Phenylpropanoids of Plant Origin as Inhibitors of Biofilm Formation by Candida albicans

  • Biofilm-related infections of Candida albicans are a frequent cause of morbidity and mortality in hospitalized patients, especially those with immunocompromised status. Options of the antifungal drugs available for successful treatment of drug-resistant biofilms are very few, and as such, new strategies need to be explored against them. The aim of this study was to evaluate the efficacy of phenylpropanoids of plant origin against planktonic cells, important virulence factors, and biofilm forms of C. albicans. Standard susceptibility testing protocol was used to evaluate the activities of 13 phenylpropanoids against planktonic growth. Their effects on adhesion and yeast-to-hyphae morphogenesis were studied in microplate-based methodologies. An in vitro biofilm model analyzed the phenylpropanoid-mediated prevention of biofilm development and mature biofilms using XTT-metabolic assay, crystal violet assay, and light microscopy. Six molecules exhibited fungistatic activity at ${\leq}0.5mg/ml$, of which four were fungicidal at low concentrations. Seven phenylpropanoids inhibited yeast-to-hyphae transition at low concentrations (0.031-0.5 mg/ml), whereas adhesion to the solid substrate was prevented in the range of 0.5-2 mg/ml. Treatment with ${\leq}0.5mg/ml$ concentrations of at least six small molecules resulted in significant (p < 0.05) inhibition of biofilm formation by C. albicans. Mature biofilms that are highly resistant to antifungal drugs were susceptible to low concentrations of 4 of the 13 molecules. This study revealed phenylpropanoids of plant origin as promising candidates to devise preventive strategies against drug-resistant biofilms of C. albicans.

Combined Effects of the Essential Oil from Eucalyptus globulus with Ketoconazole against Candida and Trichophyton Species

  • We have evaluated the combined antifungal effects of the essential oil from Eucalyptus globulus or its main component 1,8-cineole with ketoconazole. Checkerboard microtiter tests were used to analyze their effects against three Candida and six Trichophyton species. The susceptibility of the Trichophyton species to E. globulus essential oil differed distinctly. The fractional inhibitory concentration indices (FICIs) against the tested Candida species ranged between 0.09 and 0.38 for ketoconazole combined with E. globulus essential oil or 1,8-cineole, indicating significant synergism between ketoconazole and the oil samples. Similar experiments using Trichophyton species resulted in FICIs between 0.28 and 0.63, indicating relatively weaker combined effects than those observed with Candida species. Thus, the data reported here show that the anti-Candida effects of ketoconazole can be significantly improved in the presence of E. globulus essential oil or 1, 8-cineole.

  • It has been reported that the pulmonary mycosis generally results from saprophytic colonization of pre-existing lung cavities usually due to pulmonary tuberculosis etc. Culture of homogenized tissues yield pure colony of slightly brown on the Sabouraud dextrose medium. The fungus was identified as the Scophulariopsis brevieaulis which had been rarely reported in pulmonary fungal infection. Histopathological examination of excised tissues revealed a chronic granulomatous inflammation and many branched hyphal mass. Susceptibility tests were tested with ketoconazole, griseofulvin, nystatin, and amphotericin B. Of these antifungal agents, ketoconazole and griseofulvin proved to be most effective and minimum inhibitory concentration was $10{\mu}g/ml,\;50 {\mu}g/ml$, respectively.

칸디다성 구내염에 대한 표고버섯 추출물의 예방효과 (Preventive effects of shiitake mushroom extract on candida stomatitis)

  • 목적: 본 연구의 목적은 효모형 또는 균사형 Candida albicans에 대한 표고버섯 추출물의항진균효과를 살펴보기 위함이다. 연구 재료 및 방법: 표고버섯 추출물은 표고버섯을 물 또는 에탄올에 닮근 후, 상층액을 건조시켜서 얻었다. 효모형 C. albicans에 대한 추출물의 항진균활성은 마이크로플레이트를 이용한 감수성 시험을 이용하여 조사되었다. C. albicans 생물막을 CO2 배양기에서 Ham's F-12 배지를 이용하여 12-well 플레이트에 형성시키고 에탄올 추출물로 처리하였다. 또한 C. albicans 생물막을 의치상용 레진 디스크에 형성시키고 의치세정제를 에탄올 추출물이 포함 또는 포함되지 않은 조건에서 처리하였다. 두 조건의 항바이오필름 효과 시험에서의 바이오필름내 살아있는 C. albicans를 조사하기 위해서 한천고체배지에 접종한 후 집락 형성 단위(CFU) 값을 측정하였다. 결과: 효모형 C. albicans에 대해서 표고버섯으로부터 물 추출물보다 에탄올 추출물이 강한 항진균력을 보였다. 에탄올 추출물은 균사형 C. albicans 바이오필름에 대해서도 유의적인 항진균력을 보였다(P < 0.05). 또한 에탄올 추출물은 의치세정제와 의치에 형성된 C. albicans 바이오필름에 대한 항진균력에 대해서 동반상승효과를 보였다(P < 0.05). 결론: 표고버섯 에탄올 추출물은 구강 칸디다증 예방뿐만 아니라 의치관련 구내염에 대해서 예방할 수 있는 후보물질로 사료된다.

