Biological properties of $\beta$-ketoacetoanilide chlorides against late blight of tomato (베타-케토아세트아닐라이드 염화물 유도체의 토마토 역병에 대한 작용 특성)

  • [ $\beta$ ]-Ketoacetoanilide chloride derivatives containing a substituent at 4 in phenyl group (para) reduced specifically the development of tomato late blight caused by Phytophthora infestans. Among $\beta$ketoacetoanilide chloride derivatives, five (KIST163, KIST170, KIST260, KIST263, and KIST267) were selected and tested for their protective, curative, systemic, persistent activities, and disease control efficacy against tomato late blight on adult plants. They exhibited a strong l-day protective activity and $EC_{50}$ of KIST163 and KIST170 were 21.9 and $14.5{\mu}g/mL$, respectively. However, they had little curative and systemic activities. Good persistence of KIST163 and KIST170 on tomato plants were observed against P. infestans; both KIST163 and KIST170 at $100{\mu}g/mL$ showed control values more than 75% in a 7-day protective applications. In addition, the two chemicals effectively controlled the occurrence of P. infestans on adult tomato plants. These results indicate that five $\beta$-ketoacetoanilide chloride derivatives are foliar fungicides with a preventive action and KIST163 and KIST170 have a potential for the control of tomato late blight in the fields.

Antimicrobial Active Substances from Entomopathogenic Fungi (Various Applications of Entomopathogenic Fungi)

  • Insects constitute the largest and most diverse group of animals in the world. They also serve as the hosts or nutrient sources for an immense assemblage of pathogens, parasites, and predators. More than 700 fungal species from 100 genera have adopted an entomopathogenic lifestyle. Although entomopathogenic fungi were studied as only biocontrol agents against a variety of pests in various countries, it has been recently focused their additional roles in nature. They are antagonists to/against plant pathogens, endophytes, and possibly even plant growth promoting agents. The potential antimicrobial effect against fungal plant pathogens by an isolate of entomopathogenic fungi including Beauveria bassiana, Lecanicillium spp., and Isaria fumosorosea have been reported since late 1990s, but wasn't reported pathogenicity of the isolate against pests. Later, a Canadian Lecanicillium sp. isolate and L. longisporium isolated from Vertalec$^{(R)}$ showed simultaneous control effect against both aphid and cucumber powder mildew. Therefore, the antimicrobial activities of 342 fungi isolates collected from various regions and conditions in Korea were evaluated against plant pathogenic fungus Botrytis cinerea using dual culture technique on agar plate. As a result, 186 isolates (54.4%) shown the antifungal activity against B. cinerea. The culture filtrates of selected fungi completely suppressed the growth of the microorganisms, indicating that suppression was due to the presence of antimicrobial substances in the culture filtrate. Mode of action of these fungi against insect involves the attachment of conidia to the insect cuticle, followed by germination, cuticle penetration, and internal dissemination throughout the insect. During infection process, secreted enzymes, proteinous toxins, and/or secondary metabolites secreted by entomopathogenic fungi can be used to overcome the host immune system, modify host behavior, and defend host resources. Recently, secondary metabolites isolated from entomopathogenic fungi have been reported as potential bioactive substances. Generally, most of bioactive substances produced by entomopathogenic fungi have reported low molecular weight (lower than 1,000 g/mol) as peptide and, in contrast the high molecular weight fungal bioactive substances are rare. Most substances based on entomopathogenic fungi were shown antimicrobial activity with narrow control ranges. In our study we analyzed the antimicrobial substances having antagonistic effects to B. cinerea. Antimicrobial substances in our fungal culture filtrates showed high thermostability, high stability to proteolytic enzymes, and hydrophilicity and their molecular weights were differed from substance. In conclusion, entomopathogenic fungi showed pathogenicity against insect pests and culture filtrate of the fungi also shown to antimicrobial activity. In the future, we can use the entomopathogenic fungi and its secondary metabolites to control both insect pest control and plant pathogenic fungi simultaneously.

Screening for Effective Organic Farming Materials for the Control of Cucumber Scab Caused by Cladosporium cucumerinum (오이 검은별무늬병 방제에 효과적인 유기농업자재 선발)

  • This study was conducted to evaluate the control efficacy of the organic farming materials on cucumber scab caused by Cladosporium cucumerinum PT1 (KACC 48094). The antifungal activities in vitro as well as the suppressive effect of 43 organic farming materials on the spore germination and germ tube growth by inoculating spore suspension on cucumber seedlings in vivo were investigated. Thirteen organic farming materials inhibited the mycelial growth of C. cucumerinum and nine of these were microbial agents. In the screening using cucumber seedlings, six organic farming materials were very effective with control efficacy value of 90%. Among them, Bacillus amyloliquefaciens M27 provided suppressive effect on both mycelial growth and spore germination against cucumber scab. Finally, nine organic farming materials were selected to test the protective and curative effects, and all chosen organic farming materials significantly suppressed disease incidence when applied in the preventive action, in comparison with the curative action. Especially, Bordeaux mixture I and III gave excellent protective control efficacy with control values of 96.7% and 73.3%, respectively, whereas its curative control effect was significant low. Among these, only Thymus quinquecostatus+Sophora extract showed curative activity, although the control value was as low as 50%. This study suggests that cucumber scab can be controlled by some organic farming materials in the farmhouses under comparatively cold and wet condition and protective treatment is more important and efficient.

