Anticariogenic Activity of Callistemon citrinus Extract against Streptococcus mutans

  • Streptococcus mutans is known as a strong cariogenic bacterium and produces dental caries. This study was carried out to develop anticariogenic agents from Callistemon citrinus. Crude extracts and thier various solvent fractions of the plant extracts were tested for the antibacterial activity against S. mutans. The anticariogenic activity was evaluated with by paper disc diffusion method and minimal inhibition concentration (MIC). The extract from the fruit and its hexane fraction showed a strong inhibitory effect on the growth of S. mutans and the MICs were 31.2 and $3.9{\mu}g/m{\ell}$, respectively.

감국(Chrysanthemun indicum L.) 추출물의 항충치효과와 Glucosyltransferase 저해활성 탐색 (Anticariogenic Effects and Inhibition of Glucosyltransferase Activity of Chrysanthemun indicum L. Extracts)

  • 감국에 존재하는 항충치활성 물질 및 glucosyltransferase 저해 효과에 대한 정보를 얻고자 본 연구를 시도하였다. 감국 ethanol 추출물의 항충치활성을 S. mutans 및 구강세균에 대해 검색한 결과, S. mutans와 2종의 구강세균에 대하여 활성이 높게 나타났다. 감국 ethanol 추출물의 농도에 따라 GTase 저해효과는 $78.4{\sim}92.3%$의 범위로 나타나 농도가 증가할 수록 활성도 함께 증가하는 경향을 나타냈다. 감국을 ethanol로 추출, 용매분획하여 얻어진 활성성분을 silica gel adsorption chromatography, Sephadex LH-20 chromatography, TLC 그리고 HPLC 등의 방법으로 분석한 결과, 감국에 존재하는 항충치활성 물질은 약간의 극성을 띠는 중성물질로 분자량이 200-400정도 되는 저분자 물질인 것으로 확인되었다.

  • In order to find out an anticariogenic substance, antibacterial activities of some medical herbs and spices against Streptococcus mutans were investigated. The essential oils of oregano, thyme, sage, fennel, nutmeg, rosemary, calamus and cassia cortex were shown antibacterial effect against S. mutans. The essential oil of oregano among other tested medical herbs and spices showed the most active antimicrobial activity. The essential oil of oregano inhibited the activity of glucosyltransferase from S. mutans. The minimal inhibition concentration of oregano essential oil was determined as 0.05nl/ml. The activity of glucosyltransferase from S. mutans was inhibited in the presence of oregano essential oil.

구강병인균에 대한 마와 꿀풀추출물의 항균.항우식효과 (Anti-microbial and Anticariogenic Activity of Yam and Prunella Extract against Oral Microbes)

  • Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.

Combined Antimicrobial Activity of Extracts from Quercus infectoria Galls and Scrophularia striata Aerial Parts for an Anticariogenic Herbal Mouthwash

  • Objectives: Dental caries is one of the most prevalent human diseases worldwide. The disease initiates with bacterial adherence to the tooth surface followed by the formation of dental plaques. Mutans streptococci and Candida albicans are principal oral microorganisms involved in the initiation and development of dental caries. Phytochemicals have been shown to possess promising antimicrobial properties against a wide range of microorganisms and can be used for the prevention and treatment of dental caries. Herein, we reviewed literature on plants that are traditionally used for their antimicrobial properties or possess promising anticariogenic activity. We selected aerial parts of Scrophularia striata (S. striata) and galls of Quercus infectoria (Q. infectoria) and investigated their antimicrobial activity against cariogenic microorganisms. Methods: Water soluble fractions were obtained from hydroalcoholic extracts of S. striata and Q. infectoria and their antimicrobial activity against Streptococcus mutans (S. mutans), Streptococcus sobrinus (S. sobrinus), and Candida albicans (C. albicans) was evaluated separately and in combination. The extracts were then used for preparing an herbal mouthwash whose stability and tannic acid content were evaluated over 60 days. Results: Q. infectoria gall extract possesses efficient antimicrobial activity that was synergistically enhanced in the presence of S. striata extract. Mouthwash prepared using these extracts showed desirable organoleptic characteristics, antimicrobial activity, and stability. Conclusion: Extracts of S. striata and Q. infectoria galls can be used together for preparing dental products with effective anticariogenic properties. Our study highlights the importance of extensive pharmacological investigations when using herbal products alone or in combination with other chemical substances.