일개 대학병원의 임상검체에서 분리된 6년간의 효모균 분포에 관한 연구 (Study on Distribution of Yeast Isolated from Clinical Specimens for Six Years in a University-affiliated Hospital)

  • 본 연구는 한 대학병원에서 6 년간 (2006-2011) 다양한 임상 검체에서 분리된 진균의 빈도를 조사하여 임상검체에서 진균의 종류에 따른 연관성을 파악하여 이와 관련된 환자 관리에 필요한 정보를 제공하기 위함이다. 경상남도의 단일 대학병원 진단검사의학과 미생물검사실로 의뢰된 검체 163,530건을 대상으로 하였고 배양에서 양성으로 판정된 진균 5,378주(3.3%)에 대하여 환자의 일반적인 특징과 성별, 연령, 검체 종류를 분석하였다. 균주별 분리빈도는 Candida albicans가 41.9%로 가장 많았고, Candida glabrata (15.5%), Candida tropicalis (14.6%)가 그 다음 순서로 분리되었다. 분리건수는 2006년 526건에서 2011년이 1,145건으로 2배이상 증가하였다. 연령별로는 60대(27.0%)와 70대(26.5%)에서 가장 흔히 분리되었다. 검체별로는 소변이 44.8%로 가장 많았다. 성별에 따른 분리는 남성이 52.4%로 여성 47.6%보다 약간 높았다. 추후 전국적인 분포도를 연구하고, 항진균 융복합 약제 감수성을 분석한다면 더 유익한 자료를 제공할 것으로 사료된다.

Transition Metal Complexes Derived From 2-hydroxy-4-(p-tolyldiazenyl)benzylidene)-2-(p-tolylamino)acetohydrazide Synthesis, Structural Characterization, and Biological Activities

  • Mononuclear Cu(II), Ni(II), Co(II), Mn(II), Zn(II), Fe(III), Ru(III), and UO2(II) complexes of 2-hydroxy-4-(p-tolyldiazenyl)benzylidene)-2-(p-tolylamino)acetohydrazide (H2L) were prepared by direct method. The ligand and its complexes were isolated in solid state and characterized by analytical techniques such as elemental and thermal analyses, molar conductance, magnetic susceptibility measurements and spectroscopic techniques such as UV-Visible, IR, 1H-NMR and 13C-NMR. The spectral data indicated that the ligand acted as neutral/monobasic bidentate or monobasic/dibasic tridentate ligand bonded to the metal ions through the oxygen atom of ketonic or enolic carbonyl group, azomethine nitrogen atom and deprotonated/protonated phenolic oxygen atom forming either tetragonally distorted octahedral or octahedral. Antimicrobial activities of the ligand and its complexes were evaluated against Escherichia coli, Bacillus subtilis and Aspergillus niger by well diffusion method. The results of antifungal activity showed that the Fe(III) complex (10) exhibited higher antifungal against Aspergillus niger than the other complexes. However, the results of antibacterial activity revealed that Cu(II) complex (4) is the most active against Escherichia coli while the Cu(II) complex (5) and Fe(III) complex (10) exhibited higher antibacterial effect on Bacillus subtilis than the other complexes.

