Antidiabetic Effect of Glechoma longituba (Nakai) Kupr in Diabetic Rats induced by Streptozotocin (연전초(連錢草) 추출물이 Streptozotocin으로 유발시킨 흰쥐의 당뇨에 미치는 영향)

  • Objecitivies : While Glechoma longituba (Nakai) Kupr(GL) has been known as a antiinflammatory, antidiabetic and antihypertension medicine in korea traditional medicine, there has been only a few researches about it in Korea. Thus, we examined antidiabetic effect of GL in diabetic rats induced by streptozotocin Methods : To verify the effects of GL on serum glucose, creatinine, BUN, total cholesterol and triglyceride through streptozotocin-induced diabetic rats, some rats were injected 1 time with 55mg/kg streptozotocin intraperitoneally to induce diabetes, and other rats were injected with buffer I.P. as control group. Results : GL extracts significantly reduced the amount of serum glucose, BUN, creatinine, and triglyceride level in diabetics rats induced by streptozotocin. Conclusions : According to above result, Glechoma longituba was expected to be applied to the treatment of diabetes mellitus.

Antidiabetic Effect of ethanol extract of Forsythia Koreana in Streptozotocin-Induced Diabetic Rat (Streptozotocin에 의해 유도된 당뇨쥐에서 연교의 에탄올 추출물의 당뇨 개선 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of Forsythia Koreana(F.K) was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in F.K treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in F.K treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of F.K would have antidiabetic effect in STZ-induced diabetic rats.

  • This study was performed to investigate the antidiabetic effect of ethanol extract of Phelladrindron Amurense Rupr (P.A) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg,b.w. dissolved in citrate buffer. The ethanol extract of P. A was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The content of serum glucose, was significantly decreased in P.A treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased(p<0.05), and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased(p<0.05) in P.A treated group compared to those of STZ-control group, These results indicated that ethanol extract of P.A have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic Effect of Ethanol Extract on Astragali Radix (황기 에탄올 추출물의 항 당뇨 효과)

Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A. R was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in A.R treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in A.R treated group compared to the those of STZ-control group, These results indicated that ethanol extract of A.R would have antidiabetic effect in STZ-induced diabetic rats.

General Pharmacological Properties of DKY, an Antidiabetic Oriental Drug Preparation (항당뇨 천연물 복합신약 DKY의 일반약리작용)

  • DKY is an oriental drug preparation composed of 17 natural products and is known to have antihyperglycemic action at 100 mg/kg po in animal tests. The general pharmacological properties of DKY preparation were investigate in mice, rats, guinea pigs and rabbits. This preparation did neither show any effects on central nervous system, nor effects on algesia, nor epilepsia at the large doses of 3000 mg/kg po in mice or rats. However, the preparation showed hypothermic action at the doses of 330 and 1000 mg/kg po. In the guinea pig ileum, rat fundus strip and estrogenized rat uterus, DKY did not influence their tension at a concentration of 3$\times$10$^{-3}$ g/ml, and the spasmogenic actions produced by histamine, ACh and 5-HT were not blocked in the presence of DKY at 3$\times$10$^{-3}$ g/ml. The blood pressure and respiration were not considerably influenced at 10 mg/kg iv of DKY in rabbits. It did not influence the intestinal propulsion of mice and the normal gastric secretion of rats. These results may suggest that DKY preparation have little effects on central nervous, autonomic and gastrointestimal systems, except hypothermic action.

Comparison of Main Compounds and Physiological Activities of Anthriscus sylvestris (L.) Hoffm. Roots and Aerial Parts Extracts (전호 지하부와 지상부 추출물의 주요성분 및 생리활성 비교)