Biological Control of Tomato and Red Pepper Powdery Mildew using Paenibacillus polymyxa CW (Paenibacillus polymyxa CW를 이용한 고추 및 토마토 흰가루병 방제)

  • In order to improve practical utility of agro-microorganisms (AMs) which had been cultured and disseminated to promote plant growth and to control crop diseases, 51 isolates of AMs were collected from 18 agricultural extension centers in local government and screened for multi-functions such as antifungal activity, activities of phosphorus solubilization, IAA and siderophore production, nitrogen fixation, and hydrolytic enzyme activity. Finally we selected one isolate showing good antifungal activity and multi-functions related to plant growth and disease control. The selected isolate, Paenibacillus polymyxa CW, showed good inhibitory effect against plant pathogens, Pyricularia gresea, Colletotrichum acutatum, Fusarium oxysporum, Phomopsis sp., Aspergillus niger, Rhizoctonia solani and Phytophthora capsici. Suppressive effect of P. polymyxa CW against the used plant pathogens except for R. solani was much higher than that of P. polymyxa AC-1 storing in National Academy of Agricultural Science. We found P. polymyxa CW isolate showed good activity in siderophore and IAA formation, and nitrogen fixation. With P. polymyxa CW isolate, siderophore formation activity was similar to that of P. polymyxa AC-1, but IAA formation and nitrogen fixation activity was much higher than that of P. polymyxa AC-1. However neither P. polymyxa CW nor P. polymyxa AC-1 showed hydrolytic enzyme (chitinase, pectinase and cellulase) activity. The treatment of P. polymyxa CW with culture suspension of different cell density ($10^8$, $10^7$. $10^6$ cfu/ml) showed that the highest density reduced incidence of red pepper powdery mildew by 68.3% after 10 days of application. As application density of P. polymyxa CW was decreased, its control efficacy was proportionally decreased. In addition, when P. polymyxa CW was treated to control tomato powdery mildew at the same concentrations and their control effects were investigated after 7 days of inoculation, disease incidence was 0.03, 19.5, 45.7%, respectively, compared to 56.3% that of untreated check. Like red pepper powdery mildew, increase of application density of P. polymyxa CW resulted in increase of its control efficacy proportionally. P. polymyxa CW showed a density-dependent control efficacy against red pepper and tomato powdery mildews. Therefore we think that mode of action of the antagonist for suppressing two powdery mildew diseases might be antibiosis and density of more than $10^8cfu/ml$ was needed to control effectively the two diseases. On this basis, we think that P. polymyxa CW can be a promising control agent for suppressing powdery mildews of red pepper and tomato.

Control Activities of Fungicides Against Garlic White Rot Caused by Sclerotium cepivorum (마늘 흑색썩음균핵병에 대한 살균제의 작용 특성)

  • In order to control garlic white rot (Sclerotium cepivorum), which threatens garlic production in farmers fields, soil solarization (solar sterilization), sclerotia germination inducers and effective microorganisms as biological control agents, and chemical fungicides have been used. Among them, fungicide has been largely used to reduce garlic white rot. In this study, the antifungal activities of five fungicides, prochloraz(a.i. 25%, EC), tebuconazole (a.i. 25%, WP), flutolanil (a.i. 15%, EC), iminoctadine tris-albesilate (a.i. 40%, WP) and isoprothiolane (a.i. 40%, EC) with different mode of action, in mycelial growth, sclerotia germination and sclerotia production, were tested. The inhibitory effects of the 5 fungicides on the mycelial growth, and sclerotia germination and production of garlic white rot pathogen (S. cepivorum T11-2) were investigated on potato dextrose agar (PDA) and their control efficacies were evaluated on garlic flakes. There was no mycelial growth of S. cepivorum T11-2 on PDA amended with $0.8{\mu}g\;mL^{-1}$ of prochloraz or $100{\mu}g\;mL^{-1}$ of tebuconazole. Also prochloraz and tebuconazole inhibited perfectively the sclerotia germination of the pathogen at 10 and $1.0{\mu}g\;mL^{-1}$, respectively. In spite of a very low activity of isoprothiolane in mycelial growth and sclerotia germination of S. cepivorum T11-2, it showed a good inhibitory activity against sclerotia production of S. cepivorum T11-2 on PDA amended with $1.67{\mu}g\;mL^{-1}$. Prochloraz, tebuconazole and flutolanil showed above 70% of control value when they were treated at $100{\mu}g\;mL^{-1}$ using the garlic flake cutting-method.