솔잎 및 소나무 가지 추출물의 항충치 활성 및 glucosyltransferase 억제 효과 (Anticariogenic Activity and Glucosyltransferase Inhibitory Effects of Extracts from Pine Needle and Twig)

  • 솔잎과 소나무 가지의 항충치 효과와 기능성 소재로서의 활용 가능성을 조사하기 위하여 추출물과 용매 분획의 S. mutans에 대한 항균효과, GTase 활성 억제효과 그리고 열과 pH 에 대한 안정성을 측정하였다. 열수추출물 보다는 70% EtOH 추출물이, 솔잎보다는 가지 추출물이 S. mutans에 대한 생육저해 효과가 큰 것으로 나타났으며, 특히 n-hexane 분획의 MIC 는 0.6 mg/mL로서 S. mutans에 대해 가장 높은 항균효과를 나타내었다. 그리고 솔잎과 가지 추출물의 GTase 활성 억제 효과는 S. mutans에 대한 생육억제 효과 결과와 마찬가지로 열수추출물 보다는 70% EtOH 추출물이, 솔잎 보다는 가지 추출물이 GTase 활성 억제 효과가 큰 것으로 나타났다. 한편 솔잎의 n-hexane은 열처리 온도가 증가함에 따라 S. mutans에 대한 항균력이 크게 감소하였고 pH의 변화에도 크게 영향받는 것으로 나타났다.

커피박 효소분해물의 항산화 및 항충치균 활성 (Antioxidant and anticariogenic activities of enzymatic hydrolysate from spent coffee grounds)

  • 커피박을 알카리 처리한 후 Viscozyme과 Alcalase로 효소분해하여 추출물을 얻었다. 커피박을 알카리와 효소로 처리하였을때 추출물의 페놀성 화합물 함량이 증가하였고, 이에 따라 양이온라디칼과 유리라디칼에 대한 우수한 소거 활성을 나타내었다. 특히 커피박에 알카리와 Alcalase를 병행 처리하였을 때 페놀성 화합물 함량과 항산화 활성이 가장 높게 나타났고, 농도에 비례하여 Streptococcus mutans 균의 생육을 억제하였다. 결론적으로 알카리 처리된 커피박의 Alcalse 효소분해물은 우수한 항산화 및 항충치균 효과를 나타내었다.

  • This study was carried out to develop anticariogenic agents from natural products, the methanol and ethyl acetate extracts of 105 medicinal plants were tested for the antimicrobial activity against a cariogenic bacterium, Streptococcus mutans OMZ 176. Among them, the Aristolochia contorta, Dryopteris crassirhizoma, Eugenia caryophyllata, Hemerocallis fulva, Pinellia ternata, Pyrola japonica, and Scrophularia buergeriana showed significant antimicrobial activity.

Anticariogenic activity of piceatannol isolated from Callistemon citrinus fruit against Streptococcus mutans

  • Dental caries is the destruction of the enamel of teeth by Streptoccus mutans. S. mutans has been isolated from human dental plaque and is associated with the initial development of enamel lesions. We have studied the antibacterial action of the fruit of Callistemon citrin us against a cariogenic bacterium, S. mutans. From the fruit of C. citrinus, piceatannol (3,3',4',5-tetrahydroxystilbene) was isolated by repeated column chromatography with $SiO_2$ and Sephadex LH-20. Its structure was elucidated by instrumental analysis using 1D-NMR, 2D-NMR and EI-MS. This compound was isolated from the fruit of C. citrin us for the first time. The anticarcinogenic activity of this compound was determined by using agar well-diffusion method and minimal inhibition concentration (MIC).

구강질환 원인균에 대한 정제봉독의 항균효과 (Anticariogenic Activity from Purified Bee Venom (Apis mellifera L.) against Four Cariogenic Bacteria)

  • The aim of the study was performed to examine the anticariogenic potential of purified bee venom (Apis mellifera L., PBV) collected using bee venom collector from cariogenic bacteria, Streptococcus mutans, Streptococcus sanguis, Porphyromonas gingivalis, and Fusobacterium nucleatum. The anticariogenic effect of purified bee venom was evaluated by agar well diffusion method, minimum inhibitory concentraion (MIC), minimum bactericidal concentration (MBC), and postantibiotic effect (PAE). The human lower gingiva epithelial cell cytotoxicity of purified bee venom was also evaluated. Purified bee venom exhibited significant inhibition of bacterial growth of S. mutans, S. sanguis, P. gingivalis, and F. nucleatum with MIC value of 0.68, 0.85, 3.49, and $2.79{\mu}g/ml$, respectively. The MBC value of purified bee venom against S. mutans, S. sanguis, P. gingivalis, and F. nucleatum was 1.34, 1.67, 8.5, and $6.8{\mu}g/ml$. Furthermore, the results of PAE values against S. mutans, S. sanguis, P. gingivalis, and F. nucleatum showed the bacterial effect with 3.3, 3.45, 2.0, and 2.0. The concentration below 1 mg/ml of purified bee venom had no cytotoxicity in the human lower gingiva epithelial cell. These results suggested that purified bee venom have great potenial as anticariogenic agents.