  • 아쿠아포린(aquaporin)은 MIP (Major Intrinsic Protein) 패밀리에 속하는 물 수송 채널(water transport channel) 단백질로 단세포 생물인 박테리아부터 다세포 고등생물인 인간에 이르기까지 다양한 기관계에서 잘 보존되어 있다. 아쿠아포린은 정통아쿠아포린(orthodox aquaporin)과 아쿠아글리세로포린(aquaglyceroporin)으로 구분되는데, 정통아쿠아포린은 주로 세포내의 물 유입 및 수송에 관여하며 아쿠아글리세로포린은 glycerol, polyol, urea를 비롯한 작은 비극성 분자의 수송에 관여하는 것으로 알려져 있다. 최근까지 효모에서 아쿠아포린 기능이 일부 밝혀졌지만 Aspergillus 속을 포함하는 사상성 진균에서는 거의 연구가 되어있지 않은 실정이다. 본 연구에서는 A. nidulans의 유전체 염기서열 정보를 분석하여 하나의 정통아쿠아포린(aqpA)과 네 개의 아쿠아글리세로포린(aqpB-E)을 발견하였다. 이를 바탕으로 aqpA 유전자 결실돌연변이들을 만들어 그 기능을 분석하였다. aqpA 결실돌연변이는 각종 삼투 스트레스(osmotic stress)에서는 표현형의 변화가 거의 관찰되지 않았으며 이는 이들 유전자가 삼투 스트레스에 반응하지 않거나 유전자의 중복성 때문으로 여겨진다. 그러나 항진균제인 fluconazol에 대해서 그 감수성이 적어지는 것이 관찰 되었다. 이는 aqpA 유전자가 삼투스트레스 반응보다 항진균제의 감지에 더 기능을 가지고 있을 수 있음을 시사한다.

Crystal Growth, Spectral, Magnetic, Antibacterial and Antifungal Studies of Co(II) and Ni(II) Complexes of 4-Nitrobenzoic Acid

  • Single crystals of cobalt(II) and nickel(II) complexes of 4-nitrobenzoic acid have been successfully grown by gel diffusion technique. The grown crystals were characterized by elemental analysis, FT-IR and UV-Visible spectroscopy. The structure of the grown crystals was elucidated using single crystal X-ray diffraction studies. The title compounds $[Co(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 1 and $[Ni(Pnba)_2(H_2O)_4]{\cdot}2H_2O$ 2 where PnbaH=4-nitrobenzoic acid, crystallize in centrosymmetric triclinic space group P-1. Magnetic susceptibility measurements reveal that the compounds are paramagnetic in nature. The mechanical strength of the grown crystals was determined by Vicker's microhardness studies. The ligand (4-nitrobenzoic acid) and the complexes have been screened for their biological activity against various bacteria and fungi. The activity data show that the biological activity of the complexes is higher than that of the ligand.

Citronellol 및 Thymol이 Candida albicans 세포막 조성에 미치는 영향 (Effects of Citronellol and Thymol on Cell Membrane Composition of Candida albicans)

  • Phospholipids are crucially important in a cell membrane function and could thereby influence antibiotic susceptibility. In order to investigate the antifungal mechanism the total lipid was extracted from C. albicans treated with citronellol or thymol in concentration of their minimum inhibiting concentration and the changes in phospholipids composition were analyzed using ketoconazole as control. The cell growth and total lipid synthesis in cell walls of C. albicans were inhibited by treatment with citronellol. The levels of total lipids were decreased by 35.85% compared to the control. They also showed a significant decrease in the contents of phospholipid, phosphatidylcholine(PC), phosphatidyl ethanolamine(PE) and phosphatidylinositol(PI). As the result of GC assay for total fatty acid methyl esters of PC, PE and PI in C. albicans treated with citronellol, it was found that the major fatty acid composed of three phospholipid were palmitic acid, stearic acid and oleic acid. Moreover, the pattern of the fatty acid compositions of PC, PE and PI were changed by the oil. Based on the results, the anti-Candida mechanism of citronellol or thymol might be closely associated with disrupting the permeability barriers of the fungal cell wall composition or construction.

In vitro antimicrobial activity of Cassia auriculata

  • Ethanol and aqueous extracts of Cassia auriculata were tested in vitro against fungi (Candida albicans and Microsporum canis) and bacteria (Escherichia coli, Salmonella enteriditis, Staphylococcus aureus and Bacillus subtilis). M. canis showed dose-dependent susceptibility only towards ethanol leaf and bark extracts. C. albicans, were resistant to all types of plant extracts. Results were statistically smaller to antifungal drug ketoconazole and miconazole at equivalent concentration. Both ethanol and aqueous extracts of Cassia auriculata leaves and barks exhibit antibacterial activity against S. aureus and only the ethanol extracts of leaf and bark were detected against Bacillus subtilis. The results were compared to antibacterial drugs chloramphenicol, ampicillin, penicillin G, and enrofloxacin. The antibacterial activity was statistically similar to penicillin G. Based on the current findings, it can be concluded that this plant has antimicrobial activity, which is as potent as standard antimicrobial drugs.
