}$. Conclusion: The eight saponins may enrich and expand the chemical library of saponins in Panax genus. Moreover, it is reported for the first time that gypenoside IX showed moderate antiinflammatory activity.

Identification of a novel triterpene saponin from Panax ginseng seeds, pseudoginsenoside RT8, and its antiinflammatory activity

  • Background: Panax ginseng Meyer (Araliaceae) is a highly valued medicinal plant in Asian regions, especially in Korea, China, and Japan. Chemical and biological studies on P. ginseng have focused primarily on its roots, whereas the seeds remain poorly understood. This study explores the phytochemical and biological properties of compounds from P. ginseng seeds. Methods: P. ginseng seeds were extracted with methanol, and 16 compounds were isolated using various chromatographic methods. The chemical structures of the isolates were determined by spectroscopic data. Antiinflammatory activities were evaluated for triterpene and steroidal saponins using lipopolysaccharide-stimulated RAW264.7 macrophages and THP-1 monocyte leukemia cells. Results: Phytochemical investigation of P. ginseng seeds led to the isolation of a novel triterpene saponin, pseudoginsenoside RT8, along with 15 known compounds. Pseudoginsenoside RT8 exhibited more potent antiinflammatory activity than the other saponins, attenuating lipopolysaccharide-mediated induction of proinflammatory genes such as interleukin-1β, interleukin-6, inducible nitric oxide synthase, cyclooxygenase-2, and matrix metalloproteinase-9, and suppressed reactive oxygen species and nitric oxide generation in a dose-dependent manner. Conclusion: These findings indicate that pseudoginsenoside RT8 has a pharmaceutical potential as an antiinflammatory agent and that P. ginseng seeds are a good natural source for discovering novel bioactive molecules.

쿼세틴 담지 나노캡슐을 함유한 알지네이트 스펀지의 제조 및 생리활성 특성 (Preparation and Study of Bioactive Characteristics of Alginate Sponge Containing Quercetin-encapsulated Nanocapsules)

  • Quercetin is one of flavonoids widely distributed in the plants and well known to have antioxidants, antiinflammatory, antimicrobial properties. In this study, alginate sponge containing quercetin-encapsulated nanocapsules was prepared by miniemulsion polymerization, dyring/crosslinking method and their bioactive characteristics were investigated. Alginate sponge containing quercetin-encapsulated nanocapsules were evaluated using a field emission scanning electron microscope(FE-SEM), a high performance liquid chromatography, cell viability, DPPH radical scavenging activity and antibacterial activity. The study indicates that alginate sponge containing quercetin-encapsulated nanocapsules had significant antioxidant, antiinflammatory and antibacterial activities. This study suggested that alginate sponge containing quercetin-encapsulated nanocapsules can be a potential candidate for medical materials.