  • The number of people suffering from diabetes have been increased around the world. In this study, we investigated the antidiabetic and antioxidant effects of Anthriscus sylvestris(L.) Hoffm and its main compounds. It was divided into root(R) and aerial part(AP) for comparative analysis. Total polyphenol, total flavonoid content was higher in AP extract, but nodakenin content was higher in R(1169.13 ± 6.00 mg/g) extract. Antioxidant activity was also higher in AP extract. To compare antidiabetic efficacy, we analyzed the effects of R and AP extracts on ɑ-glucosidase inhibition(AGI), dipeptidyl peptidase-4(DPP-4) and protein tyrosine phosphatase(PTP)1B activity. R and AP extracts showed similar effects on AGI and DPP-4 activity in a concentration dependent manner, and there was no effect on PTP1B activation. Glucose uptake(139.51 ± 3.19%) in 3T3-L1 cells was more effective in the AP extract-treated group than the positive control, rosiglitazone, group. Both R and AP extracts were effective in protecting against pancreatic beta cell damage caused by hyperglycemia. These results suggest that Anthriscus sylvestris(L.) Hoffm. could be used as a candidate for diabetes treatment.

Antidiabetic Effect of Ethanol Extract of Liriope platyphylla in Streptozotocin Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨쥐에서 맥문동 에탄올 추출물의 당뇨개선효과)

  • The ethanol extraction yield of Liriope platyphylla(Lp) was about 30.7% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities effects of Lp in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in Lp treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in Lp treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in Lp treated group. Also the activity of glucokinase(Gk) was increaed in Lp treated group. The content of hepatic glycogen was significantly increaed in Lp treated group. These results indicated that ethanol extract of Lp would have antidiabetic effect in STZ-induced diabetic rats.

Effects of Cheonghyul-san on Blood Glucose, Hyperlipidemia, Polyol Pathway and Reactive Oxygen Species in ob/ob Mice (청혈산(淸血散)이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 Reactive Oxygen Species에 미치는 영향)

  • Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Cheonghyul-san has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs. In male ob/ob mouse with severe obesity, hyperinsulinemia, hypergiycemia, hyperlipidemia, the acting mechanisms of Cheonghyul-san were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. The effects of Cheonghyul-san extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Cheonghyul-san lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Cheonghyul-san treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the generation of ROS in the kidney. Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.

Studies on Antioxidant and Antidiabetic Effects of Fermented Cnidium officinale Makino (발효(醱酵) 천궁(川芎)의 항산화 및 항당뇨 활성(活性)에 관한 연구(硏究))

  • Objectives : We investigated the antioxidant and Antidiabetic effects of Cnidii Rhizoma Fermentata by Aspergillus oryzae and Aspergillus Kawauchi for 3days. Methods : In this study we compared Cnidii Rhizoma Fermentata with Aspergillus oryzae and Aspergillus Kawauchi that examined using reducing sugar, DPPH radical scavenging activity, ${\alpha}$-amylase and ${\alpha}$-glucosidase inhibitory activity. Also determined changes of pH and sugar content during fermentation for 3days. Results : The values for DPPH radical scavenging activity of Cnidii Rhizoma fermented by Aspergillus oryzae (86.6%) was higher than that of Aspergillus Kawauchi (77.9%). In ${\alpha}$-amylase inhibitory activity, fermented by Aspergillus Kawauchi had the highest inhibitory activity among other groups. But in ${\alpha}$-glucosidase inhibitory activity, fermented by Aspergillus oryzae had the highest inhibitory activity among other groups. While all groups of the sugar content increased During 3days fermentation, the pH was decreased. Conclusions : Based on these results, It was suggested that Cnidii Rhizoma Fermentata can be a useful and cost-effective resource for fuctional food and medicine.

2-Hydroxyquinoline and Its Structural Analogs Show Antidiabetic Effects against α-Amylase and α-Glucosidase

  • This study investigated the inhibitory activities of 2-hydroxyquinoline and its analogs against ${\alpha}$-glucosidase and ${\alpha}$-amylase. Based on the $IC_{50}$ values of 2-hydroxyquinoline analogs tested against ${\alpha}$-glucosidase and ${\alpha}$-amylase, 2-hydroxyquinoline had potent inhibitory activity (64.4 and $130.5{\mu}g/mL$, respectively), while 2-methyl-8-hydroxyquinoline showed weakly inhibitory activity (90.7 and $215.4{\mu}g/mL$, respectively). 2-Methylquinoline demonstrated no activity against ${\alpha}$-glucosidase and ${\alpha}$-amylase. In conclusion, 2-hydroxyquinoline analogs, with the existence of a methyl group and hydroxyl on quinoline, can be useful as a new diabetes treatment.
