  • Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.

  • This study was done to find out the antifungal activity of two antibacterial antibiotics, Chlorampenicol and Streptomycin sulfate, against Phytophthora n. var. Parasitica, the causal agent of Phytophthora blight of sesame, growing on artificial media. On PDA medium, Chlorampenicol at 10 ppm, Streptomycin sulfate at 25ppm highly inhibited mycerial growth and completely inhibited zoosporangial formation of Phytophthora n. var. parasitica, and Chlorampenicol at 5 ppm, Streptomycin sulfate at 10 ppm slightly inhibited the mycerial growth and zoosporangial formation of the fungus. These antibiotics showed considerably increased inhibitory effect on the fungal growth when they were mixed with other chemical. Protein content in myceria of the fungus was decreased and abnormal growth of mycerial apex was observed by treatment of these antibiotics. Phytotoxicity on sesame seedlings was not observed by application of them.

  • For the development of antibiotics from Korean mushrooms, the biological activities of extracts from 98 species of mushrooms and from 25 mushrooms were tested against 9 different Gram-negative bacteria and 8 fungi, respectively. Fruiting bodies of each mushrooms were extracted with petroleum ether (P), 80% ethanol (E), and distilled water (H) in that order. P, E, or H extracts from 20 mushroom samples exhibited the antibacterial activity against 8 different Gram-negative bacteria containing Klebsiella pneumoniae, selectively. Among the mushroom extracts with antibiotic activity, E extracts of Boletus umbriniporus, Armillariella tabescens, Rhodophyllus sinuatus, and Suillus luteus showed various antibiotic activities against several bacteria. E extracts of Abortiporus biennis, Phellinus gilvus, and Polyporus dispansus are highly active against Salmonella typhi and their minimum inhibitory concentration (MIC) was all $10\;{\mu}g/ml$. E extract of Armillariella tabescens showed the antifungal activity against Trichopyton mentagrophytes, and its MIC was $300\;{\mu}g/ml$.

  • Two bithiazole-type antibiotics were isolated from the culture broth of a Myxococcus species which isolated from the marine sediment off the coast of Cheju Island, Korea. The structures of these metabolites were determined as KR025 and melithiazole F, previously reported bithiazoles, using combined spectroscopic methods. Both compounds showed an antifungal activity. In in vivo tests, these compounds exhibited potent controlling activities against tomato late blight, wheat leaf rust, and barley powdery mildew with control values more than 80% at a concentraton of 20 $\mu\textrm{g}$/ml.

  • The aim of the present research program was to develop a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 440 isolates, a strain (NA-4803) producing antifungal substance against Trichophyton spp. Nannizzia otae and Pyricularia oryzae was selected. The strain NA-4803 was identified as strain similar to Streptoverticillium blastmyceticum with respect to morphological and physiological characteristics, lecithinase and lipolytic activity, degradation of organic compounds, resistance to antibiotics and utilization of carbon and nitrogen sources. But it showed some differences such as positive reaction of nitrate reduction, negative reaction of L-tyrosine degradation, resistance to cephaloridine, and utilization of I -rhamnose and inulin. The strain NA-4803 was named as Streptoverticillium sp. NA-4803.

Antifungal Activity of SeO2 against Pathogenic Fungus Candida albicans (SeO2의 병원성 진균 Candida albicans에 대한 항균 활성)

  • This study was carried out to determine the antifungal activity of $SeO_2$ against pathogenic fungus, Candida albicans. Concentration-dependent Inhibitory activity of $SeO_2$ against C. albicans was shown. When $400{\mu}g/disc$ of $SeO_2$ was tested, the inhibition size ranged from 20.7 to 26.8 mm (avg. 23.4 mm). MIC values of $SeO_2$ against cell growth of C. albicans ranged from 50 to $100{\mu}g/mL$ (avg. $70{\mu}g/mL$). These results could be applied to antibiotics-resistant C. albicans and other pathogenic fungi.

  • A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..

  • The effectiveness of three antifungal antibiotics amphotericin B, clotrimazole and 5-fluorocytosine was tested against 120 clinical isolated Candida species. The minimum inhibitory concentrations of amphotericin B was considerably lower than those of clotrimazole and 5-fluorocytosine. High-level resistance to 5-fluorocytosine was present in 50% of the isolates. The combination of clotrimazole and 5-fluorocytosine produced synergistic inhibition against all 20 strains of Candida albicans tested that were relatively resistant to both antifungal agents.

Antifungal Activity of Valinomycin, a Peptide Antibiotic Produced by Streptomyces sp. Strain M10 Antagonistic to Botrytis cinerea

  • A strain of Streptomyces sp. (M10) antagonistic to Botrytis cinerea was isolated from orchard soil obtained from Jeju Island, Korea. An antifungal substance (CN1) was purified from the culture extracts of the strain, and then identified as valinomycin through extensive spectroscopic analyses. Valinomycin showed potent in vitro antifungal activity against Botrytis cinerea and also in vivo control efficacy against Botrytis blight development in cucumber plants. Overall, the disease control efficacy of valinomycin was similar to that of vinclozolin, a commercial fungicide. This study provides the first report on the disease control efficacy of valinomycin against Botrytis blight.

  • An improved method for the selective isolation of high-yielding mutant strains for the production of antifungal antibiotic KRF-001 was investigated. The mutant strain U. V 4, which produces high titer of KRF-001, was selected on the high potency agar plate after ultraviolet light irradiation. The U. V 4 strain produced 2-fold more KRF-001 than the mother strain in production media. Large scale fermentation was performed using the U. V 4 strain in 100$\ell$ fermenter. The antifungal antibiotic KRF-001 secreted into culture broth was detected by HPLC in 24hrs of fermentation.
